        Drought is one of the environmental factors inhibiting plant productivity and growth, leading to oxidative damage. This study aims to identify the role of sodium hydrosulfide (NaHS) as a hydrogen sulfide (H2S) donor in drought stress tolerance in Brassica napus. Drought-induced stress symptoms appeared eight days after treatment, showing wilted leaves and a significant reduction of leaf water potential. Drought-induced increase of lipid peroxidation was significantly reduced by NaHS application. NaHS-treated plants mitigated stress symptoms under drought conditions by reducing hydrogen peroxide (H2O2) content, confirmed with H2O2 localization in situ. Furthermore, NaHS promotes photosynthetic activity by maintaining chlorophyll and carotenoid content, thereby supporting plant growth under drought conditions. Pyrroline-5-carboxylate and proline contents were significantly increased by drought but further enhanced by NaHS treatment, indicating the important roles of proline accumulation in drought stress tolerance. In conclusion, this study provides valuable insight into the roles of NaHS in alleviating drought stress by reducing oxidative stress and promoting proline accumulation. Therefore, NaHS may serve as an effective strategy to enhance crop production under drought-stress conditions.

        Background: Aflatoxin B1 (AFB1) is a toxic metabolite generated by Aspergillus species and is commonly detected during the processing and storage of food; it is considered a group I carcinogen. The hepatotoxic effects, diseases, and mechanisms induced by AFB1 owing to chronic or acute exposure are well documented; however, there is a lack of research on its effects on the intestine, which is a crucial organ in the digestive process. Dogs are often susceptible to chronic AFB1 exposure owing to lack of variation in their diet, unlike humans, thereby rendering them prone to its effects. Therefore, we investigated the effects of AFB1 on canine small intestinal epithelial primary cells (CSIc). Methods: We treated CSIc with various concentrations of AFB1 (0, 1.25, 2.5, 5, 10, 20, 40, and 80 μM) for 24 h and analyzed cell viability and transepithelial-transendothelial electrical resistance (TEER) value. Additionally, we analyzed the mRNA expression of tight junction-related genes (OCLN, CLDN3, TJP1, and MUC2), antioxidant-related genes (CAT and GPX1), and apoptosis-related genes (BCL2, Bax, and TP53). Results: We found a significant decrease in CSIc viability and TEER values after treatment with AFB1 at concentrations of 20 μM or higher. Quantitative polymerase chain reaction analysis indicated a downregulation of OCLN, CLDN3, and TJP1 in CSIc treated with 20 μM or higher concentrations of AFB1. Additionally, AFB1 treatment downregulated CAT , GPX1, and BCL2. Conclusions: Acute exposure of CSIc to AFB1 induces toxicity, and exposure to AFB1 above a certain threshold compromises the barrier integrity of CSIc.

        To understand the detrimental effects of triclosan on Java medaka (Oryzias javanicus) embryos, fertilized embryos were exposed to different concentrations (1, 10, 50, 100, 200, 400, 600, 800, and 1,000 μg l-1) of triclosan until hatching. Then, we examined the survival rate and developmental parameters as well as alterations in antioxidant constituents and DNA damage markers. The results showed dose-dependent mortality, hatching delays, and developmental abnormalities in the embryos. Additionally, there were significant increases in oxidative stress parameters and antioxidant responses, along with elevated DNA damage. These findings suggest that sublethal concentrations of triclosan induce toxic effects through oxidative stress on Java medaka embryos, as evidenced by changes in in vivo parameters and biochemical constituents.

        Background: Cadmium (Cd) is toxic heavy metal that accumulates in organisms after passing through their respiratory and digestive tracts. Although several studies have reported the toxic effects of Cd exposure on human health, its role in embryonic development during preimplantation stage remains unclear. We investigated the effects of Cd on porcine embryonic development and elucidated the mechanism. Methods: We cultured parthenogenetic embryos in media treated with 0, 20, 40, or 60 μM Cd for 6 days and evaluated the rates of cleavage and blastocyst formation. To investigate the mechanism of Cd toxicity, we examined intracellular reactive oxygen species (ROS) and glutathione (GSH) levels. Moreover, we examined mitochondrial content, membrane potential, and ROS. Results: Cleavage and blastocyst formation rates began to decrease significantly in the 40 μM Cd group compared with the control. During post-blastulation, development was significantly delayed in the Cd group. Cd exposure significantly decreased cell number and increased apoptosis rate compared with the control. Embryos exposed to Cd had significantly higher ROS and lower GSH levels, as well as lower expression of antioxidant enzymes, compared with the control. Moreover, embryos exposed to Cd exhibited a significant decrease in mitochondrial content, mitochondrial membrane potential, and expression of mitochondrial genes and an increase in mitochondrial ROS compared to the control. Conclusions: We demonstrated that Cd exposure impairs porcine embryonic development by inducing oxidative stress and mitochondrial dysfunction. Our findings provide insights into the toxicity of Cd exposure on mammalian embryonic development and highlight the importance of preventing Cd pollution.

