        돼지의 행동을 조사하여 정상행동과 비정상행동으로 구분하고 이들 중 동물복지 수준평가 기준으로 활용 가능한 행동형을 선별하였으며, 이들을 농장동물의 복지수준 평가시스템 구축을 위한 유용 파라미터로 활용하고자 하였다. 국내 대부분의 양돈장에서 채택하고 있는 임신돈 스톨과 분만틀을 사용하고 있는 관행농장과 임신돈군사와 분만틀이 없는 분만방을 사용하고 있는 동물복지형 농장에서 임신, 분만, 자돈 및 육성비육 등 4단계로 구분하여 조사, 분석하였다. 총 92가지의 행동유형을 조사하였으며, 정상행동 57가지와 비정상행동 35가지로 분류하였다. 임신돈의 정상행동과 비정상행동은 각각 14가지와 8가지였으며, 대표적인 비정상행동은 스톨 내에서의 헛씹기, 철봉 물기 및 공격 등이었고 분만돈은 정상행동이 15가지, 비정상행동은 귀물기, 경련 및 공격 등 3가지였다. 자돈은 22가지의 정상행동과 17가지의 비정상 행동으로 조사되었으며, 대표적인 비정상행동은 괴롭히기, 밀어내기, 몸빨기, 짓밟기 등이었다. 육성비육돈은 정상행동 5가지와 괴롭히기, 밀어내기, 물기, 올라타기, 싸우기 등의 대표적인 비정상행동을 나타내었다.

        Cotesia plutellae, an endoparasitoids braconid wasp, possesses a polydnaviruses (PDVs) called Cotesia plutellae bracovirus (CpBV) that encodes viral histone H4 (= CpBV-H4). This viral histone H4 shares high sequence homology (82.5%) with host`s H4 of P.xylostella, except an extended N-terminal tail consisting of 38 amino acid residues with nine lysines. Its extended N-terminal tail has been postulated to play a crucial role in suppressing host immunity, growth and development-associated genes, presumably through an epigenetic control mechanism. A suppression subtractive hybridization (SSH) analysis was analyzed in transcriptome by short-read sequencing technology and provided several target and non-target genes of a viral histone H4. In this study, we analyzed the effect CpBV-H4 on the expression of two target genes: Lysine-specific demethylase (KDM) and Serine proteinase inhibitor (Serpins). Transient expression of CpBV-H4 into non parasitized P. xylostella was performed by microinjection of a recombinant expression vector, and showed the expression up to 70 h. Under this transient expression condition, we analyzed the effect of CpBV-H4 on the expression of target genes by RT-PCR at different time points. Interestingly, the CpBV-H4 significantly inhibited the expression of these target genes, while the truncated CpBV-H4 deleting the N-terminal tail did not show this inhibitory effect. This study also showed that the viral histone H4 suppresses expressions of lysine-specific demethylase and serine proteinase inhibitor (Serpin2) to inhibit host growth and development.

        Temperature and insecticide are two important factors that affect survival, reproduction and other physiological processes of insects. To determine interactions of temperature and insecticide treatment on susceptible (S) and imidacloprid-resistant strains (IMI-R) of Aphis gossypii, adults were exposed to three temperatures (17, 22, and 28℃) or combinations of three temperatures (17, 22 and 28℃) and imidacloprid (LC20), and the expression of several genes (heat shock protein 70, cuticle protein, cytochrome P450, and elongation factor) were analyzed. The expression level at 17℃ of heat shock protein 70, cuticle protein, and elongation factor in S strain were up-regulated with increased time and higher than those of IMI-R strain. However, expression of cytochrome P450 was not affected by elevated temperature both S and IMI-R strain. Combined treatment of elevated temperature and imidacloprid were significantly up-regulated only cuticle protein in S strain and higher than those of IMI-R strain.

