        To investigate the in vivo cognitive effects of syringic acid(SA), Y-maze and passive avoidance tests were performed in amyloid-β(Aβ)-induced cytotoxicity. Learning and memory impairment by Aβ neurotoxicity was partially recovered in Institute of Cancer Research(ICR) mice orally administered SA(10mg/kg of body weight). The SA treated group showed an inhibitory effect on acetylcholinesterase(AChE) that was extracted from mice brain tissue after in vivo tests. Aβ-induced oxidative stress was also examined by malondialdehyde(MDA) and 2',7,-dichlorofluorescein diacetate(DCF-DA) assays, and lipid peroxidation of brain homogenates and cellular oxidative stress were reduced by SA. In cell viability assays using 3-[4,5-dimethythiazol-2-yl]-2,5-diphenyl tetrazolium bromide(MTT), lactate dehydrogenase(LDH) leakage, and caspase 3/7 activity, the SA treated group showed relatively effective protection against Aβ-induced neurotoxicity compared to the others. Consequently, these results suggest that SA in black soybean seed coat extract might improve cognitive function because of its neuronal cell protective effects against oxidative stress and the inhibitory effect of AChE as a cholinergic enzyme.

        본 연구에서는 제주 지역에서 재배하고 생산되는 보리의 이화학적 특성을 분석하고, 수용성 식이섬유인 β-glucan을 추출하여 타 지역에서 재배된 보리와 비교, 분석하여 제주 보리가 기능성을 갖춘 제품으로 활용될 수 있는 자료를 제 시하고자 하였다. 제주 지역에서 재배된 보리는 전반적으 로 전분 함량과 식이섬유 함량이 높았으며, 특히 수용성 식이섬유인 β-glucan의 함량이 높은 것으로 나타났다. 제주 맥주보리는 타 지역 보리와 유사한 성분 함량을 갖는 것으 로 나타났으며 도정유무가 전분, 식이섬유와 β-glucan 함량 에 영향을 주는 것으로 나타났다. 보리의 점도는 가열 처 리에 의해 상당량 증가하였는데 상온에서는 제주쌀보리가 102.5 cP의 높은 점도를 나타내었고 가열 후에는 4563.3 cP로 제주청보리가 가장 높은 점도를 나타내어 타 지역 보 리에 비해 점도가 높게 나타났다. 또한 도정 유무가 점도 에 영향을 미쳤는데, 도정되지 않은 제주맥주보리의 점도 가 도정된 제주맥주보리에 비해 현저히 감소하였다. 열수 와 효소를 이용하여 보리로부터 비교적 높은 수율의 β- glucan을 추출하였다(70-80%). 추출된 β-glucan의 재용해율 은 보리의 품종에 따라 다르게 나타났는데, 검정보리와 제 주쌀보리의 재용해율이 높은 것으로 나타났다. 도정되지 않은 제주맥주보리는 가장 낮은 재용해율을 보여 도정이 추출된 β-glucan의 물리적 특성에 영향을 미치는 것으로 나타났다. β-Glucan을 구성하는 glucose들간의 β(1→3):β(1 →4) 결합비는 1:2.3-4.49의 범위에서 측정되었으며, 정제 정도만이 β(1→3):β(1→4) 결합비에 영향을 미치는 것이 아 니라 도정율과 같은 다른 요인들도 영향을 미칠 수 있다는 것을 확인하였고, 구체적으로 어떠한 요인들이 β(1→3):β(1 →4) 결합비에 영향을 미칠 수 있는지에 대한 연구가 더 필요한 것으로 보인다. 본 연구를 통하여 제주 지역에서 재배된 보리의 일반성분과 온도에 따른 점도 특성, 추출된 β-glucan의 이화학적 특성의 결과를 바탕으로 제주 보리를 활용한 새로운 기능소재 및 제품 개발에 이용 가능성이 높 을 것으로 기대되었다.

