        Tuberculosis is a potentially deadly infectious disease caused by the Mycobacterium tuberculosis (M. tuberculosis). Tuberculosis is diagnosed by proving the M. tuberculosis in sputum samples based on the results of acid-resistant staining, culture, and nucleic acid amplification tests. However, there is a report that the detection rate of M. tuberculosis is low in acid-resistant staining using tissue specimens. It has been suspected that the cause is a potential loss of acid resistance by the organic solvents used for tissue specimen preparation. Therefore, this study was pursued to find out if Gram staining and fluorescent staining in addition to acid-resistant staining would be helpful in diagnosing tuberculosis. We used four tissue (lung, small intestine, large intestine, and lymph node) samples with chronic granulomatous inflammation observed in HE staining and positive results in real-time PCR. These detection rates and staining properties were investigated through microscopic examination using the Ziehl-Neelsen, Gram, and Auramin rhodamine staining. In this studies, M. tuberculosis were observed by Ziehl-Neelsen, Gram, and Auramin rhodamine staining in all four samples. In the evaluation of clinical microbiology proficiency testing (CMPT), the Ziehl-Neelsen and Gram staining were the same result, but the Auramin rhodamine staining was relatively low. These data indicated that Gram staining is useful for detecting M. tuberculosis in formalin-fixed tissue specimens. Therefore, if the Ziehl-Neelsen and Gram staining are combined as the M. tuberculosis staining method in tissue specimens, a better direction may be provided for tuberculosis diagnosis.

        When decommissioning a nuclear power plant, the structure must be made to a disposable size. In general, the cutting process is essential when dismantling a nuclear power plant. Mainly, thermal cutting method is used to cutting metal structures. The aerosols generated during thermal cutting have a size distribution of less than 1 μm. The contaminated structures are able to generate radioactive aerosols in the decommissioning. Radioactive aerosols of 1 μm or less are deposited in the respiratory tract by workers’ breathing, causing the possibility of internal exposure. Therefore, workers must be protected from the risk of exposure to radioactive aerosols. Prior knowledge of aerosols generated during metal cutting is important to ensure worker safety. In this study, the physical and chemical properties of the aerosol were evaluated by measuring the number and mass concentrations of aerosols generated when cutting SUS304 and SA508 using the laser cutting method. High-resolution aerosol measuring equipment (HR-ELPI+, DEKATI) was used to measure the concentration of aerosols. The HR-ELPI+ is an impactor-type aerosol measuring equipment that measures the aerosol number concentration distribution in the aerodynamic diameter range of 6 nm to 10 um in real-time. And analyze the mass concentration of the aerosol according to the diameter range through the impactor. ICP-MS was used for elemental mass concentration analysis in the aerosol. Analytical elements were Fe, Cr, Ni and Mn. For the evaluation of physical and chemical properties, the MMAD of each element and CMAD were calculated in the aerosol distribution. Under the same cutting conditions, it was confirmed that the number concentration of aerosols generated from both materials had a uni-modal distribution with a peak around 0.1 um. CMAD was calculated to be 0.072 um for both SUS304 and SA508. The trend of the CMAD calculation results is the same even when the cutting conditions are changed. In the case of MMAD, it was confirmed that SUS304 had an MMAD of around 0.1 μm in size for only Fe, Cr and Mn. And SA508, Fe, Cr, Ni and Mn were all confirmed to have MMAD around 0.1 μm in size. The results of this study show that a lot of aerosols in the range of less than 1 μm, especially around 0.1 μm in size, are generated when metal is cut using laser cutting. Therefore, in order to protect the internal exposure of workers to laser metal cutting when decommissioning NPPs, it is necessary to protect from nano-sized aerosols beyond micron size.

