        A recent study reported that Pleurotus ostreatus has the potential to be used as a β-glucan-based cream for supportive complementary therapy of atopic dermatitis. KH054 is a new herbal prescription consisting of P. ostreatus and Panax ginseng. The effects of atopic dermatitis-induced materials on the expression of cytokine genes in human monocytes (THP-1, EoL- 1) have been examined. Some reports demonstrated that P. ginseng augments the activity of natural killer cells, which plays an important role in innate immunity against infection and tumor development. Monocyte chemotactic protein 1 (MCP-1), interleukin (IL)-6, and IL-8 have important roles in mediating the infiltration of various cells into the skin of atopic dermatitis and psoriasis. The present study investigated whether KH054 on induced IL-6, IL-8, and MCP-1 secretion by house dust mite (Dermatophagoides pteronissinus) in THP-1 (human acute monocytic leukemia) and EoL-1(Human eosinophilic leukemia) cell. D. pteronissinus functions in the pathogenesis of allergic diseases, including atopic dermatitis and asthma. The inhibitory effect of KH054 on the induction of IL-6, IL-8, and MCP-1 secretion by D. pteronissinus extract in THP-1 and EoL-1 cells was examined. KH054 potently suppressed the elevated production of IL-6 and IL-8 induced by D. pteronissinus treatment in THP-1 and EoL-1 cells. Based on the present results, KH054 may be useful for developing functional foods to treat atopic dermatitis.

        Somatic cell nuclear transfer (SCNT) has been considered for preserving genetically valuable or endangered animals. Sapsaree is a Natianal Monument in Korea to maintian a pure pedigree. The aim of this study was to produce azoospermia Sapsaree using SCNT and identify normal reproductive ability of cloned azoospermia Sapsaree. Ear skin biopsy was performed on a thirteen-year-old azoospermia Sapsaree and ear skin fibroblasts were isolated for SCNT as donor cells. The fibroblasts were injected into enucleated in vivo matured oocytes, the couplets were electrical fused by two pulse of direct current (55 V for 15 μs) using titanium and platinum fusion needle and activated by calcium ionophore. Cloned embryos were surgically transferred into oviducts of natuarally estrus cycle synchronized recipient dogs. The fusion rate of platinum needle was 70%, which was higher than those of titanium needle (64.1%). Developmental rate to the 8 cells and 10 cell stages was higher in platinum needle group (24% and 16%, respectively) than those of platinum needle group (14.8% and 3.1%, respectively). Total 35 SCNT embryos were transferred into oviducts of 3 recipient dogs and one recipient finally delivered a puppy by caesarean section. As results, this study demonstrated that platinum fusion needle could be successfully make the reconstructed embryos and improve the efficiency of canine SCNT. Cloning azoospermia Sapsaree may contribute to conserve genetically valuable and unique pedigree. And further study should be confirm whether cloned live dog is azoospermia.

        Prolonged communication between oocytes and the surrounding somatic cells is one of the unique reproductive physiology in canine. Paracrine Kit ligand (KITL) signaling is a well-known communication between granulosa cells and the oocyte. KITL is a cytokine growth factor secreted by granulosa cells that signals via the c-kit receptor expressed by oocytes. Paracrine factors, including growth differentiation factor 9 (GDF9) and bone morphogenetic protein 15 (BMP15), exert their effects by binding with the kinase receptors expressed on the granulosa cells. However, the regulations of GDF9 and BMP15 in the canine KITL expression are currently poorly understood. Therefore, we investigated the effects of GDF9 and BMP15 on the expression of KITL in canine ovarian granulosa cells in vitro. In Annexin V assay recombinant GDF9 and BMP15 did not induce apoptosis in the cultured ovarian granulosa cells. When treated, FSH significantly increased KITL expression, and hCG suppressed its expression. When both FSH and hCG were treated, the expression of KITL was affected by GDF9 and BMP15 in dose and time dependent manner in the luteal granulosa cells. GDF9 (10 ng/mL) significantly decreased KITL expression after12 h. BMP15 (10 ng/mL) significantly also decreased KITL expression after 24 h. Western blot and immunochemistry results indicate that GDF9 activated Smad2/3. After blocking ALK 4/5/7 receptors by SB, GDF9 failed to activate Smad2/3, also BMP15 did not activate Smad1/5/8 after blocking ALK 2/3/6 receptors by DM. So GDF9 exerts its effects via using ALK 4/5/7 receptors to activate SMAD2/3 signaling, and BMP15 binds ALK 2/3/6 receptors to activate SMAD1/5/8 signaling. The expression of KITL was not changed by SB or DM treatment. However, the effect of GDF9 and BMP15, which decreased the expression of KITL, was suppressed by SB or DM treatment. In conclusion, this study provides the first evidence that recombinant GDF9 and BMP15 decrease KITL expression in canine ovarian granulosa cells.