        본 연구는 큰느타리버섯의 고온적응성 형질에 관련된 SCAR marker를 개발하기 위해 수행되었다. operon 사의 OPA(20개), OPB(20개), OPL(20개), OPP(20개), OPR(20개), OPS(20개) 등 총 120개 primer를 random primer(10 mer)로 사용하여 대립 계통 7종과 고온성 계통 7 종을 대상으로 RAPD를 이용한 bulked segregant analysis를 실시하여 OP-A06 primer로부터 대립 계통에는 나타나지 않고 고온성 계통에만 나타나는 특이적인 RAPD 밴드를 얻었다. OP-A06 primer를 이용한 RAPD 결과, 약 385 bp 부근에서 고온성 계통에 특이적인 DNA 밴드가 관찰되었으며 이 DNA 밴드의 염기서열 말단을 근거로 SCAR 마커로 사용할 specific primer인 OP-A06-1-F와 OP-A06-1-R를 디자인하였다. SCAR 마커 OP-A06-1-F/-1-R primer를 이용하여 PCR을 수행한 결과에서도 385 bp 부근에서 대립 계통과 구별되는 DNA 밴드가 고온성 계통에서 확인되었으며 random primer인 OP-A06 primer를 이용하여 PCR을 수행했을 때보다 재현성이 높고 진한 DNA 밴드임을 확인할 수 있었다.

        본 연구는 한려해상국립공원 동부지역에 위치한 5개의 특정도서지역(소치도, 장도, 세존도, 솔섬, 학섬)의 관속식물상을 2011년 4월부터 2012년 10월까지 총 50회에 걸쳐 조사를 실시하였다. 장도에서 관속식물의 출현은 특산식물 1분류군, 귀화식물 10분류군, 희귀식물 1종을 포함하여 총 52과 102속 11종11변종 2품종 124분류군 이 조사되었으며, 학섬에서는 44과 73속 76종 8변종 84분류군이 출현하였고 특산식물 3분류군 귀화식물 5분류군을 포함하고 있었다. 소치도에서의 관속식물은 38과 51속 51종 8변종 59분류군이 출현하였고, 귀화식물 3분류군, 희귀식물 3분류군이 조사되었다. 솔섬에서는 귀화식물 1종, 희귀식물 2분류군을 포함하여 28과 28속 24종 4변종 28분류군, 세존도에서는 귀화식물 1종, 희귀식물 1종을 포함하여 15과 19속 16종 4변종 20분류군이 출현하였다. 식물구계학적 특정식물은 I등급 46종, II등급 1종, III등급 16종, Ⅳ등급 2종, Ⅴ등급 4종이 출현하였다.

        최근 국내 포도원에서 과실 표면에 구름모양의 흰색 얼룩이 자주 발견되고 있으나, 그 원인에 대해 명확히 알려진 바가 없어 방제대책을 세우지 못하고 있는 실정이다. 흰색의 달무리 반점은 포도 과실 표면에 부정형으로 퍼져 있으며, 그 중앙에는 총채벌레가 산란할 때 만들어진 작은 구멍의 상처가 남아 있다. 이 상처부위는 시간이 지나면서 코르크화되고 표피세포와 분리되어 딱지로 남거나 떨어져 나가게 된다. 이러한 증상은 총채벌레의 섭식이나 노린재의 흡즙에 의한 상처와는 구별된다. 산란구멍에서 발견한 총채벌레 알껍질에서 DNA를 추출하여 ITS2 부위의 염기 서열을 PCR-RFLP 방법으로 분자동정을 실시한 결과 꽃노랑총채벌레의 것으로 확인되었다. 미토콘드리아 COI 염기서열은 이러한 분자 동정 결과를 재확인하여 주었다. 본 연구결과는 포도 과피에 꽃노랑총채벌레 산란에 의해 유발되는 독특한 피해 증상에 대한 정확한 정보를 제공하며 포도원에서 이 해충에 대한 방제전략 수립에 도움이 될 것으로 여겨진다.