        PURPOSES : The purpose of this study is to investigate the effects of urban road characteristics on temperature changes in urban areas using surface thermal temperature. METHODS : This study measured the surface thermal temperature of each road component from urban roads, analyzed the effect of the road component characteristics and vegetation on temperature, and estimated the regression models. RESULTS : As a result, the mean temperature was 27.3 ℃ on the roadway, 25.5 ℃ on the vegetated median, and 22.9 ℃ on the sidewalks. The roadside temperature was 26.14 ℃ with surrounding buildings and 23.82 ℃ near green spaces. The temperature with street trees was lower (24.45 ℃) than without (28.38 ℃) while it was 23.96 ℃ with vegetated median and 25.64 ℃ without. The temperatures were lower (24.70 ℃) on the permeable surface than on the impermeable surface (28.38 ℃). Model estimates show that the temperature decreases by 0.007 ℃ with an increase of 1 m² green (permeable space) space and the temperature with buildings tends to be 1.729 ℃ higher than that with green space. As green space increases by 1 m², the temperature tends to decrease by 0.017 ℃ on the roadway and by 0.012 ℃ on the sidewalk. Shade effect models show that street trees with shade affect temperature reduction by -3.884 ℃ on roadways and -3.314 ℃ on sidewalks. CONCLUSIONS : The results of this study demonstrate the differing effects of road characteristics on temperature. The roadway is more sensitive than the sidewalk to temperature changes and roadside vegetation, ambient green space, and pavement permeability contribute to temperature reduction.

        In a previous study, it was reported that enzymatic hydrolysis under pressurization could be a new method which could produce arginine dipeptide and free amino acid in anchovy hydrolysate as salty enhancer at optimal condition. Powder is more efficient than liquid in terms of transport and storage stability. For the purpose of producing spray dried powder of various salt contents was investigated the effect of different salt concentration of anchovy hydrolysate on spray dried powder properties. The anchovy hydrolysate of various salt contents(in the range of approximately 0.7- 19.8% w/w) prepared adding the fish sauce (Dae-Young fish market) at inlet drying air temperatures of 120°C and 140°C. The process yield and physicochemical properties such as moisture content, bulk density, hygroscopicity and the morphology (EDS, XPS, XRD) of the anchovy hydrolysate powder was measured. The glass transition temperatures (Tg) of the powders equilibrated under various water activities were determined using a differential scanning calorimeter (DSC). Different drying conditions and salt concentration could generate anchovy hydrolysate powders with different process yield, bulk density and moisture content. The spray-dried anchovy hydrolysate powder was confirmed by XRD to be a mixture of an amorphous substances and crystalline salts. The energy dispersive X-ray spectrometer (EDS) and X-Ray Photoelectron Spectroscopy (XPS) analysis demonstrated that the surface NaCl concentration of the powders increased with an increasing drying air temperature. Increasing moisture adsorption of the anchovy hydrolysate powders resulted in a Tg reduction. It is suggested that producing spray dried anchovy hydrolysate for the industrial use is the use of the feed salt concentration of not lower than % w/w and inlet air temperature at 120°C, 140°C

        The aim of this study was to use an in vitro method and estimate glycaemic index (GI) from porridges to determine the digestibility of porridges. Glycaemic index’s concept is to classify foods on the basis of their postprandial blood glucose response. The GI of a foodstuff is generally measured by determining the increment in blood glucose concentration after the consumption of a test meal over a set period of time and comparing it with an isoglucosidic control meal (normally white bread or glucose) and expressed as a percentage. In this study, the 5 porridges were studied for their starch digestibility. The available starch contents of the samples varied from 65~85 g/100 g dry solids. From in vitro digestion, the porridge samples were Medium glycaemic index foods with calculated GIs ranging from 56 to 67.

        The present paper proposes an categorization of commercial fluid products. A total of 21 commercial fluid products were analyzed(porridges, universal design foods, foods for special medical purposes) corresponding to 11 different commercial brands. Joint consideration has been made of viscous behavior (flow and thixotropy) and viscoelastic behavior (oscillatory testing). Rheological measurements were fitted to different rheological mathematical models, and a total of 4 parameters were generated. Our results correspond to three different classes. The three defined classes have been named in a way similar those most commonly used in the literature. Each class has been defined from the parameters allows us to know the common rheological characteristics of any of the products belonging to a given class.

