        In the current study, a total of 102 common Todarodes pacificus squid caught in the East Sea were investigated for parasitological research. The results revealed that 33 (32.35%) out of 102 squid were infected by Nybelinia surmenicola, the mean intensity was 5.58 parasites per squid, and the maximum abundance was 11. Morphological analysis using a field emission scanning electron microscope showed the characteristic features of N. surmenicola. Molecular identification based on the 28S rRNA gene confirmed the isolated parasite as N. surmenicola, while phylogenetic analysis revealed that N. surmenicola isolated in this study was clustered with N. surmenicola isolated from Japan. This is the first report of phylogenetic characterization of N. surmenicola isolated from Korea.

        The red imported fire ant (Solenopsis invicta) is a species of ant native to South America. The fire ant was inadvertently introduced into USA, Australia, New Zealand, and other Asian countries including China and Taiwan. Since the first report of the fire ant in port city of Busan, Korea in 2017, it was found in many other cities of Korea in following year. To obtain the molecular information of this invasive species, total RNA was extracted from the abdominal segment of the ants collected in Incheon, and subjected to transcriptome sequencing. By using Illumina sequencer platform, 101 base pared-end sequencing generated 2 × 50,064,081 of raw reads to obtain 2 × 45.95 Gbase of quality filtered nucleotide sequences. The in silico cDNA library was constructed by Trinity de novo assembler followed by TransDecoder ORF finder and CD-HIT clustering program to streamline the library. The final version of cDNA library contains 20,442 contigs with protein coding capability. To survey the virome of this ant, these contigs were searched against the viral reference sequences from NCBI RefSeq database with BLASTN program. As a result, contigs which showed high sequence identities with several RNA viruses including previously reported SINV-2 were found from the fire ant. This virome information might give an idea of a shift of virological environment of this newly found ant isolate or population in Korea.

        Varroa destructor is a devastating ectoparasitic mite which attacks Honeybee, Apis mellifera. V. destructor feeds on honeybee hemolymph, and often harbors small RNA viruses such as the deformed wing virus to transmit these viruses in the infested bee hive. To survey the genes of V. destructor, total RNA was subjected to high-throughput transcriptome sequencing to construct in silico cDNA library by using the Illumina HiSeq 2000 platform. Total of 2×107,748,792 paired-end short reads were obtained and quality filtered reads were subjected to Trinity de novo assembler followed by TransDecoder, and CD-HIT program to make a V. destructor reference cDNA library containing 28,023 of clustered contigs with protein coding capacity. These cDNA sequences will help us to understand the molecular biology of V. destructor.

        The squid Todarodes pacificus is a commercially important fishery species in East Asia. As East Asians consume raw or lightly cooked squids, there has been growing concern about parasitic infections associated with squid consumption. In the current study, five squids caught in the East Sea were sampled for parasitological research from the biggest fishery market in Seoul, Republic of Korea. They were dissected and examined for parasites using a light microscope. Proteocephalus sp. was isolated and identified using field emission scanning electron microscopy, PCR, and sequencing. Given that Proteocephalus spp. can be affected by water temperature and geographical characteristics, further research on Proteocephalus spp. with respect to changes in water temperature in the East Sea is important.

        Bone fractures are most often seen in racetrack horses because of the high level of intensity in racing. These issues are the main cause of decreased performance in racehorses. Mesenchymal stem cells (MSCs) have been explored to improve intra-articular therapy in racehorses. MSCs are essential for the repair and regeneration of damaged tissues. In this study, the effect of intra-articular injection of MSCs in racehorses was investigated. Before accessing the MSC therapy, synovial fluids were obtained from the fracture site of racehorses, and adipose tissue was collected for MSC isolation. Using the MSC specific marker, adipose tissue-derived MSCs were identified. The racehorses received intra-articular injection of autologous MSCs (or allogeneic) (3 × 107 cells/3 mL). After 1 or 2 weeks, synovial fluids were collected from racehorses. To test the effect of MSC injection using ELISA, we analyzed inflammatory factors from the untreated samples compared to MSC-treated samples of racehorses. The level of pro-inflammatory factors (interleukin-1β and prostaglandin E2) was significantly decreased in synovial fluids of MSC-injected racehorses, compared to before accessing the MSC therapy, whereas, the level of anti-inflammatory factor (interleukin-10) was higher than prior to accessing the MSC therapy. Further studies are needed to investigate the anti-inflammatory mechanism of MSC in racehorses.

