Background : Rehmannia glutinosa L. that is a perennial plant belonging to the family Scrophulariaceae is one of the most popular local income crops. Recently, the cultivation area of Chungnam (2017) accounts for 50.5% (106 ha) of the whole country. In July and August, which is the off-season at the highest price of R, glutinosa L. This experiment was carried out to develop early harvest cultivation technology for the improve farm income.
Methods and Results : In order to investigate the growth characteristic and yield of R, glutinosa L. by using the rhizome of 'Tokang' were transplanted four times at intervals of 10 days from mid-March to mid-April 2017 and 2018. Growth characteristics of the above and under ground part were investigated by 3 repetitions for 10 ea harvested in mid-July. Regression analysis was performed on the relationship between the planting time and fresh root weight. The highest emergence rate was 88.7% in the mid-March, and the highest above ground part growth was observed in the plant height, leaf number, leaf length, leaf width, and fresh leaves weight. The number of days to flowering tended to be shortened to 60, 53, 52, and 49 days from the mid-March to the mid-April. The growth of underground part was also highest in mid-March and the total root weight per 10 a was also 1,127.4 ㎏ in mid-March. The faster the planting time, the higher the yield. The results of the linear regression on the roots weight according to the planting time showed that there was a negative correlation relation between the planting time, the marketable roots weight, rhizome weight and total roots weight.
Conclusion : The total roots weight improved by 80% in the mid-March than mid-April. Therefore, mid-March is considered optimal planting time for off-season production of R. glutinosa L.
Background : In the year 2015, the area under rhizome of Rehmannia glutinosa in Chungnam province increased to 65% (105 ha) of domestic total cutivation area and is recognized as income crop. Since proper strorage method of rhizome has not been established, rhizome decay rate is so high during storage. This experiment was carried out to establish proper strorage method for stable production of rhizome of Rehmannia glutinosa Methods and Results : Korea jiwhang was used for this experiment and rhizome was cured under vinyl house and room temperature for 1, 2, 3 days respectively. Rhizome was stored for 140 days at –2 - 0℃ after curing, and rhizome quality and saprogen were investigated after storage. Trimming loss was 11.3 - 21.9%, higher in longer curig time and rice hull packing. Moisture content was 70.0 - 75.1%, higher in shorter curing time and no rice hull packing. Curing efficacy was better in vinyl house than room temperature, decay rate by weight and number of rhizome was lowest in 48hrs curing and no hull packing as 0.5% and 2.0% respectively. Identified saprogen during storage was Fusarium oxysporum, Fusarium tricinctum, Penicillium verrucosum, Mucor racemosus. Conclusion : Curing efficacy was better in 48hrs treatment under vinyl house with lowest decay rate and rice hull packing increased decay rate considerd due to saprogen existing in rice hull. 4 saprogens including Fusarium oxysporum were identified during storage of rhizome.

High quality jiwhang GAP production and improvement of the distribution system including magnus customers, producer association fosterage and conventional market Vitalization were the reason of rapid cultivation area increase of Rehmannia glutinosa L. in Geumsan. This study was performed to set up the standard Rhizome size for planting in Rehmannia glutinosa L. Materials and Methods ○ Experiment variety : Togang ○ Treatment(rhizome length) : ① 1cm ② 2cm ③ 3cm ④ 4cm ⑤ 6cm(control) ○ Planting date : May 6, 2015 ○ Planting distance : 30×15cm ○ Experiment place : Ginseng and Medicinal Plant Research Institute (Geumsan-gun, Chungcheongnam-do) Conclusion : In the aboveground growth was no significant difference among treatments. In the root characteristics, there was no significant difference among treatments. Take a 1∼2cm rhizome planting saved rhizome amount and planting time to 73.8%∼64.3% and 68.4%∼ 57.8% respectively than 6cm(control).