        In this research, the marine medaka Oryzias javanicus underwent a 96 h exposure to two concentrations of the red tide dinoflagellate Karenia mikimotoi (1,000 and 5,000 cells mL-1), and the temporal variations in biochemical responses related to antioxidant and immunity parameters were assessed in the liver tissue. The study revealed a significant increase in ichthyotoxicity with elevated cell concentrations of K. mikimotoi, especially evident at 96 h in marine medaka exposed to 5,000 cells mL-1. At 1,000 cells mL-1 of K. mikimotoi, the opercular respiratory rate showed a significant increase, whereas exposure to 5,000 cells mL-1 resulted in a lowered rate. The intracellular malondialdehyde content was significantly elevated in response to both cell concentrations at 96 h. Regarding glutathione content, levels were significantly increased by exposure to both cell concentrations. Catalase and superoxide dismutase enzymatic activities experienced an increase at 1,000 cells mL-1 of K. mikimotoi, while their activities were reduced at 5,000 cells mL-1 at 96 h. The analysis of two immunity parameters, alternative complement pathway and lysozyme, demonstrated significantly reduced activities in the liver tissue exposed to 5,000 cells mL-1 of K. mikimotoi. These findings aim to enhance the understanding of K. mikimotoi toxicity in marine fish by offering insights into biochemical responses associated with harmful algal blooms.

        Cardiovascular disease remains a leading global cause of mortality, demanding effective prevention and management. This systematic review explores the impact of lycopene, prevalent in various fruits, on cardiovascular health. Thirteen randomized and controlled trials were analyzed, involving 385 participants with cardiovascular risk factors. Notable findings included significant blood pressure reductions, particularly among hypertensive individuals, and favorable alterations in lipid profiles. Lycopene interventions also demonstrated potential in mitigating oxidative stress and inflammation markers. While these findings show promise, more rigorously designed studies are essential to determine optimal dosages and durations for precise cardiovascular benefits. In conclusion, lycopene shows potential in managing cardiovascular risk factors, and its role in blood pressure regulation, lipid profile improvement, and antioxidant and anti-inflammatory effects is encouraging. Nonetheless, further research is warranted to provide definitive guidance on integrating lycopene to enhance cardiovascular health.

        Despite numerous advances in in-vitro embryo production (IVP), many documented factors have been shown to influence the development of mammalian preimplantation embryos and the success of IVP. In this sense, elevated levels of reactive oxygen species (ROS) correlate with poor outcomes in assisted reproductive technologies (ART) due to oxidative stress (OS), which results from an imbalance between ROS production and neutralization. Indeed, excessive production of ROS compromises the structural and functional integrity of gametes and embryos both in vivo and in vitro. In particular, OS damages proteins, lipids, and DNA and accelerates cell apoptosis. Several in-vivo and in-vitro studies report an improvement in qualityrelevant parameters after the use of various antioxidants. In this review, we focus on OS and the source of free radicals and their effects on oocytes, sperm, and the embryo during IVP. In addition, antioxidants and their important role in IVP, supplementation during oocyte in vitro maturation (IVM), in vitro culture (IVC), and semen extenders were discussed. Nevertheless, various methods for determining the level of ROS in germ cells have been briefly described. Still, it is crucial to develop standardized antioxidant supplement systems to improve overall IVP success. Further studies should explore the safety, efficacy, mechanism of action, and combination of different antioxidants to improve IVP outcomes.

