        Phosphoprotein Enriched in Astrocytes (PEA15) is a 15kD-sized intracellular signaling protein, highly expressed in astrocytes and constitutively expressed in peripheral tissues. Recently it was found that PEA15 expression was elevated in patients suffering type 2 diabetes and suggested to be involved in the syndrome of insulin resistance. To investigate the functional role of PEA15 for the control of blood glucose level, we produced a transgenic pig over-expressing mouse PEA15 (mPEA15). As a model animal, pig has many advantages. They have a higher fecundity and a short generation time and are physiologically similar to human. Using the transgenic pig, we carried out a series of experiments to establish a link between PEA15 expression and the insulin resistance. Our results suggested that, compared with control pig, mPEA15 pig has, (1) a higher blood resistin level, (2) a lower cell membrane-embeded GLUT4 level, and (3) a lower glucose clearing ability based on oral glucose tolerance test (OGTT). When our results combined, it can be concluded that mPEA15 over-expressing pig has many symptoms of insulin resistance and these pigs will become a useful disease model to investigate diabetes mellitus in the near future.

        The objective of this study was to determine the effect of macrophages on growth of human colon cancer cells. The results showed that co-culture of colon cancer cells with macrophages inhibited the growth of colon cancer cells (HCT116 and SW620) depending on the number of macrophages, RAW 264.7 cells, and activated THP-1 cells accompanied by down regulation of pSTAT3 in cancer cells. We also found that expression and release of cancer cell growth inhibitory cytokines, IL-1 receptor antagonist (IL-1ra) and IL-10, was increased in macrophages. Blocking of the STAT3 pathway with specific inhibitor and siRNA of STAT3 abolished the growth of colon cancer cells and expression of IL-1ra and IL-10. In addition, neutralization of IL-1ra and IL-10 with antibodies resulted in reversal of macrophage-induced inhibition of cancer cell growth. These data showed that IL-1ra and IL-10 released from macrophages inhibit growth of colon cancer cells through inhibition of the STAT3 pathway.

        To perform imaging observations of optically red objects such as high redshift quasars and brown dwarfs, the Center for the Exploration of the Origin of the Universe (CEOU) recently developed an optical CCD camera, Camera for QUasars in EArly uNiverse (CQUEAN), which is sensitive at 0.7-1.1 μm. To enable observations with long exposures, we develop an auto-guiding system for CQUEAN. This system consists of an off-axis mirror, a baffle, a CCD camera, a motor and a differential decelerator. To increase the number of available guiding stars, we design a rotating mechanism for the off-axis guiding camera. The guiding field can be scanned along the 10 arcmin ring offset from the optical axis of the telescope. Combined with the auto-guiding software of the McDonald Observatory, we confirm that a stable image can be obtained with an exposure time as long as 1200 seconds.

        The dissemination process of agricultural research and development (R&D) results has somewhat different characteristics from that of typical R&D results. However, these characteristics are not adequately considered on the basis of an examination of the current performance system, the resulting management plans, and strategies for the application and dissemination of the results of agricultural R&D in Korea. The performance evaluation indicator exposed the problem of the inadequate consideration of the characteristics of each of these areas, particularly the lack of unified R&D-related institutions and the inadequacy of the system to monitor outcomes. To address these shortcomings in the agricultural R&D programs in Korea, the policies pertaining to agricultural R&D performance, results management, and dissemination in the U.S. and Japan were examined. Based on these investigations, we proposed strategies to improve the agricultural R&D policies in Korea.

        This study describes the development of a web-based system that collects all data generated in the research conducted to set pre-harvest residue limits (PHRLs) for agricultural product safety control. These data, including concentrations of pesticide residues, limit of detection, limit of quantitation, recoveries, weather charts, and growth rates, are incorporated into a database, a regression analysis of the data is performed using statistical techniques, and the PHRL for an agricultural product is automatically computed. The development and establishment of this system increased the efficiency and improved the reliability of the research in this area by standardizing the data and maintaining its accuracy without temporal or spatial limitations. The system permits automatic computation of the PHRL and a quick review of the goodness of fit of the regression model. By building and analyzing a database, it also allows data accumulated over the last 10 years to be utilized.

