Until now, in case of Palaearctic region, only one species, Nesendaeus monochrous in this genus was recorded from China. but the biology and life cycle are not clearly known, yet. This species collected from the bud of Glochidion chodoense in Korea for the first time and closely related to Heterochyromera imerodeus Kojima & Morimoto which was recorded only in Japan,. But Nesendaeus monochrous is easily distinguishable from H. imerodeus by the relatively longer elytra with bisinuately round apices. Heterochyromera imerodeus Kojima & Morimoto can be found from Eurya emarginata and Glochidion obovatum in Japan. So, this two species are closely related each other and need more taxonomic study about the generic synonymy or changing the generic position.
Cnidium officinale M. is an important crop that is widely used as a raw material for health functional foods. However, it is experiencing cultivation difficulties due to climate change and abnormally high temperatures. In response to this problem, the characteristics and main causes of the high-temperature damage occurring in C. officinale M. cultivation fields were analyzed. A survey of five farmhouse fields in Jecheon and Bonghwa, major C. officinale M. cultivation areas in Korea in 2018, indicated that about 5% to 37% of the cultivation fields in Jecheon and 5% to 15% of the fields in Bonghwa died from wilting. The high-temperature damage of the C. officinale M. fields is divided into two categories: upper leaves drying due to solar radiation and temperature, and lower leaves dying serially to the radiant heat of the vinyl mulch. Damage caused by radiant heat was typically greater. This is due to the greenhouse effect that occurs in the small space between the black vinyl mulching and the soil. The heat radiated to the surface of the ridge creating an environmental condition that greatly exceeded the atmospheric temperature especially on hot days. As a result, short plants with underground parts, such as C. officinale M., can suffer more high-temperature damage than other plants, so it is considered that it is necessary to develop related technologies such as mulching materials that can reduce pavement temperature in the future.
Wolsong unit 1, the first PHWR (Pressurized Heavy Water Reactor) in Korea, was permanent shut down in 2019. In Korea, according to the Nuclear Safety Act, the FDP (Final Decommissioning Plan) must be submitted within 5 years of permanent shutdown. According to NSSC Notice, the types, volumes, and radioactivity of solid radioactive wastes should be included in FDP chapter 9, Radioactive Waste Management, Therefore, in this study, activation assessment and waste classification of the End shield, which is a major activation component, were conducted. MCNP and ORIGEN-S computer codes were used for the activation assessment of the End shield. Radioactive waste levels were classified according to the cooling period of 0 to 20 years in consideration of the actual start of decommissioning. The End shield consists of Lattice tube, Shielding ball, Sleeve insert, Calandria tube shielding sleeve, and Embedment Ring. Among the components composed for each fuel channel, the neutron flux was calculated for the components whose level was not predicted by preliminary activation assessment, by dividing them into three channel regions: central channel, inter channel, and outer channel. In the case of the shielding ball, the neutron flux was calculated in the area up to 10 cm close to the core and other parts to check the decrease in neutron flux with the distance from the core. The neutron flux calculations showed that the highest neutron flux was calculated at the Sleeve insert, the component closest to the fuel channel. It was found that the neutron flux decreased by about 1/10 to 1/20 as the distance from the core increased by 20 cm. The outer channel was found to have about 30% of the neutron flux of the center channel. It was found that no change in radioactive waste level due to decay occurred during the 0 to 20 years cooling period. In this study, activation assessment and waste classification of End Shield in Wolsong unit 1 was conducted. The results of this study can be used as a basis for the preparation of the FDP for the Wolsong unit 1.
본고에서는 최근 중국을 중심으로 추진되고 있는 한약재 ISO 국제표준화에 대한 대응을 위해 최신 등재 현황을 소개하고 주요 추진상의 문제점과 대응방안들을 점검해 보았다. 주요 내용을 정리하면 다음과 같다.
1. 국제표준화기구 전통의학 기술위원회(ISO TC249)는 2009년 설립이후 31개의 국가가 참여하여 한약재 국제표준화를 추진하고 있다. 한·중·일은 발의 문건은 90%이상을 차지하여 사실상 아시아의 전통의학 분야를 두고 경쟁하고 있으며 간사국인 중국은 약용작물의 종자 ·종묘에서부터 약재의 생산, 품질 그리고 침구, 의료기술 등에 이르기까지 전 한방분야에서 표준화를 주도하고 있다. 현재 진행되고 있는 약용작물 재배 및 한약재 품질 관련 안건만 약 50여건에 달해 향후 5년 내에 시장 거래량 상위품목들의 국제표준화 작업이 대부분 완료될 것으로 예상된다.
