Background: This study focused on reproductive traits in Hanwoo cattle, specifically the environmental factors affecting gestation length and birth weight. Methods: The records of 1,540 cows calved at the Hanwoo Research Institute from 2015 to 2023 were examined. This study analyzed two populations, linebreeding Hanwoo (LBH) and general Hanwoo (GH), with all cows undergoing estrus synchronization and artificial insemination. The R software was used to compare the differences between the two populations and analyze the environmental factors affecting each trait. Results: The results showed that the average gestation length for LBH was 283.28 ± 5.93 days, which was significantly shorter than that of the GH, which had an average of 285.63 ± 6.21 days (p < 0.001). The average birth weight of LBH calves was 25.10 ± 3.69 kg, significantly lighter than GH calves, which weighed 27.26 ± 4.11 kg on average (p < 0.001). Analysis of environmental factors revealed significant differences in the gestation length of LBH based on dam parity, year, and season of calving. However, no significant differences were observed based on calf sex. For LBH, birth weight showed significant differences based on dam parity, year of calving, and sex of the calf, but not the season of calving. In GH, gestation length varied with dam parity and calving season, but not with calving year or calf sex. The GH birth weight showed differences based on dam parity, year of calving, and calf sex, but not the season of calving. Conclusions: Reproductive traits in the Hanwoo cattle industry are economically vital but are heavily influenced by environmental factors due to their low heritability. An accurate evaluation of the genetic potential of these traits requires an analysis of the environmental factors affecting them. The results of this study serve as foundational data for predicting the potential for genetic improvement in the gestation length and birth weight of Hanwoo cattle.
Uranium-235, used in nuclear power generation, produces a lot of radioactive waste. Among radioactive waste nuclides, I-129 is problematic due to its long half-life (1.57×107 y) with high mobility in the environment. It should be captured and immobilized into a geological disposal environment through a stable waste form. In this study, various additives including Al, Bi, Pb, V, Mo and W were added to silver tellurite glass to prepare a matrix for immobilizing iodine, and its thermal and leaching properties were evaluated. To prepare glass, the glass precursor mixture was placed in alumina crucibles and heated at 800°C for 1 h. Except for aluminum, there was no significant loss of constituent elements. The loading of iodine in the matrix was approximately 11-15% by weigh, excluding oxygen. The normalized releases of all the elements obtained by PCT-A were below the order of 10-1 g/m2, which satisfies US regulation (2 g/m2). Differential scanning calorimetry was performed to evaluate the thermal properties of the glass samples. The glass transition temperature (Tg) increased by adding such as V2O5, MoO3, or WO3. The similar relative electrostatic field values of V2O5, MoO3, and WO3 could provide sufficient electro static field to the TeO2 interacting with the non-bridging oxygen forming Te-O-M (M = V, Mo, W) links. The addition of MoO3 or WO3 in the silver tellurite glass system increased glass transition temperature (Tg) and crystallization temperature (Tc) while maintaining the glass stability.
The red imported fire ant (Solenopsis invicta) is a species of ant native to South America. The fire ant was inadvertently introduced into USA, Australia, New Zealand, and other Asian countries including China and Taiwan. Since the first report of the fire ant in port city of Busan, Korea in 2017, it was found in many other cities of Korea in following year. To obtain the molecular information of this invasive species, total RNA was extracted from the abdominal segment of the ants collected in Incheon, and subjected to transcriptome sequencing. By using Illumina sequencer platform, 101 base pared-end sequencing generated 2 × 50,064,081 of raw reads to obtain 2 × 45.95 Gbase of quality filtered nucleotide sequences. The in silico cDNA library was constructed by Trinity de novo assembler followed by TransDecoder ORF finder and CD-HIT clustering program to streamline the library. The final version of cDNA library contains 20,442 contigs with protein coding capability. To survey the virome of this ant, these contigs were searched against the viral reference sequences from NCBI RefSeq database with BLASTN program. As a result, contigs which showed high sequence identities with several RNA viruses including previously reported SINV-2 were found from the fire ant. This virome information might give an idea of a shift of virological environment of this newly found ant isolate or population in Korea.
