        농촌진흥청 국립원예특작과학원에서는 2018년 분홍색계 소 형 호접란 ‘Tiny Bell’을 육성하였다. 2010년 진분홍색 다화 성 소형 품종 Phalaenopsis ‘D07PN16’와 연분홍색 소형종 P. ‘D03PN22’를 모본과 부본으로 교배하였다. 2014년 생육이 우 수하고 화색, 화형, 꽃대수 등 개화 특성이 우수한 ‘10531-53’ 개체를 선발하여 기내 화경배양을 통해 증식하였다. 2014년부 터 2018년까지 1차, 2차특성검정을 통해 품종의 안정성과 균 일성을 확인한 후 ‘Tiny Bell’로 명명하였다. 이 품종은 밝은 보라빛 분홍색(RHS, PVGN80A)을 띄며, 가장자리에 백색무늬 를 가지는 것이 특징이다. 평피기 형태의 꽃은 길이와 폭이 각각 3.89, 4.01cm이며, 총상화서로 분지가 발생한다. 1개의 꽃대에 19.7개의 소화가 착생하여 꽃 수가 많이 달리는 다화 성이며, 꽃대 길이는 28.48cm정도이다. ‘Tiny Bell’은 생육시 잎이 수평으로 자라며 길이와 폭은 각각 13.1cm, 5.3cm였다. 신품종인 ‘Tiny Bell’은 소형 분화용으로 이용 가능할 것이다.

        농촌진흥청 국립원예특작과학원에서는 2017년 빅립 형태의 분홍색계 팔레놉시스 ‘Lovely Angel’을 개발하였다. 2009년 백색 팔레놉시스 P. ‘Timonthy’와 빅립을 가지는 분홍색 P. World Class ‘Big Foot’을 모본과 부본으로 교배하였다. 2013년 생육이 우수하고 화색, 화형, 꽃대수 등 개화 특성이 우수한 ‘09056-12’를 개체선발하여 기내 화경배양을 통해 증식하였다. 2014년부터 2017년에거쳐 1차, 2차 특성검정을 통해 품종의 안정성과 균일성을 확인하여 ‘Lovely Angel’을 육성하였다. 이 품종은 밝고 선명한 보라빛 분홍색(RHS, PVG81C)을 띄며, 줄 무늬를 가지고 있고 설판이 큰 빅립 형태가 특징적이다. 안아피는 형태의 꽃은 길이와 폭이 각각 6.1, 5.6cm이며, 1개의 꽃대에 20.6개의 소화가 착생하여 꽃 수가 비교적 많이 달리고, 꽃대 길이는 31.8cm 이다. ‘Lovely Angel’는 생육시 잎은 반하수로 자라며 길이는 18.7cm, 폭은 6.8cm이다. 신품종인 ‘Lovely Angel’은 소형 분화용으로 이용 가능할 것이다.

        The black cutworm, Agrotis ipsilon (Hufnagel, 1766) is a cosmopolitan pest that poses an economic threat to many crops such as pepper, corn, perilla, etc. A. ipsilon can be mass reared by providing Chinese cabbage, Brassica rapa under 60~70%, 16L:8D and 25±1℃ in laboratory. This study presents the mass rearing methods and the morphological characteristics of the immature stages of A. ipsilon including descriptions of the each immature stages.

        The turnip sawfly, Athalia rosae is one of the main pests that damage the leaves of cabbage, radish and other cruciferous crops. The developmental biology and morphological characteristics of the immature stages of Athalia rosae were studied in the laboratory using host plant, Chinese cabbage, Brassica rapa var. glabra. A. rosae can be mass reared in laboratory throughout the year under RH 60~70%, 16L:8D and 25±1℃. This species has six larval stages in the female and five larval stages in the male. The developmental period is about 28~29 days. Ovipositional periods and larval developmental periods were 6, 9 days, respectively. The pupal period was about 14 days. Illustrations and descriptions of the various immature stages and their behaviors are provided.

