Environmental DNA (eDNA) has emerged as a promising tool for aquatic biodiversity monitoring, yet its collection in lentic ecosystems remains technically constrained by filtration capacity and field logistics. In this study, we applied a novel eDNA concentration system, QuickConcTM, to evaluate freshwater mussel diversity in lakes, and compared its performance with the conventional GF/F filtration method. Water samples were collected from four reservoirs at surface, mid, bottom, and waterside layers, and processed using both filtration techniques. Metabarcoding of mitochondrial 16S rDNA revealed that QuickConcTM captured a higher average number of amplicon sequence variants (ASVs) and exhibited greater species richness and diversity indices (Shannon and Simpson), although the differences were not statistically significant. QuickConcTM samples showed a greater capacity to detect rare taxa and to recover higher ASV richness in certain cases, suggesting its potential to enhance biodiversity resolution. Species composition remained consistent across methods, with Cristaria plicata and Sinanodonta lauta being dominant in both cases. However, slight spatial variations in species assemblages were observed between center and waterside sampling points, highlighting the influence of habitat heterogeneity on eDNA distribution. Overall, our results demonstrate that the QuickConcTM system offers a practical and efficient alternative to traditional filtration methods for eDNA-based freshwater mussel monitoring, particularly in environments with high suspended solids. The findings underline the need for adaptive sampling strategies that consider both methodological and ecological factors when designing eDNA surveys in lentic ecosystems.
Springtails (class Collembola) play a crucial role in soil ecosystems. They are commonly used as standard species in soil toxicity assessments. According to the ISO 11267 guidelines established by the International Organization for Standardization (ISO), Allonychiurus kimi uses adult survival and juvenile production as toxicity assessment endpoint. Conventional toxicity assessment methods require manually counting adults and larvae under a microscope after experiments, which is time-consuming and laborintensive. To overcome these limitations, this study developed a model using YOLOv8 to detect and count both adults and juveniles of A. kimi. An AI model was trained using a training dataset and evaluated using a validation dataset. Both training and validation datasets used for AI model were created by picturing plate images that included adults and larvae. Statistical comparison of validation dataset showed no significant difference between manual and automatic counts. Additionally, the model achieved high accuracies (Precision=1.0, Recall=0.95 for adults; Precision=0.95, Recall=0.83 for juveniles). This indicates that the model can successfully detect objects. Additionally, the system can automatically measure body areas of individuals, enabling more detailed assessments related to growth and development. Therefore, this study establishes that AI-based counting methods in toxicity assessments with offer high levels of accuracy and efficiency can effectively replace traditional manual counting methods. This method significantly enhances the efficiency of large-scale toxicity evaluations while reducing researcher workload.
Autonomous vehicle (AV) technology is rapidly entering the commercialization phase driven by advancements in artificial intelligence, sensor fusion, and communication-based vehicle control systems. Real-world road testing and pilot deployments are increasingly being conducted, both domestically and internationally. However, ensuring the safe operation of AVs on public roads requires not only technological advancement of the vehicle itself but also a thorough pre-evaluation of the road environments in which AVs are expected to operate. However, most previous studies have focused primarily on improving internal algorithms or sensor performance, with relatively limited efforts to quantitatively assess how the structural and physical characteristics of road environments affect AV driving safety. To address this gap, this study quantitatively evaluated the compatibility of road environments for AV operation and defined the road conditions under which AVs can drive safely. Three evaluation scenarios were designed by combining static factors such as curve radius and longitudinal gradient with dynamic factors such as level of service (LOS). Using the MORAI SIM autonomous driving simulator, we modeled the driving behaviors of autonomous vehicles and buses in a virtual environment. For each scenario, the minimum time to collision (mTTC) from the moment the AV sensors detected a lead vehicle was calculated to assess risk levels across different road conditions. The analysis revealed that sharper curves and lower service levels resulted in significantly increased risk. Autonomous buses exhibited a higher risk on downhill segments, autonomous vehicles were more vulnerable to uphill slopes and gradient transitions. The findings of this study can be applied to establish road design standards, develop pre-assessment systems for AV road compatibility, and improve AV route planning and navigation systems, thereby providing valuable implications for policy and infrastructure development.

