        Rice stripe virus (RSV) is one of the serious plant pathogenic viruses for rice transmitted by small brown planthopper, SBPH, Laodalphax striatellus. RNA interference (RNAi) is an universal gene-knockdown mechanism in eukaryotic organisms which includes insects,and has been considered as an alternative strategy to control insect pests. Hence, we applied this technique to interfere the translation of target RNA genes to knockdown the virus gene on RSV-viruliferous L. striatellus. Three out of seven RSV genes, RdRp, NS3, and NCP were used as target genes and each dsRNA targeting the viral genes were delivered to the insects indirectly through the rice leaves by irrigation. As a result, not only the relative expression level of target genes decreased but also those of non-target genes and the replication of RSV genome as well. In summary, leaf-mediated dsRNA feeding methods would be useful in the knockdown of target genes on piercing-sucking insects. The genes used in this experiment can be utilized for the development of pest-resistant transgenic plants based on RNAi.

        Crystals of proteinaceous insecticidal proteins, Cry proteins, produced by Bacill us thuringiensis (Bt) have been generally used used to control insect pests. In this st udy, through the 3D structure prediction and accompanying mutagenesis study for the Mod-Cry1Ac, 7 and 16 amino acid residues from domain I and II, respectively, responsible for its insecticidal activity against larvae of Plutella xylostella, Spodopt era exigua and Ostrinia furnacalis were identified. To construct novel cry genes wi th enhanced insecticidal activity, we randomly mutated these 24 amino acid sequen ces by in vitro muti site-directed mutagenesis, resulting in totally 34 mutant cry gen es. For further characterization, these mutant cry genes were expressed as a fusion protein with polyhedrin using baculovirus expression system. SDS-PAGE analysis of the recombinant polyhedra revealed that expressed Cry proteins was occluded in to polyhedra and activated stably to 65 kDa by trypsin. When the insecticidal activit ies of these mutant Cry proteins against to larvae of P. xylostella, S. exigua, and O. furnacalis were assayed, they showed higher or similar insecticidal activity compar ed to those of Cry1Ac and Cry1C. Especially, among them Mutant-N16 showed th e highest insecticidal activity against to both of P. xylostella, S. exigua and Ostrinia furnacalis. Therefore, Mutant-N16 is estimated to have the potential for the efficac ious bioagent.

        To date there have been only two species in genus Pogonus and subgenus Europhilus of genus Agonum recognized from Korean insect fauna: Pogonus itoshimaensis Habu, 1954 and Agonum (Europhilus) bellicum Lutshnik, 1934. In the present study one additional species for each genus is newly recognized from Korea: Pogonus (Pogonus) japonicus Putzeys, 1875 and Agonum (Europhilus) gratiosum nipponicum Habu, 1972. A key to adults, redescription, diagnostic photos of adult and male and female genitalia are provided.

        The genus Glischrochilus Reitter, 1873 is characterized by reddish or yellowish markings on their elytra. This genus is belonging to family Nitidulidae Latreille,1802. Five species in this genus: Glischrochilus (Librodor) ipsoides (Reitter, 1879), Glischrochilus (Librodor) japonicus (Motschulsky, 1858), Glischrochilus (Librodor) pantherinus (Reitter, 1879), Glischrochilus (Librodor) parvipustulatus (Kolbe, 1886), Glischrochilus (Librodor) rufiventris (Reitter, 1879) have been recorded in Korea upto now. Additionally, a new recorded species, G. christophi, are recognized from korea. As the results, descriptions and illustrations of adults of totally five species with one newly recorded species in Korea were reviewed herein. A key to Korean species of genus Glischrochilus is also provided.

        Insect pollinators of the endanger orchid Cypripedium japonicum were surveyed and identified during two years, as a part of a conservation project of the orchid at Jukyeup-san and Hwaak-san (Mt.), South Korea. In total 40 individuals of 16 species in 4 families were identified. The dominant family was Halictidae, and Lasioglossum exiliceps Vachal visited the most frequently C. japonicum during the surveys. The average visiting frequency was 2.5 individuals per hour and the highest 4.3, from 12:00 – 13:00 in a day. After 15:00 insects did not visit the flowers at all. However, all of the visiting insects were found to not carry a pollinium or pollens of the orchard on their bodies; pollen carryover by any of the visiting insects did not occur at all. The orchid seems to require certain pollinators in particular body thickness due to its unique pollination mechanism. The orchid has two exit route openings, around 1 cm in diametre, where the entrapped insects can exit and an anther is situated just in front of each opening. It was inferred that a pollen carrier should be around 1 cm in body thickness. Therefore, the candidate species as the proper pollen carriers can be Tetralonia nipponensis Perez, Xylocopa appendiculata circumvolans Smith and Bombus consobrinus Dahlbom among the surveyed visitors.

