뽕잎 (Morus alba L.)을 노루궁뎅이 버섯균사체(Hericium erinaceum)로 발효시켜 열수 추출한 발효물(MA-HE)이 항산화 효소인 Cu,Zn-superoxide dismutase (SOD)와 ceruloplasmin (CP)의 산화적 변형에 미치는 영향을 관찰하였다. 세포질에서 중요한 항산화 활성을 나타내는 Cu,Zn-SOD와 CP는 모두 구리이온을 포함하는 단백질이다. MA-HE은 peroxyl radical에 의해 변형되는 Cu,Zn-SOD와 CP를 모두 보호하였다. MA-HE은 peroxyl radical에 의한 Cu,Zn-SOD의 단백질 fragmentation과 효소의 inactivation을 모두 효과적으로 억제하였다. 또한, MA-HE는 CP의 fragmentation과 inactivation도 효과적으로 억제하였다. MA-HE가 peroxyl radical을 소거할 수 있는 기능을 가지고 있는 지를 확인하기 위해 thiobarbituric acid를 이용한 활성 측정법으로 관찰한 결과, MA-HE는 농도가 증가함에 따라 비례적으로 peroxyl radical 소거 활성이 높아지는 것으로 관찰되었다. MA-HE는 100 μg/mL에 서 44.03% 소거활성을 나타냈다. 따라서, MA-HE는 peroxyl radical을 효과적으로 소거함으로서 세포 내의 항산화 효소를 산화적 손상으로부터 보호할 것으로 생각된다. 이와 같은 결과는 MA-HE가 세포 내에서 발생하는 활성산소종을 효과적으로 소거하여 산화적 스트레스에 의해 야기되는 세포 독성에 대한 보호 작용을 할 것으로 생각된다.
In this study, we examined antioxidative effects and the anti-adipogenesis effect of different parts of Cudrania tricuspidata (C), and Morus alba (M). Total polyphenol contents were highest in M-root (34.56±0.045 mg GAE/g), and there was no significant difference, between C-root and M-leaf. Total flavonoid contents of C-root were highest (23.07±0.004 mg QE/g). To examine antioxidant activities of C and M extracts, DPPH and ABTS radical scavenging activity, and FRAP assay, was used. Results show that antioxidant activities of C and M extracts increased, in a dose-dependent manner. Adipocytes are generated by preadipocyte differentiation, during adipogenesis. Matured adipocytes accumulate in abnormal and cause obesity. We investigated effects of leaf and root extracts of C and M, on lipid accumulation, in 3T3-L1 adipocytes. Changes in cell morphology, and degrees of lipid accumulation in adipocytes, were evaluated by Oil Red O staining. Root extracts of C and M, reduced lipid content in a dose-dependent manner. Therefore, root extracts of C and M, may be good candidates for managing obesity.
본 연구는 연잎, 상엽, 건조 무 혼합 추출물의 wogonin, quercetin, quercetin-3-O-glucuronide의 함량 분석 및 해당 표준물질들의 동시 분석법의 개발 및 검증을 실시하였다. 분석법은 특이성, 직선성, 정확성, 정밀성, 검출한계 및 정량한계를 통하여 분석법의 신뢰성을 검증했다. HPLC를 이용한 분석방법에서 표준용액의 머무름 시간과 연잎, 상엽, 건조 무 혼합 추출물 중 quercetin-3-O-glucuronide의 머무름 시간이 일치하였으며 동일한 spectrum을 나타내는 것을 확인해 분석법의 특이성을 검증하였다. Wogonin, quercetin 및 quercetin-3-O-glucuronide의 검량선은 상관계수 값이 wogonin, quercetin 및 quercetin-3-O-glucuronide 모두 0.9999로 우수한 직선성을 보여 분석에 적합함을 알 수 있었고 정밀성과 정확성은 농도를 아는 시료에 인위적으로 저농도, 중농도, 고농도의 표준물질을 첨가해 계산하였다. Wogonin, quercetin, quercetin-3-O-glucuronide의 정밀성은 일간 정밀성, 일내 정밀성으로 확인했으며 세 가지 성분들의 일간 정밀성은 각각 0.76~1.24%, 0.90~1.40%, 0.74~1.87% 수준으로 확인되었고, 일내 정밀성은 0.12~1.06%, 0.28~1.12%, 0.52~0.92%로 5% 이하의 우수한 정밀성을 보였다. 정확성의 측정 결과 wogonin, quercetin 및 quercetin-3-O-glucuronide의 일간 정확성은 99.99~107.97%, 99.96~115.88%, 99.73~111.91%를 나타냈고 일내 정확성은 100.36~108.88%, 99.77~114.81%, 99.91~112.44%로 우수한 수준의 정확성을 보였다. Wogonin, quercetin 및 quercetin-3-O-glucuronide의 검출한계는 각각 0.16 μg/mL, 0.09 μg/mL, 0.15 μg/mL로 측정됐으며 정량한계는 각각 0.48 μg/mL, 0.26 μg/mL, 0.45 μg/mL로 나타나 저농도에서도 검출이 가능함을 확인할 수 있었다. 분석법 검증결과, 확립된 분석법은 특이성, 직선성, 정밀성, 정확성, 검출한계 및 정량한계가 모두 우수한 분석법임이 검증됐으며, 검증된 분석법을 이용해 연잎, 상엽, 건조 무 혼합 추출물의 wogonin, quercein, quercetin-3-O-glucuronide의 함량을 분석한 결과 wogonin과 quercetin은 검출이 되지 않았지만 quercetin-3-O-glucuronide는 함량을 가지고 있는 것으로 확인됐다. 본 연구 결과 에탄올로 추출한 11번(EM11) 샘플에서 quercetin-3-O-glucuronide의 함량이 가장 높게 나왔으며 열수와 에탄올로 추출한 샘플들이 함유하고 있는 quercetin-3-Oglucuronide의 함량은 불검출~50.95 mg/dry weight g 수준으로 함유되어 있는 결과를 보였다.