Background : Generally, price of Jiwhang(Rehmannia glutinosa L.) is highest in August. In this study, Economic Evaluation and Productivity of 9 Jiwhang varieties w ere analyzed according to harvesting tim es of August and October. Materials and Methods ○ Experiment variety : Jiwhang 1, Korea jiwhang, Daegyung, Gogang, Togang, Dagang, Wongang, Hwanggang and Yeongang ○ Treatment : ①Initial-August harvest ②Mid-August harvest ③Ended-August harvest ④Ended-October harvest ○ Planting date : April 17, 2015 ○ Planting distance : 30×15cm ○ Experiment place : Ginseng and Medicinal Plant Research Institute (Geumsan-gun, Chungcheongnam-do) Conclusion : The higher roots yield at August harvest was in Gogang, Dagang and Togang. The lower roots yield at August harvest was in Korea jiwhang and Hwanggang. The higher economic feasibility at August harvest was in Gogang, Togang, Dagang and Wongang. The higher economic feasibility at ended-October harvest was in Gogang, Yeongang, Togang, Dagang and Wongang.
Background : According to medicinal plant standard culture, flower stalk should be removed at the stage of 4-5 main leaves appearance but there is no detail information on stalk removal in jiwhang (Rehmannia glutinosa L.). This study was carried out to examine effect of flower stalk removal on the increase of total yield in Rehmannia glutinosa L. Materials and Methods ○ Experiment variety : Korea jiwhang, Wongang ○ Treatment : ① Peduncle non-cutting ② Peduncle cutting ○ Planting date : April 18, 2015 ○ Planting distance : 30×15cm ○ Experiment place : Ginseng and Medicinal Plant Research Institute (Geumsan-gun, Chungcheongnam-do) Conclusion : Aerial part growth was no significant difference according to treatments in Wongang and aerial weight was havier in Peduncle cutting than in Peduncle non-cutting. Aerial part growth was no significant difference according to treatments in Korea jiwhang. The roots yield in Korea jiwhang was increased about 5.2% in Peduncle cutting. The roots yield in Wongang was increased about 24.9% in Peduncle cutting.

Materials and Methods ○ Experiment variety : Korea jiwhang ○ Treatment : ①Control ② 1 time irrigation at planting time ③1 time irrigation at planting time + transparent vinyl mulching ○ Planting date : April 30, 2015 ○ Planting distance : 30×15cm ○ Experiment place : Ginseng and Medicinal Plant Research Institute (Geumsan-gun, Chungcheongnam-do) Conclusion : There was no significant difference in the aboveground growth among treatments. The seedling emergence rate was higher in 1 time irrigation at planting time + transparent vinyl mulching(1ITVM) than control or 1 time irrigation at planting time(1I). Aerial part weight was significantly havier in 11ITVM than control or 1I. The roots yield was also more in 1ITVM than control or 1I. According to economic analysis, total income in 1ITVM compared to 1I and control was 125.1% and 54.0% higher respectively.
Resveratrol, which is both a phytoalexin with antifungal activity and a phytochemical associated with reduced cancer risk and reduced cardiovascular disease, is synthesized in a limited number of plant species including peanut. Resveratrol synthesis is catalyzed by the enzyme stilbene synthase including resveratrol synthase (RS). Resveratrol synthase gene (RS3) obtained from peanut, Arachis hypogaea, Fabaceae has been transferred into chinese foxglove, Rehmannia glutinosa by using Agrobacterium mediated transformation. RS t-DNA introduced to chinese foxglove (R. glutinosa L) by transformation and its reaction product, resveratrol-3-O-β-D-glucoside was isolated and characterized using HPLC. Also its biological effects was tested in inhibition of the lipid peroxidation of mouse LDL by glycosylated stilbenes derivatives obtained from transgenic plants. Resveratrol-3-O-β-D-glucoside isolated from transgenic R. glutinosa L. showed antimicrobial activity of the growth inhibition zone against Escherichia coli and Salmonella typhimurium. Therefore, this compound can be contributed to be useful as a phytoalexin for plant health as well as a phytochemical for human health.
