        Streptozotocin(STZ)으로 유도된 당뇨 흰쥐에게 연교의 에탄올 추출물을 1일 1회 7일간 1,000㎎/㎏.b.w의 용량으로 투여 후 glucose함량과 이에 관여하는 효소(glucokinase(GK), glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dehydrogenase(G-6-PDH)활성과 glycogen 함량, triglyceride(T.G), total cholesterol등의 지질대사에 관여하는 물질들의 함량을 측정한 결과 연교 에탄올 추출물 투여군이 glucose, T.G, total cholesterol 등의 함량과 G-6-Pase 활성이 유의적인 감소를 나타내었으며 glycogen 함량과 GK의 활성이 유의적인 증가를 나타내었다. 이와 같이 연교 에탄올 추출물을 1,000㎎/㎏.b.w을 당뇨 흰쥐에게 투여한 결과 혈당저하, 지질대사의 개선 효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

        당뇨 합병증을 일으키는 중요 매체인 산화 스트레스에 대한 매생이 추출물과 그 지표물질인 pheophorbide a (PhA)의 정소 조직 내 산화 스트레스 보호 효과를 streptozotocin(STZ)에 의해 유발된 당뇨 쥐 모델에서 확인하였다. 당뇨를 유발하는 물질로 알려진 STZ를 40 mg/kg body weight (b.w.)의 농도로 복강 투여하여 당뇨를 일으킨 후, 매생이 추출물(CFE)샘플을 각각 4, 20 mg/kg b.w. 그리고 PhA를0.2 mg/kg b.w.로 9 주간 투여하여 실험하였다. 정소 조직에서 산화 스트레스에 생성되는 체내 과산화물 지표인 혈액 nitric oxide와 지질 과산화물이 당뇨 유발군에 비해 CFE와 PhA 투여군에서 유의적인 감소를 보였으며, 체내 과산화물 축적을 제어하는 효소인 glutathione peroxidase,glutathione-S-transferase가 정상 수준으로 회복되었다. 또한, 체내 항산화 방어기작에서의 중요한 효소인 superoxide dismutase, catalase 활성 또한 샘플 투여 시 회복되는 것을 보아 CFE 투여군과 PhA투여군에서 조직 내 항산화 효소 조절과 함께 과산화물 축적 저해 효과를 통하여 당뇨 상태에서 고혈당에 의한 산화 스트레스가 일으키는 생식 조직 손상으로부터 매생이 추출물과 그 지표물질인 PhA는 보호 효과를 가지는 것을 확인할 수 있었다.

        본 동물실험은 STZ로 유도한 C57BL/6에게 5% sodium butyrate 를 급여했을 때 항당뇨 및 항염증 효과를 연구하고자 하였다. 본 연구에서 STZ로 당뇨를 유발한 마우스에게 5% sodium butyrate를 급여했을 때 체중과 식이섭취량에서는 크게 유의 적 차이가 없음을 확인하였다(p<0.05). STZ에 의한 당뇨 쥐는 인슐린의 분비가 감소되면서 당대사의 불균형을 초래하며 간 등이 비대해진다고 알려져 있으나, 본 연구에서는 간의 장 기 무게에서는 크게 실험군 간에 유의적인 차이가 없었다 (p<0.05). 또한 비장과 흉선의 무게는 0.5% sodium butyrate 첨가 식이군에서 유의적으로 낮아짐을 알 수 있었다(p<0.05). 당뇨병은 염증 상태로서 고혈당으로 인하여 monocyte에서는 여러 염증성 사이토카인이 분비가 활성화된다. TNF-α, IL-6 등은 염증성 사이토카인으로서 혈관염증의 중요한 마커로 인식되고 있고, 당뇨병 환자들은 이러한 염증성 사이토카인 이 높은 수준으로 활성화 된다. STZ 처리 시 마우스 혈청에서 의 염증성 사이토카인의 분비 및 발현이 증가되었으나, 5% sodium butyrate를 급여했을 때 염증성 사이토카인의 분비 및 발현이 저해됨을 확인할 수 있었다. 본 연구는 sodium butyrate 보충은 당뇨병이 유발된 동물모델에서 혈청지질 농도 및 혈 당 조절, 염증 상태를 개선에 다소간의 효과가 있는 것으로 나타났다. 이에 따라 당뇨병과 같은 만성적인 대사질환 개선 에 sodium butyrate가 효과적인 식이인자가 될 것으로 생각된 다. 그러나 앞으로 더 명확한 효능을 탐색하기 위해서 시료 첨가수준의 다각화 및 여러 가지 보완연구가 필요할 것으로 생각 된다.