        The change of phenolic compounds and radical scavenging activity of black soybean flours after germination and roasting treatment were evaluated as a part of the purpose of setting the quality of black soybean flours for each application. The moisture content of roasted black soybean flours decreased significantly according to roasting temperature and time, and the crude ash, protein, and fat contents increased. Water binding capacity of roasted black soybean flours without and with germination increased significantly according to roasting temperature and time, however water solubility index and swelling power decreased. The lightness of roasted black soybean flours was significantly decreased, and the redness and yellowness increased. The phenolic compounds and radical scavenging activity of roasted black soybean flours increase with increasing roasting temperature and time. The total polyphenol contents of roasted black soybean flours without and with germination were 5.43∼7.81 and 4.52∼6.17 mg GAE/g, and total flavonoid contents were 2.90∼3.50 and 2.34∼3.01 mg CE/g, respectively. DPPH radical scavenging activity of roasted black soybean flours without and with germination was 254.98∼415.05 and 171.95∼295.15 mg TE/100 g, and ABTS radical scavenging activity was 459.74∼596.37 and 422.95∼526.85 mg TE/100 g, respectively. As a result, it is necessary to set quality standards for each application considering the quality and antioxidant properties of roasted black soybean flours.

        Honey bee swarming is a natural phenomenon that occurs when the colony encounters changes in the in-hive (i.e. population size and queen condition) and environmental conditions. To better understand the molecular basis of swarming, we conducted the transcriptomic profiles of worker bees between before swarming [pre-swarming colony (PSC)] and after swarming [swarming group (SG) and remaining group (RG)]. Based on the gene set enrichment analysis (GSEA), we predicted the biological processes associated with swarming. In addition, we analyzed the composition of cuticular hydrocarbons (CHCs) by gas chromatography-mass spectrometry and compared their profiles between different bee groups. GSEA results showed that there were a little differences between PSC and RG while many of the pathways related with metabolism and protein processing were down regulated in SG relative to PSC and RG. CHCs profiling revealed a similar CHCs composition between PSC and RG but some differences in CHCs composition (i.e. heneicosane, octacosane, octacosanol) were detected between SG and RG. These differences in gene pathway and CHC composition were discussed with respect to physiological changes and social communication.

        Acetylcholinesterase (AChE) is an enzyme for hydrolyzing neurotransmitter acetylcholine. Soluble form of AChE is generated via alternative splicing and functions as a bioscavenger in Dropsophila melanogaster. In this study, effects of ethanol and acetic acid on the soluble AChE expression were investigated. Treatment of ethanol and acetic acid results in over-expression of soluble AChE in the abdomen in a dose-dependent manner. However, no apparent change in AChE expression was observed in the head. Our finding suggests that the soluble AChE is involved in chemical defense against high concentration of ethanol, which is a common by-product from fermented food,and acetic acid, the main metabolite of ethanol. Thus, high level of ethanol and acetic acid resistance in D. melanogaster appears to be evolved via the induction mechanism of soluble AChE expression.

        Even though klotho deficiency in mice exhibits multiple aging-like phenotypes, studies using large animal models such as pigs, which have many similarities to humans, have been limited due to the absence of cell lines or animal models. The objective of this study was to generate homozygous klotho knockout porcine cell lines and cloned embryos. A CRISPR sgRNA specific for the klotho gene was designed and sgRNA (targeting exon 3 of klotho) and Cas9 RNPs were transfected into porcine fibroblasts. The transfected fibroblasts were then used for single cell colony formation and 9 single cell–derived colonies were established. In a T7 endonuclease I mutation assay, 5 colonies (#3, #4, #5, #7 and #9) were confirmed as mutated. These 5 colonies were subsequently analyzed by deep sequencing for determination of homozygous mutated colonies and 4 (#3, #4, #5 and #9) from 5 colonies contained homozygous modifications. Somatic cell nuclear transfer was performed to generate homozygous klotho knockout cloned embryos by using one homozygous mutation colony (#9); the cleavage and blastocyst formation rates were 72.0% and 8.3%, respectively. Two cloned embryos derived from a homozygous klotho knockout cell line (#9) were subjected to deep sequencing and they showed the same mutation pattern as the donor cell line. In conclusion, we produced homozygous klotho knockout porcine embryos cloned from genome-edited porcine fibroblasts.
