This study was performed to investigate immune changes by comparing the proportion and function of immune cells in the blood under high-temperature period and convalescence temperature period in Holstein dairy cows. The experiment was conducted using Holstein dairy cows of five animals per group (60 ± 20 months old, 175 ± 78 non-day) from the National Institute of Animal Science at high-temperature period (THI: 76 ± 1.2) and convalescence temperature period (THI: 66 ± 1.3). Complete blood count results showed no change in the number of immune cells between groups. In the analysis using Flow Cytometry of PBMCs, no significant differences were observed among B cells, Helper T cells, cytotoxic T cells, and γδ T cells between groups. However, there was an increase in Th17 cells producing IL-17a, while Th1 cells decreased during the convalescence temperature period. The results of gene expression analysis using qRT-PCR in PBMCs revealed an increase in IL-10 during the convalescence temperature period, while a decrease in HSP70 and HSP90 was observed. In conclusion, the increased expression of IL-10 and the decrease in HSP expression suggest the possibility of a weak recovery from heat stress. However, the lack of observed changes in B cells, T cells, and other immune cells indicates incomplete recovery from heat stress during the convalescence temperature period.
This study aimed to investigate the effects of amino acid complex additives, such as protected vitamin C (VC) or detoxified sulfur (DS), on the growth and metabolism of Hanwoo cattle under high-temperature conditions. Accordingly, farms in Temperature-Humidity Index (THI) regions ranging from 78 to 89 for over 100 days were selected. The experimental groups were control, T1 (lysine + methionine + VC, 50 g/head/day), and T2 (lysine + methionine + DS, 50 g/head/day) with 70, 77, and 71 animals each. The range of the THI for 115 days was 78-89, and this occurred in most of the experiment days. The results showed that there was no significant difference in rectal temperature among the groups. The body weight increased to 786.4 and 809.0kg in the T1 and T2 groups, respectively, compared to the control group (p<0.05). Linoleic acid showed a high result of 2.01% in the T1 group compared to the control group (p<0.05). Unsaturated fatty acids were higher at 55.70 and 56.54% in the T1 and T2 groups, respectively, compared to the control group (p<0.05), and the omega 6/3 ratio was reduced to 20.10% (p<0.05). These findings indicate that T1 has a positive impact on growth, meat quality, and fatty acid composition compared to the control group. In conclusion, amino acid complex with VC improved the body weight of Hanwoo steers and the unsaturated fatty acids and essential amino acids of their meat; however, further research is needed to clarify this impact on carcass performance.
Radiant tubes heat exchangers are critical components that facilitate the heat transfer process to steel in an annealing furnace, and it addresses several engineering problems, such as thermal stress and mechanical failure due to long-term thermal cycling, which can significantly affect the longevity of the tubes and maintenance requirements. In this study, we used commercial software (ANSYS) to simulate the thermal stress and deformation of radiant tubes subjected to extreme thermal conditions and pressure loads. We evaluated both thermally induced deformation and creep deformation, which is a time-dependent deformation under constant stress over the long term. The results showed that uneven temperature conditions and pressure loads lead to significant deformation and potential failures. To mitigate these engineering challenges, we also tested several designs that include supporting brackets. This study provides valuable insights for designing radiant tube heat exchangers in annealing furnaces to extend their lifespan and ensure system safety.
살리실산은 식물의 생장 및 발달, 항산화 방어기작, 광합성 작용 그리고 생물적 및 비생물적 스트레스 조건에서 다양한 생리적 기능을 조절하는 물질로 알려져 있다. 본 연구에서는 고온・건조 스트레스 조건에서 살리실산 경엽처리가 고추의 생육, 광합성 특성 및 항산화효소 활성에 미치는 영향을 구명 하고자 하였다. 광합성 특성 측정결과 광합성 속도, 기공전도 도 및 증산 속도가 증가하였고, 3회차 처리에서 가장 높았다. 세포내 MDA와 H2O2 함량은 살리실산 3회차 처리에서 현저 하게 감소하는 경향을 보였다. APX, CAT, POD 및 SOD 활 성이 현저하게 증가하였으며, 무처리 대비 최대 247, 318, 55 및 54% 증가하였다. 고추의 생육 특성은 무처리구와 유의한 차이를 보이지 않았으나, 상품 수량은 15% 정도 증가하였다. 이러한 결과들을 종합해 볼 때, 살리실산의 경엽처리는 고추 의 광합성 특성과 항산화효소 활성을 증진시켜 고온・건조 스트 레스에 의한 피해 경감에 긍정적 효과를 유발함을 확인하였다
Environmental stresses caused by climate change, such as high temperature, drought and salinity severely impact plant growth and productivity. Among these factors, high temperature stress will become more severe during summer. In this study, we examined physiological and molecular responses of maize plants to high temperature stress during summer. Highest level of H2O2 was observed in maize leaves collected July 26 compared with June 25 and July 12. Results indicated that high temperature stress triggers production of reactive oxygen species (ROS) in maize leaves. In addition, photosynthetic efficiency (Fv/Fm) sharply decreased in leaves with increasing air temperatures during the day in the field. RT-PCR analysis of maize plants exposed to high temperatures of during the day in field revealed increased accumulation of mitochondrial and chloroplastic small heat shock protein (HSP) transcripts. Results demonstrate that Fv/Fm values and organelle-localized small HSP gene could be used as physiological and molecular indicators of plants impacted by environmental stresses.
