        EP was obtained through 20% ethanol extraction of Pueraria lobata root, and the fermented form of EP, FEP, was prepared from the EP after incubating with Lactobacillus rhamnosus vitaP1. There was no significant toxicity by EP and FEP up to 1000 ㎍/㎖ in NIH-3T3, HaCaT, and B16F10 cells. In addition to antioxidant potentials of EP and FEP determined by DPPH and ABST assays, we confirmed increase of procollagen type I and elastin synthesis by supplementation of the EP and FEP at the concentration of 50 ㎍/㎖ using ELISA kits. The protein expression levels of matrix metalloprotease (MMP)-1, -3, and -9, those are involved in the degradation of collagen or other skin matrix proteins, were remarkably suppressed while their inhibitory protein metallopeptidase inhibitor 1 (TIMP-1) was greatly up-regulated by supplementation of the EP and FEP at a concentration of 50 ㎍/㎖. Taken together, both EP and FEP supplementation could be involved in the suppression of the skin wrinkle formation through inhibiting degradation of collagen and stimulating the synthesis of collagen and elastin. The results showed that the anti-wrinkle potential of the EP and FEP will be a promising candidate for developing cosmeceutical compounds or products.

        Background : Heat stress induced from high temperature are known to crucially affecting on physiological properties and yield in Cnidium officinale. Methods and Results : The effect of foliar application of mixture including a urea, ascorbic acid and calcium chloride on high temperature injury of Cnidium officinale. Photosynthesis and leaf temperature in Cnidium officinale were investigated after cultivating for 24 hours at 35℃. Net photosyntheis rate, transpiration was measured at 1,000 μmol m-2 s-1 of photon flux density and leaf temperature was analyzed by thermal image. Net photosyntheis rate, stomatal conductance and transpiration rate in mixture traetment were 2 times of higher than in control. Water use efficience was not different significantly. Leaf temperature was lower in mixture treatment (25.3℃) than in control (29.0℃). Conclusion : This result show that foliar application of urea, ascorbic acid and calcium chloride was reducing a high temperature injury through a improving photosynthetical capacity and decreasing leaf temperature of Cnidium officinale.

        Background : Management of air temperature are known to primarily affecting on physiological properties and yield in plant. Methods and Results : The effect of air temperature on characteristics of photosynthesis and chlorophyll fluorescence in Cnidium officinal were investigated using growth chamber after cultivating for 24 hours under controlled condition. Net photosyntheis rate, transpiration was measured at 1,000 μmol m-2 s-1 of photon flux density and chlorophyll fluorescence was analyzed by OJIP method. Net photosyntheis rate was highest in treatment of 25℃. Although transpiration rate was lowest, water use efficience was also in treatment of 25℃. Stomatal conductance was mainly influenced from ambient climatric factors such as vapor pressure deficit. As results of chlorophyll fluorescence by OJIP analysis, maximum quantum yield (Fv/Fm) of photosystem II (PSII), PIabs and the relative activities per reaction center such as ABS/RC, DIo/RC were not changed at air temperature. Therefore, elevated air temperatue during short term influence the dark reaction in photosystem through controlling a water use efficience and transpiration. Conclusion : This result show that 25℃ of air temperature may be a adequate temperature to improving the efficiency of photosynthesis in Cnidium officinale.

        Background: Mahonia Nepalensis DC. (Hoang lien o ro), the specie of the family Berberidaceae, is widely distributed in the high mountainous areas at altitudes 1700 – 1900 m of Vietnam. It is found that the stem of Mahonia nepalensis indicated anti-inflammatory, anti-bacterial and antifungal activities and they are used particularly for the treatment of eczema, psoriasis, and other skin conditions. However, no study on the antioxidant and anti-cancer activities of Mahonia Nepalensis stem has been previously reported. The aim of this study was to evaluate anti-oxidant and anti-cancer activities of Mahonia Nepalensis stem. Methods and Results: The stem pieces of Mahonia Nepalensis were dried and extracted three times with 100% methanol. After that, the extract was suspended in distilled water and then partitioned with n-hexane, ethyl-acetate (EtOAc) and butanol (water saturated BuOH) fractions were then evaporated using a vacuum rotary evaporator. Evaluation of the anti-oxidative activity of Mahonia Nepalensis was carried out using a DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-producing system. The results revealed that the ethyl acetate fraction of M. nepalensis possessed higher potential DPPH radical scavenging activity (IC50, 81.88 ± 1.33㎍/㎖) than other fractions as well as BHT (2,6-Di-tert-Butyl-4-methylphenol) (IC50, 250.49 ± 1.60㎍/㎖). The reducing power assay was also investigated and EtOAc fraction showed higher absorbance values than other fractions. At 1.0 mg/ml concentration, EtOAc fraction showed absorbance of 1.72, be higher than Ascorbic acid. Cell viability was evaluated according to the MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyl tetrazolium Bromide) assay. By MTT assay, all fractions showed a significant reduction in cell viability on COLO 205 (Human colon carcinoma cell) at the highest concentration tested (200㎍/ ㎖) with over 70% decrease in cell viability was obtained, and the highest significantly inhibiting effect occurred in butanol fraction with approximately 90% reduction in cell viability. Conclusion: We demonstrated that Mahonia Nepalensis stem extract has highly potential in anti-cancer activity. Further studies are necessary in order to explore the variety of Mahonia Nepalensis stem to be applied as a valuable natural material.