Mulberry fruits were semi-dried using hot air (60-100℃) or cool air (20-40℃), and the effects of the dryingtemperature and processing time on the quality of the final driedmulberry fruits were investigated. Response surface methodologywas employed to establish a statistical model and predict theconditions resulting in minimal loss of the total phenolic content(TPC) and ascorbic acid. Thus, using overlapped contour plots,the optimal conditions for producing semi-dried mulberry fruits,which reduced the moisture residue to 45% and minimized thenutrient losses of TPC and ascorbic acid, were determined for thehot-air process (60.7℃ for 5.4h) and cool-air process (34.8℃ for23.3h). Plus, a higher drying temperature was found to lead to afaster loss of moisture and ascorbic acid, while the TPC wassignificantly decreased in the cool-air dried mulberry fruits due tothe higher activity of polyphenol oxidase between 30 and 40℃.
This study was conducted to investigate the quality characteristics of the natural dishwashing liquid added with the mixture extracts of Curcuma Longa L. (CL), Morus alba (MA) and Ecklonia cava (EC) at various concentrations. The pH and lightness of the natural dishwashing liquid were reduced by adding the mixture extracts, while the turbidity was increased. The natural dish washing liquid added with mixture extracts was shown to display strong antimicrobial activities against L. monocytogenes compared to that of control. Also, it revealed that antioxidant activity was increased depending on concentrations. However, natural dishwashing liquid added with mixture extracts showed the low detergency efficiency. In sensory evaluation, the natural dishwashing liquid containing CL 0.5%, MA 0.25% and EC 0.25% was preferred than the control and it showed negative result in skin patch test. These results suggest that the addition of CL 0.5%, MA 0.25% and EC 0.25% positively improved the qualities characteristics in the natural dish washing liquid.
The purpose of this study was to investigate the effect of dietary Morus alba L.(Bong-ip, B), Glycyrrhizae glabra(Gam-chei, C), Pinus densiflora(Sol-lp, S) and Angelica gigas(Dang-gi, D)powder on serum composition in rats(Sprague-Dawley male rats, 100-110g). Serum TG(triglyceride, p〈0.01), total cholesterol, glucose, total protein, albumin, GGT(γ-glutamyl transferase, p〈0.05) were significantly increased D group than that of nomal and other groups, but UA(uric acid, p〈0.05) was significantly decreased, and C group(p〈0.05) was significantly increased. but C group of urine(p〈0.05) was significantly decreased. Also, B and S groups(p〈0.05) of BUN(blood urea nitrogen), S group(p〈0.05) of ALP(alkaline phosphokinase, Band C(p〈0.05) of CPK(creatinine phosphokinsae, p〈0.05) were significantly increased. B, S and C groups were better than D group for lipid metabolism, and pretection to liver. Also, B and C groups of glucose were same as normal diet, so Morus alba L. was good food for lipid metabolism and hypoglycemic effect.

