        Proteinuric conditions demonstrate structural and compositional changes of the foot processes and slit diaphragms between podocytes. β-Catenin in podocytes serves as an adapter protein anchoring P-cadherin of slit diaphragms to actin filaments of the podocyte cytoskeleton. To investigate the effect of ginseng total saponin (GTS) on pathologic changes of podocyte P-cadherin/β-catenin unit induced by diabetic conditions, we cultured mouse podocytes under: 1) normal glucose (5 mM, = control); 2) high glucose (HG, 30 mM); 3) advanced glycosylation end products (AGE)-added; or 4) HG plus AGE-added conditions and treated with GTS. In confocal imaging, β-catenin colocalized with P-cadherin predominantly at intercellular junction area. However, diabetic conditions relocalized and concentrated both molecules at perinuclear cytoplasmic area. In Western blotting, diabetic conditions, especially HG plus AGE-added condition, also decreased cellular β-catenin protein levels at 6, 24, and 48 hours. GTS improved such quantitative and qualitative changes of β-catenin. These findings imply that HG plus AGE have an influence on the redistribution and amount of P-cadherin/ β-catenin unit of podocytes, which can be reversed by GTS.

        목 적: 근거리 주시시차와 사위를 측정하여 주시시차의 방향과 분포를 연구하고 주시시차에 영향을 미치는 주시시차곡선의 3가지 변수 값을 구하여 양안시 이상의 진단과 처방에 응용하고자 한다. 방 법 : 안질환이 없으며 사시와 약시가 없고 근거리에서 수직사위가 없는 남녀 대학생 85명을 대상으로 완전교정 후 사위 및 주시시차를 측정하였다. 근거리의 완전융합제거사위는 Modified Torington Card를 이 용하였고, 주시시차는 Wesson fixation disparity card를 이용하였다. 결 과 : 주시시차곡선 유형은 제 I유형 69.4%, 제 II유형 5.9%, 제 III유형 15.3%, 제 IV유형 9.4%로 나타 나 제 I유형, 제 III유형, 제 IV유형, 제 II유형의 순으로 많았다. 일부융합제거사위와 주시시차곡선의 기울기 가 주시시차에 영향을 많이 미치는 것으로 나타났다(p=0.000). 결 론 : 근거리 주시시차는 양안시 상태에서 버전스와 조절의 상호작용을 임상적으로 확인할 수 있으므로 양안시 이상의 진단과 처방에 유용한 변수이다.

        As a sensor of cellular energy status, AMP-activated protein kinase (AMPK) plays an important role in the pathophysiology of diabetes and its complications. Because AMPK is also expressed in podocytes, podocyte AMPK would be an important factor contributing to development of podocyte injury. We investigated the roles of AMPK in the pathological changes of podocyte synaptopodin induced by angiotensin II (Ang II), a major injury inducer. Mouse podocytes were incubated in media containing various concentrations of Ang II and AMPK-modulating agents, and the changes of synaptopodin were analyzed by confocal imaging and Western blotting. Ang II and compound C, an AMPK inhibitor, concentrated and re-localized synaptopodin from peripheral cytoplasm to the internal cytoplasm portion in podocytes. Ang II also reduced synaptopodin protein and mRNA, which were reversed by metformin and 5-aminoimidazole-4-carboxamide ribonucleoside. Losartan, an Ang II type 1 receptor antagonist, also recovered synaptopodin mRNA, which was suppressed by Ang II. We suggest that Ang II induces the relocation and suppression of podocyte synaptopodin by suppression of AMPK and via Ang II type 1 receptor, which would be an important mechanism in Ang II-induced podocyte phenotypical changes.

        The black soldier fly, Hermetia illucens (BSF), has a worldwide distribution in the tropics and warm temperate regions and is active in the Korea from May through October. This species colonize a wide variety of decomposing vegetable and animal matter and oviposit in a variety of decomposing materials. In this study, how the BSF molting, adult emergence and mating rate changed by seasonal condition at the artificial rearing system was investigated. The black soldier fly larvae and pupae were reared under laboratory condition (27℃, 60% R.H.). Under the laboratory condition, molting and adult emergence were not influenced by seasonal factors such as climate, radiation intensity. But it is known that the sunlight is the most important factor of the mating. In the previous study the time of day, temperature, and humidity is significantly correlated with oviposition and mating. The rearing of BSF throughout the year is restricted by sunlight. In this study, the data shows definitely different mating numbers throughout whole year. The time of the day and sunlight density are changed with season and it influence on artificial rearing. To culture the black soldier fly throughout the year in Korea needs a more deep study under the artificial rearing system.