        The phylogenetic relationships of species and genera in the subfamily Nymphalinae from Myanmar were inferred using mtDNA sequence data from 608 bp of cytochrome oxidase subunit I (COI). A total of 20 species in 10 genera were sequenced and used to construct phylogenetic trees. The base composition of COI sequences was 38.1% T, 15.6% C, 31.6% A, 14.7% G, revealing strong AT bias (69.7%). The sequence distance of 20 species of Nymphalinae ranged from 1.5% to 15.5%. The transition of nucleotide substitution was more common than transversion. The transition between T and C were higher than transition between A and G, and the transversion between A and T was the highest amongst other types of transversion. The phylogenetic trees were constructed using the neighbor-joining (NJ) and maximum likelihood (ML) methods and showed almost identical topologies. The results indicated that the tribes Junoniini and Nymphalini (sensu Wahlberg et al., 2005) formed monophyletic groups but Kallimini was not monophyletic group. Rhinoplapa polynice formed sister group to Junoniini clade with moderate support in both trees. The relationship of species in Junoniini was ((Junonia + Yoma) + Hypolimnas) and the relationship in Nymphalini was (Symbrenthia + (Vanessa + (Kaniska + Polygonia))). The clustering results were almost identical to current morphological classification.

        Calla lily is a mainstream floricultural crop but itscommercial success is limited by severe susceptibility to softrot. This disease can be controlled to some extent by imple-menting appropriate cultural practices but developing resis-tant cultivars against this pathogen is the most promisinglong-term strategy. The purpose of this work was to developnew cultivars resistant to this bacterial disease by initiatingan appropriate breeding programme. Field observations of120 resistant lines selected by a leaf-disk susceptibility testwere carried out and survival rates were measured. Twolines were selected after further evaluation of resistancelevel, flowering characteristics and following guidelines forconduct of tests for distinctness, homogeneity, and stability.Further preference testing of the selected lines by growers,consumers, researchers and agricultural advisors was car-ried out with excellent results. The selected new lines are‘Silky White’ and ‘Mont Blanc’ which emerge as the first,soft-rot resistant calla lily cultivars from our breeding program.

        본 연구는 양념 닭 가슴살의 양념 액에 귤 껍질 추출물(CPE, 2%), 양파 껍질 추출물(OPE, 2%), calcium lactate(2%), 난황 유래 phosvitin(0.1%)과 CPE, OPE 및 calcium lactate 복합처리를 첨가하였을 때 서로 다른 저장온도와 저장기간에 따른 항균 효과 및 품질 변화를 확인하고자 수행되었다. 모든 저장온도와 저장기간에서 CPE, OPE 및 복합 처리군의 총 호기성 세균 수는 대조구에 비해 유의적으로 낮게 확인되었으며, 가장 높은 미생물 생장 억제 효과가 있었던 처리군은 CPE 처리군이었다. 하지만, calcium lactate와 phosvitin 처리군의 미생물의 성장 억제 효과는 나타나지 않았다. pH의 경우, calcium lactate와 OPE 처리군은 모든 저장온도의 저장 0일차에서 대조구에 비해 유의적으로 낮은 값을 나타내었다. OPE 처리군은 모든 저장온도와 저장기간에서 다른 처리구들에 비해 유의적으로 높은 적색도를 나타내었고, CPE와 함께 저장온도에 따른 유의적 차이를 나타내지 않았다. 하지만 이러한 OPE 처리군은 풍미, 맛 및 종합적 기호도에서 다른 처리구들에 비해 유의적으로 낮은 값을 보였으며, calcium lactate와 phosvitin 처리군의 종합적 기호도는 대조구와 유의적 차이가 나타나지 않았다. 결론적으로, 양념 닭가슴살의 CPE, OPE 및 복합처리군에서 저장온도를 달리한 저장기간에 따른 미생물 억제 효과를 확인할 수 있었으나, 관능적 특성을 개선하는 방법의 개발이 필요하다고 판단된다.