        This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats

        The ethanol extraction yield of Meliae toosendan fructus(MT) was about 24.5% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of MT in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in MT treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in MT treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in MT treated group. Also the activities of glucose-6-phosphate dehydrogenase(G-6-PDH) and glucokinase(Gk) were increaed in MT treated group. The content of hepatic glycogen was significantly increaed in MT treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in MT treated group.         This study was conducted to examine the effects of Opuntia ficus-indica complex (OF) on the lipid metabolism, bile acid in feces, alanine aminotransferase (ALT) activity, aspatate aminotransferase (AST) activity, composition of urine and expression of cholesterol related mRNA in streptozotocin (STZ)-induced diabetic rats. Thirty two male Sprague-Dawley rats were randomly divided into non-diabetic control (NC), diabetic control (DC), diabetic OF of 2% (OF-2) and diabetic OF of 5% (OF-5), then each group was fed for 3 weeks. Plasma total cholesterol, non-esterified fatty acids (NEFA), total cholesterol, low density lipoprotein (LDL), very low density lipoprotein (VLDL) were decreased significantly (p<0.05) in OF-5 group compared to DC, but high density lipoprotein (HDL) was not changed. AST and ALT were also reduced and bile acid excretion was improved. Composition of urine in OF-5 was almost same in NC. The expression of cholesterol 7α-hydroxylase (CYP7A1), 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA-R), Low density lipoprotein receptor (LDL-R) mRNA indicated that feeding OF have the effects of cholesterol decreation in plasma by synthesis of bile acid from cholesterol. These results provide experimental evidence about improved lipid metabolism of the OF feeding in the STZ-induced diabetic rats.

        This study was performed to investigate the antidiabetic study effect in ethanol extract of Ailanthicortex(A.C) radici in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45㎎/㎏ dissolved in citrate buffer. The ethanol extract of A.C radici was orally administrated once a day for 7 days at a dose of 500㎎/㎏. The content of serum glucose, triglyceride(T.G), total cholesterol were significantly decreased in treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased in treated group compared to the those of STZ-control group. but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was significantly increased. These results indicated that ethanol extract of A.C would have antidiabetic effect in STZ-induced diabetic rats.

        This study was carried to investigate the antidiabetic effect of ethanol extract of Smallanthus sonchifolius(S.S) in Streptozotocin(STZ)- induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 43mg/kg dissolved in citrate buffer. The ethanol extract of S,S in leaves and root were orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in S.S treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased,and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in S.S treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of S.S would have antidiabetic effect in STZ-induced diabetic rats.

        To investigate the effects of coated liposome from Discorea rhizoma extract (DRE) in streptozotocin (STZ)-induced, we evaluated changes in body weight, fasting blood glucose, blood insulin and blood lipid concentrations in mice. Mice were divided into four groups: (DC), diabetic DRE at 10 ㎎/㎏ (DRE-1), diabetic DRE at 50 ㎎/㎏ (DRE-2), and diabetic DRE at 250 ㎎/㎏ (DRE-3). Mice had free access to water and diet (10 weeks). The DC group showed higher blood cholesterol than the DRE-1, DRE-2, DRE-3 groups. In glucose tolerance test, the DRE-1, DRE-2, and DRE-3 groups increased after 30 minutes in decremental glycemic response area under the curve. Fasting blood glucose levels in the DRE groups significantly decreased through 4 weeks. Plasma total cholesterol, triglyceride and LDL-cholesterol concentrations were also lower in the DRE groups. On the other hand, the DRE-1, DRE-2 and DRE-3 groups showed higher HDL-cholesterol and insulin levels than the DC group. Moreover, blood glucose and lipid levels significantly decreased in streptozotocin (STZ)-induced diabetic mice treated with DRE. These results indicate that DRE may reduce elevated blood glucose levels and serum lipid concentrations in hypoglycemic and diabetic mice, suggesting its usefulness as a functional food.