A recent study reported that Pleurotus ostreatus has the potential to be used as a β-glucan-based cream for supportive complementary therapy of atopic dermatitis. KH054 is a new herbal prescription consisting of P. ostreatus and Panax ginseng. The effects of atopic dermatitis-induced materials on the expression of cytokine genes in human monocytes (THP-1, EoL- 1) have been examined. Some reports demonstrated that P. ginseng augments the activity of natural killer cells, which plays an important role in innate immunity against infection and tumor development. Monocyte chemotactic protein 1 (MCP-1), interleukin (IL)-6, and IL-8 have important roles in mediating the infiltration of various cells into the skin of atopic dermatitis and psoriasis. The present study investigated whether KH054 on induced IL-6, IL-8, and MCP-1 secretion by house dust mite (Dermatophagoides pteronissinus) in THP-1 (human acute monocytic leukemia) and EoL-1(Human eosinophilic leukemia) cell. D. pteronissinus functions in the pathogenesis of allergic diseases, including atopic dermatitis and asthma. The inhibitory effect of KH054 on the induction of IL-6, IL-8, and MCP-1 secretion by D. pteronissinus extract in THP-1 and EoL-1 cells was examined. KH054 potently suppressed the elevated production of IL-6 and IL-8 induced by D. pteronissinus treatment in THP-1 and EoL-1 cells. Based on the present results, KH054 may be useful for developing functional foods to treat atopic dermatitis.
Pyrifluquinazon, as a quinazinalone chemical group, based on a new mode of biological activity. It is reported that mode of action is modifies insect behavior, rapidly stopping feeding such that insects starve to death. Time-release feature and mortality effect on M. persicae using different pyrifluquinazon nano type and non-nano type were compared. Pyrifluquinazon nano type was formulated with different molecular weight and density of used chitosan (CS 30000 0.1% and CS 3000 0.3%). In the CS 30,000 0.1%, the mortality was weakly occurred at early time, but steadily increased after 4days. Finally, we confirmed more than 70% mortality as a peak at 16days. In CS 3000 0.3%, the mortality showed about 70% until 18days as a effective controlled release. Also, We examine time-release feature and mortality effect on M. persicae according to the different pyrifluquinazon nano type(CS 30000 0.1% and CS 3000 0.3%) of concentrations. The CS 30000 0.1% bioassay results of different concentration were showed that the highest concentration(100ppm) was measured better mortality than other concentration at 0 day, but cannot confirm different effect about dissimilar concentration. However, increasing rates of M. persicae were low as treatment concentrate was high. In CS 3000 0.3% 100ppm concentration bioassay result, aphid mortality reached peak at 24 days and increasing rate also low. Additionally, for the comparing of bioassay and feeding behavior of M. persicae against pyrifluquinazon nano types and non-nano type, EPG technique was carried out. In case of non nano type, feeding inhibition efficacy was showed during 4 days after treatment, but appeared similar level with control after 10days. In CS 3000 0.3% 50ppm, residual efficacy was specially showed until 28days after treatment whereas treatments with CS 30000 0.1% were similar to the control after 22days. These result show that the change of feedinng behavior and motrality of M. persicae is correlated with the change of nano type or non nano type of pyrifluquinazon.
Several species of the genus Aphidius are used in biological control programs against aphid pests throughout the world and their behavior and physiology are well studied. But despite knowing the importance of sensory organs in their behavior, their antennal structure is largely unknown. In this study, the external morphology and distribution of the antennal sensilla on the antennal of both female and male adults of A. colemani were described using scanning electron microscopy (SEM). Generally, the filaform antennae of males (1,515.20±116.48 ㎛) are longer than females (1,275.06±116.42㎛). Antennae of this species is made up of scape, pedicel and flagellomeres. Male and female antennae differed in the total number of flagellomeres as 15 in males and 13 in females. Female and male antennae of A. colemani has samely seven types of sensilla. We classified sensilla placodea, Bohm bristles, 2 types of sensilla coeloconica, , 2 types of sensilla basiconica as with a tip pore and with wall pores, sensilla trichodea. In addition, the possible functions of the above sensilla types are discussed in light of previously published literature; mechanoreception(Bohm bristles, sensilla coeloconicaⅡ and sensilla trichodea) and chemoreception(sensilla coeloconicaⅠ, sensilla basiconicaⅠ,Ⅱ and sensilla placodea). Future studies on the functional morphology of the antennal sensilla of A. colemani using transmission electron microscopy (TEM) coupled with electrophysiological recordings will likely confirm the functions of the different sensilla identified in this study.