        산화적 스트레스는 세포 및 조직 손상을 통해 피부의 탄력 및 보습 기능 저하, 피부 노화 촉진 을 비롯한 다양한 피부질환을 일으킨다. 본 연구의 목적은 인간 피부각질세포 (HaCaT keratinocyte)에서 산화적 스트레스에 대한 붉은 토끼풀 추출물의 효능을 검토하여, 피부에 효과적으로 사용할 수 있는 기능 성 소재로서의 활용 여부를 확인하고자 하였다. 본 연구에서는 붉은 토끼풀 추출물이 인간 피부각질세포에 서 산화적 스트레스에 따른 세포사를 억제시키는 것을 확인하여, 이를 조절하는 보호기전을 규명하였다. 이는 붉은 토끼풀 추출물이 Caspase-3 비활성, 세포사 촉진단백질 Bax 발현 억제, 세포생존 촉진단백질 Bcl-2 발현 증가 및 MAPK 신호전달계 단백질의 인산화 억제를 통해 H2O2에 의해 유도된 산화적 스트레 스를 보호할 수 있다는 것을 확인하였다. 따라서 붉은 토끼풀 추출물은 피부의 산화적 손상을 감소시키는 유용한 소재로 평가되며, 이는 피부보호 및 미용을 위한 다양한 제품 및 산업에 활용 가능성이 높은 것으로 판단된다.

        Despite concerns about the significant toxicity of copper pyrithione (CuPT) at environmental concentrations, effects of CuPT on benthic organisms have received little attention. Here, we analyzed the detrimental effects of CuPT at sublethal concentrations (1/50, 1/20, and 1/10 of the 96 h-LC50 value) for 14 days in the marine polychaete Perinereis aibuhitensis. Reduced burrowing activity and significantly decreased the acetylcholinesterase activity in response to relatively high concentrations of CuPT were identified as CuPT-triggered cholinergic inhibition. The lipid peroxidation marker, malondialdehyde levels were dosedependently increased, whereas intracellular glutathione was depleted by relatively high concentrations. In the CuPT-treated polychaete, significant fluctuations in the enzymatic activities of the antioxidant defense system (catalase, superoxide dismutase, glutathione reductase, and glutathione peroxidase) were observed with significantly modulated glutathione S-transferase activity. These results indicate that even sublethal levels of CuPT would have detrimental effects on the health status of the marine polychaete.

        Diabetic encephalopathy is a major complication with cognitive impairment and neurodegeneration in patients with type 1 or type 2 diabetes mellitus (DM). DM-induced glucolipotoxicity is a risk factor for Alzheimer’s disease–like phenotype, including amyloidogenesis, tau hyperphosphorylation, and neuronal apoptosis. Although the detailed mechanism underlying the pathogenesis of diabetic encephalopathy remains unclear, mitochondrial oxidative stress is emerging as a key factor for diabetic complications and neurodegeneration. A deeper understanding of the regulatory mechanism of mitochondrial oxidative stress under hyperglycemic conditions will provide insights into the development of therapeutic strategies for diabetic encephalopathy. Here, we review the role of mitochondrial oxidative stress in diabetic encephalopathy and the regulatory mechanisms by which high glucose induces the generation of mitochondrial reactive species oxygen species in neuronal cells. This review also summarizes the mitochondrial-dependent and -independent pathways (O-linked-N-acetylglucosaminylation, calcium, and glycogen synthase kinase 3β signaling) that regulate mitochondrial oxidative stress in a DM model.

        Drought stress is a condition that occurs frequently in the field, it reduces of the agricultural yield of field crops. The aim of the study was to screen drought-adapted genotype of Italian rye grass. The experiments were conducted between the two Italian ryegrass (Lolium multiflorum L.) cultivars viz. Hwasan (H) and Kowinearly (KE). The plants were exposed to drought for 14 days. The results suggest that the morphological traits and biomass yield of KE significantly affected by drought stress-induced oxidative stress as the hydrogen peroxide (H2O2) level was induced, while these parameters were unchanged or less affected in H. Furthermore, the cultivar H showed better adaptation by maintaining several physiological parameter including photosystem-II (Fv/Fm), water use efficiency (WUE) and relative water content (RWC%) level in response to drought stress. These results indicate that the cultivar H shows improved drought tolerance by generic variation, improving photosynthetic efficiency and reducing oxidative stress damages under drought stress. These findings can be useful to the breeder and farmer for improving drought tolerance in Italian rye grass through breeding programs.

