        풀무치 (Locusta migratoria) (Orthoptrea: Acridiade)는 전 세계적으로 작물 생산에 심각한 문제를 야기하는 돌발 해충이다. 그러나 우리나라 의 경우 풀무치를 방제하기 위한 방제제 및 적용에 대한 연구는 미흡한 실정이다. 본 연구에서는 풀무치에 병원성을 갖는 풀무치병원성 진균 라이브러 리를 구축하였으며, 풀무치 방제에 이용 가능한 생물학적 방제제로서의 가능성을 평가 하였다. 먼저 갈색거저리 유충-baiting 시스템을 이용하여 다양 한 지역에서 채집된 토양에서 곤충병원성 진균을 발굴 하였다. 풀무치 병원성 검정을 진행하기 위하여 국립 농업 과학원에서 풀무치를 분양 받았으며, 고체 배양된 곤충병원성 진균을 곤충 사육 상자에 처리하여 (2 g/ box), 풀무치 약충 (3-4령충)에 대한 곤충병원성 진균의 병원성을 평가 하였다. 그 결과 곤충병원성 진균 처리 3-7일차에 풀무치의 머리, 복부, 다리 표면에서 진균이 증식하는 mycosis를 확인 할 수 있었다. 특히, Metarhizium anisopliae, M. lepidiotae, Clonostachys rogersoniana에서 높은 병원성이 나타나는 것이 확인 되었다. 확보된 34개의 풀무치병원성균주의 특성을 파악 하기 위하여 열안정성 및 포자생산성을 확인 한 결과, Paecilomyces, Purpureocillium 균주가 다른 균주에 비해 열에 대한 높은 안정성안 나타나는 것을 확인 하였으며, 대부분의 균주에서 1 × 108 conidia/gram 이상의 포자수를 생산 하는 것을 확인 하였다. 또한 온실 조건에서 비교적 병원성이 높았던 M. anisopliae 고체 배양된 균주를 토양에 처리하여 병원성을 확인한 결과, 85.7%의 높은 방제효과를 확인 할 수 있었다. 본 실험을 통하여 풀무치가 이동하면서 토양에 정착된 곤충병원성 진균에 접촉되어 치사 될 수 있을 것으로 판단되며, 효과적인 풀무치 방제가 가능 할 것이라고 판단된다.

        The aim of this study was to examine indoor fungal concentration and fungal volatile organic compounds(VOCs) in single-person households. A total of 22 houses occupied by one person were investigated in this study. 19 VOCs detected in the field were estimated as fungi-derived VOCs through a review of the literature, and 11 VOCs were confirmed as fungal VOCs by laboratory experiments. Exposure to fungal VOCs in the indoor environment has been confirmed to be highly influenced by airborne fungal concentration and indoor humidity. 3-octanone was characterized by a clear generation profile in Aspergilus versiocolor, and n-decane in Pencillum chrysogenum. ntetradecane emitted by Stachybotrys chartarum exhibited a tendency to occur consistently. The observations made in this study demonstrated that single-person households can easily be exposed to fungi, and MVOCs can be used as an indicator of fungal exposure in indoor environments.

        2009년부터 2011년, 2015년까지 변산반도 국립공원의 고등균류를 조사한 결과, 총 2문 6강 18목 61과 157속 323종 이 조사 되었으며, 주름버섯목(Agaricales)이 23과 67속 153종, 그물버섯목(Boletales) 6과 27속 45종, 무당버섯목 (Russulales) 3과 4속 40종 및 구멍장이버섯목(Polyporales) 6과 21속 28종으로 4목의 종수가 총 266종으로 전체 종수의 82.4%로 대부분을 차지한 것으로 나타났다. 가장 많은 종수가 조사된 균류는 그물버섯과로 37종이었으며, 무당버섯과(36종), 주름버섯과(28종) 및 광대버섯과(25종) 순으로 우점하고 있는 것으로 나타났다. 서식환경별에서는 외생균근성 버섯은 15과 38속 130종(40.2%)으로 독성 버섯은 46종, 식·약용 버섯 51종 및 식·독 불명 버섯 26종 등이 조사되었으며 낙엽 및 목재부후균은 33과 72속 114종(35.3%)이었고 이 중 독성 버섯은 10종, 식·약용 버섯 52종 및 식·독 불명 버섯 46종 등이, 지상균은 24과 47속 72종(22.3%)이었고 독성 버섯은 8종, 식·약용 버섯 31종 및 식·독 불명 버섯 29종 등인 것으로 나타났다. 월별 발생에서는 대부분의 독성 버섯, 식·약용 버섯 및 식·독 불명 버섯 등은 7월과 8월에 집중적으로 발생하는 것으로 나타났다. 고도별에서는 대부분의 독성 버섯, 식·약용 버섯 및 식·독 불명 버섯 등은 1~199m 지역에서 많이 발생되고 있는 것으로 나타났으며 200m 이상 지역에서는 현저히 낮아지 는 것으로 조사되었다. 독성 버섯, 식·약용 버섯 및 식·독 불명 버섯 등은 평균온도에서는 24.0~25.9℃, 최고온도 28.0~29.9℃, 최저온도 20.0~21.9℃, 상대습도 77.0~79.9% 및 강수량 300.0~499.9 mm일 때 발생이 높은 것으로 나타났다.