2. 현재 진행되고 있는 안건들은 한약재의 품질이나 품질평가에 관한 안건들이 대부분이다. 이 안건들이 국제표준으로 인정되면 세계 한약재 유통시장에는 새로운 질서와 우열기준이 생기고 유통 한약재의 시장가치 평가에 반영되어 각국 한약재 점유율 변동에도 영향을 줄 수 있다. 이들 안건들은 직접적인 재배생산 표준이 아닌 경우가 대부분이지만 성분이나 성상 등에 대한 표준을 담고 있으므로 대부분 재배생산과도 밀접한 관련이 있다. 만일 중국 중심의 표준화가 계속해서 심화된다면 장기적으로는 국내 생산농가에 부담을 줄 수도 있다. 그러므로 한약재 국제표준화 대응은 향후 국내 생산물이 표준규격을 달성할 수 있는 범위내로 기준을 유도하는 전략을 펴야 할 것이다. 이를 위해 국내 생산물의 특성을 파악하고 선반영 하려는 노력이 필요하다.
3. 한약재 표준화 대응은 중국의 추진안건이 많으므로 분야별 파급영향을 고려한 선별적인 대응이 필요하다. 종자·종묘 등 추가적인 제한 필요성이 낮은 분야보다는 향후 시장에서 한· 중 간 경쟁이 발생할 수 있는 분야에 집중하는 것이 중요하다 . 원산지 변조, 농약 잔류량·이산화황 과다검출 등의 이력이 있는 약재들은 해당 표준을 강화하고 재배환경의 영향을 받을 수 있는 도지약재들의 경우 성상이나 지표성분 등에 대한 국내약재의 특성을 최대한 반영할 필요성이 있다. 곰팡이나 해충관리 등은 아직 각국의 관리규정이나 저온저장 시설 등 현장 인프라 구축이 충분치 않은 점들도 고려되어야 한다. 아직 국제표준을 추진하기 위한 국내의 연구결과들이 충분치 않은 부분들이 많으므로 추가적인 연구와 지원이 필요할 것이다.

Resistances to tetracycline were declined in accord with reduced usage of tetracycline in live stock production. E. coli stains from chicken meat had higher multi-drug resistance ratio than strains from other meat. One extended spectrum beta lactamase (ESBL) producing E. coli and two ESBL producing Salmonella were identified in this study. ESBL producing Salmonella strains were confirmed to carry CTX-M-1 type genes.

Lepidopteran pests monitoring in adult stage was generally performed using delta or corn typed trap including rubber septa impregnated sex pheromone (lure). Sometimes, unfortunately trapped samples were severly damaged because of biotic and/or abiotic environments such as micro-organism, predator and rain, sticky material, respectively. In our case, we monitored potato tuber moth, PTM, Phthorimaea operculella distribution during 2009~2012 in Korea. However, we encountered unexpected problem, another species can be trapped in species specific sex pheromone trap. Therefore, species confirmation was needed in trapped samples. Here we developed confirmation method by direct PCR (without DNA extraction) or sequencing methods which trapped samples that cannot identified by morphologically. We designed multi-plex PCR universal primers and species specific primers in rRNA region because to check the success of PCR and species identification. This direct PCR method can be applied in other species confirmation which monitored using pheromone trap.
The green peach aphid, Myzus persicae (Sulzer), is one of the most serious pests in cabbage cultivation. Field survey was carried out to know the insecticide resistance levels and to develop the applicable insecticide resistant markers in five main cabbage cultivation regions (Pyeong-chang, Hong-cheon, Bong-wha, Mu-ju and Je-ju) during 2009 to 2011. M. persicae can resist a wide range of insecticides in five surveyed local populations. Therefore multi resistant (MR) strain was selected from these five local populations and esterase over-expression, modified AChE (MACE) and mutation(s) in para-type sodium channel were analyzed using native IEF and quantitative sequencing with five local populations. Esterase over-expression and MACE (StoF mutation) were observed in all populations including MR strain. LtoF mutation is well known as a kdr mutation in para-type sodium channel. However, even though LC50 values of MR strain noted over 2,000 times higher than that of susceptible strain against bifenthrin, LtoF mutation was not detected in para type sodium channel and also local populations. We found another mutation (MtoL) in para and that mutation highly correlated between mutation ratio and bioassay data. For preliminary resistance monitoring, we developed quantitative sequencing (QS) to detect the frequencies of point mutation as a population genotyping. These methods can apply to manage M. persicae resistant populations in field.