This study was conducted to evaluate the germination rate, chemical composition, and in vitro digestibility of sprouted barley (Hordeum vulgare L.) and wheat (Triticum aestivum L.) sprouts depending on cultivar and growth duration. Four cultivars Keunalbori1ho, Saekeumkang, Arijinheuk, and Jokyoung were tested under hydroponic and nutrient solution conditions. The germination rate was significantly higher under hydroponic conditions compared to nutrient solution treatment. Sprouts were harvested at 0, 4, 6, and 8 days for subsequent analysis. Chemical compositions, such as dry matter (DM), crude protein (CP), ether extract (EE), and crude ash (CA), were analyzed following AOAC (2005) protocols, while neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents were determined according to the method of Van Soest et al. (1991). In vitro digestibility was assessed by incubating ground samples with rumen buffer in DaisyII incubators for 48 h. At day 6 of growth, Keunalbori1ho showed the highest NDF and ADF concentrations (48.6% and 26.2%), reflecting its high structural fiber and the lowest digestibility (IVDMD 52.2%, IVNDFD 37.2%). However, it maintained consistent nutritional characteristics across all growth stages, suggesting potential as a stable forage source. In contrast, Saekeumkang exhibited the highest IVNDFD (59.1%) along with relatively low NDF (30.5%) and ADF (16.7%) values (p<0.05), indicating superior digestibility likely attributed to a simpler cell wall structure and higher NFC availability. Notably, digestibility sharply declined by day 8, implying that the optimal harvest window lies around day 6. Therefore, this study suggests that the cultivars Keunalbori1ho and Saekeumkang possess complementary strengths in terms of chemical composition and digestibility, underscoring the importance of optimizing cultivar selection and harvest timing to enhance the potential of cereal sprouts as high quality forage.
Dietary fiber plays a important role in improving gut micriobiome and reproductive performance in gestating sows. This study aimed to evaluate effects of dietary supplementation with domestically produced alfalfa, provided in pelleted or mashed form, on body weight, body condition, litter performance, fecal score and fecal microbiome in gestating sows. A total of nine crossbred F1 gilts (Yorkshire × Landrace) with an average body weight of 230.33 ± 12.63 kg at 50 days of gestation were randomly assigned to three dietary treatments (n = 3 per treatment): (1) control (CON, basal diet), (2) Pellet (basal diet + 10% pelleted alfalfa), and (3) Mash (basal diet + 10% mashed alfalfa). Results indicated that alfalfa supplementation had no significant effect on weight gain during gestation (p>0.05), although sows in both alfalfa-supplemented groups showed numerically reduced weight loss during lactation compared to the control. The mashed alfalfa improved average daily weight gain of piglets during lactation (p<0.05). Moreover, the pelleted alfalfa alleviated constipation and numerically increased the relative abundance of positive microbes such as Firmicutes (phylum level), Lactobacillaceae and Ruminococcaceae (family level), and Clostridium_sensu_stricto_1, Lactobacillus, and Ruminococcaceae_UCG_002 (genus level) (p<0.05). On the other hands, it numerically decreased the relative abundance of negative microbes such as Proteobacteria in phylum level and Escherichia_Shigella in genus level. These findings suggest that domestically grown alfalfa, particularly in pellet or mash form, may be a valuable dietary fiber source for gestating sows to support piglet performance, gut health, and welfare. However, further studies with larger sample sizes are warranted to confirm these effects.
The Indian Ocean is the second-largest tuna fishing ground in the world, accounting for approximately 1.2 million tonnes (23%) of the estimated 5.2 million tonnes of global commercial tuna catch in 2023. This study examined the relationship between tuna catches, specifically skipjack, bigeye, and yellowfin tunas, and prey biomass (Nautical Area Scattering Coefficient, NASC) estimated from acoustic surveys conducted in the southwestern Indian Ocean from 20 April to 15 May 2019. Environmental variables were derived from the Copernicus Ocean Model, and tuna length data from the IOTC. The estimated total tuna catch in the study area was approximately 166,400 tonnes, with the northwestern region showing the highest catches and NASC values. Tuna catches increased with NASC; however, the relationship was non-linear. While skipjack showed no significant correlation with NASC, bigeye and yellowfin tunas exhibited weak but significant positive correlations. Environmental analysis revealed that the northern waters had high surface temperatures, low salinity, and low oxygen levels, with mid and deep layers characterized by low temperature, salinity, oxygen, and chlorophyll. These findings offer a foundation for understanding tuna distribution in relation to prey and environmental conditions, highlighting the need for future species- and fishery-specific studies to support sustainable tuna resource management.