The porcine reproductive and respiratory syndrome virus (PRRSV) has three major structural proteins which designated as GP4, GP5, and M. They have been considered very important to arouse the humoral and cellular immune responses against PRRSV infection and proposed to be the excellent candidate proteins in the design of PRRS bioengineering vaccine. However, the PRRSV structural proteins are produced in low levels in the infected cells because it forms insoluble protein and possesses several transmembrane regions. To overcome this problem, we fused the GP4, GP5, and M with SUMO (Small ubiquitin-related modifier), and expressed the fused gene in Bm5 cells and silkworm larvae. Expression of the proteins were analyzed by 12% SDS-PAGE and western blotting using 6xHis tag and porcine anti-PRRSV antibodies. In results, SUMO fused proteins were expressed at a high level in Bm5 cells. The levels of protein using the silkworm larvae is higher than that using Bm5 cells. The fused protein was purified by Ni-NTA affinity chromatography. This study demonstrated that SUMO, when fused with PRRSV structural proteins, was able to promote its soluble expression. This may be a better method to produce PRRSV structural proteins for vaccine development.
The multicolored Asian ladybird beetle, Harmonia axyridis, is a generalist predator of aphids also, shows a high level of phenotype polymorphism in color pattern of elytra. Although, it is not sure about genetic information of color polymorphism, it has been confirmed that this phenomenon comes from their genetic traits. The color of H. axyridis elytra is mainly composed of black and red pigment. Phenoloxidase (PO) plays an important role in many insect physiological functions, i.e. sclerotization and pigmentation of cuticle and melanization of parasites. Following activation, PO catalyses the hydroxylation of tyrosine and subsequent oxidation of phenolic substance into quinines, which are further converted to melanin. However, the molecular bases of H. axyridis color pattern formation are almost unknown but it may be that the different pro-POs have different expression. In this study, total RNA samples from four each color pattern individuals, for example, succinea 1, succinea 2, conspicua and spectabilis was extracted. A cDNA enconding pro-PO was molecular cloned from each color pattern of H. axyridis and its putative amino acid sequence shared homology with pro-PO of other insects. We are pursuing to elucidate that their pro-PO sequence will be similar with those other insect PPO sequence. There are also regions of high sequence similarity, including putative activation site and two copper binding sites.

The Classical Swine Fever Virus (CSFV) is a member of the Pestivirus genus of the Flaviviridae. The genome of CSFV is a positive single-stranded RNA molecule 12.3 kb and contains a single large open reading frame (ORF). The polyprotein composed of eight nonstructural and four structural proteins (nucleocapsid protein C and three envelope glycoprotein E0, E1 and E2). E2, the most immunogenic of the CSFV glycoproteins, induces a protective immune response in swine. To determine the characteristics of the CSFV, LOM strain, we investigated the nucleotide sequence of the glycoprotein E0, E1 and E2. Comparison of the LOM with the other strains revealed nucleotide sequence identity ranging from 97 to 98%. Expression of the glycoprotein E2 was identified by SDS-PAGE and Western blot analysis using anti-CSFV E2 monoclonal antibodies in Sf21 cells. The expression levels of glycoprotein E2 were observed from day 3 and 5 days maximum. In addition, its expression efficiency by media and cell line was investigated. The result showed that High-Five cells and Grace’s insect media for Sf21 were the best conditions for the expression of the glycoprotein E2.
Aujeszky’s disease (AD), also called pseudorabies, is an infectious viral disease, caused by an alpha herpes virus and has domestic and wild pigs, as well as a wide range of domestic and wild animals, as the natural host. AD affects many countries and regions in the world, causing important economic losses, mainly due to international trade restrictions. In this study, to determine the characteristics of the Aujeszky’s disease virus (ADV), NYJ strain, which was isolated from the serum of an infected pig in 1987, we investigated the nucleotide sequence and expression of the glycoproteins gB, gC, and gD using the bBpGOZA system. We found that the glycoproteins gB, gC, and gD of NYJ consisted of 2751 bp, 1443 bp, and 1203 bp, respectively. Comparison of the NYJ with the other strains revealed nucleotide sequence identity ranging from 91.tito 99.0%. To better understand the genetic relationships between other strains, phylogenetic analyses were performed. The NYJ strain was formed a distinct branch with high bootstrap support. The expression of glycoprotein gD in insect cells was characterized by SDS-PAGE and Western blotting with an anti-ADV polyclonal antibody. Glycoprotein gD of approximately 45 kDa was detected. The results of this study have implications for both the taxonomy of ADV and vaccine development.
To determine the characteristics of the Korean porcine reproductive and respiratory syndrome virus (PRRSV), CA, which was isolated from the serum of an infected pig in 2006, we investigated the nucleotide sequence and expression of the structural ORFs (ORFs 2 to 7) using the bApGOZA system. We found that the structural ORFs 2 to 7 of CA consisted of 3188 nucleotides that were the same as those formed from VR-2332. Comparison of the CA with the other strains revealed nucleotide sequence identity ranging from 89.8 to 99.5%. To better understand the genetic relationships between other strains, phylogenetic analyses were performed. The CA strain was closely related to the other North American genotype strains but formed a distinct branch with high bootstrap support. Additionally, expression levels of the PRRSV proteins in Sf21 cells were strong or partially weak. The results of this study have implications for both the taxonomy of PRRSV and vaccine development.
