        There are a lot of types of wild vegetables such as Colocasia esculenta (L.) Schott stem in Korea. However, the consumption of these wild vegetables is restricted because their storage decreased dramatically after harvest. To maintain original quality of vegetables, pre-treatments such as blanching and drying are important. But conditions for these treatments were still not optimized for many vegetables including Colocasia esculenta (L.) Schott stem. Thus, the objective of this study was to set up an optimal pre-treatment method for freezing storage. Colocasia esculenta (L.) Schott stems were peeled and cut equally (10 cm) for sample preparation. Dried samples (D) were dried at 90℃ for 3 h. Blanched samples (B) were blanched in hot water at 100℃ for 2 min. Blanched and dried samples (BD) were blanched and dried as same protocol. Physicochemical properties were analyzed to evaluate the quality including texture, moisture content, total color difference and viable cell count. Raw sample had 6.85 kg/cm 3 of hardness and 78.75 of chewiness whereas B was 6.83 kg/cm 3 of hardness and 7.8 of chewiness. B had the similar value compared to raw samples. Moisture content of raw sample was 94.4% and that of B was 94.1%, though there were not any significant differences between them. ΔE value of B showed lower value than those of the others. Viable cell counts and total coliforms were not detected after treatment, while raw sample had 5.39 log CFU/g of viable cell count without total coliform. Therefore, pre-treatments are essential for microbial safety of samples. All results considered, it is supposed that blanching is the optimal pre-treatment to sustain its original quality of Colocasia esculenta (L.) Schott stems before freezing.

        The food components of red sea bream(Pagrus major) cultured in copper alloy(brass) net(BN) and nylon net(NN) cages for nine months were investigated. The body weight was 234.9g at the beginning of the culture(the young fish), and then 692.1g(BN group) and 549.3g(NN group) at the fish cultured for nine months(the nine-month culture fish). The moisture content of the edible portion of the fish was higher in the young fish than in the nine-month culture fish. On the contrary, the lipid content was higher in the latter (9.1-10.6%) than in the former (7.6%).  The lipid content in the nine-month culture fish was slightly higher in the BN group compared to the NN group. There was little difference in protein content between the young fish and the nine-month culture fish, or between the BN and NN groups. There was a slightly higher level of copper content in the liver (5ppm in the young fish and 5-6ppm in the nine-month culture fish) compared to the edible portions (1ppm in the young fish and 3-4ppm in the nine-month culture fish), but no difference was evident between the BN and NN groups. The prominent fatty acids were 16:0, 18:1n-9, 22:6n-3, 16:1n-7, 18:2n-6, 20:5n-3, and 18:0 in the edible portions of the young and cultured fish. The n-3 fatty acid content was slightly richer in the BN group.

        Hypocreales entomopathogenic Beauveria bassiana and Metarhizium anisopliae are well-known biological control agents worldwide and have high potential in industrialization. However their thermo-susceptibility limits long-term storage under high temperature conditions and high insecticidal activity after application to target pests. Herein we isolated highly virulent isolates, B. bassiana JEF006 and JEF007 and M. anisopliae JEF003 and 004, and produced in three grains, as substrates for solid cultures, followed by thermotolerance assays to compare the potential of the three substrates. The JEF isolates were exposed to dry and wet heat at 50°C and overall conidia were more stable under dry heat condition rather than wet heat. Of the three grains, Italian millet was superior to the other grains in the production of thermotolerant conidia. Additionally Italian millet did not severely aggregated, which enabled air to penetrate into the substrate well compared to the sorghum and millet. JEF isolates were more thermotolerant when they were kept in oil conditions. The conidia of JEF 007 and 003 wild type and AtMT-based generating random mutants were subjected to SDS-PAGE. A significant relationship between conidial thermotoelrance and detected proteins was observed. This work suggests that Italian millet can be used as an effective substrate to produce more thermotolerant conidia, thus maintaining viability for long times under unfavorable environment and biological activity against target pests.

        The development and fitness of insects depend on the temperature to which they are exposed. The thermal windows are defined as the temperature range between the minimum and maximum rate of development for individual species. The intrinsic optimal temperature for development proposed by Ikemoto is one of important factors that determine the fitness of optimum life history. The temperature requirements for development of 65 species from five orders of insects was obtained from several journals. The minimum and maximum rate of development was estimated using empirical models. The temperature tolerance range of enzyme activation was estimated using Shape-Schoolfield-Ikemoto (SSI) model. The mean and range of intrinsic optimal temperature were 20.89°C and 15.7~27.7°C. The mean intrinsic optimal temperatures of Hemiptera and Endopterygota (Coleoptera, Diptera, Hymenoptera and Lepidoptera) were 20.97°C and 20.71°C. The mean and range of thermal windows were 25.59°C and 16.69~36.13°C. The mean thermal windows of Hemiptera and Endopterygota were 25.53°C and 25.62°C. also not much different. Each species of insects had a limited temperature range for development. It is needed further studies for understanding the ecological, physiological and evolutionary response of insects to their thermal environments.