        같은 속의 복숭아순나방(Grapholita molesta)과 복숭아순나방붙이(G. dimorpha)는 기주범위와 야외에서의 성충 발생 양상이 유사하다. 또한 두 종은 성페로몬 성분으로 cis-8-dodecenyl acetate (Z8-12:Ac)와 trans-8-dodenceyl acetate (E8-12:Ac)을 공통으로 사용하고 있는데, 이 두 성분으로 구성된 상용 성페로몬 유인제에 두 종이 혼합되어 유인된다. 본 연구는 복숭아순나방붙이 트랩에 복숭아순나방이 혼합유인되는 현상을 줄이기 위해 성페로몬 미량성분의 효과를 분석했다. 복숭아순나방붙이 성페로몬인 Z8-12:Ac/E8-12:Ac = 85/15에 cis-8-tetradecenyl acetate(Z8-14:Ac)를 첨가했을 때, 복숭아순나방붙이의 유인은 유지되면서 복숭아순나방의 유인은 유의하게 억제되었다. Dodecyl acetate (12:Ac)와 tetradecyl acetate (14:Ac)의 첨가는 두 종의 유인력에 영향을 주지 않았다. 한편 Z8-12:Ac/E8-12:Ac/Z8-14:Ac = 85/15/10의 세 성분 성페로몬에 trans-8-tetradecenyl acetate (E8-14:Ac)를 첨가했을 때, 복숭아순나방붙이의 유인이 오히려 유의하게 억제되었다. 이상의 결과로부터 Z8-12:Ac/E8-12:Ac/Z8-14:Ac = 85/15/10의 조성이 개선된 복숭아순나방붙이의 성페로몬으로 제시되었다.

        This study aims to investigate the antecedents to customer retention and brand loyalty of Internet Service Providers in Thailand. The findings reveal that customers’ commitment and value are influenced by information support, privacy and security. Additionally, the positive relationships between information quality and privacy and behavioural loyalty were revealed. Customers’ commitment and value were positively associated with behavioural loyalty. However, surprisingly, there was no support for the hypothesised positive association between customers’ value with behavioural loyalty. Practical implications that can be drawn from this research will form a foundation for service providers in the residential internet market to develop new retention strategies. These providers would l be able to reduce the current issues relating to the high customer churn rate. By making customers more central in company operations, these strategies can potentially reduce the expenses associated with acquiring new customers.

        This study examines the effect of brand-specific transformational leadership (BSTL) on employee-based brand equity (EBBE) with the mediating mechanism of empowerment. We explore the newly emerging concept of ‘employee based brand equity’ particularly from the perspective of brand-specific transformational leadership in the service industry. Furthermore, we explore the perspective of empowerment, structural and psychological empowerment, as the mediating mechanism between BSTL and EBBE. We highlight the direct and indirect effects of BSTL on EBBE and employs hierarchical linear modeling (HLM). 578 questionnaires were collected from multilevel data of subordinates and their supervisors at 58 well-known service brand units in Taiwan. Our research results implicate that: (1) BSTL is positively related to employee base brand equity; (2) Structure empowerment is a mediator factor of BSTL and employee base brand equity relationship; (3) Psychological empowerment is a mediator factor of BSTL and employee base brand equity relationship. This study presents three key contributions. This study provides the following research contributions: (a) In contrast to previous studies that focused on customer brand equity, we initiate the quantitative research of research on frontline employee based brand equity. (b) This study suggests and confirms that BSTL influences EBBE through the mediation of structure empowerment and psychological empowerment, which further contributes to EBBE research. (c) Finally, this study applies a hierarchical linear model to conduct a cross-level analysis of EBBE. Consequently, this study provide evidence supporting the previous assertion that comprehensive insight into service organizational behaviors can be achieved only by employing cross-level analysis and mediating factors.

        Endothelin 2 (EDN2) induces follicular rupture by constricting periovulatory follicles. In this study, it was investigated the mechanisms of EDN2 action on follicular rupture with respect of receptor using the conditionally granulosa cell specific EDN2 receptor type A (ETa) KO mice (gcETaKO; ETaflox/-․Amhr2Cre). It was generated the gcETaKO mice by breeding with ETaflox/- mice after mono-alleic ETa knockout by ZP3Cre and Amhr2Cre mice. Fertility, ovulation and maturation rates of ovulated oocytes after super ovulation were investigated in the gcETaKO mice compared with wild-type mice (ETaflox/flox and ETaflox/-) as a control group. In the gcETaKO mice, normal fertility after breeding with male mice was shown compared with wild-type mice. And, there was no significant differences in ovulation rates after super ovulation, however its maturation rates was lower than that of wild type mice. These findings show that EDN2 in follicular rupture for ovulation is related with an other ETa not in granulosa cells. Further studies are needed to investigate how EDN2 is acted in ovarian follicular rupture for ovulation.