        The oocyte undergoes various events during maturation and requires many substances for the maturation process. Various intracellular organelles are also involved in maturation of the oocyte. During the process glucose is essential for nuclear and cytoplasmic maturation, and adenosine triphosphate is needed for reorganization of the organelles and cytoskeleton. If mitochondrial function is lost, several developmental defects in meiotic chromosome segregation and maturation cause fertilization failure. The endoplasmic reticulum, a store for Ca2+, releases Ca2+ into the cytoplasm in response to various cellular signaling molecules. This event stimulates secretion of hormones, growth factors and antioxidants in oocyte during maturation. Also, oocyte nuclear maturation is stimulated by growth factors such as epidermal growth factor. This review summarizes roles of organelles with focus on the Golgi apparatus during maturation in oocyte.

        Ceramics biomaterials are useful as implant materials in orthopedic surgery. In this study, porous HA(hydroxyapatite)/β-TCP(tricalcium phosphate) composite biomaterials were successfully fabricated using HA/β-TCP powders with 10-30 wt% NH4HCO3 as a space holder(SH) and TiH2 as a foaming agent, and MgO powder as a binder. The HA/β-TCP powders were consolidated by spark plasma sintering(SPS) process at 1000 oC under 20 MPa conditions. The effect of SH content on the pore size and distribution of the HA/β-TCP composite was observed by scanning electron microscopy(SEM) and a microfocus X-ray computer tomography system(SMX-225CT). These microstructure observations revealed that the volume fraction of the pores increased with increasing SH content. The pore size of the HA/β-TCP composites is about 400-500 μm. The relative density of the porous HA/β-TCP composite increased with decreasing SH content. The porous HA/β-TCP composite fabricated with 30%SH exhibited an elastic modulus similar to that of cortical bone; however, the compression strength of this composite is higher than that of cortical bone.

        β-cyclodextrin has an ability to protect compounds from oxidative reaction by collecting them within its ring-like structure. So, In harsh condition (40oC), marker compound, quercetin, was dramatically reduced in Hovenia dulcis fruit extract containing dextrin at 4 and 8 week compared to 0 week, but not that containing β-cyclodextrin. To evaluate the effects of dextrin and β-cyclodextrin on protective effect of H.dulcis fruit extract against alcohol- induced liver damage, The mice were orally injected alcohol, H. dulcis fruit extract/dextrin (HD) and H. dulcis fruit extract/β-cyclodextrin (HCD), respectively, for 7 days. The mice orally administrated with alcohol significantly enhanced the serum concentration of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and the activity of lactate dehydrogenase (LDH) in serum compared to the control group. HD and HCD significantly decreased the levels of serum ALT and AST and serum LDH activities compared to alcohol group. And also alcohol group significantly increased the level of total cholesterol compared to the control group, but HD and HCD significantly reduced it compared to the alcohol group. However, the levels of TG in blood were not significantly changed in all groups. The activities of alcohol dehydrogenase (ADH) were significantly increased in HD and HCD group although those of aldehyde dehydrogenase showed an increasing tendency. This data suggested that HD and HCD were able to induce alcohol degradation in the liver tissues. All together, the results showed that HCD demonstrated their ability to protect liver from alcohol-induced damage on equal terms with HD.

        본 연구에서는 꽃송이버섯 건물과 powder를 각각 Yeast Extract broth와 MRS broth에서 Lactoballius 4종(L. plantarum, L. acidophilus, L. helveticus, and L. delbrueckii)과 함께 발효시켰다. 발효액의 final pH는 pH 3.9±0.02로 유산균의 일반적인 final pH 값과 일치함을 통해 유산균의 발효를 확인하였고, 유산균에 의한 꽃송이버섯 발효의 효과를 β-glucan 함량을 통하여 확인하였다. Yeast Extract broth 발효액은 YSB-2LP 1.65 g/100 g으로 β-glucan을 거의 함유하고 있지 않았지만 MRS broth 발효액은 Yeast Extract broth 발효액의 10배에 달하는 MPB-1LP 10.84 g/100 g을 함유하고 있었다. MPB-1LP는 꽃송이버섯 powder와 L.actobacillus plantarum으로 발효한 발효액으로 pH stress로 인해 만들어진 L. plantarum의 β-D-glucosidase에 의해 분해된 고분자량의 수용성 β-glucan이라 사료된다. 꽃송이버섯을 powder 상태로 L. plantarum과 함께 발효시켰을 때 꽃송이버섯의 수용성 β-glucan 추출 효율을 높일 수 있으며, 본 연구의 결과는 꽃송이버섯 유산균 발효물의 면역촉진 작용 연구를 위한 기초연구자료로 활용될 수 있을 것이다.