        Pluripotent stem cells can be derived from both pre- and post-implantation embryos. Embryonic stem cells (ES cells), derived from inner cell mass (ICM) of blastocyst are naïve pluripotent and epiblast stem cells (EpiSCs) derived from post-implantation epiblast are primed pluripotent. The phenotypes and gene expression patterns of the two pluripotent stem cells are different each other and EpiSCs thought to be in a more advanced pluripotent (primed pluripotent state) than mouse ES cells (naïve pluripotent state). Therefore, we questioned whether EpiSCs are less potential to be differentiated into specialized cell types in vitro. EpiSCs were isolated from 5.5~6.5 day post coitum mouse embryos of the post-implantation epiblast. The EpiSCs could differentiate into all tree germ layers in vivo, and expressed pluripotency markers (Oct4, Nanog). Interestingly, EpiSCs also were able to efficiently differentiate into neural stem cells (NSCs). The NSCs differentiated from EpiSCs (EpiSC-NSCs) expressed NSC markers (Nestin, Sox2, and Musasi), self-renewed over passage 20, and could differentiate into two neural subtypes, neurons, astrocytes and oligodendrocytes. Next, we compared global gene expression patterns of EpiSC-NSCs with that of NSCs differentiated from ES cells and brain tissue. Gene expression pattern of brain tissue derived NSCs were closer to ES cell-derived NSCs than EpiSC-NSCs, indicating that the pluripotent stem cell-derived somatic cells could have different characteristics depending on the origin of pluripotent stem cell types. * This work was supported by the Next Generation Bio-Green 21 Program funded by the Rural Development Administration (Grant PJ 008009).

        Neural stem cells (NSCs) are self-renewing tripotent cell populations and have capacity of neuronal (neurons) and glial (astrocytes and oligodendrocytes) differentiation. Many researchers have reported that NSCs have therapeutic effects in neurological disease by transplantation. However, it is not easy to obtain NSCs in vitro. Recently, Yamanaka and colleagues showed that somatic cells could be reprogrammed into pluripotent state by enforcing reprogramming factors. Induced pluripotent stem (iPS) cells undergo unlimited self-renewal and have differentiation potential into various types of cells like embryonic stem cells. Direct differentiation into a specialized cell types from iPS cells hold considerable promise for regenerative medicine as well as basic research. Here, we induced differentiation of iPS cells into NSCs in vitro and in vivo, which were compared with embryonic stem (ES) cell-derived NSCs and brain derived NSCs. NSCs from ES and iPS cells were morphologically indistinguishable from brain derived NSCs and stained positive for NSCs markers Nestin and Sox2. ES cells derived NSCs were transcriptionally distinguishable from brain derived NSCs. However, global gene expression pattern were similar but distinct between iPS derived NSCs and brain derived NSCs. Moreover, iPS derived NSCs were spontaneously aggregated upon passaging, formed ES cell like colonies, and finally reactivated Oct4-GFP. The spontaneously reverted GFP-positive cells (iPS-NSC-iPS) expressed similar levels of pluripotency markers (Oct4,Nanog) to ES and iPS cells, and could form germ line chimera. One possible explanation for this phenomenon is that spontaneously re-reprogramming was associated with transgene re-activation when iPS cells were differentiated into NSCs. However, NSCs from dox-inducible iPScells could not be reprogrammed into pluripotent state without doxycycline.         Cytokines are known to function as regulatory molecules that can be produced by virtually every nucleated cell type in the body, including lymphocytes, monocytes/macrophages, epithelial cells, fibroblasts, and many others. Cytokines include lymphocyte-derived factors (lymphokines), monocyte-derived factors (monokines), hematopoietic factors (colony-stimulating factors), connective tissue/ growth factors, and chemotactic chemokines. Cytokines released in response to infection can affect tumor development in different ways. When exposed to infectious agents, cytokines are secreted by sentinel cells, such as macrophages and dendritic cells. These cytokines include interleukin 1 (IL-1) and tumor necrosis factor-α, as well as others, such as IL-6, IL-12, and IL-18. When released in sufficient quantities, these molecules can cause inflammation. Chronic inflammation is highly associated with tumor initiation, promotion, and progression. In this article, we review the roles and mechanisms of cytokines in tumor development.

