본 연구는 양식산 참돔과 넙치를 Cochlodinium polykrikoides 적조에 노출시켜 노출 시간에 따 른 생존율, 호흡수, 혈중 스트레스 지표 및 조직학적 변화를 비교 조사하였다. 대조구는 자연해 수를 사용하였고, 실험구는 C. polykrikoides 밀도를 5,500±200 cells/ml로 설정하였다. 그 결과, 참돔은 적조 노출 1시간 이내, 넙치는 적조 노출 5시간 이내 전량 폐사하였다. 생리학적 반응 을 분석한 결과, 참돔은 적조 노출 후 혈중 Glucose 농도가 감소하였으며, 혈중 GOT 및 GPT 농도는 증가하였고, 혈중 SOD 농도는 감소하고, CAT 및 GPx 농도는 증가하는 경향을 보였다. 넙치는 적조 노출 후 혈중 Cortisol 및 GOT, GPT 농도가 증가하였고, 혈중 Glucose 농도는 적조 노출 1시간째 증가한 이후 감소하였으며, 혈중 SOD, CAT, GPx 농도는 노출 1시간째 감소한 이후 증가하였다. 조직학적 분석 결과, 참돔과 넙치의 아가미에 구조적인 손상이 발생하였다. 결론적으로 5,500 cells/ml 밀도의 C. polykrikoides 적조 노출은 양식산 참돔과 넙치에게 산화 적 스트레스로 작용하여 체내 항산화 방어 기작을 활성화하고, 간과 아가미의 손상을 발생시 키는 것으로 나타났다.
대왕자바리의 대량생산을 위한 적정 염분을 구명하였다. 각 염분별 노출시킨 대왕자바리의 생 존율은 0 psu에서 노출 4일째 모두 폐사하였으며, 염분 5 psu 이상에서 생존율은 100%였다. 성 장률은 염분 5 psu 이상에서 염분 상승에 따라 체중과 체장은 모두 증가하였으며, 염분 30 psu (대조구)에서 성장률은 가장 높았다. 먹이섭취량은 염분이 하강함에 따라 감소하는 경향을 보 였다. 염분 3 psu에서 먹이섭취는 없었으며, 염분 20, 25 및 30 psu 간에 유의한 차이는 없었다. 염분별 혈액 삼투질농도는 염분 5~30 psu에서 341~368 mg Osmol/㎏였다. 염분변화에 따른 산소소비율은 30 psu(대조구)에서 163.6±22.3 mg O2/㎏ fish/h으로 유의하게 높았다. 염분변화 에 따른 SOD, CAT 및 GSH-PX는 염분 15 psu에서 가장 높았다. 따라서 대왕자바리 생존 최저 임계염분은 5 psu이며, 양성을 위한 적정 염분은 20~30 psu으로 추정된다.
메기 (Silurus asotus)를 대상으로 210일간 기아 시 계측 형질 및 체 절단면 형질을 측정하여 그 기아 효과를 조사하였다. 실험 종료시 섭이군의 생존율은 92.2±0.47% 였으며, 기아군에서는 74.4±2.59%의 생존율을 보였다. Ls, DALPO/Ls, DADOP/Ls, ED/Ls, CH/Ls, BDAA/Ls, BDMA/Ls 및 BWAA/Ls의 8개 항목에서 유의한 차이를 보 였으며 (p<0.05), 다른 16개 항목 DALAD/Ls, DPDPL/ Ls, HPLAA/Ls, HALAV/Ls, HALOP/Ls, DAUPO/Ls, DADAV/Ls, DADAA/Ls, Max BL/Ls, Man BL/Ls, LD/Ls, HWOP/Ls, DAUF/HL, DAUS/HL, IW/HL 및 DAUE/HL 에서는 초기군, 섭이군 및 기아군의 세 그룹 간 유의한 차이가 없었다 (p>0.05). 섭이군은 각 체 절단면에서 기아군 보다 더 높은 전체 높이 (THX), 더 넓은 너비 (WX) 및 더 넓은 면적 (AX)을 보였다. 본 연구는 메기의 기아 상태에 대한 외형 및 체 절단면 형질에 대한 정보를 제공한다. 이는 메기에 대한 먹이 조절에 대한 방법을 보조하고 영양상태를 나타내는 기본 기초자료로 활용될 수 있을 것이다. 또 한, 본 연구에서 사용한 조사 분석 방법을 타 어종에서 적용 시 그 영양 상태에 대한 정보를 파악하는 데 유용하게 활용될 수 있을 것으로 사료된다.