        The green peach aphid, Myzus persicae, is one of the most important pests of vegetable crops worldwide. This species cause direct damage by feeding on plant nutrients and indirect damage as transmits many virus vectors. The control of aphid has relied on the use of chemical insecticides and their intensive use over many years has led to the development of resistance. To surmount or avoid these problems, we screened the entomopathogenic fungi against green peach aphid and evaluated their virulences. Several entomopathogenic fungal isolates were selected with high virulence to green peach aphid. Additionally, the antimicrobial activity of the selected fungal isolates was tested and analyzed to phytopathogens. Consequently, these entomopathogenic fungi would be used effectively for dual control agents for the green peach aphid and phytopathogen.

        Spodoptera exigua (Hübner) is an important pest with wide host range including cabbage, beans, potato etc. We evaluate the virulence of Beauveria bassiana ARP14, B. bassiana GHA, and Isaria japonica with concentration of 1×108 conidia/mL against larval stage of S. exigua using glass scintillation vial. Lethal median time (LT50) of these three fungi was not found significantly different in both fourth and second instar. Infection rate was higher in fourth instar than second instar. Infection rate was highest by B. bassiana ARP14 (84.0%) followed by B. bassiana GHA (63.6%) and I. japonica (45.8%) in fourth instars of S. exigua. Our results suggest that B. bassiana ARP14 can be a new component of integrated pest management of S. exigua.

        Entomopathogenic fungi, widely distributed in nature, have been developed as biopesticides of agricultural pests. However, little is known about isolation method for obtaining a variety of highly virulent strains from the soil. In this work, the previously established method using mealworm (larvae of Tenebrio molitor) as a baiting system was used to efficiently isolate fungal strains from soil, and the isolated fungal library in Korea was established. To isolate entomopathogenic fungi, soil samples from 10 regions of Korea such as Iksan, Imsil, Gunsan, Yeosu, Suncheon, Gwangyang, Jinju, Hamyang, Boseong and Jangseong were collected. Of the collected soils, 21 and 34 isolates of entomopathogenic fungi were isolated from Iksan and Gunsan, respectively. The isolates were classified as genera of Metarhizium, Beauveria and Isaria. This work can suggest an effective insect pathogenic fungal isolation method

        Entomopathogenic fungi have been known as promising candidates in biological control. However, a large number of conidia with higher stability should be produced for industrialization. In this work investigated the possibility of several grains for the production of thermotolerant Isaria javanica and I. fumosoroseus conidia. The fungal conidia were produced on ten grains for 14 days in Petri dish conditions. Of the ten granular substrates, millet, perilla seed and barley showed the high conidial production, 15.7 ± 8.4, 17.3 ± 5.2 and 13.5 ± 1.9 × 109 conida/g in I. javanica and 15.9 ± 0.6, 15.0 ± 0.5 and 15.0 ± 0.7 ×109 conida/g in I. fumosoroseus, respectively. However, the two strains showed relatively low thermal stability regardless of substrates, when exposed to 45℃ for 1, 2 and 3 hours. This work suggests a possible substrate for conidial production of entomopathogenic fungi with low cost.

        Entomopathogenic fungi have been widely studied for their potential as the effective biological control agents. They produce variety of secondary metabolites with insecticidal activities, and it is reasonable to assume that entomopathogenic fungi might produce secondary metabolites modulating juvenile hormone for their survival against defense mechanisms of host insect. In this study, acetone extract of the Lecanicillium spp. cultured on unpolished rice medium showed juvenile hormone antagonist (JHAN) activity in the yeast-two hybrid β-galactosidase assay and high insecticidal activity against Aedes albopictus and Plutella xylostella. In addition, to compare bioactivities of secondary metabolites from solid and liquid culture, the Lecaniciilium spp. strain cultured on unpolished rice medium or PDB medium were serially extracted with acetone and ethyl acetate respectively. Both extracts showed JHAN activity and high insecticidal activity against A.albopictus. Theses results suggested that secondary metabolites of entomopathogenic fungi could be useful for development of novel IGR insecticides.