        The Riptortus (stinkbug) has a specialized symbiotic organ, M4 midgut, to harboring symbiont Burkholderia. M4 midgut is located in abdomen and surrounded with insect hemolymph. Recently our group demonstrated that symbiotic Burkholderia showed different physiology after adapting in M4 gut compare with in vitro cultured Burkholderia. And population of symbiotic Burkholderia in the M4 midgut is regulated by special organ. However, the molecular mechanism to prevent spreading and migrating symbiont bacteria to other host tissues from symbiotic organ is not clear. Therefore, we assumed that symbiont Burkholderia are susceptible to host humoral immunity after established infection in M4 midgut to prevent spreading and migrating into the other host tissues through Riptortus hemolymph. To prove this assuming, we tested the susceptibility and survival rate of symbiont Burkholderia in hemolymph of Riptortus in vitro and in vivo. We also examined the susceptibility of symbiont Burkholderia using purified antimicrobial peptides (AMP), pyrrhocoricin-like, thanatin-like and defensin-like AMPs. Finally, we tested inducing ability for AMPs by systemic infection of symbiotic Burkholderia. Gene expression of purified AMPs was not different after systemic infection of both symbiont and in vitro cultured Burkholderia. Surprisingly, in vitro cultured Burkholderia resisted on bacteria injected hemolymph and purified AMPs but symbiont Burkholderia were highly susceptible in bacteria injected hemolymph and purified AMP. These results suggest that symbiont Burkholderia can't survive in the hemolymph after escaping symbiotic organ. Moreover, humoral immunity of host Riptortus is important to prevent spreading and migrating symbiont Burkholderia into the other host tissue or organ from symbiotic organ.

        This study was carried out to know some regional gene difference in carabid beetle, Coptolabrus jankowskii. There are eight subspecies in this species from Korea and it was too difficult to identify by morphological similarity. The mitochondrial ND5 (NADH dehydrogenase subunit 5) gene of Coptolabrus jankowskii from four regions of (Sangju, Daegu, Mt. Jumbong and Jeju island) was compared. The results showed the differences of the base sequence of total 57 sites and the amino acid variation of the 25 sites. The Neighbor-joining tree and Maximum parsimony tree were established based on sequence data of the ND5. In NJ tree, the Jeju area except Songdangri, and Mt. Jeombong region showed close relationship group. In the case of Maximum parsimony tree, the result divided to three separated group, the first connected group was Jeju Songdangri area and the other the Korean peninsula region except Mt. Jeombong. Others were Jeju area except Songdangri, and Mt. Jeombong region.

        Amur silver grass, Miscanthus sacchariflorus is one of the promising biofuel crops. A damage of noctuid pest, Leucapamea askoldis was firstly observed from Amur silver grass in Hwasun silver grass plantation during the survey of insect pests of Amur silver grass in Iksan, Hwasun, and Sancheong plantation areas in Korea. The host of L. askoldis was not known yet in Korea. Thus, M. sacchariflorus was the first known host in Korea. The L. askoldis damage was observed as larval feeding on newly grown shoots of M. sacchariflorus close to soil surface from early April to early May in 2013. Investigated larval density was 1.6 ± 1.1 per m2 on April 4 and damage rate of shoots was 0.8% ± 0.4 per hundred plants on May 4, 2013. The larvae bore into shoots of M. sacchariflorus and then feed inside of plant. The damaged shoots are easily pulled out and distinguished by the boring hole on the shoots.

        Pansies butterflies belong to the genus Junonia are widely distributed throughout Myanmar and 6 species of Junonia have been reported in Myanmar. Molecular comparison of the cytochrome c oxidase subunit I (COI) nucleotide sequences among the species of Junonia was conducted. The nucleotides divergence of COI sequences between species were 2.9 to 7.9% and sequences identity was 92.5-97.7%. The phylogenetic trees were constructed by using Neighbour-joining (NJ) and Maximum likelihood (ML) methods. In both methods, phylogentic trees were showed identical and were revealed that all sequences of Junonia species from Myanmar were in the same cluster with those of the same species from other countries.