        Background : Haskap berries commonly refer to fruits of Lonicera caerulea L., recognized by the Japanese aborigines as the “The elixir of life.”. Due to their recent arrival on the North American market, haskap berries have not yet been positioned among other berries and compared in terms of their phytochemical content. And haskap berries have higher ascorbic acid and anthocyanin content than other berries known for their health-promoting benefits, such as blueberries. However, no study has reported on the antioxidant and anti-cancer activity of Lonicera caerulea stem. The purpose of this study is to present the current research on the chemical content, antioxidant and anti-cancer activities of Lonicera caerulea stem. Methods and Results : The stem of Lonicera caerulea L. ware dried in the shade at room temperature and extracted with 100% methanol. The extract was suspended in deionized water and partitioned sequentially with n-hexane, chloroform, ethyl-acetate and butanol (water saturated BuOH) fractions. Antioxidant activities were measured by determination of antioxidants, DPPH (2,2-diphenyl-1-picrylhydrazyl). Cell viability was determined by the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assay. All cell lines were purchased from the Korean Cell Line Bank (Seoul, Korea). All results were performed with three replications were processed statistically. By DPPH assay, the Lonicera caerulea L. the highest activity was obtained from the ethyl-acetate fraction (IC50=15.46 ㎍/㎖). By MTT assay, the chloroform fraction showed a significant growth inhibiting effect on MCF-7 (Human breast cancer, IC50=225.91 ㎍/㎖), COLO 205 (Human colon cancer, IC50=179.55 ㎍/㎖), but on AGS (Human stomach cancer) and other fractions it did not show effect. Conclusion : We demonstrated that Lonicera caerulea L. stem extract and fractions has antioxidant and antiproliferation activity in vitro. Further studies should identify the active constituents in Lonicera caerulea L stem to evaluate the potential in vitro antioxidant and antiproliferation activities of the extract.

        Background : Astilboides tabularis (Hemsl.) Engl. is a perennial herbaceous plant, distributed in the northern high mountains of the Korean peninsula and China. It is an excellent ornamental plant currently at risk of overharvesting and therefore, is designated as an endangered wild plant Class II by the Ministry of Environment. Physiological research on A. tabularis has not be reported. Therefore, in this study, using A. tabulari extracts, antioxidant and Anti-inflammatory effects were determined. Methods and Results : The antioxidant and free radical scavenging activities of A. tabularis extracts were evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. The results showed that the ethyl acetate fraction of A. tabularis possesses potent DPPH radical scavenging activity (2.90±0.08㎍/㎖), similar to the scavenging activity of ascorbic acid (2.19±0.06㎍/㎖), and better than the powerful antioxidant α-tocopherol (10.60±0.40㎍/㎖) as well as BHA (butylatedhydroxy anisole)(6.12±0.27㎍/㎖). The ethyl acetate fraction possessed a significantly higher concentration of total phenolic (549.70±2.72㎎GAE/g) and flavonolic content (154.58±1.04㎎QE/g). It was also found that the ethyl acetate fraction exhibited high reducing power and inhibition of ROS (Reactive Oxygen Species) formation. Different fractions of A. tabularis were tested for anti-inflammatory activity using LPS stimulated Raw 264.7 cells. The n-hexane and ethyl acetate fractions exhibited a high inhibitory effect on NO (Nitrite oxide) production (22.43±1.06%, 19.30±0.45%, respectively) at 200㎍/㎖ concentration. The mRNA of IL-1β, iNOS and COX-2 gene expression was decreased by treatment with the ethyl acetate fraction. These results showed that A. tabularis extracts can be used as natural substances to control inflammation. Conclusion : These result showed that A. tabularis extracts can be used in a variety of antioxidant and other functional product research and development processes as valuable natural materials.