        The growth, differentiation, development and fecundity of insects are influenced by temperature. The relationship between the development rate of insect and temperature has been studied since 1978 in Korea. The relationship between temperature and insect development was published in the Korean Journal of Applied Entomology (71 papers). The contents about Hemiptera, Lepidoptera, Coleoptera, Diptera, Hymenoptera, Acarina and Neuroptera were published in 27, 16, 7, 6, 4, 5, and 2 papers of the journal. Approximately 33 functions from many international journals were published to figure out the relationship between temperature and development rate of insects. The functions have been developed based on two principal ways – simplified analytic method and biophysiological approach. The empirical models are based on the law of total effective temperature and heat summation. The biophysiological models are based on the equations of Arrhenius and Eying. The thermal constant and lower temperature threshold are estimated using linear functions. The minimal and maximal development rate are presented by nonlinear equations. The Sharpe-Schoolfield-Ikemoto (SSI) model showed the intrinsic optimal temperature. Cumulative proportion of development completion for each life stage of insects was analyzed using Weibull and sigmoid functions. We discussed the application and implication of linear and non-linear temperature dependent development models.

        The effects of seed soaking treatment with the solutions of plant growth regulators IAA, GA3 and BAP on seed germination and shoot and bulb growth of Allium victorialis var. platyphyllum (Korean wild garlic) were determined. A significant variation in the seed germination rate was recorded at all treatments for various soaking periods. Maximum seed germination was obtained when seeds were soaked in IAA or GA3 solution at 200 mg L-1. The MAP treated seeds started to germinate after 3 months. Among treatments, IAA was found to be most effective in improving seed germination, but further seedling growth was not correlated to the soaking time. Seed soaking in IAA or GA3 solution enhanced further growth of seedlings compared with water control treatment. Shoot and bulb growth was highest in GA3 treatments.

        A positional scanning synthetic peptide combinatorial library (PS-SCL) was screened in order to identify antimicrobial peptides against the cariogenic oral bacteria, Streptococcus mutans. Activity against Streptococcus gordonii and Aggregatibacter actinomycetemcomitans was also examined. The library was comprised of six sub-libraries with the format O(1-6)XXXXX-NH2, where O represents one of 19 amino acids (excluding cysteine) and X represents equimolar mixture of these. Each sub-library was tested for antimicrobial activity against S. mutans and evaluated for antimicrobial activity against S. gordonii and A. actinomycetemcomitans. The effect of peptides was observed using transmission electron microscopy (TEM). Two semi-mixture peptides, RXXXXN-NH2 (pep-1) and WXXXXN-NH2 (pep-2), and one positioned peptide, RRRWRN-NH2 (pep-3), were identified. Pep-1 and pep-2 showed significant antimicrobial activity against Gram positive bacteria (S. mutans and S. gordonii), but not against Gram negative bacteria (A. actinomycetemcomitans). However, pep-3 showed very low antimicrobial activity against all three bacteria. Pep-3 did not form an amphiphilic α-helix, which is a required structure for most antimicrobial peptides. Pep-1 and pep-2 were able to disrupt the membrane of S. mutans. Small libraries of biochemically-constrained peptides can be used to generate antimicrobial peptides against S. mutans and other oral microbes. Peptides derived from such libraries may be candidate antimicrobial agents for the treatment of oral microorganisms.

        The study was to examine the variation and pattern of colony components of Vollenhovia emeryi. The total of 38 colonies was collected from 10 locations, including 35 colonies from 9 locations in South Korea and 3 colonies from 1 location in USA. To know colony components variation, we studied the approximate colony size, the approximate number of reproductives: queens, gyne and male, the approximate number of brood during 7 months. The observation period was from September 2012 to March 2013 over the breeding season from July to August. As a result, the study has found that colony components of the 38 colonies are not constant. This also indicates that gynes and males appear during non-breeding season at least in the laboratory condition. There are 4 types of gynes and males emergence: both(31.6%), only gynes(2.6%), only males(28.9%), absence(28.9%). The number of gynes and males of each colony also shows variance. It indicates that Vollenhovia emeryi may show colony components continuum. In this species, fertilized eggs become worker females and males; worker females are diploid but males are haplid because of maternal genomic elimination. Unfertilized eggs become queens which are diploid by clonal reproduction. Therefore, each individual of the sex can exceptionally transfer its own genes. This unique haplodiploid sex determination may create the conflict between queen and male and cause colony components continuum.