The porcine reproductive and respiratory syndrome virus (PRRSV) has six structural proteins which encoded by ORFs 2 to 7 are designated as GP2, 3, 4, 5, M and N, repectively. In this study, we determined the expression of each protein using novel transfer vector, pBmKSK4 which has the polyhedrin promoter of BmNPV and 6xHis tag. The recombinant transfer vector was co-transfected into Bm5 cells along with bBpGOZA DNA. Recombinant virus was purified by plaque assay and amplified in Bm5 cells. Expression of each protein was identified by SDS-PAGE and Western blot analysis using anti-6xHis monoclonal antibody. The expression levels of the structural proteins in Bm5 cells were stronger than the expression system using pBacPAK9 transfer vector in Sf21 cells. As expected, GP5 was expressed at low levels from its structural properties and its toxicity for cells. In addition, each recombinant protein was purified using Ni-NTA spin columns. The ability to produce each protein in the baculovirus system indicates that these could be major candidates for the development of a vaccine against PRRSV.
Background : Insect pests damages are increasing on the field of Lycium chinense under eco-friendly organic management and especially, pest damage of Gelechiidae (Ilseopsis parki Povolny) is serious. Currently, various eco-friendly organic materials are registered, but insecticidal activity against Gelechiidae was not verified. Therefore, this experiment was carried out to select eco-friendly organic materials showing insecticidal activity in Gelechiidae. Methods and Results : To test insecticidal activity, the Gelechiidae lavas were collected from the field and Nine eco-friendly organic materials were collected from the market. method. The insecticidal activity assay method was, putting Gelechiidae lavas on the 90mm petridish then water-diluted eco-friendly organic materials were sprayed onto the laver. Result of insecticidal activity was observed with a microscope 10 minutes, 2 hours and 18 hours after the treatment. Microscopic observation results, insecticidal activity was shown only in 5 out of 9 materials. Conclusion : As a result, Insecticidal activity against Gelechiidae was not effective in all tested materials, but the 5 eco-friendly organic materials showed the same effect in repeated experiments. In this study, we selected 5 materials out of 9 eco-friendly organic materials against Gelechiidae, then future studies are planned to select the most suitable materials by field testing the 5 eco-friendly organic materials.
Background : This study was conducted to serve as a basis for the production of high quality organic cultivation Platycodin grandiflorum Ridix roots by analyzing the content of saponins according to organic fertilizer for organic farming. Methods and Results : Platycodin grandiflorum Ridix was treated by with biodegradable film, rice husks, and sawdust, non-mulching (hand weeding). A root of Platycodon grandiflorum were analyzed saponins using HPLC harvested on October. Content of Plantycodin D3 was an average of 68.5 ㎎/100g, which appeared as 111.2 ㎎/100g highest content in the sawdust treated. Deapioplatycodin D showed the highest 48.1 ㎎/100g in the sawdust treated. The content of Platycodin D was shown to 246.2∼373.8 ㎎/100g, the highest values were observed in non-mulching, it showed the lowest value in the rice husks treated. The content of Polygalacin D showed up to 156.0 ㎎/100g in the non-mulching as the biggest difference, the lowest value were 30.1 and 32.2 ㎎/100g in sawdust and rice husks treated. Large differences between fertilizers was not showed, but the overall saponin content of Platycodin grandiflorum Ridix were low in value compared to the control than organic fertilizers. Conclusion : Large differences between mulching materials was showed, thus the overall saponin content of Platycodin grandiflorum Ridix was highest in value compared to non-mulching than sawdust treatment. Mulching Platycodin grandiflorum Ridix with sawdust was an effective method for improving saponin contents.