        Sepsis is a clinical syndrome defined as a systemic inflammatory response to infection. Eritoran is a synthetic lipid A derivative that competes with lipopolysaccharide in binding to the identical site of myeloid differentiation-2/toll-like receptor 4 complex. Eritoran is effective in decreasing the septic mortality of Gram-negative bacteria-infected animals. Eritoran has been highlighted as a candidate drug for treatment of endotoxemia in phase I clinical studies with healthy human volunteers. A phase II trial of eritoran has been conducted in patients with severe sepsis. Intravenous infusion of eritoran reduced the mortality rate, as compared with the placebo group, in sepsis patients at a high risk of mortality according to acute physiology and chronic health evaluation-II scores. A phase III study of eritoran was completed in 2011. The results appear to be disappointing as no statistically significant difference in all-cause mortality was observed between the eritoran treatment group and the placebo group on day 28 in sepsis patients with a high risk of death. In this review, we focus on the rationale for the use of eritoran in treatment of sepsis as well as its clinical applications.

        A steady-state analysis and a simple dynamic model as simplified methods are developed, and results of energy consumption loads are compared with results obtained using computer to evaluate the analytical solution. Before obtaining simplified model a mathematical model is formulated for the effect of wall mass on the thermal performance of four different houses having various wall construction. This analytical study was motivated by the experimental work of Burch et al. An analytical solution of one-dimensional, linear, partial differential equation for wall temperature profiles and room air temperatures is obtained using the Laplace transform method. Typical Meteorological Year data are processed to yield hourly average monthly values. This study is conducted using weather data from four different locations in the United States: Albuquerque, New mexico; Miami, Florida; Santa Maria, California; and Washington D.C. for both winter and summer conditions. The steady state analysis that does not include the effect of thermal mass can provide an accurate estimate of energy consumption in most cases except for houses #2 and #4 in mild weather areas. This result shows that there is an effect of mass on the thermal performance of heavily constructed house in mild weather conditions. The simple dynamic model is applicable for high cycling rates and accurate values of inside wall temperature and ambient air temperature.

        The present case study highlights the effects of a novel Comprehensive Hand Repetitive Intensive Strengthening Training (CHRIST) on morphological changes and associated upper extremity (UE) muscle strength and motor performance in a child with spastic quadriplegic cerebral palsy (CP). The Child, a 10-year-old girl with spastic quadriplegic CP, was treated with CHRIST for 60 minutes a day, five times a week, for 5 weeks. The CHRIST was designed to improve motor function and strength. Clinical tests including the modified Wolf Test, Jebsen-Taylor Hand Function Test, and Pediatric Motor Activity Log questionnaire were used to determine motor function. Ultrasound imaging was performed to determine the changes in the cross-section area (CSA) of the extensor carpi radialis (ECR) and triceps brachii (TRI). Muscle strength was measured with a dynamometer at pretest, and post-test, and 3-month follow-up. Ultrasound imaging data showed that the CSAs of both ECR and TRI muscles were enhanced as a function of the intervention. These changes were associated with muscle strength and motor performance and their effects remained even at a 3-month follow-up test. Our results suggest that the CHRIST was effective at treating muscle atrophy, weakness and motor dysfunction in a child with spastic quadriplegic CP.

        Ascotis selenaria, a major geometridae moth in citrus trees, annually damages the citrus leaves and fruits. The surface of young citrus fruit were usually fed by 1st larva of A. selenaria after landing or stepping onto the citrus fruits. To protect the larval damages of citrus fruits needs to predict the accurate occurrence time of the 1st larva for spraying. Because larval occurrences is dependent on the oviposition of adult female and the eggs were not found in/on citrus trees, oviposition model of A. selenaria linked with the egg development model will be helpful of protecting larval damages on citrus fruits. Adult longevity, survival and fecundity of A.selenaria was investigated at 13, 16, 20, 24, 28, 30, 32, and 35℃. The longevity decreased as the temperature increased and the female development rates (1/median longevity) were well described by a modified sigmoid model, which was used to calculate the adult physiological age. Description of the total fecundity was used by a non-linear model: The maximum fecundity of A. selenaria was estimated as 2490 eggs and peaked temperature was 19.7℃ according to the fecundity model. The cumulative age-specific oviposition rate and the age-specific survival rate was well described by 2 parameters Weibull function and a reverse logistic curve respectively. Total fecundity model, age-sepecific oviposition model, and age-specific survival model were incorporated into the oviposition model.