A MnSOD gene was cloned from the fall webworm, H. cunea. The MnSOD cDNAs encode precursor proteins of 215 amino acid residues. The deduced amino acid sequences of the H. cunea MnSOD cDNA showed 76% identity to B. mori MnSOD and 62-56% to MnSOD sequences from other organisms. MnSOD and Cu/ZnSOD in H. cunea is expressed from all tissues. MnSOD expression is changed at a trace level in infected larvae, while Cu/ZnSOD expression is strongly changed against bacteria, and fungi. The expression level of Cu/ZnSOD increased by different artificial photoperiod (24L:0D), UV irradiation (312nm), and starvation condition, while the expression level of MnSOD only increased by starvation condition. Also, expression of MnSOD and Cu/ZnSOD showed no significant change in 0L:24D condition. In addition to expression levels of Cu/ZnSOD in H. cunea significantly increased by temperature stress and injection with paraquat, but reduced by injection with 10% H2O2. The expression level of MnSOD significantly increased by temperature stress and reduced by injection with 10% H2O2 and paraquat.
Apolipophorin-III (apoLp-III) is a hemolymph protein whose function is to facilitate lipid transport in an aqueous medium in insects. Recently, apolipophorin-III in Galleria mellonella and Hyphantria cunea was shown to play an unexpected role in insect immune activation. We show here a novel possible function/role of the apoLp-III in insects. To investigate the genes which have a relationship with apoLp-III in fall webworm larvae, we reduced endogenous Hc apoLp-III mRNA levels in larvae via RNA interference (RNAi). The RNAi-mediated Hc apoLp-III reduction resulted in the reduction of antioxidants, like MnSOD, catalase, and glutathione S transferase as well as immune proteins. In particular, expression of MnSOD commonly decreased in fat body, midgut, and hemocytes following the knockdown of Hc apoLp-III, which induced an elevated level of superoxide anion in Hyphantria cunea larvae. The observed effect of Hc apoLp-III RNAi suggests that Hc apoLp-III is related to the action/expression of antioxidants, especially MnSOD.
A new insect member of the STAT family of transcription factors (HcSTAT) has been cloned from the lepidopteran, Hyphantria cunea. The domain involved in DNA interaction and the SH2 domain are well conserved. The gene is transcribed at a low level during all stages of development, and transcribed in hemocyte, fat body, midgut, epidermis, and Malpighian tubule. Especially, hemocyte and Malpighian tubule showed transcriptional activation of HcSTAT upon Gram-negative and -positive bacteria challenge. Gram-negative and -positive bacteria challenge specifically results in nuclear translocation of HcSTAT protein and induction of DNA-binding activity that recognizes a STAT target site in H. cunea hemocyte. In vivo treatment with sodium orthovanadatetranslocates HcSTAT to the nucleus in hemocyte cells.
Root knot nematode species, such as Meloidogyne hapla, M. incognita, M. arenaria and M. javanica are economically most notorious nematode pests, causing serious damage to the various crops throughout world. In this study, DNA sequence analyses of the D1-D3 expansion segments of the 28S gene in the ribosomal DNA were conducted to characterize genetic variation of the four Meloidogyne species obtained from Korea and United States. PCR-RFLP (Polymerase Chain Reaction-Restriction Fragment Length Polymorphism), SCAR (Sequence Characterized Amplified Region) marker and RAPD (Random Amplification of Polymorphic DNA) also were used to develop the methods for exact and rapid species identification. In the sequence analysis of the D1-D3 expansion segments, only a few nucleotide sequence variation were detected among M. incognita, M. arenaria, and M. javanica, except for M. hapla. The PCR-RFLP analysis that involves amplification of the mitochondrial COII and lrRNA region yielded one distinct amplicon for M. hapla at 500 bp, enabling us to distinguish M. hapla from M. incognita, M. arenaria, M. javanica reproduced by obligate mitotic parthenogenesis. SCAR markers successfully identified the four root knot nematode species tested. We are under development of RAPD primers specific to the three root knot nematodes found in Korea.
