        볏짚 대체 배지를 개발하기 위해 밀짚 혼용 비율에 따른 볏짚 대체 효과를 구명한 결과는 다음과 같다. 후발효 배지의 화학적 성분을 분석한 결과 밀짚을 20% 혼용한 배지의 질소 함량과 C/N율은 각각 2.2%와 16.7이고, 볏짚 단용 배지는 2.0%, 17.9로 조사되었으며, 볏짚 단용 배지보다 초발이소요일수가 빠르고, 수량도 30% 증수되어 대체 효과를 기대할 수 있었다.

        Recently, round bale silage is very popular preservation method in Korea. In autumn, many farmers usually make silage from fresh rice straw, but the quality of rice straw silage was not good. This study was conducted to find out the silage quality of fresh rice straw in the mid-western costal area in Korea. The average DM (dry matter) content was 40.5% and pH was 5.0. Forage quality of "A" and "D" farm silage was higher than that of others area. Silage quality was also higher in "A" and "D" farm silage of west costal area. The content of lactic acid of "A" and "D" farm silage was high and acetic and butyric acid was low. The result of this experiment indicated that the quality of fresh rice straw silage made by farmers was somewhat low. So we must guide farmers to making good quality of fresh rice straw silage.

        Mycotoxin producing moulds are commonly in contact with forage in the field before harvest, during harvesting, drying and transport and during storage. Therefore, this study was to isolate and identify fungi contaminated rice straw in the earlier spring of Korea. The pure fungi colony was isolated from normal rice straws in the tree dairy farms. The fungi were cultured on MEA and PDA agar plate and confirmed by sequencing the ITS region. Fungal contamination was found in all samples of rice straws and mold counts averaged 4.6×10⁴ per gram. The mycotoxigenic fungi found were Penicillium roqueforti and Fusarium culmorum. Other several fungi were determined Gibberella moniliformis, Ascomycota sp., Myrothecium sp. When the rice straw samples were analyzed mycotoxins by HPLC, it was not found aflatoxins, zearalenone, deoxynivalenol, fumonisin, ochratoxins A and T-2 toxin. The results suggest t㏊t the mold contaminated rice straw is not activated in the spring of cool season but is able to develop in the coming warm and humidly summer, specially mycotoxigenic fungi such ase Penicillium roqueforti and Fusarium culmorum.

        In previous studies, we reported that sow which was transferred OPS-freezing embryos not able to deliver a piglet (Kim et al, 2004). This study was conducted to investigate a possibility of gilt as recipients which produce piglets after transfer of OPS-freezing embryos. All transferred embryos were prepared by in vitro production (IVP) system. In vitro culture (IVC) medium used glucose-free NCSU23 supplemented with 5mM sodium pyruvate, 0.5 mM sodium lactate and 4 mg/ml bovine serum albumin for 2 days at . From day 3 of IVC, 10% fetal bovine serum albumin was added to the culture medium. In preparing of freezing embryos, embryos were treated with 7.5 cytochalasin-B for 30 min and centrifuged at for 13 min. And then, embryos were exposed sequentially to an ethylene glycol (EG) solution, aspirated into open pulled straw (OPS), and plunged or thawed into the liquid nitrogen. In embryo transfer (ET), we used two kinds of type (surgical method vs. non-surgical method). In surgical method of embryo transfer, embryo were transferred in both uterine horn of two recipient gilts by plastic straw. Non-surgical method which is like artificial insemination was performed on three gilts. Each 140 frozen embryos were transferred to two gilts and 40 fresh embryos to one gilt. Pregnancy establishment was shown one recipient at 45 days after ET. However, the one recipient was also aborted at 58 days after ET. These results suggest that gilts can be considered as a candidate of recipients for OPS-freezing embryo transfer.