        본 연구는 큰느타리버섯의 베타글루칸 고함유 형질에 관련된 SCAR marker를 개발하기 위해 수행되었다. operon사의 OPA(20개), OPB(20개), OPL(20개), OPP(20개), OPR(20개), OPS(20개) 등 총 120개 primer를 random primer(10 mer)로 사용하여 대립 계통 9종과 베타글루칸 고함유 계통 9 종을 대상으로 RAPD를 이용한 bulked segregant analysis를 실시하여 OP-R03 primer로부터 대립 계통에는 나타나지 않고 베타글루칸 고함유 계통에만 나타나는 특이적인 RAPD 밴드를 얻었다. OP-R03 primer를 이용한 RAPD 결과, 약 91 bp 부근에서 베타글루칸 고함유 계통에 특이적인 DNA 밴드가 관찰되었으며 이 DNA 밴드의 염기서열 말단을 근거로 SCAR 마커로 사용할 specific primer인 OP-R03-1-F와 OP-R03-1-R를 디자인하였다. SCAR 마커 OP-R03-1-F/-1-R primer를 이용하여 PCR을 수행한 결과에서도 91 bp 부근에서 대립 계통과 구별되는 DNA 밴드가 베타글루칸 고함유 계통에서 확인되었으며 random primer인 OP-R03 primer를 이용하여 PCR을 수행했을 때보다 재현성이 높고 진한 DNA 밴드임을 확인할 수 있었다.

        The ubiquitous Na, K-ATPase is a membrane-bound ion pump located in the plasma membrane in all animal cells and plays an essential role in a variety of cellular functions. Studies in several organisms have shown that this protein regulates different aspects of embryonic development and is responsible for the pathogenesis of several human diseases. Na, K-ATPase is an important factor for retinal development, and combinations of the isoforms of each of its subunits are expressed in different cell types and determine its functional properties. In this study, we performed RT-PCR assay to determine temporal expression and in situ hybridization to determine spatial expression of Na, K-ATPase β2 isoform (atp1b2) in Xenopus laevis. Focusing on retinal expression to distinguish the specific expression domain, we used retinal marker genes sox4, sox11, vsx1, and pax6. Xenopus atp1b2 was expressed from late gastrulation to the tadpole stage. Using whole mount in situ hybridization, we showed that Xenopus atp1b2 was expressed broadly in the eye, the whole surface ectoderm, and gills. In situ hybridization on sections revealed detailed and specific expression in the outer nuclear layer of the retina, which consists of two major classes of photoreceptors, rods and cones, surface ectoderm, pharyngeal epithelium, and gills. These findings indicate that atp1b2 may play an important role for the development of Xenopus retina.

        Recombinant thymosin β4 (rTβ4) has been reported to migrate and promote vascularization, wound-healing, and hair growth in a mouse hindlimb ischemia model of peripheral vascular disease. C57BL/6 mice (11-weeks-old) were anesthetized and an ischemic model was made by cutting the right aorta femoralis. The ischemic group was intraperitoneally administered with saline (300 μL/mouse) and the muscular administration group received rTβ4 (150 μg in 300 μL of saline) or rTβ4 (150 μg in 300 μL saline) to the abdominal cavity at 3-day intervals for 21 days. Myoatrophy of the ischemic group was observed compared to the normal control group. Generation of adjacent vessels was carried out in the rTβ4 administration group compared to the ischemic group. The biopsy results showed significant fibrosis around the muscular undersurface and perimysium in the musculus quadriceps femoris of the ischemic group, whereas partial fibrosis was observed in the perimysium and endomysium in the rTβ4 administration group. Immunostaining indicated that expression levels of hypoxia-inducible factor-1α (HIF-1α), vascular endothelial growth factor-1 (VEGF-1), and endothelial nitric oxide synthase (eNOS) in the rTβ4 group were higher than those of the ischemic group. Western blotting showed that expression levels of HIF-1α, VEGF-1, and eNOS in the rTβ4 group were higher than those of the ischemic group. In conclusion, rTβ4 increases expression levels of HIF-1α, VEGF-1, and eNOS, resulting in angiogenesis.