        The primary objective of the present study is the characterization of the somatic hybrids of dikaryon-monokaryon (di-mono) crosses in mushroom breeding. We employed this technique for developing superior strain from Pleurotus ostreatus strains with 56 intraspecific hybrids of 14 combinations between six Pleurotus ostreatus strains and one Pleurotus florida strain. In this study, the results of analysis on hybridization rate, nuclear DNA patterns, and colors and morphology of fruit-bodies, are presented as follows. In di-mono crosses, somatic hybrids among Pleurotus strains showed 100% of crossability as seen in those among Pleurotus strains indicating that nuclei of a dikaryon migrated to a recipient. 89.3% of the somatic hybrids among Pleurotus strains were similar to the donor dikaryons, and 10.7% had combined DNA patterns of both parents. In the 14.3% di-mono cross between P. ostreatus and P. florida, the nuclear DNA patterns of the all hybrid strain showed the same or similar patterns compared to the donor dikaryons. 75.0% of the hybrid between P. ostreatus and P. ostreatus were similar to the donor dikaryons; 10.7% had combined DNA patterns of both parents. 82.2% of fruiting body morphology of the hybrids among Pleurotus strains were similar to the dikaryons, and 17.8% had combined DNA patterns of both parents. All hybrid strains between dikaryon P. florida and monokaryon P. ostreatus showed the fruiting body whose colors were similar to those of the dikaryon, while the hybrids between dikaryon P. ostreatus and monokaryon P. florida were all showed combined colors of both parents but are more similar to the dikaryon. Therefore, the fruiting body color of P. florida tends to be generally dominant. The present study was able to find out and suggest superior hybrid strains by identifying the nuclear DNA patterns of hybrids between Pleurotus strains as well as the characteristics of their fruiting bodies. This study expects that the advantages of the di-mono crossing are needs to be fully utilized in mushroom breeding and it is better to develop superior strains of Pleurotus strains.

        느타리(Pleurotus ostreatus)의 이핵-단핵 계통간(di-mono)교잡주의 DNA 유전에 관한 특성을 구명하기 위하여 느타리 6계통 및 사철느타리 1계통으로 이핵-단핵 계통간 12조합 48교잡주를 얻어 교잡율, 교잡주의 핵 DNA패턴 양상과 유연관계도, 자실체의 형태, 갓 색깔을 분석하였다. 이핵-단핵 계통간 교잡에서 느타리와 느타리간, 느타리와 사철느타리간 교잡은 모두 교잡율 100%로 나타났다.이핵-단핵 계통간 교잡주는 공여체(donor) 이핵체의 핵이수용체(recipient) 단핵체로 전이되었다. 이핵-단핵 계통간교잡주의 DNA 패턴은 이핵체와 유사하거나 동일한 것이87.5%, 양친의 중간 패턴이 12.5%였다. 즉, 느타리 이핵주와 느타리 단핵 계통간 교잡주는 이핵체와 유사한DNA 패턴이 70.9%, 양친의 핵이 공존하는 중간 패턴이12.5%였으며, 사철느타리와 느타리간 이핵-단핵 계통간교잡주는 16.6%로 모두 사철느타리 핵 DNA 패턴과 유사하거나 동일하였다. 교잡주의 핵 DNA 패턴은 교잡조합에 따라 차이가 나타났는데 12교잡조합 중에서 4조합에서만 단핵주와 유사하거나 중간 형태를 나타내었고 나머지는 이핵주와 동일한 양상이었다. 교잡주의 자실체 형태는 이핵주 형태가79.2%, 양친의 중간형태 또는 단핵체 모군주의 형태가20.8%였다. 하지만 이핵체 형태라 하더라도 자실체 색깔은 다소 달랐다. 사철느타리 이핵-느타리 단핵주간 교잡주의 자실체 갓 색깔은 모두 이핵체 사철느타리와 유사하거나 동일하였다. 느타리 이핵-사철느타리 단핵 계통간교잡주는 양친의 중간 갓 색깔로 모두 나타났으며 다소이핵체와 유연관계가 가까운 색깔이었다. 따라서 사철느타리가 다소 우성으로 나타나는 경향이었다. 이러한 결과로 보아 교잡주는 3종류의 핵이 모두 공존하는 세포가 많을 것으로 생각되며, 이핵-단핵 계통간 교잡 방법은 우수한 계통을 육성하는 훌륭한 방법으로 이용될 수 있을 것으로 기대된다.