        본 연구에서는 서로 다른 지역에서 재배된 장뇌삼 8 품종과 밭에서 재배된 재배인삼 등 총 9개체에 대해 장 뇌삼 품종들 간의 유연관계를 분석하였다. RAPD분석에 사용한 random primer는 총 40종류(OPA 1~20, OPB 1~20)였으며, 이 중 34개가 각기 다른 지역에서 재배된 품종간의 집단을 형성하여 9개 품종간 유연관계를 밝히 는데 유용하였다. 유연관계를 분석한 결과, 홍천과 재배 인삼, 의성과 금산, 진안과 풍기와 강화, 상주와 안동 4개 의 그룹으로 구분되었으며 이는 다시 2개의 그룹「홍천- 재배인삼-의성-금산」,「풍기-강화-상주-안동」으로 구 분 되었다. 9개 지역에서 수집한 품종간의 유사도 값은 최저 0.30, 최고 0.53으로 나타났다. RAPD방법은 사용법 이 간단하고 소요시간이 적게 소요될 뿐만 아니라 적은 양의 DNA시료로도 분석이 가능한 검정법으로 형태적인 차이가 구별되지 않는 장뇌삼 품종 간의 유전적 유연관 계를 밝히는 데 유용한 방법임을 알 수 있었다. 그러나 품종별 계통유연관계를 보다 정확히 분석하기 위해서 AFLP 또는 Molecular Maker를 이용하거나 계통분류에 널리 이용되는 핵, 엽록체 DNA의 유전자 염기서열 비교 와 같은 정밀한 분자계통학적 연구가 수행되어져야 할 것으로 사료된다.

        A multiplex polymerase chain reaction (PCR) was developed for the simultaneous detection and differentiation among Nosema apis and Nosema ceranae in honeybee. Three sets of primers were selected from different genomic sequences to specifically amplify a 831 bp amplicon within the SSU rRNA gene, specific for both N. apis and N. ceranae (MSSR primer); a 375 bp amplicon within the SSU rRNA gene, specific for N. apis (NA primer); and a 1,131 bp amplicon within SSU rRNA gene, specific for N. ceranae (NC primer). Using the primers in conjunction (multiplex PCR) we were able to N. apis and N. ceranae and to differentiate between them. The sensitivity of this PCR assay was approximately 102 spores per milliliter. We proposed that the multiplex PCR was sensitive, specific and rapid tool that can serve as a useful differential diagnostic tool for detecting N. apis and N. ceranae in honeybee.

        This study was conducted to investigate the distribution pattern, ecological characteristics and life cycle of the Black Soldier Fly (Hermetia illucens, BSF). The BSF was widely distributed throughout Korea. The insect was mainly found in the vicinity of and in cattle sheds, manure sheds, living waste dump grounds, and food waste dump grounds. Developmental characteristics of the BSF are as follows: the egg was long oval shaped of 887㎛ in the major axis and 190㎛ in the minor axis; it weighed 24㎍. Female oviposited ca. 1,000 eggs on average; eggs hatched in 81 hours under laboratory condition (27℃, 60% R.H.). The duration of the larval stage was approximately 15-20 days. The size of the last instar larvae was 21mm. The cuticle of the pupae gradually acquired red-brown color and the size of them was 19 mm. The pupal stage was shorter for females (16 days) than males (15 days). Adults were sized about 13-20mm long and black-colored. The life span of adult insects was 5-8 days for the first generation (June-July), 7-10 days for the second generation (Aug.-Sept.), and 13-18 days for the third generation (Sept.-Oct.). Mating started on the next day of emergence and actively occurred at the third day after emergence. Mating mostly occurred between 10:00 and 16:00 during which light intensity is highest. Egg-laying started on the third day and was most frequent from the fourth to the sixth day after emergence. Similar to mating time, females oviposited mostly between 10:00 and 16:00.