        The cancer and Parkinson's disease associated protein DJ-1 is multifunctional protein that involves in diverse cellular process. DJ-1 protein has a cellular protective role and promoted cell survival under an oxidative stress. However, the cellular protective mechanism of DJ-1 is not fully understand, and we needs to be further study their functions in novel organisms. In the present study, we investigated the protective role of DJ-1 against induced oxidative stress in canine cell line. On the basis of these experiments, canine DJ-1 overexpressing and null cell lines were established. The stable overexpression and down regulation of DJ-1 efficiency confirmed by the western blot analysis. Subsequently, the DJ-1 gene transfected cell lines and control cells were subjected to induced the oxidative stress, and then cell viability, cell proliferation assay, cellular apoptosis detection analysis (Annexin V and TUNEL assay), intracellular ROS and mitochondrial activity were measured appropriately. The results showed that DJ-1 overexpressed cells were up-regulated cell viability under oxidative stress conditions induced by the rotenone and hydrogen peroxide (H2O2), whereas loss of DJ-1 cells were down-regulated the cell survival activity. Additionally, overexpression of DJ-1 cells increased cell resistance to oxidative stress and inhibited the elevation of cell death and cellular ROS induced apoptosis. Moreover, DJ-1 overexpressed cells was increased mitochondrial functions by using confocal microscopy with MitoTracker staining. On the contrary to this, DJ-1 null cells show defective cellular protection and mitochondria activity against oxidative stress conditions. Our data indicate that canine DJ-1 protein attenuates cellular apoptosis and ROS generation, enhances the cellular survival activity and promote mitochondrial function under the oxidative stress, likewise other mammalian cells. Importantly, DJ-1 overexpression may be an important part of a protective strategy as a sensor for oxidative stress.

        The spotted-wing drosophila (SWD), Drosophila suzukii (Diptera: Drosophilidae), is an economically damaging pest that feeds on most thin-skinned fruits. In this study, we sequenced portions of the mitochondrial (mt) COI and ND4 genes from a total of 195 individuals collected mainly from Korea. A total of 139 haplotypes were obtained from the concatenated COI and ND4 sequences. A dataset combining GenBank sequences with our own data identified a total of 94 worldwide COI haplotypes with a maximum sequence divergence of 5.433% (32 bp). A rough estimate of genetic diversity in each country showed higher diversity in ancestral distributional ranges, but the invasion over Asian countries seems to have been substantial because haplotype diversity was only 2.35-3.97-fold lower in the USA, Canada, and Italy than that in the populations ancestral ranges.

        cording to histopathological characteristics. One is composite tumor, and the other is collision tumor. Composite tumor is a lesion showing the combined histopathological characteristics of two or more previously recognized tumor and/or cysts of different categories. Two constituent parts are mixed with each other, and histological transition is often observed. On the other hand, collision tumor is a lesion showing the independent histopathological characteristics of two or more previously recognized tumor and/or cysts of different categories. Two constituent parts are separated each other by connective tissue, and histological transition is not observed. Their origin is still unclear. In this study, a collision tumor was reported. 77-year old woman was referred to CNUH with a neck mass in right submandibular area. Incisional biopsy was carried out, and the patient was diagnosed as collision tumor of squamous cell carcinoma and neurofibroma. After reviewing studies reporting collision tumor from head and neck area, formation of a collision tumor was most frequent from a cervical area and average age is 59.6. A collision tumor commonly lead to misdiagnosis, and the prognosis is not always transparent. The variety symptom of collision tumor has to be kept in mind in the diagnosis and treatment of the lesions in head and neck area.