        Protein and carbohydrate are the two most important macronutrients that have profound consequences for the fitness of insects. Many insects are capable of balancing the intake of multiple nutrients to minimize the fitness costs associated with ingesting diets that are imbalanced with respect to protein and carbohydrate. It has been hypothesized that insects will redress the imbalance of their nutrient state through increasing the appetite for specific nutrients that are ingested in deficit. We tested this possibility using a mealworm beetle, Tenebrio molitor (Coleoptera: Tenebrionidae). Newly emerged beetles were confined to one of two nutritionally imbalanced foods that contained only protein or carbohydrate (P:C ratio = 0:42 and 42:0, expressed as % dry mass) for 16 days, after which they were given an opportunity to choose between two nutritionally imbalanced diets (0:42 versus 42:0). Over the first few days of the food choice, beetles that had previously experienced protein-limitation preferred protein to carbohydrate while the reverse was true for those that had experienced carbohydrate-shortage. Such contrasting patterns of diet preference observed between the two groups of beetles diminished subsequently as the insects recovered from nutrient imbalance. Our results provide strong support for the long-standing idea that attaining the right balance of nutrients is the main motive for foraging in insects.

        Cell-free fetal RNA has been highlighted as useful tools for the fetal sex determination or other genetic inherent disorder. However, there is no knowledge about the sex determination using cell free fetal RNA in bovine field. Thus, the present study aimed to evaluate the presence of transcripts of DDX3Y, USP9Y and ZRSR2Y genes in maternal plasma of pregnant cows to determine the sex of the fetus using real-time quantitative polymerase chain reaction assay, and verify its accuracy, sensitivity and specificity compared with the molecular testing and the calf sex at birth. Transcripts of USP9Y and DDX3Y genes were expressed in the all plasma of males and females both the control group and the experimental group. However, ZRSR2Y gene was matched up with the molecular testing and the true sex in control group and has an overall accuracy of 82.6%, a sensitivity of 75%, and a specificity of 100% in experimental group. Therefore, these results indicated that real time PCR technique, as a noninvasive and cost-efficient method, is possible to determination fetal sex in the bovine species using circulating cell free RNA in maternal plasma and especially ZRSR2Y gene could be a good candidate for the RNA based sex determination work.

        본 연구에서는 입경·비중 겸용 밭작물 곡물 선별기 개발을 위한 전 단계로서, 밭작물 곡물의 완전립과 불량립의 기하학적, 공기역학적, 마찰 특성을 측정하고, 선별기 설계에 적용하기 위한 선별 요인을 분석하였다. 측정된 곡물의 기하학적 특성 결과(길이, 폭, 두께)를 이용하여 스크린의 치수와 형상을 결정하여 선별기를 제작하여 선별시험을 한 결과 선별효율은 콩의 경우 94.7%, 커피는 92.0%, 아몬드는 74.5%, 옥수수는 90.0%로 나타났다. 측정된 곡물의 마찰 특성 결과를 이용한 고무벨트 선별 시험 결과 콩의 경우 벨트 속도 0.5 m/s, 벨트 각도 5˚ 일 때 선별효율이 88.2%로 가장 높게 나타났다. 측정된 곡물의 공기역학적 특성 결과, 종말속도는 콩의 경우 4.4∼6.0 m/s, 커피는 3.2∼4.4 m/s, 아몬드는 3.5∼5.1 m/s, 옥수수는 4.7∼6.0 m/s의 범위로 나타났다.