        This study was carried to investigate the antidiabetic metabolism effect of water extract Cordyceps Militalis(C.M.) in Streptozotocin(STZ) induced diabetic rats. Diabetes were induced by intravenous injection of STZ at a dose of 42mg/kg,b.w. dissolved in citrate buffer. The water extract of C.M. was orally administrated once a day for 7 days at a dose of 500mg/kg,b.w(body weight). or 1,000mg/kg.b.w. The content serum glucose was significantly decreased in C.M. treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in C.M. treated group compared to the those of STZ-control group. These results indicated that water extract of C.M. would have antidiabetic metabolism effect in STZ-induced diabetic rats.

        To identify the treatment effect of lactic acid bacteria for diabetes, the treatment effects of a single administration of acarbose (a diabetes treatment drug) or lactic acid bacteria, and the mixture of acarbose and lactic acid bacteria on diabetes in a type 1 diabetes animal model, were studied. In this study, streptozotocin was inoculated into a Sprague-Dawley rat to induce diabetes, and sham control (Sham), diabetic control (STZ), STZ and composition with live cell, STZ and composition with heat killed cell, STZ and composition with drugs (acarbose) were orally administered. Then the treatment effect on diabetes was observed by measuring the body weight, blood glucose, and serum lipid. For the histopathological examination of the pancreas, the Langerhans islet of the pancreas was observed using hematoxylin and eosin staining, and the renal cortex, outer medullar, and inner medullar were also observed. The induced diabetes decreased the body weight, and the fasting blood glucose level decreased in the lactic-acid-bacteriaadministered group and the mixture-administered group. In addition, the probiotic resulted in the greatest decrease in the serum cholesterol level, which is closely related to diabetes. Also, the hematoxylin and eosin staining of the Langerhans islet showed that the reduction in the size of the Langerhans islet slowed in the lactic-acid-bacteria-administered group. The histopathological examination confirmed that the symptoms of diabetic nephropathy decreased in the group to which viable bacteria and acarbose were administered, unlike in the group to which dead bacteria was administered. The mixture of lactic acid bacteria and acarbose and the single administration of lactic acid bacteria or acarbose had treatment effects on the size of the Langerhans islet and of the kidney histopathology. Thus, it is believed that lactic acid bacteria have treatment effects on diabetes and can be used as supplements for the treatment of diabetes.

        The present study was performed to determine beneficial effects of donkey meat extract supplementation (15 and 30 mg/kg B.W.) on streptozotocin (STZ) induced diabetic rat. Rump muscle of donkey was extracted by boiling in water for 3 hrs and supplemented to STZ induced diabetic rats for 6 weeks. Feed and water intake of STZ induced diabetic rats were higher than those of normal rats, while no significant difference was found in DME treated rats as compared to the rats in control group. STZ-induced diabetic rats did not improved their body weight as compared to that of rats in the control group. However, blood glucose level of 30 mg DME supplemented rats was found to be significantly reduced after 4 weeks of supplementation (p<0.05) when compared to the control animals. Administration of DME did not show any improving effect on serum lipids (HDL-cholesterol, total cholesterol, and triglyceride). No significant changes were noticed in terms of weight of all the visceral organs of animals fed on DME except liver. It is therefore, concluded that supplementation of DME with dose of 30 mg/B.W. resulted in decreased blood glucose levels, however, blood lipid profile of diabetic rats, having more than 400 mg/dl glucose level, did not change.