        Protease from various sources have been studied biotecnologically. For biotechnological applications, one highly preferred enzyme is protease. There have been no reports of cloned genes encoding digestive proteases in the Laccotrephes japonenis, Ranatra unicolor, Muljarus japonicus. These insects are considered to be a predator of aquatic insects. RT-PCR was used to amplify cDNA fragments for digestive proteases from total RNA the hole body of the insects. The flanking sequences of the 5'- and 3'- end of the these genes were characterized by RACE-PCR. Sequence analysis showed that these genes contained complete ORF. The deduced amino acid sequences of these protease showed 62% identity to the serine protease of Creontiades dilutus, 58% to Lygus loneolaris trypsin-like serine proteinase, 54% to Triatonatoma infestans salivary trypsin. To generate Laccotrephes japonensis serine protease, the DNA fragment coding for serine protease is cloning into suttle vector pBACⅠ, named pBAC1-JG and infected to Spodoptera frugiperda (sf9) insect cell. The cDNA encoding JG was expressed as a 32-kDa polypeptide in baculovirus infected insect cells and the recombinant protein showed activity in the protease enzyme assay using gelatin as a substrate.

        In order to establish artificial indoor rearing techniques for the black soldier fly (BSF), we developed indoor rearing instructions and collection manual for each developmental stage of the fly. The fly collection was conducted between June and October at 1 month interval. Calf feed and food waste were most effective to attract adults. Collection efficiency is higher from the trap installed in a shady spot than that in an open spot. It was highest to collect flies in August and September. As the egg-laying medium for the artificial egg collection, calf feed and food waste were most effective. The optimal number of the medium (W*D*H=60*40*15cm) was 8 for 2000 adults (male:Female=1:1) in the egg-laying net (W*D*H=4*2*2m). Flower foams and wooden blocks with holes were used as egg-laying sites. Adult females preferred the holes on average 3~5mm in diameter and 7~10mm in depth for oviposition. Larvae reared in low density (2~4 individuals/㎠) showed superior practical traits than those reared in an overcrowded environment (6~10 individuals/㎠). It is important to place the fly pupae in moist sawdusts (humidity: 20~40%), since a pupa tends to hide in a refuge. Adult insects should be employed for laying eggs for food waste processing immediately after emergence. When treated at 10℃ for 10 days after pupation, emergence rate of the insect was still 93.3%. By keeping pupae at the low temperature, emergence timing could be manipulated with about 10 day flexibility.

        This study was conducted to investigated the distribution, ecological character and life cycle of Black Soldier Fly(BSF), Hermetia illucens. The distribution of BSF was defined in all parts of the country in Korea. Its main habitat was found to be areas near cattle sheds, manure sheds, living waste dump grounds, and food waste dump grounds. Observed characteristics of BSF by developmental stage may be summarized as follows: eggs were a long oval shape of 886.9±19.7 ㎛ in major axis and 190.1±9.7 ㎛ in minor axis; they were 24.0±1.6 ㎍ in weight. One adult insect laid 1001±247 eggs in quantity; days to hatch from eggs (27℃, 60% R.H.) were 81.3±12.5 days. Last instar larva ranged from 20.7±1.1 mm in size, the length of larval stage was approximately 15~20 days. Pupae exhibited red brown, 19.2±1.1 mm in size; pupal state lasted 15.5±1.4 days for female, 14.7±1.4 days for male, exhibiting the tendency of males having shorter period than females. Adult insects were sized about 13~20 mm and colored black. Mating started 2 days after emergence and was most active during the 3rd day. Mating mostly occurred between 10:00 and 16:00 during which light intensity is high. Laying eggs started 3 days after emergence and was most frequent during days 4~6. Time of laying eggs during the day was similar to copulation time, showing the highest laying rate between 10:00 and 16:00.