        Entomopathogenic fungi are expected to play a role as great biological control agents in the global bio-pesticide market in the future. The first step is the collection of fungal isolates and it should be a platform for the development of highly effective biopesticides. In this work, we constructed a fungal library using a mealworm pathogenecity-based fungal collection method and further characterized some isolates with high virulence. A phylogenetic tree was generated, and of the isolates 17 isolates’ biological features were characterized, such as morphology, virulence against several insect, stability of conidia for heat, production of biologically active materials, such as enzymes. This work reports an attractive entomopathogenic fungal library including the information of effective isolates in pest management.

        Entomopathogenic fungi are potential pesticides as biological control agents. However, many previous studies have focused on basic researches to control pest using insect-pathogenic fungi. Herein, the molecular characteristics of insect defense mechanisms against fungal pathogens and fungal pathogenicity to insects were investigated. The whole genome of Beauveria bassiana (Bb) JEF-007 was sequenced (36,538,394 bp) using Pacbio sequencing. Based on this data, the transcriptomes of bean bug and fungus were analyzed using RNA-seq. In the transcriptome analyses, six and five genes were validated to be involved in the fungal pathogenicity and insect defense, respectively. This work can provide an understanding of the insect-pathogen interactions

        Virulence of six entomopathogenic fungi was assessed against the 1st instar larva (<5 h old) of oriental fruit moth Grapholita molesta (Busck) in laboratory which is considered as an important pest for stone fruits. Three strains of Beauveria bassiana and each strain of Metarhizium robertsii, Metarhizium bruneum, and Isaria farinosa were evaluated with the concentration of 1×108 conidia/mL at 95.9% RH and 25.4°C by glass scintillation vial assay. Among all, two strains of B. bassiana, ARP14 (87.5%) and A (unpublished) (72.7%), showed highest virulence with 9.8 and 12.6 h of LT50 within 16 h of exposure. However, B. bassiana A showed highest infection rate of 90.0% compare to ARP14 (70.0%) after 14 days. From these results, the two strains of B. bassiana, ARP14 and A, can be used for the control of G. molesta.

        Supplemental substrates for mushroom cultivation media are potent sources for fungal contamination of indoor environment in cultivation houses. The present study was performed to investigate the presence of harmful fungi in the supplemental materials (beet pulp, cotton seed meal, kapok meal, peat moss, wheat straw) imported from China, Egypt, Germany, Indonesia and Pakistan. Fungal concentration in the supplemental materials ranged from 1.3×102 to 1.8×103 cfu/g. A total of 11 genera and 21 species were identified. Two genera and two species were found in Germany peat moss. In Pakistan wheat straw, three genera and four species were detected. In Egypt beet pulp and China cotton seed meal, four genera and five species and seven genera and ten species were shown, respectively. Among the identified fungi, Acremonium sp., Aspergillus chevalieri, Cryptococcus adeliensis, Eurotium amstelodami, Paecilomyces variotii and Rhodotorula mucilaginosa were known as human pathogens. This study provided evidence that pathogenic yeast and filamentous fungi are present in imported agricultural byproduct materials for mushroom cultivation.

        Four fungal species, during indoor air monitoring for fungi that possibly affect the field testing of a newly bred shiitake cultivar in cultivation houses located in Cheongyang, Chungnam Province and Jangheung, Jeonnam Province. Of these species, two are known to be plant pathogens and the other two are saprobes but potent contaminators in the mushroom cultivation environment. This study reports the morphological characteristics and phylogenetic relationships of these four species based on nucleotide sequences of the internal transcribed spacer (ITS) and 18S rDNA region, including their known information.

        Honey production from approximately 1.7 million colonies owned by around 21 thousand Korean beekeepers was almost 36 thousand M/T in 2014. Honey farmers have mainly obtained commodities such as honey, beeswax, pollen and royal jelly. Honeybees gather the nectars and pollens from flowers of plants or trees. Bee pollen is rich in various nutrients and called as natural complete food. Bee pollens of Quercus acutissima, Actinidia arguta, Amorpha fruticosa, Oryza sativa, Chrysanthemum morifolium and Salix koreensis were mainly collected by Apis mellifera in Korea. But stiff pollen wall hinders dissolution of polysaccharides and lowers extraction efficiency. Thus, we measured antioxidant activity as DPPH radical scavenging assay system on six kinds of pollens inoculated with fungi to confirm the husk removal effect. The antioxidant activity was evaluated by measuring the free radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH). Briefly, extracts prepared from different extraction conditions were mixed with freshly prepared DPPH solution. After shaking, the reaction mixtures were stand for 30 min at room temperature in dark places. The radical scavenging activity was determined by measuring the absorbance at 517 nm. The relative radical scavenging activity (%) was calculated as [1-absorbance of solution with sample and DPPH / absorbance of solution with DPPH] x 100. The scavenger activities of Quercus acutissima, Actinidia arguta, Amorpha fruticosa, Oryza sativa, Chrysanthemum morifolium and Salix koreensis were 42.73±1.25, 8.98±1.46, 4.38±3.53, 36.09±0.74, 17.28±1.95 and 62.76±2.07 at 500㎕/㎖, respectively.

