곤충병원성 선충은 인체나 환경에 매우 안전하며 표적해충에만 기생하여 치사시키고, 작물생육에는 전혀 피해를 주지 않는 대량생산이 가능한 환경친화적 천적자원으로 현재 우리나라에서는 7종이 분포하고 있다. 곤충병원성 선충은 인공영양배 지원(in vitro)과 곤충자원(in vivo)을 이용하여 대량 증식하고 있는데, 인공영양배지원을 이용할 경우 누대배양에서 병원성이 다소 떨어지는 경향을 보이나 대량증식이 용이하다. 곤충자원을 이용할 경우 병원성은 매우 높으나 기주곤충의 대량 확보가 어려울 뿐만 아니라 기주곤충으로부터 이탈하는 선충을 수확하는 일이 쉽지가 않다. 따라서 본 결과는 곤충자원과 버미큐라이트, 목재 칩을 이용하여 곤충병원성 선충을 보다 간편하게 대량 증식시킬 수 있는 방법에 대해 소개하고자 한다. 곤충병원성 선충의 실용적 생산에 적합한 곤충은 꿀벌부채명나방 유충이었으며, 다음으로는 곤충자원의 대량 확보가 용이한 갈색거저리 유충으로 유충 당 선충 500마리 접종 시 98,333마리가 증식되었다. 동애등에는 자원의 확보는 용이하나 선충의 감염과 증식력이 낮아 곤충병원성 선충의 증식용으로 부적합한 것으로 나타났다. 곤충과 버미큐라이트를 이용한 곤충병원성 선충의 입상형 대량증식에서는 버미큐라이트 소립형에 수분 20% 중량이 가장 좋은 것으로 나타났다. 곤충과 목재 칩을 이용한 곤충병원성 선충의 원스톱 증식에서도 선충의 증식력은 매우 높은 것으로 나타났다.

국내에서 곤충병원성 선충(entomopathogenic nematode)을 이용하여 원예작물 해충의 생물적 방제 연구가 본격적으로 시작된 것은 1995년 이후로 초기에는 대표적인 나방류 해충인 담배거세미나방(Spodoptera litura), 파밤나방(Spodoptera exigua), 배추좀나방(Plutella xylostella), 목화바둑명나방(Palpita indica) 등의 살충효과 및 포장 효과시험이 수행되었다. 2000년 이후에는 배추흰나비(Pieris rapae), 도둑나방(Mamestra brassicae), 흰띠명나방(Hymenia recurvalis), 왕담배나방(Helicoverpa armigera) 등 나방류 해충 뿐만 아니라 벼룩잎벌레(Phyllotreta striolata), 좁은가슴잎벌레(Phaedon brassicae) 등 잎벌레, 작은뿌리파리(Bradysia difformis), 긴수염버섯파리(Lycoriella mali) 등 근권 및 버섯파리에 대한 실내 및 포장시험에 관한 연구가 활발하게 이루어졌고, 꽃노랑총채벌레(Frankliniella occidentalis) 번데기, 복숭아혹진딧물(Myzus persicae) 등 미소해충 에 대한 살충력 시험도 수행되었다. 그리고 곤충병원성 선충의 토양 처리에 의한 뿌리혹선충(root-knot nematode)의 저지효과에 대한 연구도 수행되었다. 또한 곤충병원성 선충의 다른 천적에 대한 영향과 복합 처리에 의한 방제의 상승효과에 관한 연구가 수행되었거나, 진행되고 있다. 가장 최근에는 곤충병원성 선충과 유아등을 활용하여 블루베리의 뿌리를 가해하여 피해가 많은 풍뎅이를 효과적으로 방제하였다. 따라서 현재 다양한 작물 및 해충의 생물적 방제에 이용되고 있는 곤충병원성 선충의 원예작물 해충 방제 사례를 소개하고, 향후 생물농약으로서의 이용 확대방안에 대하여 발표하고자 한다.