        This study was conducted to investgate the effect of gamma irradiation on the lipid oxidation, olfactory properties, fatty acid isomer and volatile compound profiles of five different oil sources. Three plant oils, canola oil, corn oil and soy oil, and two animal fat, tallow and fish oil, were irradiated with 0, 5, 10, and 20 kGy level of dose by Co-60 as a radiation source. Lipid oxidation parameters, FFA, POV and TBARS, were determined according to the AACC and AOCS method. Olfactory property of irradiated oil sources was analyzed using electronic nose (FOX3000, AlphaMOS Co., France) with 12 metal oxide sensors and the result was interpreted using principle component analysis program. Fatty acid isomer profiles and volatile compound profiles of irradiated oils were also measured by GC and GC-MS, respectively. Although the free fatty acid level of oil were the highest by high dose level (20 kGy) of irradiation, changes of both TBARS and POV values by gamma irradiation were varied between oil sources and irradiation level of dose. Electronic nose analysis showed a distinct difference among dose levels of irradiation. Although there was no new volatile compound produced by gamma irradiation, amounts of each volatile compounds were increased by irradiation. Only cis isomer, 9c12c15c of linolenic acid was dose-dependently decreased as irradiation level increased but there was no difference among other isomer profiles of linoleic acid and linolenic acid by gamma irradiation. This study showed that changes in physico-chemical properties of both plant and animal oil were remarkable by gamma irradiation. Therefore, a further study should be needed to scrutinize the interaction among lipid oxidation, production of volatile compounds and off-flavor, and isomerization of fatty acid by gamma-irradiation.

        제주지역 초 • 중 · 고생 1383명을 대상으로 하여 안경 장용율, 시력검사 장소, 안경 교체시기 · 비용 · 이유, 안경장용과 거주지역간의 상관성 등에 관하여 설문조사를 실 시하여 다음과 같은 결론을 얻었다. 1. 조사 대상자의 573 명 (41 .43%) 에 해당하는 학생이 시력교정을 위해서 안경을 장 용하는 것으로 조사되었다. 이들 안경장용 학생의 학년별 분포를 조사한 결과, 학년이 높아질수록 안경장용율이 증가하고 있음을 확인하였다(초등학생 21.40%, 중학생 45.33%, 고등학생 50.73%). 2. 안경교체시기에 대해 초 · 중 · 고생 공통적으로 6개월 이상 -1 년 미만이 가장 많 은 비율을 차지하고 있었다(초등학생 43.83%, 중학생 52.02%, 고등학생 59.21%). 3. 안경교체이유는 시력변화가 54.62%, 테 · 렌즈 파손이 28.97%, 안경을 바꾸고 싶 어서가 14.66%, 기타 1.75% 순으로 조사되었다. 4. 안경 교체시 사용비용은 전체 학생의 48.17% 에 해당하는 학생이 30,000원 이상 50,000원 미만의 금액을 사용하는 것으로 나타났다. 이들 학생들은 해당 학년에 서, 초등학생은 32.87%, 중학생은 43.50%, 고등학생은 55.96%로 나타났다. 5. 안경 구입시 안경태와 안경렌즈의 상대적인 중요도에 관한 질문에 대해서는 둘 다 중요하다고 응답한 경우가 가장 많았다. 6. 안경을 장용한다고 응답한 학생들을 대상으로, 현재 사용중인 안경의 시력검안 장소에 관한 질문에 대해 초등학생은 안과에서, 중 · 고생은 안경원에서 시력검 사를 받는 학생의 비율이 더 높게 나타났다. 7. 안경 장용 학생의 지역별 분포를 조사한 결과, 제주시가 45.03%로 가장 많았으 며 서귀포시 (26.35%) , 북제주군 (14.66%) , 남제주군 (13.96%) 순으로 나타났다.

        The purpose of this study was to evaluate the quality of fresh-cut lettuce after a washing treatment with aqueous thyme extracts of differing concentration. Four different concentrations of thyme extract were employed: 0.001% (w/v, TEA), 0.005% (w/v, TEB), 0.01% (w/v, TEC), 0.05% (w/v, TED), while distilled water was used as a control. Measurements of O2 concentration, CO2 concentration, total aerobic bacteria, CIE L*, a*, b*, browning index, total phenolic contents and enzymatic activities were investigated on day 0, 1, 3, 5, and 7 at 10 . The O2 concentration in TEB were higher than those in other samples during storage, and the CO2 concentration in TEB and TEC were significantly lower than in other samples on day 7. There was no significant difference in the total aerobic bacteria counts between treatments (p<0.05). The samples treated with TEB and TEC showed higher L* (lightness) values, but lower browning indexes, total phenolic compound levels, and enzymatic activities (polyphenol oxidase, peroxidase) than other samples during storage. Specifically, on day 7, the browning index of Cont reached 0.25, while those in the TEB and TEC were about 0.15. The L* in Cont decreased from 69.50 to 58.92, while TEB and TEC were values of 65.61 and 63.20, respectively. These results reveal that 0.005-0.01% thyme extract was effective as a washing treatment inhibited the browning of fresh-cut lettuce and is thus expected to be a useful natural extract for maintaining the quality of fresh-cut lettuce.