Methods and Results : Carotenoids are secondary metabolites found in most of the higher plants. In the current study, the levels of carotenoids were much higher in tetraploid of M. alba L. ‘Cheongil’ than diploid of M. alba L. ‘Cheongil’. Particularly, the levels of lutein, 13z-β-carotene, α-carotene, E-β-carotene, and 9Z-β-carotene were 2.51, 5.53, 7.68, 4.43, and 4,46 times higher in tetraploid of M. alba L. ‘Cheongil’, respectively. Additionally, hierarchical cluster analysis (HCA) was carried out using Pearson’s correlation results in order to determine the relationships between metabolites of diploid and tetraploid in Morus alba L. ‘Cheongil’. The results of HCA revealed the degree of correlation among 42 metabolites, which were identified and quantitated by GC-TOFMS. Among TCA organic acids, succinic acid was positively correlated with fumaric acid (r = 0.8250, p < 0.0001) and malic acid (r = 0.9436, p < 0.0001). Among carbohydrates, fructose was positively correlated with glucose (r = 0.9398, p < 0.0001) and mannose (r = 0.9394, p < 0.0001).

Background: Inflammation plays an important role in various diseases, including ulcerative colitis, Behcet's disease, and rheumatoid arthritis. In this study, we investigated the anti-inflammatory effects of Morus alba L. extracts obtained using different extraction methods (water extraction or high temperature extraction) on RAW264.7 cells.
Methods and Results: Extracts from the central part (including the heartwood, sapwood, cambiun, and phloem) and bark (including the periderm and cortex) of Morus alba L. were obtained using either water or high temperature extraction. The following extract were obtained: MA1, water extract from the central part of Morus alba L., MA2, high temperature extract from the central part of Morus alba L., MA3, water extract from the bark of Morus alba L., and MA4, high temperature extract from the bark of Morus alba L. None of these extracts was observed to be cytotoxic to RAW264.7 cells. The MA2 extract reduced the production of LPS-induced NO (nitric oxide), PGE2 (prostaglandin E2), TNF-α, IL-6, and IL-1β production in LPS-stimulated RAW264.7 cells.
Conclusions: These results indicated that the inflammatory response was moderated by MA2. Treatment with MA2 could be used as a natural medicine for treating diseases involving inflammation. However, further experiments are required to determine how the high temperature extraction method alters the active ingredients in the extract and influences the anti-inflammatory effects of Morus alba L..
Background : 1-Deoxynojirimycin (DNJ) is a representative compound of the antidiabetic constituent in mulberry leaves (Morus alba L.). The hot water extracts of mulberry leaves were fermented with lactic acid bacteria in order to analysis the changes of the DNJ contents and α-glucosidase inhibition. Methods and Results : The mulberry leaves were extracted with hot water (121℃, 3 hr). The extracts were fermented with nine strains of lactic acid bacteria such as Lactobacillus acidophilus, in order to increase the contents of DNJ and α-glucosidase inhibition. DNJ contents in the fermented extracts were determinated by HPLC analysis. The α-glucosidase inhibition was mesured by comparing dose-inhibition curves of α-glucosidase and IC50 value. The DNJ contents after fermentation have increased in the almost fermented extracts. Especially, DNJ of the extracts fermented with L. acidophilus was increased from 8.38 ㎍ ㎖ -1 to 21.77 ㎍ ㎖-1. IC50 values of the α-glucosidase inhibition were shown to be decreased in the fermented extracts. The extracts fermented with L. casei KCTC 3109 was determined at 290.04 ㎍ ㎖-1, resulting it is lower about 140 ㎍ ㎖-1 than 429.76 ㎍ ㎖-1 of the control. Conclusion : From the above results, we may suggest that the lactic acid fermentation of mulberry leaves extracts can more enhance the hypoglycemic activities such as DNJ contents.