        Grapholita molesta occur four times a year and Carposina sasakii occur twice a year, and both pests do damage on stone fruits such as peach, apple, plum, apricot, etc. Grapholita molesta is worldwide distributed in temperate and subtropical areas including South Korea. But, Carposina sasakii distributed in South Korea, Japan, China and Asia, and has been managed as an important import quarantine pest by the authorities of United States, Canada and Taiwan. Forecasting of both pests in Korea is currently done through the investigation of 1,000 fruits per 10 trees (100 fruits / tree) in designated peach orchard. However, this method is very difficult to observe the pest by bagging of peach and require too much time and labor. Therefore, we tried to carry out a new forecasting method by using of sex pheromone traps for newly standardized method as an alternative. Using sex pheromone trap, attractiveness of G. molesta was proved to be 2.5 > 1.5 > 0.5 m by the height and the border => outside > center by the position. Attractiveness of C. sasakii made no difference in height, but, more trapped at the center and border than outside in position at peach orchard.

        The spotted-wing drosophila (SWD), Drosophila suzukii (Diptera: Drosophilidae), originally distributed across a few Asian countries, including South Korea, has invaded North America and Europe, but is absent from Australia. In order to export the South Korean grape cultivar Campbell Early to Australia, its potential to serve as oviposition and development medium for SWD must first be determined. In this study, we determined the oviposition and development potential of SWD on Campbell Early, after elucidating the SWD life cycle and establishing an artificial diet-based mass-culturing system. An investigation of the life cycle under five temperature regimes (16, 19, 22, 25, and 28°C) showed that the durations of the egg, larval, and adult stages were shorten when temperature was increased from 16, 19, 22, 25, and 28°C, but pupal duration was shortest at 25°C and extended again at 28°C. A test of oviposition and development potential of SWD on Campbell Early grape clusters showed oviposition of 30.8 ± 6.8 eggs per cluster of injured grapes and 157.7 ± 16.2 eggs on a culture dish of artificial diet. However, in a similar experiment using uninjured grape clusters, only a single egg was deposited on the grape skin, which soon dried. In light of these results, newly harvested grapes left at vineyards during daily harvests are unlikely to serve as an oviposition and development medium for SWD, as long as the grapes remain uninjured.

        Odontogenic cyst and odontogenic tumor shares developmental source. However, they have different histopathologic features, and they are classified respectively. Odontogenic cyst and tumor can share same physical region. It is called a hybrid lesion, a lesion showing the combined histopathological characteristics of two or more previously recognized odontogenic tumor and/or cysts of different categories. In this study, a hybrid lesion was researched. 61-year old man was referred to our department with a multilocular radiolucency in right mandibular angle. Incisional biopsy was carried out, and the patient was diagnosed with ameloblastoma. Odontogenic keratocyst was found with the tumor, and it was thought to be evolved via neoplastic transformation from lining epithelium of the keratocyst. After reviewing studies reporting hybrid lesions from odontogenic cyst and tumor, formation of a hybrid lesion was most frequent from a dentigerous cyst and an adenomatoid odontogenic tumor. A hybrid lesion commonly lead to misdiagnosis, and the prognosis is not always transparent. The close relationship between the odontogenic cyst and tumor has to be kept in mind in the diagnosis and treatment of the lesions in maxillofacial area.

        We present a case of intraparotid plexiform neurofibroma in a 7-year-old man. The lesion was determined as plexiform neuroma in histopathological findings, but final diagnosis was plexiform neurofibroma considering his familiy history of neurofibromatosis and café au lait spots on his body. Currently we are executing follow-up after removing the tumor with surgery. Plexiform neurofibroma can develope at any point along a nerve and spread out either just under the skin or deeper in the body. According to the literature, the most common site of plexiform neurofibroma is mouth and face in the head and neck region. Also, plexiform neurofibroma occurs at 8.8-year-old in neurofibromatosis typeⅠ patient with familial history. Because of interlacing with adjacent normal tissue and the invasive nature we have difficult resecting the mass completely. So when the tumor turns symptomatic or disfiguring leading to an aesthetic problem, surgery had better be undertaken.