        The synthesis of octenyl succinyl β-gucan (OSA-β-glucan) was carried out and its interfacial properties at the oil-water interface and in emulsion systems were investigated. An aqueous ethanol system as a reaction media was used to facilitate the synthesis process; 10% (w/w) ethanol found to be the best as it showed a maximum degree of substitution (DS: 0.0132). FT-IR showed a characteristic absorption spectrum at 1736cm-1, indicating the esterification of octenyl succinyl groups to β-glucan backbone. As for interfacial tension measurements, it was decreased with increasing concentration of OSA-β-glucan in the aqueous phase and when NaCl was added to aqueous OSA-β-glucan solution in the range of 0.01 M to 0.1 M and also when pH was raised (pH 3 ~ pH 9). In systems of emulsion stabilized with OSA-β-glucan, fat globule size found to decrease with increasing concentration of OSA-β-glucan, showing a critical value of about 0.32μm at 0.5 wt%. When the OSA-β-glucan emulsions were stored, it was found that fat globule size was increased with storage time and particularly pronounced increase was observed in emulsion with 1% OSA-β-glucan, possibly due to depletion flocculation. Results of creaming stability evaluated by light scattering technique showed that it was more stable in emulsions containing smaller fat globule size. Surface load of OSA-β-glucan in emulsions increased with increasing concentration of OSA-β-glucan, suggesting a multilayer adsorption.

        DEAE Sephadex column chromatography에 의해 Bacillussp. 유래 β-mannanase의 정제를 수행하여 비활성 21.57units/mL 정제배율 95.33배를 나타내었다. SDS-PAGE에 의한 단일밴드를 확인하였고, 분자량은 38.9kDa으로 결정되었다.정제효소에 의해 Picea abies galactosyl glucomannan을 가수분해하여 activated carbon column chromatography에 의해 당가수분해물을 분리 회수하여 TLC, FACE 및Timell’s method에 의해 중합도 8, 10으로 결정되었으며,Penicillum purpurogenum유래 정제 β-mannanase와 α-galactosidase를 이용한 enzymatic sequential action에 의해 2가지 가수분해산물 모두 hetero type galactosylglucomannooligosaccharides로 확인되었다. B. longum, B.bifidum, B. infantis, B. animalis, B. breve, B. adolessentis,B. auglutum의 생육활성에 대한 중합도 8, 10의 영향을 검토하기 위하여 modified-MRS 배지상에 탄소원으로 중합도8, 10를 대체하여 생육활성을 비교한 결과 B. animalis에서는 중합도 8 galactosyl glucomannooligosaccharide를 탄소원으로 대체한 경우 표준 MRS 배지와 비교하여 19.5배, 중합도 10에서 18.7배의 가장 우수한 생육활성을 나타내었으며, B. bifidum에 대해서는 중합도 8의 경우 15.3배와 중합도10에서 14.3배 그리고 B. longum에서는 중합도 8 galactosylglucomannooligosaccharide를 탄소원으로 대체한 경우 표준MRS배지와 비교하여 중합도 8에서 15.2배, 중합도 10에서13.9배의 상대활성을 우선적으로 나타내었다.

        팽화홍삼에 존재하는 major ginsenoside를 minorginsenoside로 생물전환하기 위하여 β-glucosidase 활성이높은 유산균주를 탐색하였다. 된장으로부터 분리한 YLB 8균주가 β-glucosidase 활성이 가장 높았으며 Leuconostocmesenteroides로 동정되었다. L. mesenteroides YLB 8을MRS 배지에서 배양하였을 경우 최대의 β-glucosidase 효소 활성을 나타내기 위한 최적 배양 조건은 glucose 첨가량 1%(w/v), 초기 pH 8.0, 배양온도 30oC, 배양시간 16시간이었다. L. mesenteroides YLB 8을 이용하여 팽화홍삼추출액을 생물전환하였을 경우 최적 온도는 30oC었으며,팽화홍삼 추출액의 농도는 12oBx이었다. 팽화홍삼 추출액내에 존재하는 major ginsenoside인 Rb1과 Rb2는 minorginsenoside인 Rg3로 전환되었으며, Re는 Rg1으로 전환되었으며, 이 중 일부는 Rh1과 Rh2로 전환되었다.