        Pyrifluquinazon, as a quinazinalone chemical group, based on a new mode of biological activity. It is reported that mode of action is modifies insect behavior, rapidly stopping feeding such that insects starve to death. Time-release feature and mortality effect on M. persicae using different pyrifluquinazon nano type and non-nano type were compared. Pyrifluquinazon nano type was formulated with different molecular weight and density of used chitosan (CS 30000 0.1% and CS 3000 0.3%). In the CS 30,000 0.1%, the mortality was weakly occurred at early time, but steadily increased after 4days. Finally, we confirmed more than 70% mortality as a peak at 16days. In CS 3000 0.3%, the mortality showed about 70% until 18days as a effective controlled release. Also, We examine time-release feature and mortality effect on M. persicae according to the different pyrifluquinazon nano type(CS 30000 0.1% and CS 3000 0.3%) of concentrations. The CS 30000 0.1% bioassay results of different concentration were showed that the highest concentration(100ppm) was measured better mortality than other concentration at 0 day, but cannot confirm different effect about dissimilar concentration. However, increasing rates of M. persicae were low as treatment concentrate was high. In CS 3000 0.3% 100ppm concentration bioassay result, aphid mortality reached peak at 24 days and increasing rate also low. Additionally, for the comparing of bioassay and feeding behavior of M. persicae against pyrifluquinazon nano types and non-nano type, EPG technique was carried out. In case of non nano type, feeding inhibition efficacy was showed during 4 days after treatment, but appeared similar level with control after 10days. In CS 3000 0.3% 50ppm, residual efficacy was specially showed until 28days after treatment whereas treatments with CS 30000 0.1% were similar to the control after 22days. These result show that the change of feedinng behavior and motrality of M. persicae is correlated with the change of nano type or non nano type of pyrifluquinazon.

        Several species of the genus Aphidius are used in biological control programs against aphid pests throughout the world and their behavior and physiology are well studied. But despite knowing the importance of sensory organs in their behavior, their antennal structure is largely unknown. In this study, the external morphology and distribution of the antennal sensilla on the antennal of both female and male adults of A. colemani were described using scanning electron microscopy (SEM). Generally, the filaform antennae of males (1,515.20±116.48 ㎛) are longer than females (1,275.06±116.42㎛). Antennae of this species is made up of scape, pedicel and flagellomeres. Male and female antennae differed in the total number of flagellomeres as 15 in males and 13 in females. Female and male antennae of A. colemani has samely seven types of sensilla. We classified sensilla placodea, Bohm bristles, 2 types of sensilla coeloconica, , 2 types of sensilla basiconica as with a tip pore and with wall pores, sensilla trichodea. In addition, the possible functions of the above sensilla types are discussed in light of previously published literature; mechanoreception(Bohm bristles, sensilla coeloconicaⅡ and sensilla trichodea) and chemoreception(sensilla coeloconicaⅠ, sensilla basiconicaⅠ,Ⅱ and sensilla placodea). Future studies on the functional morphology of the antennal sensilla of A. colemani using transmission electron microscopy (TEM) coupled with electrophysiological recordings will likely confirm the functions of the different sensilla identified in this study.

