The present study was to survey the site environment, vegetation structure and soil characteristics in the wild habitats of Acer tegmentosum Maxim. and offers basic information for habitats conservation and restoration. Most of the wild habitats were located at altitudes between 605~1,413m with inclinations ranged as 8~30°. The bare rock rate were 8~50%. The vegetation structure by the PC-ORD based on the Two Way Cluster Analysis were divided into three groups CommunityⅠ(Acer tegmentosum – Quercus mongolica), CommunityⅡ(Acer tegmentosum - Carpinus cordata), CommunityⅢ(Quercus mongolica – Tilia amurensis). The species diversity(H') was highest in CommunityⅡ as 1.474, CommunityⅠwas 1.471, Community Ⅲ was 1.219. The soil textures were Clay loam, The average soil pH was 4.8, Soil organic matter was 15.15% and available phosphorus was 2.33ppm. Ordination analysis result by soil characteristics, community, characteristic species showed that the greatest effect factors were slope, altitude, tree and shrub’s cover rate, organic matter, total-nitrogen, calcium, magnesium. Correlation analysis between environment factor result showed that O.M. - (T.N., K+, Mg2+, CEC, EC), T.N. - (K+, Mg2+, CEC, EC) were positive correlations.
산겨릅나무(Acer tegmentosum Max)는 단풍나무과의 약용식물로 기내 대량증식 및 발근에 적합한 최적 배지와 식물생장조절물질의 농도를 구명하고, 순화조건을 검증하여 대량생산조건을 확립하기 위해 수행되었다. 산겨릅나무의 종자 소독 후 1.0mg l-1 GA3가 첨가된 MS배지에서 발아시켜 기내식물체를 확보하였고, 생장에 적합한 최적배지 구명을 위해 7종류의 다양한 식물배지에서 4주간 배양한 결과, WPM 배지가 가장 효과적이었다. 액아배양을 통한 대량증식에 최적 조건을 구명하기 위하여 4종의 cytokinin의 다양한 농도별 처리를 통해서 8주 동안 배양한 결과, 1.0mg l-1 Zeatin이 첨가된 배지에서 평균 3.3개의 다경줄기가 유도되었다. 기내발근에는 0.5mg l-1 2.4.-D가 단독 첨가된 배지에서 뿌리 수가 높았고, 다신초 개체는 기외의 토양으로 이식하여 환경적응을 유도한 후 배양묘 대량생산 조건을 확립하였다.
Mushroom is cultivated as one of the major economical crops in many areas in Korea. The total production has steadily increased from approximately 186,400 M/T in 2007 to 190,104 M/T in 2009. The oyster mushroom, Pleurotus ostreatus, showed the 2th production to 46,598 M/T in 2011. This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Mycelial growth of Pleurotus ostreatus showed the highest growth at the media of 10% Acer tegmentosum and was decreased to increase of Acer tegmentosum addition ratio. But mushroom mycelial growth at Acer tegmentosum media was slower than that of the control. Total nitrogen and carbon source of Acer tegmentosum was 0.19% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrates mixed with Acer tegmentosum were 2.7∼3.0 and 4.8∼5.0, respectively. The contents of P2O5 and K2O were decreased by increasing Acer tegmentosum but the contents of CaO, MgO and Na2O were no significant difference. The contents of Fe and Mn were decreased according to increasing of Acer tegmentosum, but the contents of Zn and Mn were no significant difference. Yields of fruiting body and diameter and thick of pileus were the highest at 10% Acer tegmentosum. The L value of pileus was the highest at the 30% Acer tegmentosum during mushroom harvest, but there was no significant difference in the a-value and the b-value. The contents of P2O5,, CaO, MgO and Na2O of fruiting body were no significant difference, but contents of Cu was decreased and Fe was increased according to increasing of Acer tegmentosum.
This study was conducted in order to investigate the effects of Acer tegmentosum Maxim. extract (ATE) in prevention of liver injury. After oral administration of ATE to SD rats, liver injuries were induced by treatment with CCl4, galactosamine, or ethionine. Aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglyceride, and cholesterols in blood were used as indicators of liver damage. When acute hepatitis was induced by CCl4 or galactosamine, ATE-fed rats showed a lower level of AST and ALT in plasma than ATE-unfed rats. In the case of ethionine-induced fatty liver, triglyceride levels in plasma were reduced in ATE-fed rats, compared with ATE-unfed rats. These results indicate that Acer tegmentosum Maxim. extract protected against hepatic failure.
In this study, we were performed to elucidate the antioxidant and anticancer activity by leaves extracts from Acer tegmentosum (AT-L). In DPPH, ABTS radical scavenging activity, the AT-L revealed the high scavenging activity. Especially, the AT-L measured the highest ABTS radical scavenging activity, which is higher than ascorbic acid. The types of human cancer cells for evaluating the anticancer activity were colorectal cancer (SW480), prostate cancer (PC-3), breast cancer (MCF-7), pancreatic cancer (AsPC-1), lung cancer (A549) and liver cancer (HepG2). Human cancer cell viability was measured using MTT assay. Treatment of the AT-L decreased the cell viability and induced apoptosis in SW480 cells. These results suggest that extracts of the AT-L can be used as supplementary material for developing the natural antioxidant and anticancer drug for human cancer cells.