        Background : We studied the anti-oxidant activity and anti-inflammatory effects of Rhododendron lapponicum (L.) Wahlenb. var. parvifolium (Adams) Herder extract (RLE). Methods and Results : The RLE was prepared using methanol. The antioxidant effects of RLE was evaluated for its DPPH (1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity, reducing power. Subsequently, using the RAW 264.7 cells, the cell viability of RLE was evaluated with or without LPS (lipopolysaccharide), and the anti-inflammatory effects of RLE was also estimated by nitric oxide (NO) and using real-time polymerase chain reaction (RT-PCR). The extract showed antioxidant activity (DPPH free-radical scavenging activity) with RC50 value of 57.67 ㎍/㎖. The reducing power of the extract was Abs 0.77 at 250 ㎍/ ㎖. The result indicated that RLE would have significantly high anti-oxidative effects. Cell viability was determined using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. To evaluate anti-inflammatory activity, we examined the inhibitory effects on LPS-induced NO production in RAW 264.7 cells. The NO inhibition rate was 85.44% at 200 ㎍/㎖ RLE. At the same concentration, the expression of pro-inflammatory genes such as inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 also decreased. In RLE 50 ㎍/㎖ concentration showed the highest decrease. Conclusion : This result suggest that RLE is a novel resource for the development of foods and drugs that possess anti-oxidant and anti-inflammatory activity. Also, RLE can be developed as an inflammatory agents for cosmetic bases in the future.

        Background : This study aimed to investigate the antioxidant and anti-inflammatory activities of water chestnut (Trapa japonica Flerow) extract. Methods and Results : The antioxidant and anti-inflammatory activities of 100% methanol extract of water chestnut were investigated. The methanol extract was evaluated for its total phenolic and flavonoid content, DPPH•(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity,reducing power, andeffect on nitric oxide (NO) production and cell viability using real-time polymerase chain reaction (qPCR). The total phenolic content was 438.31 ㎍ allic acid equivalent (GAE)/㎎ extract and the total flavonoid content was 61.40 ㎍ quercetin equivalent (QE)/㎎ extract. In addition, results revealed the extract possessed antioxidant activity (DPPH• free-radical scavenging activity) with IC50 value of 5.28 ㎍㎖ The reducing power of the extract was assayed spectro photometrically and showed Abs of 0.71 at 100 ㎍㎖ Furthermore, extracts of water chestnut exhibited no cytotoxicity in RAW 264.7 cells. In addition, the NO assay revealed that LPS-induced NO production was significantly inhibited following treatment with water chestnut extracts. The expression of pro-inflammatory proteins such as inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 decreased in a concentration-dependent manner. The water chestnut extract also decreased tumor necrosis factor α (TNF-α) release. Conclusion : Therefore, the present findings provide scientific evidence for the nutritional potential, chemical composition, and biological activities of Trapa japonica Flerow anddemonstrate its potential use as a functional food forapplication in the pharmaceutical industry

Therefore, to derive the electricity production efficiency due to organic concentration and ammonia nitrogen concentration in order to apply a microbial fuel cell (MFC) livestock wastewater treatment process in this study, and to derive the energy production potential with livestock waste water through the study. Lab. scale Reactor fabrication and operation to try to derive the reaction factor of the optimum operating conditions in accordance with the livestock wastewater applied through the evaluation of trends and removal efficiency of organic matter and nutrients in the microbial fuel cell. In addition, from the final research results, I try to present the direction of future research for the improvement of application possibilities and microbial fuel cell power generation efficiency of microbial fuel cell in the livestock wastewater treatment facilities.