        The ant species, Vollenhovia emeryi Wheeler (Hymenoptera: Myrmicinae) is endemic in East Asia and has invaded into North America. In this species, the queen caste shows polymorphism in its wing morphology; long-winged queen and short-winged queen, and two morphs are thought not to coexist in nature. This research is conducted to 1) deduce the phylogeographical structure of the two wing morphs in South Korea and to trace the distribution pattern from East Asia to North America, and 2) investigate the Wolbachia and WO phage infection frequency of the species. Either individuals or colonies of V. emeryi were collected from 80 locations, encompassing 68 locations in South Korea, 11 in Japan, and one in USA. Among the collected samples in South Korea, the long-winged morph is dominant and considered as the ancestral type, while the short-winged morph is very rare and derived character. The origin of the US population is neither Korea nor Japan at least in this study. However, we do not exclude the possibility that its origin is the other parts of Japan or the other countries. All of the long-winged morph are infected with Wolbachia, while the short-winged seems to be geographically partially infected. It suggests the possibility that the short wing trait is linked with the evolution of resistance to Wolbachia infection. Bacteriophage WO infection status has no correlation with host insect lineage.

        CD63, a member of tetraspanin membrane protein family, plays pivotal role in cell growth, motility, signal transduction, host-pathogen interactions and cancer. In this work, the cDNA encoding CD63 homologue (TmCD63) was cloned from larvae of coleopteran beetle, Tenebrio molitor. The cDNA is comprised of an open reading frame of 705 bp, encoding putative protein of 235 amino acid residues. In silico analysis shows that the protein has four putative transmembrane domains and one large extracellular loop. The characteristic ‘Cys-Cys-Gly’ motif and ‘Cys188’ residues are highly conserved in the large extracellular loop. Phylogenetic analysis of TmCD63 revealed that they belong to the insect cluster with 50-56% identity. Analysis of spatial expression patterns demonstrated that TmCD63 mRNA is mainly expressed in gut and Malphigian tubules of larvae and the testis of the adult. Developmental expression patterns of CD63 mRNA showed that TmCD63 transcripts are detected in late larval, pupal and adult stages. Interestingly, TmCD63 transcript was upregulated the maximum 4.5 fold in response to DAP-type peptidoglycan during the first 6 h, although other immune elicitors also made significant increase in the transcript level at later time-points. These results suggest that CD63 might contribute to T. molitor immune response against various microbial pathogens.

        The bumblebee, Bombus terrestris, has played an important role as one of the alternative pollinators since the outbreak of honeybee collapse disorder. Recently, pathogens and parasites such as viruses, bacteria and mites, which affect the life span and fecundity of their host, have been discovered in B. terristris. In order to detect the microsporidian pathogen, Nosema Spp. in the field populations of B. terristris, we collected adults and isolated their genomic DNA for diagnostic PCR. The PCR primers specific for Nosema Spp. were newly designed and applied to gene amplification for cloning. Only small subunit ribosomal RNA(SSU rRNA) gene of N. ceranae was successfully amplified and sequenced among examined genes, which indicates that N. ceranae mainly infects the examined field population of B. terristris. To detect of SSU rRNA gene, two regions of SSU rRNA gene were selected by primary PCR analysis and further analyzed in quantitative real-time PCR(qRT-PCR). The qRT-PCR analysis demonstrated that SSU rRNA of N. ceranae was detected at concentrations as low as 0.85 ng/μl genomic DNA. This result suggests that the detection via qRT-PCR can be applied for the rapid and sensitive diagnosis of N. ceranae infection in the field population as well as risk assessment of B. terristris.