        The inhibitory activities of the Cordyceps pruinosa butanol fraction (Cp-BF) were investigated by determining inflammatory responses of lipopolysaccharide (LPS)-treated RAW264.7 macrophage cells and by evaluating HCl/ethanol (EtOH)-triggered gastric ulcers in mice. The molecular mechanisms of the inhibitory effects of Cp-BF were investigated by identifying target enzymes using biochemical and molecular biological approaches. Cp-BF strongly inhibited the production of NO and TNF-α, release of reactive oxygen species (ROS), phagocytic uptake of FITC-dextran, and mRNA expression levels of interleukin (IL)-6, inducible NO synthase (iNOS), and tumour necrosis factor-alpha (TNF)-α in activated RAW264.7 cells. Cp-BF also strongly down regulated the NF-κB pathway by suppressing IKKβ according to luciferase reporter assays and immunoblot analysis. Furthermore, Cp-BF blocked both increased levels of NF-κB-mediated luciferase activities and phosphorylation of p65/p50 observed by IKKβ overexpression. Finally, orally administered Cp-BF was found to attenuate gastric ulcer and block the phosphorylation of IκBα induced by HCl/EtOH. Therefore, these results suggest that the anti-inflammatory activity of Cp-BF may be mediated by suppression of IKKα and its downstream NF-κB activation. Since our group has established the mass cultivation conditions by developing culture conditions for Cordyceps pruinosa, the information presented in this study may be useful for developing new anti-inflammatory agents.

        Double-stranded RNA(dsRNA) had been used to specitically suppress target gene expression at post-tanscription level. Injection of dsRNA to hemocoel is the most efficient to knockdown target mRNA. However, some insects have shown to be susceptible to feeding dsRNA. Spodoptera exigua was susceptible to dsRNA at oral treatment. Especially dsRNA specific to β-integrin was potent to survival of S.exigua larvae. This study advanced our dsRNA application technology by generating recombinant E.coli expressing dsRNA specific the β-integrin. A recombinant vector L4440 was constructed with a partial β-integrin gene under T7 RNA polymerase promoter. The recombinant vector was used to transform HT115 competent cells of E.coli. The transformed E.coli expressed the dsRNA. The production of dsRNA was proportional to the bacterial number. By feeding the recombinant E.coli, S.exigua underwent significant mortality. By adding E.coli expressing Cry1Ca Bt toxin to E.coli expressing dsRNA, S.exigua exhibited highly enhanced mortality. This study suggests a possibility to use a recombinant E.coli expressing dsRNA to control S.exigua.

        본 연구는 남한지역 백두대간 마루금 일대의 고도별 식물 다양성 분포 패턴 및 기후인자, 면적, 기하학적 억제인자, 순일차 생산량이 고도별 식물 다양성 분포 패턴에 미치는 효과를 구명하기 위해 수행되었다. 백두대간 마루금 일대를 따라 위치한 1,100개의 조사구에 대한 식생조사 결과, 총 97과 342속 802종의 식물종이 관찰되었으며, 고도에 따른 식물종의 α 다양성을 나타내는 종풍부도와 Shannon-Wiener 지수는 단봉형 패턴을 나타내는 반면, β 다양성을 나타내는 Jaccard와 Chao-Jaccard 유사도 지수는 고도에 따라 일반적으로 증가하는 경향을 나타내었다. 또한, 이러한 분포 패턴을 제어하는 인자로서 기하학적 억제인자가 α 다양성을 설명하는 가장 중요한 인자로 나타났으며, β 다양성은 기후인자가 가장 높은 설명력을 가지는 인자로 나타났다. 본 연구는 동일한 분류군 내에서도 다양성 측정방법에 따라 상이한 고도별 분포 패턴 및 제어인자를 가진다는 것을 제시하며, 백두대간 마루금 일대의 식물 다양성을 제어하는 인자가 단순히 하나가 아닌 몇 가지 인자의 복합작용에 의해 나타나는 현상임을 의미한다.