        Recently there are increasing numbers of people participating in sports activities. Accordingly, the rate of sports injuries is increasing as well. In this light, this study was conducted to investigate the extent of acute damage and its forms by examining the current state of patients with sports injuries. This research study was conducted between January of 2008 and December of 2010 in Chosun University Hospital through retrospective review of the medical records of patients with sportsrelated injuries. The total number of patients with sports injuries who visited the emergency center was 887. The number of male patients was 726 (81.85%), far more than the number of female patients (18.15%). More than half of the patients, accounting for 419 (47.23%), had injuries on their face; second and common parts, in which injuries occurred, were upper and lower body, accounting for 146 (16.45%) and 193 (21.75%), respectively. The majority of patients did not undergo surgical treatment. Only 113 (12.73%) patients underwent surgical treatment. In addition, more than 80% of them were not hospitalized but 170 of them needed to be hospitalized. Most sports injuries occurred in young male patients. This can interrupt participation in sports activities for a long time and result in lower quality of life. Hence proper preventative measures and scientific treatment methods are needed.

        Genetically modified (GM) papaya (Carica papaya L.) line 55-1 (55-1), which is resistant to papaya ringspot virus infection, has been marketed internationally. Many countries such as the European Union, Japan, and Korea have a mandatory safety assessment, approval and labeling regulations for GM foods. Thus, there is a need for specific methods for detecting 55-1. In this study, we established a real-time PCR detection method applicable to 55-1 for a variety of papaya products. The limit of detection was possible for fresh papaya fruit up to dilutions of 0.005% and 0.01% (weight per weight [w/w]) for homozygous SunUp and heterozygous Rainbow cultivars, respectively, in non-GM papaya. The 55-1 event-specific detection method observed parallelism (r2>0.99) between the concentration of line 55-1 cultivars and Ct values obtained in amplification plots at concentrations of 0.005-10% for SunUp DNA and 0.01-10% for Rainbow DNA. The method was applicable to the qualitative detection in various types of processed products (cocktail fruit, dried fruit, juice, etc.) containing papaya as a main ingredient. Monitoring papaya products for the presence of GM papaya were demonstrated using a P35S and T-nos real-time PCR detection method but no amplification signals were detected.

        Mungbean (Vigna radiata) is a fast-growing, warm-season legume crop that is primarily cultivated in developing countries of Asia. We constructed a draft genome sequence of mungbean to facilitate genome research into the subgenus Ceratotropis and to enable a better understanding of the evolution of leguminous species. The draft genome sequence covers 80% of the estimated genome, of which 50.1% consists of repetitive sequences. In total, 22,427 high confidence protein-coding genes were predicted. Based on the de novo assembly of additional wild mungbean species, the divergence of what was eventually domesticated and the sampled wild mungbean species appears to have predated domestication. Moreover, the de novo assembly of a tetraploid Vigna species (Vigna reflexo-pilosa var. glabra) provided genomic evidence of a recent allopolyploid event. To further study speciation, we compared de novo RNA-seq assemblies of 22 accessions of 18 Vigna species and protein sets of Glycine max and Cajanus cajan. The species tree was constructed by a Bayesian Markov chain Monte Carlo method using highly confident orthologs shared by all 24 accessions. The present assembly of V. radiata var. radiata will facilitate genome research and accelerate molecular breeding of the subgenus Ceratotropis.

        Tongil (IR667-98-1-2) rice, developed in 1972, is a high-yielding rice variety derived from a three-way cross between indica and japonica. Tongil contributed to staple food self-sufficiency of Korea, an achievement that was termed the ‘Korean Green Revolution’. In this study, we analyzed the nucleotide-level genome structure of Tongil rice and compared it to those of the parental varieties. A total of 17.3 billion Illumina Hiseq reads, 47× genome coverage, were generated from Tongil rice. Three parental accessions, two indica and one japonica types, of Tongil rice were also sequenced for approximately 30x genome coverage. A total of 2,149,991 SNPs were detected between Tongil and Nipponbare; the average SNP frequency of Tongil was 5.77 per kb. Genome composition based on the SNP data by comparing with the three parental genome sequences on sliding window of Nipponbare genome sequence revealed that 91.8% of the Tongil genome originated from the indica parents and 7.9% from the japonica parent, different from the theoretical expectation in a three-way cross, i.e., 75% indica and 25% japonica parental origins on average. Copy number of SSR motifs, ORF gene distribution throughout the whole genome, gene ontology (GO) annotation, yield-related QTLs or gene locations, and polymorphic transposon insertions were also comparatively analyzed between Tongil and parents using sequence-based tools. The results indicated that each genetic factor was transferred from parents into Tongil in proportion to the whole-genome composition. The Tongil rice is the first successful superior cultivar derived from indica × japonica hybridization in Korea. Defining of genome structure demonstrates that the Tongil genome is composed mostly of the indica genome with a small proportion of japonica genome introgression.         The objective of this study is to analyse the dynamic stable and unstable behaviours of a space truss using an accurate solution obtained by the high-order Taylor method. Because numerical solutions can lead to incorrect analyses in the case of a space truss model due to the being parameters large, we analyse the solution’s behaviour using essentially an analytical solution obtained by the multi-step high-order Taylor method. In detail, the dynamic instability and buckling characteristics of the SDOF model under step, sinusoidal and beating excitations are investigated.