        고준위방사성폐기물 처분장의 완충재로 고려되고 있는 자연산 벤토나이트에 대해서 기존의 물리·화학적 및 광물학적 성질 외에 생물학적 특성을 살펴보았다. 국내산 ‘경주벤토나이트’를 대상으로 만든 현탁액을 영양배 지 세럼병에서 일주일 이상 숙성시키며 시간에 따른 벤토나이트의 변화를 관찰하였다. 영양배지에서 활성화 된 벤토나이트는 고체 시료뿐만 아니라 용액도 함께 변하였다. 용존황산염 수용액으로부터 검은색의 미립자 황화물이 생성되기 시작하였으며, 시료를 채취하여 배양한 결과 4 종류의 황산염환원박테리아(SRB)가 자체 생존하고 있음이 확인되었다. 이러한 결과는 벤토나이트 분말시료 내에 황산염환원(혹은 금속환원)박테리아 가 고착 및 서식하고 있음을 말해주는 것으로, 이는 지하의 환원환경 조건하에서 완충재 내외부에 장기적으로 생지화학적 영향이 발현될 가능성이 있음을 의미한다.

        Wolbachia is one of the most common endosymbionts best known to induce several reproductive alterations in its insect hosts. In some cases, the insect hosts harbor more than two strains of the bacterium. The Vollenhovia emeryi ant lives in dead trees and is morphologically subdivided into the long-winged and the short-winged. Interestingly the short-winged morph is free of Wolbachia, but only the long-winged morph is multiple- infected with the Wolbachia bacterium. We sampled four populations of the long-winged morph in Korea and performed pyrosequencing in Multilocus Sequence Typing (MLST), to determine the bacterial strain diversity. Six different gene regions (coxA, fbpA, ftsZ, gatB, hcpA and wsp gene) were targeted and amplified. However, the result shows that diversity of haplotypes is very high. The pyrosequencing approach in MLST, a new method of discriminating Wolbachia strains, is promising to effectively detect multiple infections and rare haplotypes.

        Staphylococcus species are one of prevalent pathogens found in hospitals. Microbes that are a primary cause of nosocomial infection were isolated from a dental and medical environment it may assist the reader to explain what this is and how it differs from the ‘dental health care providers and ward health care providers’. To investigate the distribution of staphylococcus species in this environment, we used vitek II to measure drug sensitivity, and further performed biochemical testing. The isolation rate of staphylococcus species from the dental and medical environment was 100% but from dental health care providers and ward health care providers were 44.4% and 33.3%, respectively. In the analyses, staphylococcus species showed resistance to diffusion of cefoxitin and oxacillin discs. These staphylococci may be sufficiently positive for the mecA gene. Our results suggest that staphylococci might be an important cause of nosocomial infection in the dental clinic.

        This study was conducted in order to examine the effects of alcohol-free cetylpyridinium chloride drinking water additive and oral gel on clinical parameters related to periodontal disease in beagle dogs. This study was conducted with healthy 15 beagle dogs. Following a professional teeth cleaning procedure, dogs were divided into three groups. Dogs in the control group received nothing, those in the drinking water additive (DWA) group received 800 ml water with 15 ml of alcohol-free cetylpyridinium chloride drinking water additive daily, and those in the Oral gel (OG) group were treated with oral gel containing alcohol-free cetylpyridinium chloride and 0.05% chlorhexidine gluconate daily. Clinical parameters, including plaque index (PI), calculus index (CI), and gingivitis index (GI) were evaluated at two and four weeks. Dogs in the DWA and OG groups had significantly less plaque than dogs in the control group at two and four weeks (P<0.01, P<0.05). And, at four weeks, CI was significantly lower in the OG group compared to the control group (P<0.05). On GI, similar scores were recorded for all groups during the experimental period. No significant difference was observed between the DWA group and the OG group. The effect of alcohol-free cetylpyridinium chloride drinking water additive was similar to the result for alcohol containing cetylpyridinium chloride mouthwash reported in a previous study. The effect in control of periodontal disease was better in the OG group because of additional chlorhexidine gluconate. However, use of drinking water additive will be more convenient for owners; thus, it will be more effective for achievement of long-term results.

        The influence of the culture media on the growth and catechins and caffeine production was investigated in adventitious root cultures of tea tree. The growth rate of adventitious roots was higher than that of solid medium. Growth rate of adventitious roots was noted to be optimal in N6 liquid medium. Yields of (-)-epicatechin gallate (ECG) and caffeine were the highest when adventitious root cultures were maintained in Nitsch and B5 liquid medium, respectively. The production of (-)-epicatechin (EC) and (-)-epigallocatechin gallate (EGCG) from adventitious roots was maximal when cultured in 1/2MS liquid medium. The adventitious root extracts of tea tree produced on catechins as EC (20.77 mg/g) and EGCG (1.84 mg/g) in 1/2MS medium; EGC (24.39 mg/g) and caffeine (12.97 mg/g) in B5 medium and (-)-epigallocatechin (ECG) (2.16 mg/g) in Nitsch liquid medium.
