The purpose of this thesis is to build a dirt-walled house based on the principle of temperature/humidity control in the underground storage room and examine the constant temperature/humidistat effects. The result of the experiment showed that (1) the temperature inside the dirt-walled house was remained lower than the exterior temperature by 3.5-8 'C. And, the daily amount of temperature fluctuation turned out to be 2.5 - 5 'C, which was not significant. (2) The change rate of the humidity was maintained relatively steady at around 4 - 9%.

Food quality indicator is a simple device to communicate information of food quality to consumers. Change of specific gas composition in the headspace of food packaging can be considered as a food quality indication. For examples, level of carbon dioxide (CO2) increases in the headspace of packaged kimchi which is fermented over time during storage. The elevated ammonia (NH3) level inside a food package is an indication of spoilage. In this study, CO2 indicators and NH3 indicators were developed to monitor food quality and their performance were evaluated. Especially, the correlation between the food quality change and the indicator change were studied during storage. Two types (chitosan-based and whey protein isolate-based) of CO2 indicators was developed to monitor the CO2 concentration in the headspace of a food package. The CO2 indicators showed a visual change depending on CO2 level in the package headspace and were installed on the packaged kimchi and the packaged makgeolli in order to notify their ripening stage. NH3 indicators were developed to monitor the headspace NH3 concentration in the packaged fishery products such as fermented skate. The NH3 indicators showed a prominent color change depending on NH3 level as the skate was fermented over time. In addition, the polylactic acid (PLA)-based package box installing NH3 indicator was developed for the fermented skate and the color change of the indicator was evaluated at different storage temperatures over time. In conclusion, the gas indicators incorporated into the package was able to simply inform food quality over time.

We developed the smart mosquito counter device (height 1080 mm x width 560 mm x diameter 320 mm, 220V 60Hz 30 W), which can attract the blood sucking female mosquitoes by emitting CO2 gas (300ml/min), can count the number of the captured mosquitoes by an infra-red beam area sensor, can identify the some mosquito species by analyzing mosquito image patterns using RGB (Red, Green, and Blue), and can send the captured mosquitoes’ number and pictures through the CDMA module at the real time. We operated the smart mosquito device and we could know when any mosquito peak time was in a day as well as what the mosquito species are and the captured mosquito numbers are. We found that the accuracy of the smart device was about over 95% compared the real mosquito data including numbers and images and transmitted data by CDMA. Because we can know the mosquito occurrence peak times, locations, and mosquito species by this device, we can efficiently control mosquitoes at the right time, and we can alert the public for any mosquito-born diseases based on the mosquito occurrence data. We can improve the quality of mosquito vector control with the functions of this device and we will develop the smart device to identify more mosquito species.

Entomopathogenic fungi infect some arthropod pest and have been used for biological control. Some entomopathogenic fungi have high potential in insect pest management worldwide, and most of researches were given to Hypocreales order of Ascomycota, but little in Zygomycota, such as Entomophthorales in our country. We have identified some species belonging to Entomophthorales and investigated cultural features and ecology of Entomophaga aulicae. E. aulicae in sweet potato fields, where Aedia leucomelas was a dominant pest, had caused epizootics from 2002–2005. E. aulicae-infected A. leucomelas larva were mostly found from August to October and its occurrence was significantly related to the precipitation. E. aulicae were mainly observed in dead A. leucomelas larval populations (infection rate = 41.3% in 2002), rather than other lepidopteran pests. E. aulicae was morphologically and genetically identified and its virulence was characterized in laboratory conditions. Additionally, pest infection by Zoophthora radicans, Neozygites floridana and some Entomophthorales have been identified. On the basis with this information, we need research to predict the prevalence and to develop biological control agent using Entomophthorales including E. aulicae that contribute to regulation of host populations.

Insects impact human health through vector-borne diseases and cause major economic loses through damaging crops and stored agricultural products. Insect-specific growth regulators (IGR) represent attractive control agents because of their safety to the environment and humans. Here, we report identification of plant compounds that are antagonists of the insect-specific juvenile hormone (PJHANs), using the yeast two hybrid system transformed with the mosquito JH receptor as a reporter assay. We show that these compounds act by inhibiting larval growth and reproduction in mosquitoes. We also demonstrate that PJHANs affect the JH receptor, Methoprene-tolerant (Met), by disrupting its complex with CYCLE, formation of which is required for mediating JH action. We isolated five diterpene secondary metabolites with JH antagonist activity from two plants, Lindera erythrocarpa and Solidago serotina. They are effective in causing mortality of mosquito larvae at relatively low LD50 values. Two of these diterprenes affect Met function, leading to reduction in expression of Met target genes and causing retardation of follicle development in mosquito ovaries. Developing potent compounds counteracting JHaction (JH antagonists) would find a wider range of control applications. However, so far such JH antagonists have not been developed. Here, we report the discovery of potent JH antagonists in plants, which represents an innate resistance mechanism of plants against insect herbivores. These newly discovered plant JH antagonist compounds could be used as the starting material for developing novel insecticides.