A nematode, which showed entomopathogenicity, was isolated from Exomala orientalis in Korea, and then identified as Rhabditis blumi. Pathogenicity of R. blumi was evaluated against major cruciferous insect pests, including Artogeia rapae, Mamestra brassicae, and Plutella xylostella. In Petri-dish tests, insect mortality was dose and time dependent, which increased with dose (0–80 dauer juveniles/larva) and time increments. In greenhouse tests, P. xylostella larvae were most susceptible to nematodes, with insect reduction rate of 88.0 %. The rate varied with vegetable species and persistence time of live nematodes on vegetable leaves after spraying. The aeration rate was an important parameter for cultivation of the nematode R. blumi. A sufficient DOT level in the culture using an air-lift bioreactor should be maintained. The nematode growth rate increased with an increasing DOT level and/or bacteria uptake rate. The maximum nematode yield of 1.75 x 105 per ml was obtained under an aeration rate of 6 vvm.

Parasitism of horsehair worm in mantids were investigated for two years, from 2012 and 2013. The mantids were collected from forty-nine sites from August to November. Twenty-one sites (42.9%) of the collected mantids were parasitized by horsehair worms. C. japonensis was recorded from T. angustipennis, T. aridifolia and C. fukuii. The C. japonensis parasitism of T. angustipennis and T. aridifolia were 27.0% and 10.7%, respectively. The infested number of C. japonensis was 1-9 per host. The body length of C. japonensis showed tendency to decrease with number of parasites, but not significantly different. Horsehair worms did not affect body, abdomen length and digestive system of host. However, egg possession of host was different, unparasitized T. angustipennis and T. aridifolia had more eggs (63.7%) than parasitized ones (2.6%). C. japonensis was parasitized as immature stage in late Aug. immature and adult stage in early Sept. and only adult in late Oct. Parasitism was the highest in late Sept.

Various insect pest damage agricultural crops and forest. Several control strategies have been tried against these pests. Using chemical pesticides is one of the major control strategies. However, in recent years, eco-friendly control measures have received a big attention. Entomogenous nematodes are best known as potential biocontrol agent which are promising alternatives of chemical pesticides. During the survey of insect-parasitic nematodes as well as their parasitism in Korea, three new species, Parasitylenchus cyrtogenii n. sp., Parasitylenchus oscavus n. sp., Bathymermis sancheongensis n. sp. and seven other new records for the Korean nematode and nematomorpha fauna were identified, which are: Howardula phyllotretae, Parasitaphelenchus uncinatus, Hexamermis albicans, Hexamermis sp., Heterorhabditidoides chongmingensis, Pristionchus pacificus, and Spinochordodea tellini, respectively. Parasitism of nematodes ranged from 0.7 to 87.5%. The highest parasitism was recorded in P. cyrtogenii n. sp isolated from Cyrtogenius brevior as 87.5%, however the lowest parasitism found in Hexamermis sp. isolated from Oxya chinensis as 0.7%.

Entomopathogenic nematodes (EPNs) of the genus Steinernema are pathogenic to the insects and well known as ideal models for understanding parasite-host interaction. EPNs harbor a number of bacterial symbionts in their gut belonging to the noble genus Xenorhabdus which are capable of killing insects by themselves or by combination with nematodes by suppressing insect immune defense. Here, we report host range of Steinernema monticolum and its symbiont Xenorhabdus hominickii. S. monticolum has a diverse host range including lepidopteran and coleopteran insects although they showed higher pathogenicity to the lepidopteran insects. Especially, X. hominickii suppressed insect immune responses. A target insect, Spodoptera exigua, exhibited both cellular and humoral immune responses by expressing antimicrobial peptides and forming nodules in response to heat-killed X. hominickii. However, live bacteria significantly suppressed the immune responses. An addition of arachidonic acid to the bacterial infection significantly rescued the immune responses, suggesting eicosanoid biosynthetic pathway as a pathogenic target of X. hominickii.

The genus Apolygus China (Hemiptera: Heteroptera: Miridae), comprising 64 species in the world, is one of the most diverse groups in the subfamily Mirinae, including well-known insect pests such as A. spinolae and A. lucorum. A total of 56 species are known in the Palaearctic Region up to date, and most of them have been described from China (28 species), Japan (26 species) and Russia (13 species) respectively. In this talk, taxonomic study on the genus Apolygus from the Korean Peninsula is presented with three Apolygus sp. nov. and two newly recorded species. Photographs and illustrations of diagnostic characters and biological information such as host plants and distributions for each species, and a key to the Korean species are provided. Besides, COI barcoding as a diagnostic tool for this group is discussed emphasizing on the low genetic distances between some species.