        The two-spotted spider mite, Tetranychus urticae, is one of the most important pests of crops. However, chemical acaricides are not always effective and their continuous use has resulted in resistance among two-spotted spider mite populations. To reduce or replace use of chemical acaricides, the entomopathogenic fungi having acaricidal activity were investigated. As results, 12 isolates could be selected from 342 isolates of fungi against two-spotted spider mite. To show the multiple roles of entomopathogenic fungi against plant pathogen, the antimicrobial activity of selected 12 isolates were evaluated to plant pathogenic bacterium Ralstonia solanacearum and fungus Botrytis cinerea. Various antimicrobial activities were observed against these plant pathogens. Consequently, these entomopathogenic fungi would be offered an alternative for dual control agents against the two-spotted spider mite and plant diseases.

        Alternative control agents, hopefully overcoming the present issues and problems in agrochemicals, should be considered and finally applied to the vector controls. Entomopathogenic fungi can used as one of the possible novel control agents and great considerations are given to the control of soil- or water-dwelling stages of vectors, such as mosquitoes and ticks. In our research group, the entomopathogenic fungal library has been constructed using the mealworm-based isolation system, which showed a variety of opportunities of their use in vector control. Important key production technologies including granular formulation have been developed to increase their industrialization. Some entomopathogenic fungal isolates showed high biological performance in the control of mosquitoes and ticks in field stands. R&D of down-stream process should be seriously considered as the key step.

        Various arthropods have directly and indirectly threatened human life by transmitting human pathogens such as malaria and Zika virus as well as by damaging crops such as direct feeding and transmission of plant pathogens. According to WHO, vector-borne diseases account for over 17% of all infectious diseases and there are over 1 million deaths from the diseases such as malaria, dengue, etc., globally. About 13% of total crop production is destroyed by agricultural pests during crop production and storage. To control medical or agricultural pests chemical pesticides have been used, but recently because of concerns about environmental pollution and human health the demands for eco-friendly control method are increasing. Insect pathogens are good alternative candidates instead of chemicals. Over 50 entomopathogens including viruses, bacteria, fungi and nematodes are now commercially used as microbial pesticides. Entomopathogenic fungi have also been evaluated as control agents for a diverse insect pests such as aphid, moth, thrip, whitefly, mosquitoes, tick, etc. We will discuss a development of microbial pesticides using, especially, entomopathogenic fungi to control noxious arthropods to agricultural crops and human.

        Nuruk, a Korean fermentation starter for Makgeolli, contains a variety of microflora according to province and natural environment. However, the metabolites profiles in traditional Korean Nuruk were not defined yet. In this study, Nuruk were prepared with 20 different fungi originated from Gangwon province (Rhizomucor variabilis, Lichtheimia corymbifera, and Aspergillus species) and their metabolites were profiled / compared with commercial strains (Aspergillus kawachii) through metabolomics analysis. Metabolites were extracted with solvent mixture (Isopropanol: Methanol: Distilled water) and put into MS spectrometry analysis. After processing of full mass spectra data from LC-MS/MS, the characteristics of each strain were compared and analyzed by multivariate statistical analysis such as hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal partial least squares discriminant analysis (OPLS-DA).Interestingly, strain specific classification was observed by HCA and PCA among samples and significant differences were also indicated from commercial strain Aspergillus kawachii. As a results, 26 metabolites were successfully screened with a validity from a total of 189 metabolites by using of VIP plot through OPLS-DA. These results suggested the uniqueness of Korean traditional home brewed liquor compared to commercial liquor and could be useful information for Korean brewing industry.

        Basidiomycetous fungi are one of the most potent biodegraders because many of its species grow on dead wood or litter, in environments rich in lignocellulose. For the degradation of lignocellulose, basidiomycetes utilize their lignocellulytic enzymes, which typically include laccase (EC 1.10.3.2), lignin peroxidase (EC 1.11.1.14), xylanase (EC 3.2.1.8), and cellulase (EC 3.2.1.4). In recent years, the practical applications of basidiomycetes have ranged from the textile to the pulp and paper industries, and from food applications to bioremediation processes and industrial enzymatic saccharification of biomass. Recently, spent mushroom substrates of edible mushrooms have been used as sources of bulk enzymes to decolorize synthetic dyes in textile wastewater. In this review, the occurrence, mode of action, general properties, and production of lignocellulytic enzymes from mushroom species will be discussed. We will also discuss the potential applications of these enzymes