Background and Purpose: The Ankle-Brachial Index (ABI) is the ratio of blood pressure in the lower legs to that in the arms. The intima-media thickness (IMT) of extracranial carotid arteries determined by B-mode ultrasound is a measurable index of the presence of atherosclerosis. A low ABI and a high carotid IMT are independently related to increased risk of cardiovascular events. This study examined the association between carotid IMT and ABI in patients with ischemic stroke. Materials and Methods: Retrospectively, 116 hospitalized patients with ischemic stroke were recruited. Using a pulse wave velocity ABI device along with carotid duplex sonography, we measured carotid IMT and ABI and investigated the correlation between average values. Results: There was a significant difference in carotid IMT between the normal and abnormal ABI groups (P=0.0262). The group with an abnormal ABI was more than five times as likely to have increased carotid IMT as the group with a normal ABI (age, sex-adjusted OR 5.67 (95% CI 1.85~17.38)). The ABI and carotid IMT showed a weak inverse linear correlation in patients with ischemic stroke (correlation coefficient −0.378 after adjusting for age and sex). Conclusion: Our study suggests that an abnormal ABI is associated with a high carotid IMT in patients with ischemic stroke.

As a preceding study for investigating the influence of sound wave stimulus on Arabidopsis thaliana metabolomics, the polar secondary metabolomes of the plant were determined using high performance liquid chromatography coupled with tandem mass spectrometry. A total of 10 polar secondary metabolomes were characterized and quantified. Among them, 4 metabolomes, p-coumaroylagmatine isomer (7 and 8), p-coumaroylagmatine isomer (9 and 10) were identified in the plant for the first time. The validation was conducted in terms of linearity, recovery, precision, limit of detection (LOD) and limit of quantification (LOQ). The validated method was applied to the simultaneous quantification of the 10 polar secondary metabolomes.

Runx2 and Osterix, the transcription factors for osteoblast differentiation, are known as fundamental factors to regulate the development of calcified tissues. However, the biological functions of these factors in the development of the periodontal tissues remain unclear. In this study, we investigated the distribution of Runx2 and Osterix during periodontal tissue development of the mice. Mandibles from 14-day-old mice were prepared for paraffin section. Serial sections of the mandible containing 1 st molar tooth germs were obtained as a thickness of 7 μm. Some sections were stained with hematoxylin and eosin. Others were used for immunohistochemistry for PCNA, Runx2, and Osterix. Epithelial cells in growing end of Hertwig’s epithelial root sheath (HERS) and mesenchymal cells adjacent to the growing end of HERS expressed PCNA. Undifferentiated mesenchymal cells and hard tissue forming cells like cementoblasts and osteoblasts in early stage of differentiation expressed Runx2. Fully differentiated cementoblasts and osteoblasts secreting matrix proteins expressed Osterix. However, the cells terminated the matrix formation did not express Osterix. Periodontal ligament cells expressed Runx2 and Osterix. Pulp cells expressed Runx2 only. These results suggest that Runx2 and Osterix might regulate the differentiation of cementoblasts in the same manner as osteoblasts. Runx2 might participate in the process of cementoblast differentiation in early stage, whether Osterix might regulate the maturation and matrix synthesis of the cells.

Nematicidal activity of aliphatic compounds was tested against pien wood nematode, Bursaphelenchus xylophilus. There was a significant difference in nematicidal activity among function groups. In a test with alkanols and 2-alkenols, compounds with C8-C11 chain length showed 100% nematicidal activity at 0.5 mg/mL concentration. C6-C10 2-alkenals exhibited >95% nematicidal activity, but the other compounds with C11-C14 chain length showed weak activity. Nematicidal activity of alkanoic acids with C7-C11 chain length was strong. Whole compounds belonging to hydrocarbons, alkanals and alkanoic acetate showed weak nematicidal activity at 0.5 mg/mL concentration. Nematicidal activity of compounds which showed strong nematicidal activity at 0.5 mg/mL concentration was tested at a lower concentration. At 0.25 mg/mL concentration, whole compounds except C8 alkanol, C8 2-alkenol and C7 alkanoic acid showed >80% nematicidal activity. C9-C11 alkanols, C10-C11 2-alkenols, C8-C9 2-alkenals and C9-C10 alkanoic acids showed >80% nematicidal activity at 0.125 mg/mL concentration. Only C11 alkanol exhibited strong nematicidal activity at 0.0625 mg/mL concentration