A mycoinsecticide, Beauveria bassiana (strain GHA) marketed as BotaniGard® ES, was evaluated as a biological control agent against Riptortus pedestris (Fabricius) (Hemiptera: Alydidae) a major soybean pest. It was tested on commercial soybean fields with one time spray of both high (1×109) and medium (1×108spore/ml) concentration and double spray of the medium concentration. Nymph population decreased in all treated fields regardless of dose and frequency of spray. But, adult and egg populations were not affected. Alive nymphs collected from the fields and kept in laboratory showed infection rate ranging 35-68% in treated plots while 0% in control plots. It also reduced damage on soybean pods and seeds by 17.7-21.2 and 18.0-22.5% compare to control, respectively. Additionally, B. bassiana reduced other pests found in soybean fields including brown marmorated stink bug Halyomorpha halys (Stål) and two striped leaf beetle Medythia nigrobilineata Motschulsky. Also there was no negative effect on the parasitism of released non-viable eggs of R. pedestris. These results suggest that, in field condition, one time spraying of B. bassiana in the concentration of 1×108spore/ml can help reducing crop damage as well as R. pedestris nymph population.

Ionizing irradiation can be used as an alternative to chemical fumigants for disinfestation of cut flowers, agricultural products, seeds, foods, medical products, and spices. In this study, we investigated the effect of electron beam irradiation on reproduction and development in Spodoptera litura. When irradiated to the adults, there was no difference in fecundity. However, egg hatching was considerably decreased. When irradiated to the pupae, fecundity was decreased as dose increased and wing deformity of newly emerged adults was increased as time passed. When irradiated to the larvae, developmental period and pupa deformity was significantly increased. In order to investigate the molecular mechanism of sterility and abnormal development, we performed quantitative real-time PCR and SDS-PAGE.

The subfamily Prostemmatinae (Hemiptera: Heteroptera: Cimicomorpha: Nabidae) is known as one of the predatory groups in true bugs, containing 5 genera and approximately 33 species in the Palaearctic region. All Prostemmatinae are ground-inhabiting (in litter layer, under stones, etc.), whereas most Nabinae are herbicolous and arboricolous. In Nabidae, most species are predators on various stages of insects (e.g., aphids, leafhoppers and lepidopteran eggs and young caterpillars), but several genera within the subfamily Prostemmatinae (e.g., Prostemma, Alloeorhynchus and Pagasa) are specialized predators of true bugs, especially of the family Lygaeidae. In this study the subfamily Prostemmatinae is revised from Korea. Among the four species recognized in this study, one species is a new record to Korean fauna, and Alloeorhynchus sp. nov. is described from the Chungcheongnam-do Province, South Korea. The morphological information such as description and diagnoses, and biological information for each species are provided.

Alfalfa weevil is an invasive pest of several leguminous plants. In the world preservative, there is a lack of information about the level of differentiation among separate populations. We ran a comparative molecular and ecological analysis to assess diversity, phylogenetic relationship and also simplify the taxonomic decisions. By analyzing a total of 147 specimens from Middle East, Europe and America plus a few sequences in GenBank we improved our current knowledge of world phylogeography of Hypera postica. The result of mitochondrial genes (Summation of COI and CytB) revealed a deep divergence between two previously defined clades (5.6 – 6.7% nucleotide divergence) within sympatric distribution. The Eastern lineage comprised 60 haplotypes in Iran and 3 haplotypes in Italy with respectively 0.07 – 2.2% and 0.2 – 3.4% sequence divergence range. On the other hand, the Western lineage which was detected in Poland, Czech and Western America had relatively lower haplotype diversity presenting a total of 11 haplotypes and 0.1%-1.1% sequence divergence. The coexistence of two lineages in some regions led to the huge overlap in their fundamental nich, but also a certain level of divergence can be detected via ecological nich modeling analysis.