The objective of this study was to investigate anticytotoxic and antioxidatative capacities of ethanol extracts from Acer tegmentosum Maxim (A. tegmentosum) stem in vitro. The extract at concentration of 200 ug/mL inhibited 10 and 20 ug/mL arsenic trioxide-induced cytotoxicity of HepG2 cells by 79.3 and 57.5%, respectively. The extract at concentration of 200 ug/mL inhibited 0.2 and 0.5 mM t-BHP-induced cytotoxicity of HepG2 cells by 66.3 and 35.7%, respectively. Antioxidative effects of the extract were examined via measurement of ABTS, superoxide, and peroxyl radical scavenging activities. ABTS radical scavenging activity of the extract was higher than that of α-tocopherol. Superoxide scavenging activity of the extract was higher than that of catechin. Oxygen radical absorbance capacity of the extract was higher than that of ascorbic acid. Cupric reducing antioxidant capacity of the extract was higher than that of α-tocopherol. The extract at concentrations of 100 and 500 μg/mL inhibited 10 mM t-BHP-induced lipid peroxidation of HepG2 cells by 38.2 and 80.7%, respectively. The extract prevented supercoiled DNA strand breakage induced by hydroxyl or peroxyl radical. Total phenolic and flavonoid contents of the extract at concentration of 100 μg/mL were 71.3 nmol/mL gallic acid and 18.8 nmol/mL catechin equivalents, respectively. Thus, strong cytoprotective and antioxidant effects of A. tegmentosum stem extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in polyphenolic contents.
Here we report the protective activity of cultured Acer tegmentosum cell extract against liver damage in rat intentionally instigated by D-galactosamine. Local fat degeneration and infiltration of inflammatory cells were significantly decreased in cultured A. tegmentosum cell extract administered rat. In addition, acutely increased AST, ALT, LDH, ALP activities and lipid peroxidation and lipid content by liver damage were recovered in experimental rat administrated with A. tegmentosum extract. These results showed that cultured A. tegmentosum cell extract has a role in blood enzyme activation and lipid content restoration within damaged rat liver tissues. Moreover expression rate of TNF-α which accelerates inflammation and induces tissue damage and necrosis was significantly decreased. Also activities of antioxidant enzymes were more effectively upregulated comparing to those of the control group induced hepatotoxicity. All data that cultured A. tegmentosum cell extract has a preventive role against liver damages such as inflammation, tissue necrosis in rats by improving activities of blood enzymes, antioxidant enzymes and modulating expression of inflammation factor, suggest that cultured Acer tegmentosum cell extract is an effective medicinal resource for restoration of hepatotoxicity.
본 연구에서는 소종과 외상출혈 치료에 사용되며, 주로 간암, 간경화 등의 간 질환 치료제로 사용되어 온 산겨릅나무 추출물에서 식품 및 음료 등과 같은 기능성 식품으로서의 다양한 산업적 응용분야에 널리 응용될 수 있는 기초연구의 일환으로 산겨릅나무 추출물을 이용하여 항산화 활성, 항지질과 산화 활성, 항미생물 활성, 항보체 활성을 실험하였으며, 또한 유용한 생리활성 성분을 분리, 동정하고자 하였다. 그 결과 산겨릅나무 추출물은 DPPH free radical 소거법에 의한 항산화 활성 실험에서는 ehtyl acetate 분획 (RC50= 3.15 μg/ml), butanol 분획 (RC50= 5.17 μg/ml)에서 강한 항산화 활성을 보였다. 또한 분리된 2개의 화합물도 대조군인 α-tocopherol 이나 BHA보다 유사하거나 강한 항산화 활성을 나타냈다. linoleic acid에 대한 항지질과산화 활성 실험은 15일 째에 물층을 제외한 추출물, 분획물 및 화합물에서 높은 활성을 나타냈으며 특히 화합물 2 (85%)는 항산화 물질로 알려진 catechin (85%)과 유사한 활성을 나타내었다. 박테리아에 대한 향균실험은 Staphylococus aureus 에서만 활성을 보이지 않았을 뿐 다른 피검균에서는 높은 활성을 보였다. 하지만 대부분의 피검균에 대하여 강한 활성을 보인 ethyl acetate 분획에서 분리된 화합물 1, 2는 모든 피검균에 대해 활성을 보이지 않았다. 다만 fungal strain인 Candida albicands에 대해 각각 250 μg/ml. 500 μg/ml의 생육 억제 농도를 나타내었다. 보체계 활성화능을 측정한 결과 물 층 24%를 제외한 다른 분획물은 10% 이하의 낮은 억제효과를 보이거나 활성이 나타나지 않았다. 산겨릅나무 ethyl acetate 분획을 silica gel이 충전된 grass open column에 넣은 후, toluene, acetone, methanol의 혼합용매로 순차용출 (stepwise)시킨 후, ODS와 silica gel을 이용하여 column chromatography를 실시함으로서 2종의 단일 물질을 얻었다. 이 화합물은 lH- 및 13C-NMR, IR, UV, EI/FAB-Mass spectrum을 이용하여 기기분석한 결과 compound 1은 catechin으로, compound 2는 σ-Hydroxyphenethyl alcohol 1-O-β-D-(6'-O-galloyl)-glucopyranoside로 동정되었으며, 특히 compound 2의 경우 산겨릅나무에서 처음 분리되었다. 위 결과 산겨릅나무 추출물은 항산화 활성, 항지질과산화 활성, 항미생물 활성이 우수하였다. 또한 산겨릅나무 추출물을 이용한다면 식품 및 음료 등과 같은 기능성 식품으로서의 다양한 산업적 응용분야에 널리 응용될 수 있으리라 사례된다.