        This study was carried out to investigate the effects of cryoprotectants, warming solution and removal of lipid on open pulled straw vitrification (OPS) method of porcine embryos produced by nuclear transfer (NT) of fetal fibroblasts. All solutions used during vitrification were prepared with holding medium consisting of 25 mM Hepes buffered TCM199 medium containing 20% fetal bovine serum (FBS) at 38.5℃. The blastocysts derived from NT with or without lipid were vitrified in each medium of different concentrations of dimethyl sulfoxide (DMSO) and ethylene glycol (EG). Also, blastocysts after cryopreservation were warmed into different concentrations of sucrose in warming solution. The optimal concentrations of cryoprotectants in vitrification solution were 10% DMSO + 10% EG in vitrification solution 1 (VS1) and 20% DMSO + 20% EG in vitrification solution 2 (VS2). The optimal concentrations of sucrose were 0.3 M sucrose in warming solution 1 (WS1) and 0.15 M sucrose in warming solution 2 (WS2). Lipid removal from oocytes before NT enhanced the viability of NT embryos after vitrification. Our results show that use of the OPS method in conjunction with lipid removal provides effective cryopreservation of porcine nuclear transfer embryos.

        본 연구는 OPS 기법에 의한 돼지 수정란의 동결-융해 시 수정란의 발달 단계와 superoxide dismutase (SOD)가 수정란의 생존능력에 미치는 영향을 검토하였다. 돼지 체외수정 배반포는 OPS방법에 의해 동결 후 융해하여 0~10unitsml의 SOD 존재 하에 48시간 체외배양하였다. 동결-융해 후 형태학적으로 정상적인 수정란의 비율은 초기, 중기 및 확장배반포간에 유의적인 차이는 인정되지 않았다(30.8~38.6%). 그러나 발육단계가 높을수록 형태적으로 정상인 수정란의 비율이 높은 경향을 나타냈다. 수정란의 융해 후 48시간 추가 배양했을 때, 발육이 진행된 수정란은 후기배반포기에 동결한 수정란이 38.7%로 유의적으로 높았으며(P<0.05), 1 unit/ml의 SOD를 첨가한 경우 비교적 높은 생존율을 나타내었다. 본 연구의 결과로부터, 수정란의 OPS방법에 의한 동결-융해 후 생존성의 향상을 위해서는 후기배반포기 단계에 동결하는 것이 유리하며, SOD의 첨가는 수정란의 손상을 어느 정도 방지할 수 있을 것으로 사료된다.

        The aim of this study was to evaluate the effects of different straw volume (0.5 and 5.0 ㎖) on cryopreservation in boar semen. Semen quality was evaluated the motility, viability, abnormality, acrosome intactness and membrane integrity. And there were also examined the development rates of IVM/IVF embryos using frozen-thawed boar semen. Boar semen were frozen until 5℃ for 3 hours using cell freezer and making the straws, and then freezing by lowing the straws into styrofoam box on the 8 cm above the LN2. In different straw volume (0.5 and 5.0 ㎖), sperm viability and abnormality were not differ in 0.5 and 5.0 ㎖ straws, but sperm motility were significantly higher in 0.5 ㎖ straws (61.3%) than in 5.0 ㎖ straws (56.3%) (p<0.05). In the Coomassie Brillient Blue (CBB), Hoechst 33258/Propidium Iodide (H258/PI) staining and Hypoosmotic Swelling Test (HOST), the acrosome intactness and sperm membrane integrity were not differ in 0.5 and 5.0 ㎖ straws, but sperm survival rate was significantly higher in 0.5 ㎖ straw (65.0%) than in 5.0 ㎖l straw (55.0%) (p<0.05). Employing the Chlortetracycline (CTC)/Hoechst33258 (H258), all treatment group were not differ in characteristic of uncapacitated acrosome-intact sperm (F), capacitated and acrosome-intact sperm (B-type) and acrosomereaction seprm (AR-type). In the developmental rate of IVM/IVF embryos using frozen-thawed boar semen in different straw volumes, the developmental rate of morula plus blastocysts were 19.9% in 0.5 ㎖ straw and 18.8% in 5.0 ㎖ straw, respectively. These results indicated that straw volume affects the semen motility and sperm survival rate, but not other semen characteristic and developmental rate of IVM/IVF embryos.