Riptortus pedestris is a major agricultural pest in Korea. To manage this pest, it is necessary to understand the complete life history of R. pedestris. However, most studies have focused on the summer period when the pest attacks crops. Therefore, we conducted laboratory and field experiment to identify overwintering behavior of adult R. pedestris. First, we conducted two laboratory experiments to examine overwintering habitat preferences and spatial distribution of R. pedestris in the habitat. Second, we conducted field survey to characterize overwintering sites of R. pedestris in diverse landscapes. In the laboratory conditions, R. pedestris almost exclusively showed overwintering behavior in the leaf litter arena (63%) among the structures tested (rocks: 3%; rotten wood: 0%). And spatial distribution of R. pedestris showed solitary behavior in leaf litters. In the field, we located 12 individuals of overwintering adults from the urban areas, whereas no individuals were found from the high-elevation mountains and agricultural landscapes. Our results suggest that R. pedestris mainly use leaf litter as an overwintering structure, with solitary behavior, and low-elevation forested areas near urban landscapes seem to be more preferred overwintering sites.

Leafhopper subfamily, Typhlocybinae (Hemiptera: Auchenorrhyncha: Cicadellidae), caused direct damage by sap-sucking or indirect damage by transmission of viruses or diseases on the host plants, so typhlocybinae is one of the important pest groups in the world. Typhlocybinae distribute worldwide and a total of 471 genera and more than 4,300 species have been described. so far, 38 genera and 102 species were recorded in the Korean Peninsula. In this talk, we provide several taxonomic studies for the Korean typhlocybene fauna based on the genus-level reviews of the subfamily from the Korean Peninsula.

Information on spatial distribution and peak occurrence is important for site and time specific pest management. So, we investigated seasonal occurrence and edge distribution of major soybean pests by visual count in six commercial fields located in Andong from July 16 to October 23 in 2015. We found a total of 19 species in 10 families. The major pest during vegetative stage was Medythia nigrobilineata (Motschulsky) while Riptortus pedestris (Fabricius) and Halyomorpha halys (Stal) showed peak occurrence during reproductive stage of soybean. Aphis glycines Matsumura was found throughout the season. Seasonal density of M. nigrobilineata was 0.20 and 0.38/plant in edge and interior without significance, respectively. The infestation of A. glycine was also found similar between edge and interior. Immatures of R. pedestris and H. halys distributed more in edge (0.21 and 0.36/plant) than interior (0.12 and 0.12/plant), respectively. The application of these informations on spatial distribution pattern of soybean pest was discussed.

The larvicidal activities and repellency of Magnolia kobus extract against C. pipiens and Ae. albopictus were examined. M. Kobus methanol extraction showed 100% larvicidal activity both two mosquitoes at 500 ppm after treated 24 h. At a concentration of 50 ppm, the hexane fraction showed 100% and 94% mortality against C. pipiens and Ae. albopictus at 24 h, respectively. Purification of the biologically active constituents from the hexane extraction with larvicidal activity was done using silica gel open column chromatography and HPLC. H2 fraction gave >90% mortality to both mosquitoes at 10 ppm after treated 48 h. H21 fraction was determined 100% larvicidal activity to C. pipiens at 10 ppm. However, very low larvicidal activity was obseved to Ae. albopictus. This is considered a synergistic effect between substances. It also appears that there is a susceptibility difference of substance against mosquito species. H21 fraction was analyzed as the lignan, sesamin (C20H18O6) by GC-MS and NMR. Mosquito repellent and olfactory response was using a patch test.

Plant-feeding insects are a ubiquitous feature of terrestrial life. It follows that understanding the evolution of insect herbivory and its diversity is fundamental to understanding the evolution of terrestrial ecosystems. During my Ph. D. study, I found phylogenetic conservatism in the feeding mode (internal vs. external feeding) of Yponomeutoidea, one of the primitive lepidopteran superfamilies, as well as in their diet breadth and in the growth form and taxonomic affinities of host plants they used. In spite of their importance in terrestrial ecosystems, the evolutionary history of Lepidoptera has been poorly known. This mostly is attributable to their poor fossil record. My Ph. D. study provided better understanding to the lepidopteran fossils. Fossils are an essential resource in divergence time estimation using molecular clock methods. Once reliably resolved, dating data can be used in tracing the evolutionary history of Lepidoptera/plant associations. I introduce my works to take such advantages from fossils and phylogenetic studies.