잉어, Cyprinus carpio를 대상으로 고수온 스트레스 발생 시 코티졸 호르몬 변화 및 누적 사망 률을 조사하였다. 수온 변화에 따른 20℃, 24℃ 및 28℃ 실험군의 코티졸 변화는 수온 변화 후 6시간에 코티졸 함량이 증가하다가 24시간 이후부터 감소하여 20℃ 및 24℃ 실험군은 실험 전 의 코티졸 함량에 가까운 수치로 감소하였다. 48시간의 28℃ 실험군 코티졸 함량은 실험 전의 코티졸 함량보다 높았으나 24시간의 코티졸 함량보다는 유의한 차이를 보이며 감소하였다. 반면, 32℃ 실험군은 수온 변화 후 6시간부터 코티졸 함량이 증가하여 24시간에 코티졸 함량 이 최대치로 증가하였으며 48시간까지 코티졸 함량이 감소하지 않고 유지되는 경향이 나타났 다. 20℃, 24℃ 및 28℃ 실험군은 수온 변화 후 12시간까지 누적 사망률이 증가하다가 그 이 후부터 사망하는 개체가 발생하지 않았으나, 32℃ 실험군은 실험 종료 시(48시간)까지 지속적 으로 누적 사망률이 증가하는 경향이 나타났다. 본 연구로 도출된 결과를 이용하여 호르몬 분 석을 통해 특정지역의 폐사 위험 정도를 확인할 수 있는 방안으로 활용 가능할 것이다.
To investigate effects of starvation on physiological changes, stress response, and survival of cobitid loach (Misgurnus anguillicaudatus) exposed to sodium nitrite (NaNO2), a 4- week experiment was conducted. Fewer fish survived in the starved group than those in the fed group during the experiment. Starvation resulted in growth retardation, leading to differences in body length and body depth between fed and starved groups. The fed gorup continued to grow and remained in good condition. Blood chemical analysis (plasma cortisol and glucose) showed significant differences in stress response to nitrite exposure between fed and starved groups (p < 0.05). These results suggest that all parameters employed in this study to assess effects of starvation with NaNO2 stress are useful information for researching nutritional status in cobitid loach.
This study investigated cytogenetic and hematological and histological characteristics between diploid and spontaneous triploid on Cyprinidae (Crucian carp, Carassius auratus; crucian carp, C. cuvieri and common carp, Cyprinus carpio) in four major rivers of Korea. In our results, DNA contents of triploid Cyprinidae were 50% more than those of diploid Cyprinidae. Also, erythrocyte size of triploid Cyprinidae was 50% larger than those of diploid Cyprinidae. In all sampling rivers, sex ratios of C. auratus were biased to female, and especially, triploid groups of C. auratus were all female groups (p <0.05). In principle, sex ratios of C. cuvieri and common carp were equivalent between male and female.
The purpose of this study is to examine fluctuating asymmetry of eye diameter, maxilla length, operculum length, and the number of pectoral fin ray and pelvic fin ray between ploidy and sex in diploid and triploid marine medaka, Oryzias dancena. In all experimental groups, eye diameter and maxilla length showed no significant difference between left side and right side (p>0.05). Results of operculum length in triploid male group and pectoral fin ray's number in diploid male group showed similarity ones with results of operculum length in triploid female group and pectoral fin ray’s number in diploid female group (p<0.05). However, the operculum length in diploid male group and pectoral fin ray's number in triploid male group showed consinderable difference with those of operculum length in diploid female group and pectoral fin in triploid female group. Findings of pelvic fin ray's number in all groups were similar to those of pectoral fin ray’s number in all groups (p<0.01).
The morphometric truss characteristics and classical dimensions of the marine medaka, Oryzias dancena, that might distinguish diploid and triploid fish were examined. Significant differences in all the classical and truss dimensions of the diploid and triploid fish were observed in both sexes (p<0.01). All the dimensions of the triploid fish were greater than those of the diploid fish. The triploid marine medaka shows sexual dimorphism in these characters, and the sexual dimorphism of the triploid marine medaka is similar to that of the diploid marine medaka. Thus, when their classical dimension and truss dimension was measured, the growth of triploid marine medaka is faster than that of the diploid fish, and it displays clear sexual dimorphism, with male fish having longer dorsal and anal fins than female fish.
The aim of study is to contribute to this knowledge-base by investigating the respiratory function, the metabolic rate and the difference of physiological responses un-der low water temperature (20℃→15℃) stress be-tween diploid and triploid far eastern catfish, Silurus asotus. During the 48 hrs of water temperature stress exposure time, the respiratory frequencies, CO₂ and NH₄+ concentrations of diploid had higher values than those of triploid (p<0.05). However, pH of triploid was higher than those of diploid (p<0.05), and oxygen consumption rate was not different between diploid and triploid (p>0.05). The level of plasma cortisol and plasma glucose of triploid was lower than those of the diploid (p<0.05). However, in case of lactic acid, there were not significant between triploid and diploid (p>0.05). These results suggest that diploid was more sensitive for low water temperature stress response than triploid in this species.