The purpose of the study was to investigate an effect of water temperature on a non-specific immune response and mortality of tilapia, Oreochromis niloticus, following a bacterial infection. Seventy five tilapia acclimated to 25℃ were then transferred at 16 and 36℃, and examined for non-specific immune responses over 12-96 h. Respiratory burst activity was reduced significantly in the group of fish cultured at 16 and 36℃ over 24-96 h, whereas phagocytic activity decreased significantly in the group of fish reared at a low temperature (16℃) over 12 and 24 h and high temperatures (36℃) over 12-96 h. Lysozyme activity diminished significantly in the group of fish transferred to 16℃ over 12-48 h, but increased significantly in the group of fish at 36℃ over 48 and 96 h. Alternative complement pathway (ACH50) decreased significantly when transferred to 16℃ after 12 h, but increased significantly when transferred to 36℃ after 24 h. In a challenging test, 30 tilapia reared at 25℃ were injected intraperitoneally with Streptococcus iniae at a dose of 2x107 cfu/fish, and then reared onward at water temperatures of 15, 25 (control), and 36℃. Over 12-96 h, the cumulative mortality of S. iniae-injected fish held in 16 and 36℃ was significantly higher than that of injected-fish held in 25℃ In conclusion, transfer of tilapia from 25℃ to low temperature (16℃) after 12 h, and transfer of fish from 25℃ to high temperature (35℃) reduced their immune capability. Furthermore, tilapia under temperature stress at 16 and 36℃ from 25℃ decreased its resistance against S. iniae
This paper investigates the effect of prolonged high temperature exposure on concentric laminated Al2O3-ZrO2 composites. An ultrafine scale microstructure with a cellular 7 layer concentric lamination with unidirectional alignment was fabricated by a multi-pass extrusion method. Each laminate in the microstructure was 2-3μm thick. An alternate lamina was composed of 75%Al2O3-(25%m-ZrO2) and t-ZrO2 ceramics. The composite was sintered at 1500˚C and subjected to 1450˚C temperature for 24 hours to 72 hours. We investigated the effect of long time high temperature exposure on the generation of residual stress and grain growth and their effect on the overall stability of the composites. The residual stress development and its subsequent effect on the microstructure with the edge cracking behavior mechanism were investigated. The residual stress in the concentric laminated microstructure causes extensive micro cracks in the t-ZrO2 layer, despite the very thin laminate thickness. The material properties like Vickers hardness and fracture toughness were measured and evaluated along with the microstructure of the composites with prolonged high temperature exposure.
Reactive oxygen species (ROS) are produced by oxidative stresses which cause various chronic diseases such as diabetes and obesity. Ginseng (Panax ginseng C.A. Mayer) has been reported to contain various biological activities such as anti-cancer, anti-diabetic, neuroprotective, radioprotective, anti-amnestic and anti-aging effects. In this study, we investigated the effects of Panax ginseng, treated with high temperatures and high pressures, on oxidative stress in C2C12 myoblasts and 3T3-L1 adipocytes. Oxidative stress was induced in the C2C12 cells through the introduction of H2O2 (1 mM), and cells were then treated with various ginseng preparations: dried white ginseng (DG), steamed ginseng (SG) and high temperature and high pressure treated ginseng (HG). In addition, 3T3-L1 preadipocytes were treated with various ginsengs for up to 8 days following standard induction of differentiation. Our results show that HG treatment significantly protected oxidative stress in both cell lines and enhanced gene expression of antioxidant enzymes. Therefore, in this study, we investigated the protective effects of ginseng on the oxidative stress of adipocytes and muscle cells.