Background : The hypoglycemic effects of mulberry leaves extract were evaluated by comparing the abilities on glucose uptake in C2C12 myotubes and 3T3-L1 adipocyte. Glucose uptake of the extracts were identified to be enhanced by bio-conversion using cellulolytic enzyme like Viscozyme. Methods and Results : The mulberry (Morus alba L) leaves were extracted with 30% ethanol or hot water. The hypoglycemic compounds such as Moracin C and, Quercetin and 1-Deoxynojirimycin were identified from the extracts of mulberry leaves. The extracts were fermented using kinds of celluolytic enzymes, which were vicozyme, pectinase, β-glucosidase and xylanase, in order to increase the contents of hypoglycemic constituents in the extracts. The hypoglycemic effects of the fermented extracts were evaluated by comparing the abilities on glucose uptake in C2C12 myotubes and 3T3-L1 adipocyte. The extracts of mulberry leaves fermented with only Viscozyme were identified to increase glucose uptake in C2C12 myotubes and 3T3-L1 adipocyte by supplement of the concentration of 10 μM extracts, compared to insulin as control. However, bio-conversion effects by other enzymes were not shown in the treatments, suggesting hypoglycemic constituents in the extracts of mulberry leaves can be conversed to more active compounds by cellulolytic enzyme treatment like viscozyme. Conclusion : From the above results, we may suggest that the hypoglycemic constituents in mulberry leaves extracts can be conversed to more active compounds by cellulolytic enzymes.
Background : Recently, many studies to seek for medicinal herb compounds from natural products have been attempted capable of being free from harmful side effects and reducing the economic burden. This study was conducted to investigate whether Morus alba L.(MAL) water extracts has a biological safety, and an inhibitory potential against the mutagenicity induced by cigarette smoke condensates(CSC). Methods and Results : MAL was extracted with 70% ethanol and the yield of the extract was 35.1%. The 70% ethanol MAL extract was fractionated sequentially by diethylether, chloroform, dichloromethane, and water, respectively. Crude MAL 70% ethanol extract itself and its solvent fractions did not show any mutagenic effect up to 1 mg/plate toward Salmonella typhimurium TA 98 with or without the microsomal S-9 mixture of liver metabolic activations. On the other hand, the crude MAL extract showed an inhibitory activity against the mutagenicity of CSC with S-9 mixture. Diethyl ether layer among five solvent fractions showed the highest inhibitory activity at the same dose. The inhibitory activity of diethyl ether layer was also increased dose-dependently and the inhibitory rate was about 97.1% at the concentration of 1 mg/plate. Conclusion : In this study, we concluded that all of the crude MAL 70% ethanol extract and its solvent layers are potentially safe for mutagenicity and the diethyl ether layer among the crude MAL 70% ethanol extract has an inhibitory effect against the mutagenicity of CSC.
본 연구에서는 상백피의 소화효소 저해활성과 3T3-L1 전지방세포의 분화 억제능을 기반으로 항비만 효능소재로 서의 활용가능성을 평가하였다. 상백피의 에탄올 추출물 (MRE)은 α-amylase와 α-glucosidase, pancreatic lipase를 활 용한 소화효소 저해활성 평가 실험에서 각각 7.86±0.36, 0.12±0.03, 7.93±0.11 mg/mL 의 IC50 값을 보이며 우수한 억제 활성을 나타냈다. 또한 3T3-L1 전지방세포를 활용한 세포분화억제효능실험에서 MRE 처리군의 세포내 지방 축 적율은 농도 의존적으로 감소되었다. 상백피의 항비만 작 용 기전을 구명하기 위하여 adipogenesis 및 lipogenesis와 관련된 유전자 발현양상을 분석한 결과, 상백피 추출물 처 리군에서는 생체내 지방대사 조절에 중요한 역할을 하는 FAS와 ACC 뿐 아니라 adipogenesis와 lipogenesis와 관련된 주요 전사요소인 PPARγ와 C/EBPα, SREBP-1c의 유전자 발현이 현저하게 억제되었다. qRT-PCR 분석 결과, 상백피 추출물의 anti-adipogenesis 효능은 전사단계에서의 관련 유 전자 발현억제에 기인한다고 판단되었다. 본 실험결과 상 백피 추출물은 전지방세포의 분화와 세포내 지질합성을 저해하고 비만과 관련 된 소화효소에 대한 저해활성을 나타 내었다. 이러한 결과를 기반으로 상백피의 비만 예방 소재 로서의 잠재적인 가능성을 확인하였다.