        Anatatus orientalis is an important natural enemy parasitizing eggs of lanternfly Lycorma delicatula White. In order to effectively utilize this parasitoid, the parasitism rates of lantern-fly and emergence of its parasitoid were surveyed in different areas. Results showed that the emergences of wasp Anatatus orientalis from different areas had significant differences. Those parasitoids emerged from lantern-fly eggs collected from Yangling of Shaanxi province, Qinhuangdao of Hebei province, and Dagang District of Tianjin City came out focus on May only, while wasps from Yantai of Shandong province showed 2 obvious emergence peaks in May and September respectively. Wasp populations with emergence asynchrony in Yantai area were analyzed by different periods. Results revealed that the emergence percentages in spring (May) were significantly lower than that in fall (September), but the sex ratios did not differ significantly between the two seasons. The emergence asynchrony of parasitoid may be induced by the diapause of species, which is a long-term coevolutional outcome of parasitoid with host for maintaining its population. These findings provided some scientific bases for the rational utilization of this parasitoid in practice.

        The Asian honeybee, Apis cerana, is a native honeybee species in Korea which is important in agriculture for pollination and honey production. For better understanding of the physiology of A. cerana, high-throughput Illumina transcriptome sequencing was performed to analyze the gene expression profiles of queen, worker and larva. A total of 219,799,682 clean reads corresponding to 22.2Gb of nucleotide sequences was obtained from the whole body total RNA samples. The Apis mellifera reference mRNA sequence database was used to measure the gene expression level with Bowtie2 and eXpress software and the Illumina short reads were mapped to 11,459 out of 11,736 A. mellifera reference genes. Total of 9,221 genes with FPKM value greater than 5 of each sample group were subjected to evolutionary genealogy of genes: Non-supervised Orthologous Groups (eggNOG) with BLASTX for gene ontology analysis. The differential gene expression between queen and worker, and worker and larva were analyzed to screen the overexpressed genes in each sample group. in the queen and worker sample group, total of 1,766 genes were differentially expressed with 887 and 879 genes overexpressed over two folds in queen and worker, respectively. In the worker and larva sample group, total of 1,410 genes were differentially expressed with 1,009 and 401 genes overexpressed over two folds in worker and larva, respectively.

        Rice stripe virus (RSV) is one of the serious plant pathogenic viruses for rice and mediated by small brown planthopper, Laodalphax striatellus. So far, the studies have been mainly focused on the interaction between the host plant and the virus. In this study, for better comprehension of the interactions among Rice stripe virus, rice and small brown planthopper, transcriptomes of the RSV-viruliferous (RVLS) and non-viruliferous L. striatellus (NVLS) were comparatively analysed. For this, non-viruliferous L. striatellus were collected from non-infected rice field and fed RSV-infected rice for 5 days. With the RNAs prepared from the RSV-viruliferous and the non-viruliferous small brown planthoppers, we conducted Illumina RNA sequencing (Hiseq 2000) and then two transcriptome databases were generated from RVLS and NVLS, respectively. The transcriptome of RVLS and NVLS were campared to figure out how the gene expression of the insects affected by Rice Stripe Virus. RSV-dependently regulated genes analysed from this study may have important functions in the transmission and replication of RSV.

        The present study was carried out to revise the species of Chlaeniini in Carabidae from South Korea, using both classic taxonomy and molecular analysis. Another aim is to decide the taxonomic position of Chlaenius micans Fabricius of which subgeneric status in the genus has been in question. As a part of the results, Chlaenius spathulifer Bates, 1873 was newly recognized to Korean insect fauna. With this new addition Korean Chlaeniini contains now in total 34 species and two genera. Molecular analyses using CO1 sequences of representative species for six subgenera in Chlaenius were conducted to determine the position of C. micans. The result of the analyses suggested that C. micans did not belong to the subgenus Achlaenius where C. micans had been doubtfully positioned despite distinct morphological differences from the other four species of the variicornis-group (C. sericimicans, C. variicornis, C. kurosawai and C. ocreatus) in the subgenus. This study provids a key to species, photos of adult habitus, photos of male genitalia and redescriptions of 28 Korean species in Chlaeniini.