        Proper oral hygiene is required to maintain oral health and prevent oral disease. Toothbrushing is central to proper oral hygiene. Mechanical tooth cleaning is the mainstay of plaque control. A variety of toothbrushing techniques have been developed and evaluated for their efficacy. However, these evaluations are subjective. To adequately evaluate toothbrushing objectively, a novel toothbrushing monitoring system was developed. The system involves user-monitored brushing patterns including toothbrush motions using a 3-axis gyroscope, 3-axis accelerometer, 3-axis magnetic sensor, one load-cell and Bluetooth devices. To confirm the efficacy of this toothbrushing monitoring system, eight periodontist performed tooth brushing in a dental model, and their brushing motion was monitored and recorded, and evaluated by statistical means. The proposed monitoring system can be used to aid dental care personnel in toothbrushing instruction.

        Recently an outbreaked pest belongs to Hemiptera: Recaniidae, Ricanula sp. is greatly concerned about the outspreading throughout the South of Korea by wide range of host, including Cornelian cherry, Jujube, and peach trees. In Chungbuk province, this pest was first occurred at Jincheon and Okcheon in 2012, Cheongju in 2013 and now found out at Eumseong and Goesan in 2015. Ricanula sp. was oviposited directly into one-year twig, did damages on fruit-bearing formation and finally withered the host. This study was performed to understand the ovipositional characteristics and to develop the standardized forecasting method. Oviposition by Ricanula sp. was abundant in tree than in bush, adult laid eggs on new inner twigs and then covered with wax compound. Total no. of oviposited egg-mass was 10 to 318, and that of on new twig was 5 to 185 per tree, with different to host trees. Thickness of oviposited twigs were done within 2 ~ 5.5 mm and the height was mostly founded with range of 1 to 2 m, founded with highest height over 3 m. Oviposited no. of egg-mass within 30 cm twig was appeared differently from 2 to 7 every host. From based on this investigated result, we provide this for standardized forecasting method. This pest will need to control when egg-mass will occur over 2 at new twig, within 30 cm from the tip, set as total 25 point/ 5 plants (5 point per plant).

        Flowering responses under night interruption (NI) condition were determined in Doritaenopsis ‘Fuller’s Sunset ’, ‘Green Beer’, ‘Little Gem Stripes’, and ‘Queen Beer’ during their reproductive stages. Plants were grown under 10/14 h ambient light/dark (control), 10 h ambient light with night interruption (22 : 00-02 : 00 HR) at low light intensity of 3-7 μmol • m−2 • s−1 (LNI) and 10 h ambient light with NI at high light intensity of 120 μmol • m−2 • s−1 (HNI) conditions after flower spike emergence. Time to visible bud and flowering decreased more in the plants grown under HNI than those grown under LNI or control conditions. The number of buds or flowers increased in the plants grown under both LNI and HNI conditions compared to those grown under control condition for all cultivars. The tallest plants at flowering were in the HNI group in ‘Queen Beer’, ‘Green Beer’, and ‘Fuller’s Sunset’, however, the plant height was not significantly different among the treatments in ‘Little Gem Stripes’. NI with low light intensity or high light intensity can be used effectively to promote flower opening, but not with low light intensity at flower development stage. To obtain early flowering with high quality plants using NI treatment during their reproductive stage, high light intensity strategies could be suggested in Doritaenopsis cultivation.

        We present a case of cellular angiofibroma arising from right neck in a 46-year-old woman. Surgical excision was performed and the patient was disease free till now. Cellular angiofibroma usually arised in the inguinoscrotal of vulvovaginal regions. Only 3 cases of cellular angiofibroma involving maxillofacial region have been reported in the english written literature. Cellular angiofibromas are rare benign tumor characterized by bland spindle shaped cells arranged in a stroma with wispy collagen and numerous vessels. Immunohistochemical stains of the tumor cells showed the positivity for CD34, PR, EMA, but negativity for neurofilament, desmin and actin. The tumor should be differentiated from aggressive angiomyxoma and angiomyofibroblastoma, angiofibroma because of its clinical and histological similarity. We report a rare case of benign cellular angiofibroma involving right neck and study other published articles.