본 연구는 오디를 이용한 고부가가치의 기능성와인을 개발하기 위한 연구의 일환으로 먼저 4가지 베리류과실(오 디, 블루베리, 딸기, 복분자)와인의 항당뇨, 항노화 및 항산 화활성을 α-glucosidase, tyrosinase 및 DPPH radical를 사용 하여 측정한 후 α-glucosidase 저해활성이 가장 강한 오디와 인으로부터 저해제를 분리 및 동정하였다. 또한, 3 가지 뽕나무 품종(익수뽕, 대성뽕, 청일뽕) 오디를 이용하여 오디 와인을 제조하여 품종과 발효에 따른 α-glucosidase 저해제 의 함량 변화를 측정한 결과는 다음과 같다. 4가지 베리류과실 와인의 항산화, 항당뇨 및 항노화 활성 을 측정한 결과, 오디와인이 가장 높은 α-glucosidase 저해활 성을(69.37%) 나타내었으며, DPPH 라디컬 소거능은 블루 베리(64.57%) 및 오디와인(62.48%)이 유사하게 높았으며, 딸기와인이 가장 높은 tyrosinase 저해활성(59.63%)을 나타 내었다. 이 중 α-glucosidase 저해활성이 가장 높은 오디와인 으로부터 효소저해제를 분리 및 동정하기 위해 오디와인을 먼저 감압농축하여 에틸아세테이트로 분획한 다음 Diaion HP-20, ODS-A 및 Sephadex LH-20 column chromatography 를 순차적으로 실시하여 4가지 분획을 얻었으며, 이 중 α -glucosidase 저해활성이 가장 강한 Fr. 4로부터 UV 및 NMR 분석을 통해 quercetin를 동정하였다. 마지막으로 뽕나무 품종 및 발효에 따른 오디와인의 α-glucosidase 저해제의 함량을 측정한 결과, 대성뽕 오디와인의 quercetin 함량 (16.83 ppm)이 가장 높았으나, 익수뽕 및 청일뽕 오디와인 의 quercetin 함량은 각각 14.85 및 8.92 ppm으로 낮았다. 그리고 발효 및 숙성에 따라 오디와인의 quercetin 함량은 크게 감소하였다. 이상의 연구 결과로부터 4가지 베리류 과실와인 중 오디와인이 가장 높은 α-glucosidase 저해활성을 나타내었으며, 주된 저해물질은 quercetin임을 기기분석을 통해 확인할 수 있었기에 향후 오디와인은 당뇨예방용 기능성와인으로서 크게 각광을 받을 것으로 사료된다.
To determine the appropriate harvest maturity of mulberry to prolong the freshness during distribution, mulberries were classified into three groups (80%, 90%, and 100%) based on their degree of maturity and then compared for their firmness, color, anthocyanin, soluble solids content, pH, titratable acidity, viable cell, mold and sensory quality. They were then investigated for quality changes by maturity under different distribution temperature at 20℃, 10℃ and 0℃. Immediately after harvest, the 80% matured mulberries were shown to be the firmest and 1.74 and 2.64 times firmer than 90% and 100% matured mulberries respectively. The more mature the mulberries were the lower the “a” value was while anthocyanin content was higher. The soluble solids content and pH of mulberries increased as they matured. Less mature the mulberries were the higher titratable acidity was and lower microbial levels in mulberries were. In terms of sensory quality, color of the 100% matured mulberries was the highest but the 90% matured mulberries were most preferred based on other sensory indexes. In comparison to storability of mulberries by maturity, although the firmness of the 80% matured mulberry decreased rapidly, they were the firmest during distribution periods. The level of mold of the 100% matured mulberries was higher than others. Moreover, the more mature mulberries were the quicker they deteriorated. According to quality indexes, depending on maturity and sensory evaluation of mulberry distributed at different temperatures, the storability of the 80 and 90% matured mulberries were higher than that of the 100% matured mulberries. In conclusion, judging by their sensory quality, the 90% matured mulberries were more appropriate for harvesting than the 80% matured mulberries.