        The flower buds of Syzygium aromaticum (clove) have been used as traditional medicine for the treatment of male sexual disorders in Asian countries. Recently, there are some reports about the effects of the clove on reproductive activities in mammals. Therefore, its effect on testicular function was examined in male golden hamsters whose reproductive activity is inhibited by photoperiod such as winter climate. The male animals were given by daily oral administrations (56 consecutive days) in three doses (4 mg, 20 mg, and 100 mg/kg BW) of the alcoholic extract of the clove. Generally lower dose (4 mg) of the extract continued to keep the reproductive activities of testes. The both middle and high doses (20 mg and 100 mg) of the extract completely inhibited the testicular activity in some animals. Taken together, these results suggest a possible biphasic action of alcoholic extract of Syzygium aromaticum flower bud on testicular function.

        Recently, human mesenchymal stem cells (MSCs) are attracting attention as a useful source for regenerative therapy. Controlled production of cell therapy requires the establishment and management of an accurate isolation, characterization and monitoring for quality assurance of developing MSCs mediated. In this study, we were confirmed maintenance of potency of isolated and cultured human umbilical cord (hUC)-MSCs during ex vivo expansion or after cryopreservation. Expression of their cell specific marker was analyzed by flow cytometry and the differentiation potency was confirmed by guided differentiation of adipocyte, osteocyte, chondrocyte and hepatocyte after expanding over 15 doublings in vitro. Safe production of developing a cell therapy was proved by testing for microbial, mycoplasma, endotoxin, and adventitious agents. Also stability of cells in cultivation, preservation and/or differentiation was determined chromosomal assay. In developing using hUC-MSCs, cells showed an accurate isolation and stable expansion in ex vivo condition. The results of several management assay showed that the stem cell marker expression of CD31, CD34 and CD45 were under 10%, however CD90 was over 90% by FACS analysis. Any contamination and mutation in all tests weren't detected in specific points for safe or stable production of hMC-MSCs. Also the proliferation and differentiation potency maintains during in vitro culture and after cryopreservation of hUC-MSCs. These results could be used as standard methods of maintenance of hUC-MSCs for cell therapy products and clinical application.

        Although much effort has been made to find agronomically important loci in the soybean plant, extensive linkage disequilibrium and genome duplication have limited efficient genome-wide linkage analyses that can identify important regulatory genes. In this respect, recombination block-based analysis of cultivated plant genomes is a potential critical step for molecular breeding and target locus screening. We propose a new three-step method of detecting recombination blocks and comparative genomics of bred cultivars. It utilizes typical reshuffling features of their genomes, which have been generated by the recombination processes of breeding ancestral genomes. To begin with, mutations were detected by comparing genomes to a reference genome. Next, sequence blocks were examined for likenesses and difference with respect to the reference genome. The boundaries between the blocks were taken as recombination sites. All recombination sites found in the cultivar set were used to split the genomes, and the resulting sequence fragments were named as core recombination blocks (CRBs). Finally, the genomes were compared at the CRB level, instead of at the sequence level. In the genomes of the five Korean soybean cultivars used, the CRB-based comparative genomics method produced long and distinct CRBs that are as large as 22.9 Mb. We also demonstrated efficiency in detecting functionally useful target loci by using indel markers, each of which represents a CRB. We further showed that the CRB method is generally applicable to both monocot and dicot crops, by analyzing publicly available genomes of 31 soybeans and 23 rice accessions.

        “Hojoong”, a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA, Iksan, Korea, during the period from 2006 to 2012.         “Baechal”, a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA, Iksan, Korea, during the period from 2006 to 2012. It showed lower gluten content (9.6%), SDS-sedimentation volume (38.2㎖) and amylose content (6.5%) than “Keumkang”. It showed higher high viscosity (643BU), water absorption (213%), expansibility of cooking (357%).