        The purpose of this study was to determine the effects of swiss ball program training on lower extremity function of old elderly with mild cognitive impairment. The subjects were 30 mild cognitive impairment eldrly people aged between 65 and 80, who were divided into the swiss ball program training group(n=15) and the control group(n=15). The swiss ball program training group engaged in a 50-minute exercise session using. Lower extremity function scale of the two groups were measured before and after the intervention. The results are as follows. According to the swiss ball program training conducted to examine the effects of the training on lower extremity function, Moreover, the two groups showed significant differences in lower extremity function. These results indicate that virtual reality training is effective in improving lower extremity function. To conclusion, swiss ball program was found to have a positive effect on elderly lower extremity function. Swiss ball program training can be proposed as a form of fall prevention exercise for the mild cognitive impairment. Swiss ball program may be helpful to reduce the incidence of dementia and behavioral complications.

        Bacterial soft rot is one of the major disease of Zantedeschia species (Calla lily) caused by Erwinia carotovora. The objectives of this study were i) to screen the most efficient method to determine resistance level against E. carotovora and ii) to evaluate the genetic variability in Zantedeschia genotypes by inoculation of Ecc NHRI-3 and PD 1784 isolates. Four screening tests i.e. leaf disk, petiole, tuber and tuber slice tests were used to determine the resistance level in calla lily. Eleven genotypes from section Zantedeschia were used for variation studies against E. carotovora by using the leaf disk test. It was observed that all genotypes showed variation in resistance level and could be categorized in 3 groups on the basis of their resistance level. Four of the genotypes were resistant against this pathogen whereas, 6 genotypes were moderately resistant and only one genotype was found susceptible. Within section Aestivae, ‘Galaxy’, ‘Florex Gold’, ‘Treasure’ and ‘Mango’ were found very susceptible cultivars, whereas ‘Coral Sunset’, ‘Hazel Marie’ and ‘Neroli’ were less susceptible genotypes. Most of the cultivars from section Aestivae were susceptible to bacterial soft rot and the cultivar ‘Florex Gold’ was identified as susceptible control. It is suggested to perform pre-screening through leaf disk method which proved to a non-destructive test. The pre-screening evaluation can discriminate susceptible cultivar and resistant cultivar. Tuber slice test is more useful in screening subsequent genotypes at a later growth stage in any breeding program.

        The purpose of this study was to test the effect of Gastrocnemius and Low Back-muscle isotonic exercise on static•dynamic standing balance during the period of 4 weeks. This study was two groups pretest-posttest design. Nineteen subjects who were over 22 years old were randomly assigned to either the experimental group that received the Gastrocnemius muscle exercise(n=9) or the low back muscle exercise(n=10) : The former group performed isotonic exercise(plantar flexion), the latter group performed isotonic exercise(trunk extension) a total of 18 times for three times per week for four weeks. Two groups also performed static and dynamic balance before the exercise and 4weeks after the exercise. The data were analyzed by using the paired t-test and independent t-test. The results were as follows: As compared with change of dynamic balance performance capacity at two groups, a significant difference was shown in the test(p<.05), but not in static balance(p>.05). Also, a significant difference of balance between groups was not shown in the test. In this study indicated that gastrocnemius and low back muscle isotonic exercise will have positive impact on standing balance.

        The fact that flip-flops, one of many different types of unstable shoes, are light and relatively easy to put on, accounts for their popularity among people. But because flip-flops rely heavily on the support of a single thong between your first and second toes, they impose a huge amount of pressure onto lower leg. Thus in the following experiment we tried to examine the different effects of flip-flops and running shoes in terms of their effect on muscle activity and fatigue of tibialis anterior and gastrocnemius during walking. In order to measure an electromyogram we used Free EMG system. 10 men and 10 women in running shoes ran on treadmills for 15 minutes at 4.8km/h, 2 days later the same experiment was carried out, but this time, in flip-flops. p value turned out to be greater than .05 and thus there was no considerable difference between the effects of flip-flops and running shoes on muscle activity and fatigue during walking. Therefore we conclude that despite the fact that flip-flops are considered unstable, their effects on muscle activity and fatigue of tibialis anterior and gastrocnemius are negligible.