        The objective of this study was to determine phosphorus effects on drought stress-induced oxidative stress in Kentucky bluegrass. Drought stress was induced by reducing of water to plants in pots. Two types of phosphorus were applied as potassium phosphate (P) or potassium phosphonate (PA). Application of phosphorus was efficient to ameliorate the adverse effects of drought. Osmotic potential, total chlorophyll and carotenoid content were significantly decreased by drought stress, but was relieved by P or PA application. Superoxide (O2 •−) concentration was significantly increased more than 14-fold under drought-stressed plants, was accompanied with increase of hydrogen peroxide (H2O2) and lipid peroxidation (MDA). However, malondialdehyde (MDA) was much less in P or PA applied plants under drought stress condition. Activities of catalase (CAT), ascorbate peroxidase (APX) and guaiacol-peroxidase (GPX) were largely increased by drought stress and its increase rate was much higher in P or PA applied plants except APX. These results indicate that drought stress-induced oxidative stress is alleviated by P or PA application due to the increase of activities of antioxidant enzymes.

        Mesenchymal stem cells (MSCs) have been recognized as a therapeutic tool for various diseases due to its unique ability for tissue regeneration and immune regulation. However, poor survival during in vitro expansion and after being administrated in vivo limits its clinical uses. Accordingly, protocols for enhancing cell survivability is critical for establishing an efficient cell therapy is needed. CDDOMe is a synthetic C-28 methyl ester of 2-cyano-3,12-dioxoolean-1,9-dien-28-oic acid, which is known to stimulate nuclear factor erythroid 2-related factor 2 (Nrf2)- antioxidant response element (ARE) pathway. Herein, report that CDDO-Me promoted the proliferation of MSCs and increased colony forming units (CFU) numbers. No alteration in differentiation into tri-lineage mesodermal cells was found after CDDO-Me treatment. We observed that CDDO-Me treatment reduced the cell death induced by oxidative stress, demonstrated by the augment in the expression of Nrf2-downstream genes. Lastly, CDDO-Me led to the nuclear translocation of NRF2. Our data indicate that CDDO-Me can enhance the functionality of MSCs by stimulating cell survival and increasing viability under oxidative stress.

        본 연구는 미역(Undaria pinnatifida) 추출물과 쇠미역(Costaria costata) 추출물의 항산화 활성 및 HepG2 세포의 알코올로 인한 산화 손상에 대한 보호 효과를 연구하였다. 미역과 쇠미역 추 출물의 총 폴리페놀과 플라보노이드 함량은 70% 에탄올 추출물에서 가장 높았다. 또한 미역과 쇠미역 70% 에탄올 추출물의 DPPH (IC50 0.33±0.21, 0.48±0.47 mg/ml), ABTS (IC50 0.34±0.30, 0.47±0.17 mg/ml) 라디칼 소거 활성이 열수 추출물 및 10% 에탄올 추출물 보다 높았다. 추출 물의 간 보호 효과를 확인하기 위하여 HepG2 세포에 알코올 산화 스트레스를 유발하여 MTT 분석을 이용하여 세포 생존력을 측정하였다. 미역 및 쇠미역 열수 추출물은 알코올 처리군 (73.95%) 대비 세포 생존율은 각각 89.91~97.63% 및 84.99~90.54%로 농도 의존적으로 증가시 켰다. 본 연구는 미역과 쇠미역 추출물이 간 보호 및 항산화 효과를 나타냄을 확인하였고 알코 올 산화 손상에 대한 간 보호 소재로 활용될 수 있을 것으로 사료된다.

        Pollution of agricultural soil by alkaline salts, such as Na2CO3, is a critical and long-lasting problem in cultivable land. The aim of the study was to examine the putative role of citric acid (CA) in alleviating Na2CO3-stress in alfalfa. In this study, Na2CO3 significantly induced leaf chlorosis, inhibited plant growth and photosynthesis related parameters, increased hydrogen peroxide (H2O2) and reduced major antioxidant enzymes (SOD ,CAD, APX) in alfalfa. However, the presence of CA these negative effects of Na2CO3-stress largely recovered. Interestingly, expression of antioxidant and ion transporter genes (Fe-SOD, CAT, APX, DHAR and NHX1) involved in Reactive oxygen species (ROS) homeostasis and oxidative stress tolerance in alfalfa. These findings suggest that CA-mediated Na2CO3 stress alleviation is an ecofriendly approach that would be useful to local farmer for alfalfa and other forage crop cultivation in alkaline soils.