The water extract of paecilomyces japonica were orally administrated once a day for 7 days at a dose of 500mg/kg or 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in PJ treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in PJ treated group compared to the those of STZ-control group. These results indicated that water extract of paecilomyces japonica would have antidiabetic and lipid metabolism effect in STZ-induced diabetic rats.

        Our objective of current study was to investigate the development of bone and heart in association with diabetes mellitus (DM). DM was induced by administering an intraperitoneal injection of streptozotocin (STZ; 60 mg/kg) to 4‐gweek‐gold Sprague‐gDawley rats. Body weight and blood glucose were monitored, and rats were sacrificed after 2 or 5 weeks. The left ventricle (LV), including the interventricular septum, was weighed, and body weight and tibial bone length were assessed. Young diabetic rats showed reduced growth in terms of tibial length and body weight compared to controls. Moreover, diabetic males showed more significant growth suppression and reduced LV size than diabetic females. Morphometric analysis of tibiae from diabetic rats revealed suppressed bone growth at 2 and 5 weeks, with no difference between genders. STZ‐ginduced diabetes decreased bone growth and retarded pre‐gpubertal heart development. As a result, diabetes may increase cardiovascular risk factors and lead to eventual heart failure. Therefore, new therapeutic approaches are required for diabetic children exhibiting growth retardation. Heart growth factor, exercise, and cardiopulmonary physical therapy may be required to promote heart development and physiological function.

        Many biological systems are regulated by an intricate set of feedback loops that oscillate with a circadian rhythm of roughly 24 h. This circadian clock mediates an increase in body temperature, heart rate, blood pressure, and cortisol secretion early in the day. Recent studies have shown changes in the amplitude of the circadian clock in the hearts and livers of streptozotocin (STZ)-treated rats. It is therefore important to examine the relationships between circadian clock genes and growth factors and their effects on diabetic phenomena in animal models as well as in human patients. In this study, we sought to determine whether diurnal variation in organ development and the regulation of metabolism, including growth and development during the juvenile period in rats, exists as a mechanism for anticipating and responding to the environment. Also, we examined the relationship between changes in growth factor expression in the liver and clock-controlled protein synthesis and turnover, which are important in cellular growth. Specifically, we assessed the expression patterns of several clock genes, including Per1, Per2, Clock, Bmal1, Cry1 and Cry2 and growth factors such as insulin-like growth factor (IGF)-1 and -2 and transforming growth factor (TGF)-β1 in rats with STZ-induced diabetes. Growth factor and clock gene expression in the liver at 1 week post-induction was clearly increased compared to the level in control rats. In contrast, the expression patterns of the genes were similar to those observed after 5 weeks in the STZ-treated rats. The increase in gene expression is likely a compensatory change in response to the obstruction of insulin function during the initial phase of induction. However, as the period of induction was extended, the expression of the compensatory genes decreased to the control level. This is likely the result of decreased insulin secretion due to the destruction of beta cells in the pancreas by STZ.

        The investigation assessed the influence of Compositae plants consumption on the protein profile in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats by injection of STZ (45 mg/kg body weight) into tail vein. The rats were randomly assigned to five groups: normal and STZ-control fed an AIN-93 diet, and groups whose diets were supplemented with l0% Compositae powder containing Artemisia iwayomogi (A. iwayomogi), Atracηlodes lancea (A. lancea) or Taraxacum mongolicum (T mongolicum). To observe the effects of Compositae plants in the animal model, the levels of protein in liver, kidney, lung, pancreas, and brain were determined after 4 weeks. The level of protein in kidney increased significantly in rats receiving the A. iwayomogi- and T mongolicum-supplemented diet compared to the STZcontrol group. The level of protein in lung was increased significantly in the A. iwayomogi-supplemented group. Bl00d glucose level correlated well with brain protein level but did not correlate with other protein levels. Also, blood glucose correlated inversely with kidney, lung and brain protein levels. It is suggested that supplementation with A. iwayomogi in diabetic rats leads elevates protein in kidney and lung.
