An appropriate strain of Bacillus spp. derived from traditional Korean fermented foods was selected for fermentation of black rice bran, and the fermented bran’s nutritional components were analyzed. Among 18 isolated Bacillus spp. strains, the five (KU3, KU24, KU28, KU611, and KU612) that exhibited both amylolytic and proteolytic activities were selected for fermentation. Among these, Bacillus sp. KU3 showed notable dual enzymatic activity. During fermentation in black rice bran medium, Bacillus sp. KU3 increased from 5.83 to 7.83 Log CFU/mL after 24 h. The KU3 strain was identified as Bacillus subtilis using an API 50 CHB kit and 16S rRNA sequencing. Black rice bran (4% w/v) was fermented with B. subtilis KU3 at 37°C, 150 rpm for 24 h. Following fermentation, the main component of the fermented black rice bran was carbohydrate (77.13%). An increase in ash content was observed, while other nutritional components showed no significant changes. These results suggest that B. subtilis KU3 is a viable strain for black rice bran fermentation.
프로바이오틱스는 다양한 환경에서 분리되어 왔으며, Bacillus 균주는 유산균과 같은 일반적인 프로바이오틱스의 한계를 보완할 수 있는 내생포자 형성 능력으로 인해 프로바이오틱스 균주 중에서 유리하다. 본 연구의 목적은 한국에서 오염되지 않은 환경으로 알려진 울릉도 토양에서 분리된 Bacillus 균주의 프로바이오틱스 잠재성을 조사하는 것이다. 토양 시료는 울릉도 각지에 서 채취하였으며, 항생제 내성과 효소 활성을 평가하였다. 항생제에 내성을 나타내지 않는 6개 의 Bacillus 균주를 확보하였고 이후에 효소 활성을 검사하였다. 인체에 악영향을 미칠 수 있 는 β-glucuronidase 효소는 활성이 나타나지 않았으며, 시중에서 판매되고 있는 Bacillus 프로바 이오틱스와 유사한 효소의 활성과, 추가적으로 펩타이드 가수분해효소인 Leucine arylamidase 의 활성이 나타난 NB-1 균주가 확인되었다. 이에, 추가적인 효능 연구 및 안정성 실험을 통해 프로바이오틱스 균주로서의 가능성을 모색해야 할 필요가 있다고 판단된다.
DEAE Sephadex column chromatography에 의해 Bacillussp. 유래 β-mannanase의 정제를 수행하여 비활성 21.57units/mL 정제배율 95.33배를 나타내었다. SDS-PAGE에 의한 단일밴드를 확인하였고, 분자량은 38.9kDa으로 결정되었다.정제효소에 의해 Picea abies galactosyl glucomannan을 가수분해하여 activated carbon column chromatography에 의해 당가수분해물을 분리 회수하여 TLC, FACE 및Timell’s method에 의해 중합도 8, 10으로 결정되었으며,Penicillum purpurogenum유래 정제 β-mannanase와 α-galactosidase를 이용한 enzymatic sequential action에 의해 2가지 가수분해산물 모두 hetero type galactosylglucomannooligosaccharides로 확인되었다. B. longum, B.bifidum, B. infantis, B. animalis, B. breve, B. adolessentis,B. auglutum의 생육활성에 대한 중합도 8, 10의 영향을 검토하기 위하여 modified-MRS 배지상에 탄소원으로 중합도8, 10를 대체하여 생육활성을 비교한 결과 B. animalis에서는 중합도 8 galactosyl glucomannooligosaccharide를 탄소원으로 대체한 경우 표준 MRS 배지와 비교하여 19.5배, 중합도 10에서 18.7배의 가장 우수한 생육활성을 나타내었으며, B. bifidum에 대해서는 중합도 8의 경우 15.3배와 중합도10에서 14.3배 그리고 B. longum에서는 중합도 8 galactosylglucomannooligosaccharide를 탄소원으로 대체한 경우 표준MRS배지와 비교하여 중합도 8에서 15.2배, 중합도 10에서13.9배의 상대활성을 우선적으로 나타내었다.
To evaluate antimicrobial activities of chongkukjang slime fermented by different strains, growth characteristics were compared using various standard microorganisms with addition of chongkukjang slime. Chongkukgang slime was prepared by fermenting cooked soybean after inoculating with Bacillus circulars K-1, Bacillus spp N-1 and Bacillus subtilis CH-1, respectively. Significant antimicrobial activity was observed by chongkukjang slime on gram positive bacteria (Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, Micrococcus luteus), gram negative bacteria(Escherichia coli O157:H7, Salmonella Typhimurium, Pseudomonas fluorescens), and yeast (Pichia membranaefaciens, Saccharomyces cerevisiae, Candida albicans). In case of B. cereus growth inhibition of 80% was achieved by the addition of chongkukjang slime; on the contrary, to Escherichia coli O157:H7 only 20% inhibition was observed. Slime from Bacillus subtilis CH-1, in parkicular, inhibition of 40% toward bacteria and yeast, whereas slime from Bacillus circulars K-1, Bacillus spp N-1 showed only 20% inhibition.

In this study, we tried to screen the Bacillus strain having safety probability by isolation of strains from traditional fermented food, measurement of probiotic properties, and the fermentative characteristics of Cheonggukjang. We isolated 400 Bacillus-like isolates from traditional fermented foods. Selected strains examined on the prevalent characteristic such as extracellular enzyme and antibacterial activities, and their safety probability was confirmed by biogenic amine productivity, hemolytic, and harmful substances and enzyme productivity. We selected the 5 strains by analysis of biogenic amine, antibacterial and B. cereus toxic associated gene. Five selected strains were examined on cell surface hydrophobicity, and bile and acid tolerance, and we selected the SRCM100730 as the final strain. SRCM100730 was confirmed B. amyloliquefaciens by 16S rRNA sequencing, and named the B. amyloloquefaciens SRCM100730 (KCCM11966P). Finally, we manufactured Cheonggukjang using SRCM100730 for confirmation of fermentation properties. Manufactured Cheonggukjang did not contain B. cereus, and showed that γ-PGA and extracellular enzyme activities were superior to commercial Chunggukjang. Amino nitrogen content was 544.02 mg% and 26 free amino acid were detected, and the bitterness-related amino acid content was lower than commercial Cheonggukjang. Especially, the amount of GABA was 3 fold higher than commercial Cheonggukjang. These results suggest that SRCM100730 have high availability in commercial probiotics market and fermented food industry.
This study was carried out to research microorganisms having the antifungal activity against ginseng Alternaria blight pathogen Alternaria panax and ginseng anthracnose pathogen Colletotrichum gloeosporioides. Eleven Bacillus strains. were isolated from Korean traditional soybean paste and Kimchi. Among the 11 isolates, DJ5, KC1, KC2 and KC4 showing antagonistic activity on the mycelial growth of A. panax and C. gloeosporioides in pairing culture were finally selected as the antagonistic microorganisms. Based on 16s rRNA sequence and phylogenetic tree analysis, they were identified as Bacillus spp.. The selected microorganisms were investigated antagonistic activity by measured leaf-segment colonization in pot test. When Bacillus sp. were injected after A. panax treatment, KC1, KC2 and KC4 showed similar effect to chemical pesticides treated control. To measure preventive effect of Bacillus sp, antagonistic microorganisms were injected and C. gloeosporioides was treated in pot. When measuring the effectiveness for the prevention of Anthracnose, All Bacillus spp. showed approximately 83~90 % degree of superior preventive effect. In general, The four Bacillus spp. isolated from Korean traditional fermented foods showed therapeutic effect of Alternaria blight and preventive effect of Anthracnose.