Rehmannia glutinosa shows a high level of resistance to the non-selective herbicide paraquat. To characterize the antioxidant enzyme system of R. glutinosa, we comparatively examined the responses of antioxidant enzymes to UV, wounding and a general elicitor yeast extract in R. glutinosa and soybean. The levels of enzyme activities of the two plant species were drastically different between those per fresh weight (general activity) and per protein (specific activity) bases. The general activities of superoxide dismutase (SOD), peroxidase (POX), catalase (CAT), and glutathione reductase (GR) were lower, but that of ascorbate peroxidase (APX) was higher in R. glutinosa than in soybean. The specific activities of the enzymes, however, were about two- to seven-fold higher in R. glutinosa than in soybean, except that of CAT, which was about 12-fold higher in soybean. The general and specific enzyme activities of R. glutinosa relative to those of soybean showed a consistent increase in responses to the stresses only in SOD. The specific activities of SOD and APX were higher in R. glutinosa in all stress treatments. The results might suggest a relatively higher contribution of SOD and APX to the stress tolerance.

The objectives of this study were to establish the genetic transformation system of stilbene synthase in Rehmannia glutinosa. Resveratrol, which is both a phytoalexin with antifungal activity and a phytochemical associated with reduced cancer risk and reduced cardiovascular disease, is synthesized in a limited number of plant species including peanut. Resveratrol synthesis is catalyzed by the enzyme stilbene synthase including resveratrol synthase (RS). Stilbene synthase gene (RS3) obtained from peanut, Arachis hypogaea, Fabaceae has been transferred into chinese foxglove, Rehmannia glutinosa by using Agrobacterium mediated transformation. PCR analysis with RS3 primer confirmed that the targeted gene was introduced into the plant genome, 904 bp in size. Further analyses of identification of transformation using developed other molecular techniques and transgenic plants that RS t-DNA introduced to chinese foxglove (R. glutinosa L) and its reaction product, stilbene such as resveratrol will be isolate and characterize using NMR, MS, and HPLC.
Using Agrobacterium-me야ated transformation method the auxin-regulated cotton GST (Gh-5) constructs were used to transform Rehmannia glutinosa L. The PCR analysis was conducted to verify transgenicity. Based on the PCR analysis, there was verified that the 988 bp DNA band had showed in transgenic plant genomes in PCR anaJysis using Gh5-1 and Gh5-2 primers. The effects of cocultivation with Agrobacterium tumefaciens, regeneration and selection conditions on the transformation efficiency of Chinese foxglove (Rehmannia glutinosa L.) were investigated. Factors such as cocultivation period, use of acetosyringone, postcultivation in darkness, and different kanamycin concentrations for selection were assessed. In vitro regeneration, the number of leaves, shoot lengths and numbers on MS medium were superior to on B5 and WPM medium, and the shoot formation rate was highest level of 95% in cultured base part containing leaf stalk. Addition of acetosyringone at concentration of 200μM to cocultivation medium and 3-day of cocultivation improved transformation frequencies. Exposure of explants to darkness for 4 weeks on selection medium resulted in further increased the regeneration frequency of transgenic shoots. In PCR analysis, the amplified fragments of Gh5 gene were detected (988 bp), and GST-expressing transgenic R. glutinosa L. plants had approximately three-fold higher activity in leaf extracts compared with control plant.
흑색 비닐피복과 재식밀도에 따른 지황의 생육과 수량에 미치는 영향을 구명하기 위해 지황 1호를 시험재료로 흑색 비닐피복과 재식밀도를 달리하여 재배 시험을 수행하였던 결과를 요약하면 다음과 같다. 출현율은 무피복 78.0%에 비하여 흑색 비닐피복에서 3.1%더 높았고, 재식밀도 간에는 차이가 없었다 생육은 흑색 비닐피복과 소식에서 생육이 왕성하였으나, 단위 면적 당 엽 중, 근경 수는 밀식 할수록 무겁거나 많았다. 건근경중은 무피복 402 kg/10a에 비하여 흑색 비닐피복재배에서 28% 증수되었으며, 개체 당 건근경중은 소식할수록, 단위면적 당 수량은 밀식 할수록 증가하였다.