        Pseudococcus longispinus, a notorious cosmopolitan pest species of mealybugs, known to be distributed indoors in Korea since 2002, is found to be mixed with another species, Pseudococcus orchidicola. Finding P. orchidicola as a pest of tropical plants in Korea is rather unexpected because of their main distribution in Pacific area and South Asia. However, all the available information from morphology, molecule and advices on identification from mealybug specialists indicated this is best matching P. orchidicola. Morphological, molecular and some biological notes on P. orchidicola are provided with some adult and nymphal images, and compared with P. longispinus. A full discussion mainly on identity and distribution of P. orchidicola is given.

        We have used bulked segregant analysis to screen the strain-specific DNA marker associated thermophilic strain of Pleurotus eryngii. Bulked genomic DNAs of Pleurotus eryngii were amplified by randomly amplified polymorphic DNA (RAPD) using OP-A, OP-B, OP-L, OP-P, OP-R and OP-S primers to screen the strain-specific DNA marker. A unique DNA fragment of 500 bp was amplified with OP-A11 primer from the psychrophilic strain and sequenced. A sequence characterized amplified region (SCAR) marker was designed on the basis of the determined sequence and named as OP-A11-1. The PCR analysis with the OP-A11-1 primer showed that this SCAR marker clearly distinguish the psychrophilic strains from the control strains.

        The green pale plant bug, Apolygus spinolae was one of the main insect pests that damaged leaves and fruit in grapes and its damage status was firstly reported in 2000 in grape orchards. This research was conducted to evaluate the distribution and difference in damage rate depending in management type of grapevine orchards (domestic sale farm vs export farm) in the export complex area of Korea (Hwangsung in Gyeonggii, Sangju and Yeongcheon in Gyeongbuk, Namwon in Junbuk and Yeongdong in Chungbuk) from 2010 to 2012. Damage by A. spinolae occurred in all 62 survey farms and damage rate differed depending on locality and individual farms in the same area. Damage rate was lower in export farms than in domestic sale farms, and damage rate of leaves was highly correlated with damage rate of new shoots. 15 species of hemipteran insect were attracted to sticky traps and A. spinolae was the dominant species. The attracted number of A. spinolae in the sticky traps differed depending on locality, and more occurred in domestic sale farms than expert farms. A. spinolae was continually attracted to sticky traps in the harvest period in grapevine orchards.

        The mannanase, cellulase and xylanase-producing thermophilic bacteria, designated EJ3, was isolated from fresh spent mushroom (Pleurotus eryngii) substrates taken from the DOJUN farm located in Keyongnam, Korea. The isolate EJ3 was facultatively anaerobic and grew at temperature ranging from 20℃ to 60℃ with an optimal temperature of 37℃. The DNA G+C content of the isolate EJ3 was 45 mlo%. The major fatty acids were anteiso-15:0 (38.9%), 17:0 (7.4%), and iso-15:0 (38.2%). The 16S rRNA gene sequence similarity between the isolate EJ3 and other Bacillus strains varied from 97% to 99%. In the phylogenetic analysis based on these sequences, the isolate EJ3 and Bacillus subtilis clustered within a group and separated from other species of Bacillus. Based on the physiological and molecular properties, the isolate EJ3 was classified within the genus Bacillus as Bacillus subtilis EJ3. The optimal growth condition of B. subtilis EJ3 were pH 6.0 and 55℃, respectively.

        This study was conducted to evaluate the feeding value of the spent mushroom (Pleurotus eryngii) substrates (SMS) in laying hens (Hy-Line Brown). The fresh spent mushroom (Pleurotus eryngii) substrates collected from the DOJUN farm were fermented with Bacillus subtilis EJ3 for 2 weeks. A total of twenty-four laying hens were fed corn-soy based experimental diets containing 0% (control), 5% (T1), 10% (T2) and 15%(T3) fermented SMS for 7 weeks. There were no significant differences among the treatments in egg production, egg weight, egg mass, feed conversion and viability during the experimental period. Feed intake was significantly lowered in control (118.3 g) than T1 (121.9 g), T2 (120.3 g) and T3 (122.4 g). There were no significant differences among the treatments eggshell breaking strength, thickness and haugh unit, whereas the yolk color of T1, T2 and T3 were significantly heavy than T0. The palatability of boiled meat was significantly better in the T3 laying hens than in the T0 laying hens. In conclusion, fermented SMS can be used as resource of feed in laying hen feed at 5.0-15% level without effect on performance and egg qualify.