        The purpose of this study is to present the results as basic data for establishing proper storage conditions and distribution conditions of actual farms at point of increasing concern about hygiene and palatabiltiy of consumers to food. In this study, three farmhouses of dried persimmons prepared using different storage conditions were selected in Sangju (Korea). The dried persimmons were stored for 90 days. Changes in temperature and humidity were measured with a temperature and humidity recorder under each storage condition, and physicochemical analysis and sensory evaluation were performed. The average temperatures of farmhouse A, B and C were approximately -22--23℃, -19--18℃ and -25℃ respectively. The humidities of A, B and C were 62-63%, 59-60%, and 66-67%, respectively, and the moisture contents of all farmhouses increased during the storage period, with farmhouse B showing the most rapid increase. Free sugars increased, except for those from farmhouse C. Persimmons from farmhouse B showed the greatest changes in chewiness and hardness. The values of a* and b* were significantly decreased in persimmons from farmhouse B, and the color difference value of farmhouse B was dramatically increased. Sensory evaluation showed that the color preference tended to decrease compared with the initial value. Only farmhouse B showed decreased overall acceptability. Moreover, farmhouse B had the highest storage temperature and lowest humidity. Therefore, our results showed that storage at a low temperature and high humidity was important for manufacturing high-quality dried persimmons.

        Aralia elata Seemann (AE) has long been used as a folk medicine for the treatment of various diseases including diabetes mellitus, anti-arthritic, and anti-gastric ulcer agent in Korea, Japan, and China. This study was performed to establish a simple and reliable HPLC/UV analytical method for determination of most active anti-hypertensive compound, a 3-O-α-L-rhamnopyranosyl(1→2)-α-L-arabinopyranosyl hederagenin 28-O-β-D-xylopyranosyl(1→6)-β-D-glucopyranosylester (HE) for the standardization of the shoot extract of AE as a health functional food ingredient. The quantitative analytical method of HE was optimized by HPLC analysis using reverse-phase C18 column at 40°C with H2O and acetonitrile (70:30, v/v) as an isocratic mobile phase at a flow rate of 1.0 mL/min and detection wavelength of UV 205 nm. This HPLC/UV analytical method showed good specificity and high linearity in the tested range of 0.03125-2.0mg/ml with excellent coefficient of determination (R2) of 0.9999. The limit of detection and limit of quantification were 12.0 μg/mL and 36.5 μg/mL, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 0.2% and 0.1%, respectively. These results indicate that the established HPLC/UV analytical method is very simple, specific, precise, accurate, and reproducible and thus can be useful for the quantitative analysis of HE as a functional anti-hypertensive compound in AE extract.

        In this study the physicochemical properties and antioxidant capacities of ginger (Zingiber officinale Rosc) Jungkwa with different kinds of sweeteners were determined. Jungkwa made with different sugars (sugar, xylitol, honey or oligosaccharides) were compared in aspect of physiochemical properties, antioxidant activities, total phenolic contents and sensory evaluation. Moisture contents Jungkwa treated different kinds of oligosaccarides showed highest value, in the order of honey, xylitol and sugar. L* value of Jungkwa treated with xylitol was the highest, a* value of Jungkwa treated with honey, sugar Jungkwa were higher then others. Free sugar contents of Jungkwa treated with sugar showed the highest value in sucrose, glucose and galactose. Jungkwa with xylitol showed lowest value in all free sugar contents. Hardness and chewiness of Jungkwa treated with xylitol showed the highest value. The antioxidant activity measured by DPPH and ABTS radical scavenging activity and total penolic content were the highest in Jungkwa treated with honey, followed by Jungkwa treated with xylitol, oligosaccharides and sugar. Appearance and color of oligosaccarides and honey treated Jungkwa were preferred. In ginger taste, sweetness, chewiness were highest in sugar treated Jungkwa (not significant difference in treatments). As a result, honey treated Jungkwa has higher antioxidant activity and quality than other sugar treatments.