Background : Angelica dahurica Bentham et Hooker, Angelica gigas Nakai, Ostericum koreanum Maximowicz and Peucedanum japonicum Thumberg are a major medicinal plant in north Geungbuk province. Using medicinal plants are impotant it`s ingredient. Dry condition and stroage method are not standard manual. The ingredient variation of dry condition and stroage method were not researched. Methods and Results : Using plant material were cutivated on Gyongsangbukdo Bonghwa area. It were studied ingredient variation after dry and storage condition by HPLC methods. Major ingredient of Angelica gigas Nakai are decurusin, decurusinangelate. Heated air bulk dry get more decursin than natuarl dry and decurusinangelate of natural bulk dry was higher than heated air bulk dry. Major ingredient of Ostericum koreanum Maximowicz are imperatorin and isoimperatorin.. Imperatorin of Ostericum koreanum was highest peak on 50℃ heated-air dry after plastic bag sorage and isoimperatorin was highest peak on 40℃ heated-air dry after mountain cultivation. Imperatorin is a major ingredient Angelica dahurica Bentham et Hooker. Heated air bulk dry get more decursin and decursinangelate than natuarl dry and small heated-air dry. Peucedanol-7o_glucoside is a major ingredient Peucedanum japonicum Thumberg. Natural bulk dry get more peucedanol-7o_glucoside than heated-air bulk dry. Conclusion : Ingredient of Angelica dahurica Bentham et Hooker, Angelica gigas Nakai, Ostericum koreanum Maximowicz are different under various cutivation, drying method, storage. Diffent Ingedients of Angelica gigas Nakai, Ostericum koreanum Maximowicz were not accord it’s optical conditon.
Background : The purpose of this study is to investigate the effect of the ridge and mulching cultivation and adapted sowing(planting)time in stable medicinal herbs production of Lithospermum erythrorhizon Siebold et Zuc. And it accomplished one’s purpose to GAP guide manual established standard method of cultivation technology, contribute to the enlargement of farm’s income. Methods and Results : Method of cultivation type with the ridge and covering material were conducted under two condition compose to open cultivation (level row : 120㎝ ridging, black polyethylene mulching) and plastic greenhouse cultivation (high row : 90㎝ ridging, non-woven fabric mulching). Sowing with Daehong varieties in April 17, transplanting July 25 by growing pot seedling for 40 days in 2015. Planting density were spaced 30㎝ apart in inter-rows 10 ㎝ apart intra-row. Fertilizing level of N-P-K-Compost (fermented mixed oil cake fertilizer) applied at 6-4-6-300 ㎏․10 a−1, 60% of basal fertilizer in March 14, 40% of top dressing were four times application in April 14 ~July 15. Plot design randomized block 3 repetition. Conclusions : Compared to level ridging on black polyethylene mulching open cultivation for growing plastic greenhouse with the high ridging non woven mulching cultivation, seed yields havesting 1.4 times, the dried roots yields increased in 0.7 times. Also, Growth characteristics by sowing time in April 16. compared to planting time in July 25. Length of stem and main root of above-ground, subterranean part growth increment rising highest. Seed and dried roots yields with increased by 71%, 30%. respectively.
Background : The root of Platycodin grandiflorum has been widely used as a crude drug or a dependent on the cultivation area and cultivation method. Therefore, this study was conducted to know the effect of organic fertilizer on the changes of antioxidents and antioxidant activity of Platycodin grandiflorum Ridix. Methods and Results : As basal fertilizer were treated before transplanting Platycodin grandiflorum Ridix with chemical fertilizer(Super21, 21-17-17, Namhae Chemical), mixed organic fertilizer, bacterial culture, fermented oil cake and manure in each trial of nitrogen based on 3㎏/10a criteria. The total polyphenol content was highest values were 15.5mg/g appear in the mixed organic fertilizers, the lowest were 9.1mg/g appeared in the manure and control. Flavonoid content showed the highest 15.3 mg/g in the mixed organic fertilizer, and investigated at the lowest 5.0 mg/g in the fermented oil cake. ABTS radical scavenging activity was the highest in was 65.7 mgTE/g in the chemical fertilizer, showed the lowest value in the control to 13.0 mgTE/g. Therefore, both flavonoid and polyphenol content showed a high value on organic fertilizers. Conclusion : Therefore, both flavonoid and polyphenol content showed a high value on organic fertilizers. It is determined that to have researches for a variety of antioxidants by the organic fertilizers made in the future.

The content of Platycodin D was shown to 327.4∼373.8 ㎎/100g, the highest values were observed in mixed organic fertilizer, it showed the lowest value in the control. The content of Polygalacin D showed up to 336.7 ㎎/100g in the manure as the biggest difference, the lowest value were 54.5 and 74.2 ㎎/100g in bacterial cultures and control. Large differences between fertilizers was not showed, but the overall saponin content of Platycodin grandiflorum Ridix were low in value compared to the control than organic fertilizers. Conclusion : For the increasing the yield that Pharmacological effect, organic fertilizer is effective to increase the number of fine-root high which high contented with saponins.