        To identify DNA markers linked to a elytra polymorphism, amplified fragment length polymorphism (AFLP) analysis was performed on DNA samples from four each colour pattern individuals (2 females and males), for example, succinea 1, succinea 2, conspicua, and spectabilis. As a result of performing AFLP analysis with the restriction endonuclease combination EcoRⅠ and Mse I, total of 2,269 AFLP fragments which were specific to succinea, conspicua and spectabilis was identified using 24 different AFLP primer combinations. Among these 2,269 fragments, 16 bands which were the most specific to one color patterns were isolated, cloned and sequenced. Subsequent UPGMA cluster analysis revealed that population of H. axyridis was divided four major group and these genetic tree showed that H. axyridis elytra colour diversity was affected by genetic polymorphism. It is considered that these genetic analyses may be facilitated the understanding of molecular genetic mechanism related with the wing colour pattern formation in this species.

        Human saliva contains a large number of proteins and peptides whose composition may alter as a consequence 。f disease. To date. however‘ the proteins and peptides that routinely populate thi s ora l fluid a re largely unknown To provide a catalogue of saliva proteins, we have surveyed the unstimulated human whole saliva by us ing shotgun proteornics. For the shotgun approach, whole saliva proteins were digested into peptides with ChemDigestD ‘ and the resul ting peptide fragments were separated by RP-HPLC, followed by each fraction was t ryptic digestion. ChemDigestD-Trypsin digested peptides were analyzed by tandem mass spectrometry(MS/MS) us ing a nano-LC eq 버 pped quadrupole-time of f1ight mass spectrometer, and the obtained spectra were searched against human protein seq uence data base using MASCOT. Shotgun proteomics allowed a total of 291 human pr。 teins to be confidently assigned . The largest group(17 .2%) of the identified proteins sorted into functional catego ries was included in t he signal t ransduction funct ion except for the hypothetical 0 1' unknown function. This work provides a valuable s ta rting point for the analysis of human salivary proteins and their biological functions and candidates from human whole sali va that may prove to be of diagnostic and therapeutic significance

        The human ELAV(embryonic lethal abnormal vision)-like protein HuR stabilizes a certain group of cellular mRNAs that contain AU- ri ch elements in theil‘ 3’• untranslated region, Dysregulation of mRNA s tability may be relevant in tumor biology and may lead to abnormal expression of several proteins in malignant tumors, The aim of this study is to identify the differentially expressed proteins according to the functi onal activi ty of HuR Methods : We used stabl e expression of small interfering RNA(siRNA) of HuR gene to inhibit the expression of HuR in human ora l car cinoma cell lines‘ KB cell line and YD10B cell line‘ and compared the proteomic changes between s iRNA-treated and cont rol cell line using two-dimensional gel electrophoresis , Flow cytometry caliber scan(FACS) was employed to investigate the effects of HuR on cell apoptosis and proliferation , Results: Seventeen differentially expressed proteins between the two cell lines were identified by electrospray ioruzati on quadrupole time-of- fl ight mass spectrometry(ESI-Q-TOF MS) and database searching, Among them there are eleven proteins which are significant ly up- regulated in siRNA- treated cell line, which include heat shock protein 10(influencing nucl eocytoplas rnic transpo 다, cell dedifferentiation , and inhibition of apoptosis) , keratin 19(basal cell differ ent iat ion) ‘ and nucleoside diphosphate kinase B(G protein activator), etc Enolase 1 and Ml of pyruvate kinase a re the representatives of signifi cantly down-regulated proteins in siRNA-treated cell 11l1e Conclusion : Our data suggest that HuR participate in mRNAs stability of proteins that have the counter effects in the carcinogenesis of oral ca ncer , And the functiona l proteomics a re needed to elucidate the detailed interactions between HuR and t hese molec ules