        The aim of this study was to carry out the comparative analysis of the brushing force following various brushing techniques bytoothbrush mounted pressure sensing unit. The study group consisted of 10 dental hygienist participants. The brushing forces (on buccal area of each first molar) were monitored on 8 different kinds of brushing techniques; Fones, Bass, Rolling, Scrub, Charters, stillman, Modified bass and Modified stillman. In Bass, Charters, Fones and Scrub method, force distributions showed a small gap of maximum and minimum value (Max/Min) while a big difference was noted in Modified bass, Modified stillman, Rolling and Stillman methods. Especially, the biggest difference of Max/Min value was observed in the area of lower left first molar. In conclusion, highly delicate manual skill is needed in showing big error range of force distribution. It means that careful force needs to be focused during toothbrushing instruction when a delicate manual skill was carried out.

        The spotted-wing drosophila (SWD), Drosophila suzukii (Diptera: Drosophilidae), was originally observed in a few Asian countries, but is now found even in North America and Europe. Genetic information on geographic variation and relationship may broaden our understanding of origin and migration. As a first step, in this study, a portion of mitochondrial COI gene was sequenced to understand genetic relationship and diversity in Korea. Sequencing of 104 individuals provided 57 haplotypes, with the maximum sequence divergence of 1.5%, suggesting high haplotype diversity and moderate sequence divergence. Comparison to GenBankregistered D. suzukii haplotypes (possibly from Spain, Portugal, USA) has shown 100% sequence identity to most of the haplotypes found in this study, but two USA sequences were found to be independent haplotypes, with the sequence divergence ranging from 0.5% ~ 1.4% from our samples in the 553-bp comparison. Phylogenetically, no separable group was found, but, population genetically, the only Chinese population, Sandong, was significantly differentiated (p < 0.05) from all Korean populations, without sharing any haplotype. Among 28 pairwise comparisons of Korean populations only two comparisons showed a significant genetic differentiation, indicating that no population in Korea is completely isolated. Geographically, one haplotype (SWDBA06) was relatively widespread (five among nine localities) and a few haplotypes were found in more than one locality, but most haplotypes were restricted in a locality as a single individual. Overall, high rate of per generation female migration (Nm = 0.75 ~ infinite) and low level of geographic separation (FST=0~0.40) among localities were characteristic. Current data is limited mainly to Korean localities, thus, an expanded study may provide further scrutinized analysis for the fly.

        The aim of this study was to perform a comparative analysis of tooth brushing patterns before and after education with a newly invented toothbrush. All participants performed toothbrushing on left lower first molar teeth for 10 seconds, using Modified bass method with the Newly invented toothbrush before education. After recording the brushing pattern, the participants were randomly classified into 1) Tell, 2) Tell-Show and 3) Tell-Show-Do group. After their education, toothbrushing patterns were recorded again. A comparative statistical analysis with a professional group was done by a one sample t-test. Each motion of toothbrushing was represented by Roll, Pitch and Yaw directions and pressure. After toothbrushing education, the Pitch, Roll direction and the value of pressure in all groups showed significantly similarity to the professional group. The Yaw direction appears to be improved after education, but the delineated motion was not considered to represent the Modified bass method. The patterns of each group were similar to the professional group as follows; Tell < Tell-Show < Tell-Show-Do. There was general evidence of higher educational effects by telling, monitoring and practical training. Finally, toothbrushing patterns were able to be analyzed by the Newly invented toothbrush correctly and quickly. This will contribute to the establishment of correct toothbrushing habits through public use.

        Heavy metals such as cadmium and mercury are a highly toxic metal that affects a variety of cellular events, such as cell proliferation, differentiation and survival in animals as well as in human. The exposed heavy metals significantly affected the development and health in pubertal period. However, it is not clear how the toxicity of heavy metals in pubertal affects comparing in adults. To determine the effects of heavy metals on pubertal and adults, heavy metals such as cadmium and mercury were exposed to in pubertal and adults mice for 48 and 72h. Results showed that mice exposed to heavy metals for several hours induced overall tissue and organ damage. Especially, blood vessels of the most organ were more increased in adult mice compared to pubertal mice. And for morphologic alteration, secretary organs such as salivary gland and kidney were affected the most. Taken together, exposing to heavy metal in mice altered the blood vessels. In addition, the adverse effects of cadmium and mercury were more severe in adult mouse than puberty mouse. Further study is needed to focus on endothelial cells for more precise its mechanism