Zr-4 used for a cladding and an end plug of reactor component has creep deformation under operation at high temperature. Creep is regarded as the time dependent deformation of a material under constant applied stress. Although the major source of the deformation of zirconium component in water-cooled reactors is irradiation creep, the thermal creep may give a rise to significant deformation in reactor component especially at relatively high temperatures and at various constant stresses, and therefore it must be predicted accurately. Stress relaxation is the time dependent change of stress at constant strain and it is a process related intimately to creep. In this paper, the creep behavior and stress relaxation of Zr-4 is examined at the temperature of 500℃ that is 40% of the absolute melting temperature of Zr-4 under the stress below yield stress and under the various constant strains. The results obtained are summarized as follows: 1) With an increase of stress, the steady state creep rate increases and the creep rupture time decreases. 2) The steady state creep rate ε(%/s) for the stress Σsub(c) (kgf/mm super(2)) of Zr-4 increases outstandingly. All the empirical equations computed for Zr-4 increases outstandingly. All the empirical equations computed for Zr-4 are in accord with Norton's model equation(ε=KΣ sub(c) super (n)). The constants of materials computed are as follows: K=3.9881×10 super(-5), n=1.9608 3) The rupture time T sub(r) (hr) decreases linearly with the increase of stress on the log-log scaled graph. The empirical equations computed for Zr-4 are in accord with Bailey's model equation (T sub(r)=K sub(1)Σsub(c) super(m)). The constants of materials computed are as follows: K sub(1)=1.2875×10 super(16), m=-3.467 4) It seems clear that the strain could be quantitatively dependent on the high temperature creep properties such as creep stress, rupture time, steady state creep rate and total creep rate. It is found that these relationships are linear on the log-log graph. 5) In stress relaxation test, as the critical constant strain that can be allowed to the specimen is larger, stress relaxation becomes more rapid, and as the constant strain is smaller, the stress relaxation becomes slower.
Background : Due to changes in climate and cultivation conditions, the growth monitoring is an essential factor in improving crop productivity. With the recent development of image analysis technology incorporating ICT, it has become possible to constantly monitor the crop growth. As a medicinal crop specialized in Gyeongsangbuk-do, Korea, Cnidium officinale Makino was examined for the possibility of growth diagnosis through image analysis for stable production.
Methods and Results : The IP camera was installed at 2.5 m height in experiment field. The RGB image of every 06:00 was captured from July 1 to July 30 and used for analysis. The captured images were analyzed using the image analysis tool, Image J. The greeness was estimated by the average value of the green histogram. The canopy size was determined by the color range (red: 0-255, green: mean value-255, blue: 0-255) and was calculated as the ratio of pixels number of the entire image to those of the selected area. The growth temperature during investigation period was measured by Hobo MX2300. High temperature, excess of 28℃, was compared to stress response such as decrement of canopy size. The greeness and the canopy size are respectively represented by the quadratic function greeness = -0.0722GD2 + 6248.9GD – 1e + 08 (GD, growing day; R2 = 0.46) and canopy size = -0.0462GD2 + 3996.7GD – 9e + 07 (R2 = 0.93). From July 11, it began to exceed the growth limit temperature of 28℃, and the canopy size began to decrease from this period. Between the canopy size (C) and the accumulated temperature exceeding 28℃, there was a negative correlation, C = -0.13ATEC + 56.75 (R2 = 0.87) during the decreasing period.
Conclusion : Extraction of color information in Cnidium officinale Makino using RGB image should be preceded by standardized setting, but it is considered to be useful tool for analyzing the change of quantitative characteristics over time. In the future, it is necessary to make a comparative study with the actual growth rate in the image diagnosis.
Global warming means not only increase in average air temperature but also increase in frequency of extreme weathers such as extremely high temperature over 45oC. Crops are generally sensitive to high temperature during their reproductive growth stage. This experiment was thus conducted to investgate physiological responses of rice (Oryza sativa cv. Chucheong) and soybean (Glycine max cv. Sinpaldal) to high temperature (HT) stress at their reproductive stage. Rice and soybean were exposed to different degrees of high temperature stress by keeping them in a growth cabinet deisnged to maintain air temperature up to 45oC for at least 5 hours in a day for different durations, 0, 1, 2, 3, and 4 days. HT stress treatment delayed heading and flowering of rice and increased the sterility of its' main panicle with increasing duration of HT treatment; 19.7 43.4, 68.1, 81.5 and 91.1% at 0, 1, 2, 3 and 4 days HT treatment, respectively. Increasing sterility due to HT treatment thus resulted in significant rice yield loss; 5.07 4.27, 4.32, 4.51 and 3.62 g/plant at 0, 1, 2, 3 and 4 days of HT treatment, respectively. Soybean was also significantly affected by HT stress in its pod formation and sterility, particularly along the vertical stem. Increasing pod sterility with increasing HT treatemnt thus resulted in significant soybean yield loss.