        A mathematical model is formulated to study the effect of wall mass on the thermal performance of four different houses of different construction. This analytical study was motivated by the experimental work of Burch et al. An analytical solution of one -dimensional, linear, partial differential equation for wall temperature profiles and room air temperatures is obtained using the Laplace transform method. Typical Meteorological Year data are processed to yield hourly average monthly values. These discrete data are then converted to a continuous, time dependent form using a Fast Fourier Transform method. This study is conducted using weather data from four different locations in the United States: Albuquerque, New mexico; Miami, Florida; Santa Maria, California; and Washington D.C. for both winter and summer conditions. A computer code is developed to calculate the wall temperature profile, room air temperature, and energy consumption loads. Three sets of results are calculated one for no auxiliary energy and two for different control mechanism -- an on-off controller and a proportional controller. Comparisons are made for the cases of two controllers. Heavy weight houses with insulation in mild weather areas (such as August in Santa Maria, California) show a high comfort level. Houses using proportional control experience a higher comfort level in comparison to houses using on-off control. The result shows that there is an effect of mass on the thermal performance of a heavily constructed house in mild weather conditions.

        Somatostatin (SST) is a known neuromodulator of the central nervous system. The substantia gelatinosa (SG) of the trigeminal subnucleus caudalis (Vc) receives many thinmyelinated Að-fiber and unmyelinated C primary afferent fibers and is involved in nociceptive processing. Many studies have demonstrated that SST plays a pivotal role in pain modulation in the spinal cord. However, little is yet known about the direct effects of SST on the SG neurons of the Vc in adult mice. In our present study, we investigated the direct membrane effects of SST and a type 2 SST receptor agonist, seglitide (SEG), on the SG neurons of the Vc using a gramicidin-perforated current clamp in adult mice. The majority (53%, n = 27/51) of the adult SG neurons were hyperpolarized by SST (300 nM) but no differences were found in the hyperpolarization response rate between males and females. When SST was applied successively, the second response was smaller (76±9.5%, n=19), suggesting that SST receptors are desensitized by repeated application. SST-induced hyperpolarization was also maintained under conditions where presynaptic events were blocked (75±1.0%, n=5), suggesting that this neuromodulator exerts direct effects upon postsynaptic SG neurons. SEG was further found to induce membrane hyperpolarization of the SG neurons of the Vc. These results collectively demonstrate that SST inhibits the SG neuronal activities of the Vc in adult mice with no gender bias, and that these effects are mediated via a type 2 SST receptor, suggesting that this is a potential target for orofacial pain modulation.

        Poly propylene carbonate (PPC)와 cloisite 20B (PPC/C-20B)을 solution method를 통하여 합성하였고, 이를 통해 합성된 나노 조성물의 morphology, 열적 특성, 수분 흡수의 특성을 평가하였다. 나노 조성물의 구조는 X-ray diffraction (XRD) 로 확인하였고, 열적 특성은 thermal gravimetric analysis (TGA)와 differential scanning calorimetric (DSC)를 이용해 분석하였다. TGA와 DSC의 결과를 통해 나노 조성물은 기존의 PPC에 비해 높은 열적 안정성을 가짐을 확인할 수 있었다. DSC측정에서 5 wt%의 clay를 포함한 nanohybrid의 유리전이온도는 순수한 PPC의 21℃에서 30℃로 9℃ 증가하였고, TGA 측정을 통해 확인한 열분해온도(Td50%)는 순수 PPC에 비해 23℃가 높아졌음을 확인하였다. 또한 PPC 나노 조성물의 수분 흡수량은 기존의 PPC에 비해 상당히 감소하였다. 이는 clay의 PPC matrix 구조 내에 존재함으로 인해 수분 흡수를 감소시킨 것으로 해석할 수 있다. 수분 흡수는 코팅 막의 분해를 유발하고 물리적, 기계적 성능에 영향을 미친다. 따라서 나노 조성물로 인한 PPC의 열적 안정성, 수분흡수도 향상은 PPC의 사용과 실제 공정에 중요한 변수로 작용할 수 있다.

        The Notch signaling pathway regulates cell proliferation, apoptosis and cell fate decision. Recent preclinical and clinical evidence supports a pro-oncogenic function for Notch signaling in several solid tumors including breast and prostate cancer. Consequently, there is increasing interest in targeting Notch signaling therapeutically in cancer patients. Notch inhibitors, particularly gamma-secretase inhibitors, are being investigated as candidate cancer therapeutic agents. However, rational targeting of Notch signaling in cancer will require a systematic exploration of several areas that remain incompletely understood. Therefore, a clear understanding of the Notch signaling and its cross-talk with other signaling cascade will increase our ability to design rational combination regimens for cancer therapy.