        Somatic cell nuclear transfer in cattle has limited efficiency in terms of production of live offspring due to high incidence of fetal failure after embryo transfer to recipients. Such low efficiency of cloning could possibly arise from abnormal and poorly developed placenta. In the present study the placental proteome in late pregnancy established from in vitro fertilization (IVF) and nuclear transfer (NT) was analysed. Proteome alternation was tested using two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI- TOF). Comparing placenta from NT embryos to those from IVF counterparts, significant changes in expression level were found in 18 proteins. Of these proteins 12 were not expressed in NT placenta but expressed in IVF counterpart, whereas the expression of the other 6 proteins was limited only in NT placenta. Among these proteins, cytokeratin 8 and vimentin are considered to be involved in regulation of post-implantation development. In particular, cytokeratin 8 and vimentin may be used as makers for placental development during pregnancy because their expression levels changed considerably in NT placental tissue compared with its IVF counterpart. Data from 2-DE suggest that protein expression was disorientated in late pregnancy from NT, but this distortion was eliminated with progression of pregnancy. These findings demonstrate abnormal placental development during late pregnancy from NT and suggest that alterations of specific placental protein expression may be involved in abnormal function of placenta.

        본 연구에서는 SWAT 모형과 random forest를 이용하여 미래 기후변화에 따른 한강유역(34,148 km2)의 수생태계 건강성을 평가하였다. 국립 환경과학원에서 8년간(2008~2015년) 봄철(4~6월)에 모니터링한 부착돌말류 지수(TDI), 저서형 대형무척추동물지수(BMI), 어류평가지수(FAI)는 0~100점, A~E등급으로 평가되며, 이를 본 연구에서 사용하였다. 수생태 건강성에 영향을 미치는 변수로는 수질(T-N, NH4, NO3, T-P, PO4)과 수온을 선정하였으며, 수질 오염도가 낮은 경우에는 수생태계 건강성 점수가 광범위하게 분포되지만 수질 오염도가 높은 경우 수생태계 건강성 점수가 낮아지는 역상관관계를 확인하였다. 기계학습의 분류 분석 기법 중 하나인 random forest 모델을 이용한 세 개의 수생태 건강성 지수 등급 분류 결과 정밀도, 재현율, f1-score 모두 0.81 이상의 예측 정확도를 나타내었다. 기상청의 HadGEM3-RA RCP 4.5와 8.5 시나리오를 적용한 미래 SWAT 수문, 수질 결과 기저유출의 증가로 인해 질소 계열 수질 농도는 기준년도 대비 최대 43.2% 증가하였고, 지표유출 감소로 인해 인 계열 수질 오염도는 최대 18.9% 감소하는 것으로 분석되었다. 미래 FAI, BMI의 등급은 개선되는 경향을 보이지만 TDI는 등급이 악화되는 것으로 나타 났다. 이를 통해 TDI는 질소 계열 수질에 민감하고 FAI, BMI는 인 계열 수질에 더 민감하다고 판단하였다.

        This study aimed to analyze difference in clinical findings, including coronary artery complications, in patients with Kawasaki disease and respiratory symptoms with several respiratory infections. We studied 182 pediatric patients diagnosed with Kawasaki disease. Examinations for respiratory viral polymerase chain reaction were conducted in the group of patients with respiratory symptoms. Echocardiography was perfomed by a pediatric cardiologist, and laboratory findings were evaluated. Clinical manifestations and laboratory findings based on medical records were compared. There were no differences between patients with and without respiratory viral infections with respect to age, male-female ratio, coronary artery complications, Kawasaki disease-specific clinical manifestations, duration of fever, duration of hospitalization, or recurrence rate. There was a significant difference in C-reactive protein levels (55.6 vs. 73.9 mg/L) between the two groups, but the other laboratory findings. The rate of respiratory infections in pediatric patients with Kawasaki disease was similar to those reported in previous studies, and clinical manifestations and laboratory findings were not significantly different between the groups.