The first record of Melon thrips, Thrips palmi Karny, was in 1993 in Korea, and the species has become severe pest in agricultural industry. We used two different SDMs(Species Distribution Model) which have different approaches to analyse potential distribution of the pest species in climate change scenario, MaxEnt and CLIMEX. The MaxEnt model uses historical occurrence records with environmental variables to estimate the realized niche, and CLIMEX model simulates the fundamental niche of the object based on the seasonal phenology. In MaxEnt simulation, we reduced the number of variables to avoid multi-collinearity problem until we had no pairs with an absolute Pearson correlation coefficient higher than 0.8. BIO1(Annual Mean Temperature), BIO2(Mean diurnal range), BIO3(Isothermality), BIO4(Temperature seasonality) were finally selected as predictor, and we used 10 fold cross validation option to replicate. The averaged results were used to index analysis. The CLIMEX results, The Ecoclimate Index(EI), were also normalized in 0 to 1 scale to analysis. Under RCP 8.5 climate change scenario, in 2070s, the distribution of Thrips palmi was predicted to expand their territory overall agricultural area in Korea.

Arctiinae are one of the most species-rich subfamily in the family Erebidae (or classified as family Arctiidae). Around 11.155 species of 750 genera have been described from all over the world (Heppner, 2005) of which 5.931 inhabit the Neotropics (Watson & Goodger, 1986; Heppner, 1991; Scoble, 1995). Taxonomic study of the Arctiinae in Cambodia has been done by a few foreign entomologists. As the results of this study, 182 species of 68 genera are reported on, of which described in recent pages 135 species of 44 genera are reported from Cambodia for the first time, five species are new to science (Bayarsaikhan & Bae, 2015; Bayarsaikhan et al., 2015; Bayarsaikhan & Bae, 2016): Danielithosia wooshini; Stictane cambodiensis; S. khmerensis; S. bokorensis; Cyana angkorensis. The materials for identification are based on the collection of Incheon National University, which were collected from 2009-2015 in Cambodia by Cambodian and Korean researchers.

The penetration-enhancing effect of a binary mixture of rosemary essential oil constituents, and its correlation to increased toxicity were investigated. GC-MS analyses revealed that the major constituents of rosemary essential oil were 1,8-cineole and camphor, and a binary mixture of them showed synergistic insecticidal activity via topical application in third instar larvae of the cabbage looper, Trichoplusia ni. In the mixture, penetration of camphor was significantly enhanced both in vivo and in vitro, whereas that of 1,8-cineole did not change much compared to individual application. Injection into hemocoel of fifth instar larvae indicated that camphor was more toxic than 1,8-cineole, although it was less toxic than 1,8-cineole when applied topically. A possible mechanism for enhanced penetration of camphor by 1,8-cineole is reduced surface tension of the mixture. Contact angles of 50% acetonic solutions on a layer of beeswax showed that the surface tension of 1,8-cineole was lower than that of camphor. Another probable reason is a change of camphor from solid to liquid status, when mixed with 1,8-cineole. This is the first report attempting to explain the synergistic insecticidal interaction of essential oil components, from a physiochemical perspective.

For reviewing Korean species of Denticollinae and reconstructing phylogenetic relationships between subfamilies in Elateridae, three kinds of studies were conducted: 1) taxonomic review based on morphology; 2) DNA barcode for Denticollinae; 3) molecular phylogenetic study of Elateridae using 12 genetic markers. In taxonomic study, a total of 54 species were reviewed including 13 new species and then unrecorded species. In DNA barcoding, six cryptic and/or pseudocryptic species were confirmed by their sympatric or allopatric distribution. We propose a conservative threshold of 3.6% for convenient molecular operational taxonomic unit identification in Denticollinae. From this study, we confirmed that COI sequence is useful for reassessing species diversity for polymorphic and polytypic species occurring in sympatric and allopatric distributions. In molecular phylogenetic study, the phylogenetic inference constructed robust phylogeny with almost very strongly supported nodal values(>90%) and showed that Hypnoidinae, Oxynopterinae and Denticollinae are monophyletic; Elaterinae, Hemiopinae and Melanotinae are a monophyletic group; and two tribes in Denticollinae are paraphyletic, it is needed to reclassify their systematic accounts.