        본 연구에서는 가물치(Channa argus) 추출물의 신경세포 분화와 산화 스트레스에서의 효능을 분석하기 위하여 녹차와 효소를 이용한 다양한 추출 방법(상온 추출물, RE; 녹차 상온 추출물, GRE; 효소 상온 추출물, ERE; 녹차 효소 상온 추출물, GERE)을 사용하여 제조 된 추출물의 아미노산 조성과 항산화 활성을 비교 분석하였고, 신경성장인자 (NGF) 유도 신경세포 분화 및 과산화수소 처리에 의해 유도된 PC12 세포 독성에 대한 보호효과를 규명하고자 하였다. 총 아미노산 함량은 RE 및 GRE보다 효소 추출물인 ERE 및 GERE에서 훨씬 더 높았다. 효소 가수 분해물 (ERE 및 GERE)에서 ABTS 라디칼 소거 활성은 RE 및 GRE보다 높았다. 또한, RE와 ERE는 PC12 세포에서 neuronal growth factor (NGF) 매개 신경 돌기 성장뿐만 아 니라 growth associated protein (GAP)-43 및 synapsin-1의 발현을 현저하게 향상 시켰다. 과산화수소(H2O2)에 의해 손 상된 PC12 세포에 4가지 유형의 Channa argus 추출물을 첨가한 후 PC12 세포의 생존율을 측정하였다. PC12 세포 의 생존율은 RE, GRE, GERE에서 각각 77.5±1.9%, 84.0±0.8%, 81.1±0.9%이였다. 이러한 세포 생존율은 H2O2 만을 처리 한 음성 대조군(70.0±2.0%)에 비해 더 높았다. H2O2 처리에 의해 유도 된 세포 독성도 RE, GRE 및 GERE 처리에 대한 반응으로 상당히 완화되었다. 종합하면, Channa argus 추출물은 산화 스트레스와 신경 손상을 감소시키는 기능성 물질로 유용하다는 것을 시사하며, 향후 이들 소재를 활용한 다양한 기능성 제품의 개발이 필요할 것으로 판단된다.

        Larvae from the marine medaka fish Oryzias javanicus were exposed with polystyrene microplastics (MPs) for 24 h. Exposure to waterborne fluorescent MPs showed clear ingestion and egestion in feces. Under constant MPs, the concentration of dissolved oxygen significantly decreased in 24 h compared to the control. Significant intracellular reactive oxygen species and malondialdehyde contents were detected in larvae, indicating oxidative stress and lipid peroxidation. Significant elevations in mRNA expressions of heat shock protein 70 and antioxidant defense system genes (glutathione reductase, glutathione peroxidase, catalase, and superoxide dismutase) were measured with increases in enzymatic activity of oxidative stress-related proteins. Taken together, the alterations to the molecular and biochemical components suggested that waterborne MPs had an oxidative stress effect on marine medaka larvae.

        Dimethachlor is a synthetic herbicide, belonging to the chloroacetanilide group, that inhibits the undesirable growth of weeds via the suppression of very longchain fatty acid synthesis. Although dimethachlor has been shown to run off from agricultural fields into aquatic ecosystems, the toxicity of dimethachlor on aquatic invertebrates and vertebrates is unknown. In our study, we assessed the toxicity of dimethachlor on developing zebrafish embryos by analyzing viability, hatching ability, and phenotypic changes. Embryonic viability decreased from 48 h post-fertilization (hpf) at the highest concentration of dimethachlor. Decreased hatching ratio, shortened body length, and pathological changes in the eye, heart, and yolk sac were observed at sub-lethal concentrations. Additionally, dimethachlor increased the number of apoptotic cells and level of reactive oxygen species 120 hpf. Our results indicate that dimethachlor may act as an anti-developmental toxicant when accumulated in an aquatic environment.

        This study examined the bioactive compound content and the antioxidant activities of bitter melon (Momordica charantia Linn.) leaves. The content of vitamin C, beta-carotene, and total carotenoids was 69.77, 45.68, and 65.08 mg/100 g, respectively. To investigate the antioxidant capacity, bitter melon leaves were extracted using various concentrations of ethanol (60, 80, or 100%). Highest content of total polyphenols (18.07 mg gallic acid equivalent/g) and flavonoids (4.53 mg cathechin equivalent/g) was found in the 100% ethanolic extract of the leaves (E100). Also, the E100 extract showed the highest levels of 2,2'azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and α-α-diphenyl-β-picrylhydrazyl (DPPH) free radical scavenging activities. Reducing power was also the highest (39.21 mg Trolox equivalent/g) in E100 extract. The E100 extract effectively inhibited lipid peroxidation by 91.45% compared to the control group. Also, the E100 extract showed a cytoprotective effect against oxidative stress in HepG2 cells and decreased the generation of intracellular reactive oxygen species. These results suggest that bitter melon leaves could be regarded as a potential source of natural antioxidants.