The objective of this study was to determine the mitotic intervals (τ0) of two consecutive cell divisions and synchronous embryonic cleavage in grass puffer, Takifugu niphobles at different water temperatures (18, 20, 22, and 24℃). The color of the fertilized egg was light yellowish. The egg type was demersal and unadhesive. Egg weight was 0.09±0.002 mg. The sizes of unfertilized eggs were smaller than fertilized eggs in major axis and minor axis at 20℃ (p<0.05). The size of the fertilized egg of 18℃ water temperature group at the blastodisc stage was the smallest (p<0.05), but no significant differences were observed in the other water temperatures group except 18℃ water temperature group (p>0.05). The first cleavage stages at 18, 20, 22, and 24℃ were at 75, 90, 105, and 120 mins, respectively. As water temperature was increased, embryonic development and formation time of the first cleavage furrow were accelerated. There were negative correlation between τ0 and water temperature for grass puffer (Y=–1.225X+70.05, R2=0.988, n=10, where Y was τ0 and X was temperature). This study confirmed that successful hatching of grass puffer was related to water temperature. Chromosome manipulation will be helpful for this species using cleavage frequency and τ0.
The purpose of this study is to investigate the differences fatty acids and hormonal parameters in the spawning and non-spawning season between the diploid and induced triploid Far Eastern catfish, Silurus asotus. The measured triploids were produced by cold shock for 50 min at 4℃ in May 2014, the spawning season of diploid was in May, and the nonspawning season was designated in January. Estradiol and testosterone and gonadosomatic index of diploid were higher than those of induced triploid in spawning season (P<0.05), and those of diploid in spawning season were higher than nonspawning season. On the other hand, thyroid stimulating hormone and thyroxine of induced triploid were higher than those of diploid in spawning season (P<0.05). Erythrocyte count of diploid was higher than that of induced triploid in spawning season and non-spawning seasons. Mean corpuscular volume and mean corpuscular hemoglobin of induced triploid were higher than those of diploid in both seasons (P<0.05). Percentages of total saturated fatty acids and n-3 polyunsaturated fatty acids of induced triploid were higher than those of diploid in spawning season, but those of diploid were higher in non-spawning season (P<0.05). Percentages of total mono unsaturated fatty acids and total n-6 polyunsaturated fatty acids of diploid were higher than those of induced triploids in spawning season, while those of induced triploid in non-spawning season were higher (P<0.05). Therefore, induced triploids in the spawning season tend to concentrate on growth and lipid-synthesization, whereas, diploids concentrate on reproduction and gonadal maturation rather than on growth. In non-spawning season, growth and lipid- synthesization were not significantly different between diploid and induced triploid.
In a 12-week experiment, the rock bream, Oplegnathus fasciatus, and olive flounder, Paralichthys olivaceus, were investigated to determine the effects of starvation on their physiological parameters. The protein and DNA contents of the starved fish were significantly higher than the initial values and those of the fed fish. The RNA contents and RNA/DNA ratios of the fed fish were significantly higher than those of the other groups (P<0.05). The hematocrit, hemoglobin, red blood cells (RBC), and mean corpuscular volume (MCV) of the fed rock bream were significantly higher than at baseline (P<0.05), whereas the mean corpuscular hemoglobin concentration (MCHC) of the fed fish was lower than at baseline (P<0.05). The hematocrit, hemoglobin, RBC, and MCHC of the starved group were significantly lower than the baseline values, whereas the MCV of the starved group was significantly higher than the baseline value (P<0.05). No significant difference in alanine aminotransferase was observed between the fed fish and baseline, whereas the starved fish value was significantly higher than the baseline value (P>0.05). There were no significant differences in cortisol levels. However, the glucose level in the fed group was significantly higher than the baseline level and that in the starved group was significantly lower than the baseline level (P<0.05).
The influence of triploidization on histological characteristics of retina, trunk kidney, liver and midgut tissue, and cell cycle of tail fin and gill tissue in far eastern catfish, Silurus asotus were analyzed. In the infertile triploid fish, the nucleus and/or cell size of secondary proximal tubule cells of trunk kidney, hepatocyte and midgut epithelium are much larger than those of the corresponding cells in the diploid fish (P<0.05). However, triploid tissue showed fewer number of outer nuclear layer in retina and nuclei in secondary proximal tubule of trunk kidney than those for diploid tissue. The mean percentages of the Gl-, the S- and the G2+M-phase fractions were 92.5%, 3.2% and 4.3% in tail fin tissue of diploid, and 93.4%, 2.6% and 4.0% in those of triploid, respectively. There were no significant differences in the percentages of each cell cycle fraction between diploid and triploid. The mean percentages of each phase fractions were 75.1%, 11.1% and 13.8% in gill tissue of diploid and 85.2%, 8.9% and 5.9% in those of triploid, respectively. The differences of cell cycle between tail fin tissue and gill tissue were statistically significant in diploid and triploid (P<0.05). Also, the differences between diploid and triploid were statistically significant in tail fin tissue and gill tissue (P<0.05). Cyclin D1 and cyclin E expressions were not significantly difference between gill tissue and tail fin tissue, and protein expressions of induced triploid were higher than those of diploid. Results from this study suggest that some characteristics in the triploid exhibiting larger cell and nucleus size with fewer number of cell than diploid can be used as an indicator in the identification of triploidization and ploidy level in far eastern catfish.