        The purpose of the study was to investigate an effect of water temperature on a non-specific immune response and mortality of tilapia, Oreochromis niloticus, following a bacterial infection. Seventy five tilapia acclimated to 25℃ were then transferred at 16 and 36℃, and examined for non-specific immune responses over 12-96 h. Respiratory burst activity was reduced significantly in the group of fish cultured at 16 and 36℃ over 24-96 h, whereas phagocytic activity decreased significantly in the group of fish reared at a low temperature (16℃) over 12 and 24 h and high temperatures (36℃) over 12-96 h. Lysozyme activity diminished significantly in the group of fish transferred to 16℃ over 12-48 h, but increased significantly in the group of fish at 36℃ over 48 and 96 h. Alternative complement pathway (ACH50) decreased significantly when transferred to 16℃ after 12 h, but increased significantly when transferred to 36℃ after 24 h. In a challenging test, 30 tilapia reared at 25℃ were injected intraperitoneally with Streptococcus iniae at a dose of 2x107 cfu/fish, and then reared onward at water temperatures of 15, 25 (control), and 36℃. Over 12-96 h, the cumulative mortality of S. iniae-injected fish held in 16 and 36℃ was significantly higher than that of injected-fish held in 25℃ In conclusion, transfer of tilapia from 25℃ to low temperature (16℃) after 12 h, and transfer of fish from 25℃ to high temperature (35℃) reduced their immune capability. Furthermore, tilapia under temperature stress at 16 and 36℃ from 25℃ decreased its resistance against S. iniae

        In the present study, the genetic diversity of 69 Ganoderma species from various regions was determined by different molecular markers, including the internal transcribed spacer (ITS) rRNA, a partial β-tubulin gene, and mitochondrial small-subunit ribosomal (SSU) rDNA gene as well as randomly amplified polymorphic DNA (RAPD) analysis. The size of the ITS rDNA regions and mitochondrial SSU rDNA gene from different Ganoderma species varied from 625 to 673 bp and 656 to 2,040bp, respectively, and those of the partial β-tubulin gene sequence were 419 bp. Phylogenetic analysis based on the ITS region, the partial β-tubulin gene, and the mitochondrial SSU rDNA gene reveal that Ganoderma species are largely divided into two groups. Interestingly, most of the Ganoderma lucidum strains could be classified into 1 group, while other Ganoderma species divided into several groups (4 or 5 groups) in phylogenetic tree. One fragment unique to G. lucidum was selected from the RAPD profile and then sequenced. One primer pair (designated as KGS-F and KGS-R) based on this specific fragment was designed to amplify a 559 bp DNA fragment within the sequenced region. A single band with the expected size of 559 bp was observed from G. lucidum, except for G. lucidum strains from China, Canada, and Taiwan. This specific marker for G. lucidum from RAPD analysis, also supported by the phylogenetic analysis of the ITS, partial β-tubulin gene, and mitochondrial SSU rDNA sequences, will be useful for the PCR-based identification of G. lucidum in research applications as well as in the market.

GU901158.1) encodes 11 cadherin repeats and a transmembrane domain. The PxCad1 gene was expressed in all developmental stage specifically in gut tissue by RT-PCR analysis. Expression of PxCad1 gene was suppressed by feeding of its specific dsRNA PxCad1 in 4th instar larval stage. The suppression of PxCad1 expression did not significantly feeding of its specific dsRNA PxCad1 in 4th instar larval stage. The suppression of PxCad1 expression did not significantly influence on pupal and adult development of P. xylostella. However, the larval treated with dsRNA PeCad1 (150 ng/larva) significantly reduced susceptibility to B. thuringiensis Cry1Ac (4.83 μg/ml). By contrast, the dsRNA PxCad1 -treated larvae did not show any change in susceptibility to B. thuringiensis Cry1Ca (0.24 μg/ml). These results suggest that PxCad1 is a specific receptor of Cry1Ac toxin from B. thuringiensis in P. xylostella.

        The oriental fruit moth (Grapholita molesta) and the plum fruit moth (G. dimorpha) share the same major sex pheromone components, Z8-dodecenyl acetate (Z8-12Ac) and E8-dodecenyl acetate (E8-12Ac) with different ratio. However, these two congener male species were cross-attracted to the counter sex pheromone traps. For development of the specific monitoring lures, the minor sex pheromone components were added to the major components. G. molesta females emit two minor components of Z8-12OH and 12OH and G. dimorpha females emit four minor of 12Ac, 14Ac, Z8-14Ac, and E8-14Ac. For a specific monitoring lure of G. molesta, only Z8-12Ac major component attracted only G. molesta males, but did not any G. dimorpha. For a specific monitoring lure of G. dimorpha, the addition of Z8-14Ac to the major component (Z8-12Ac:E8-12Ac = 85:15) attracted G. dimorpha males with less than 5% G. molesta males. Other with components (12Ac, 14Ac, and E8-14Ac) was not effective in both trapping efficiency and selectivity.
