호접란은 세계적으로 분화뿐만 아니라 절화로도 판매되는 주 요 화훼작물이다. 상업적 종묘 대량생산은 조직배양 기술에 의 해 이루어지고 있으나 우리나라는 아직까지 균일하고 우수한 발근묘 생산 기술이 확립되지 않았다. 생육이 고르고 우수한 발근 배양묘를 생산하기 위한 적정 배양 신초수를 찾기 위해 용적 500mL 유리 배양병에 호접란 2품종(‘Lovely Angel’과 ‘UniVivace’)의 신초를 1, 4, 7, 10개씩 배양한 후 3개월에 지상부 및 지하부 초기 생육 특성을 조사한 결과, 7개의 신초를 배양하였을 때 신초의 생육이나 뿌리 유도 및 생육에 효과적이 었다. 또한, 배양병 재질이 발근묘 생육에 미치는 영향을 확인하 고자 용적이 500mL로 동일한 유리 배양병과 플라스틱 배양병 에 신초 7개 배양 3개월 후 2품종의 생육 특성을 비교한 결과, 2품종 모두 플라스틱 배양병 보다는 유리 배양병이 유의적으로 생육에 효과적인 것으로 확인되었다. 기내 발근묘 생산을 위한 배양병 재질의 영향은 이후까지 지속적으로 영향을 미치는 것으 로 확인되었다. 결론적으로, 균일한 호접란 발근묘 생산은 플라 스틱 재질보다는 유리병 재질의 배양병이 적합하며, 특히, 용적 이 500mL인 유리 배양병의 경우 신초를 7개(묘당 재식면적 5.4㎠) 이내로 배양하는 것이 바람직한 것으로 확인되었다.
Haemaphysalis longicornis는 사람과 동물에게 여러 심각한 병원체를 전달하는 주요 매개체로, 한반도에 널리 분포하고 있다. H. longicornis는 Rickettsia spp., Borrelia spp., Francisella spp., Coxiella spp., 그리고 중증열성혈소판 감소증후군 바이러스 (SFTS virus) 등을 매개하는 것으로 알려져 있다. 국내에 서식하는 H. longicornis의 미생물 군집과 관련된 연구는 많이 진행되지 않은 것으로 확인되었다. 이 연구는 한반도 내 다양한 지역에서 채집된 H. longicornis의 미생물군집 다양성을 지역별, 성장 단계 및 성별에 따라 분석하였다. 2019년 6월부터 7월까지 질병관리청 권역별기후변화매개체감시거점센터 16개 지역에서 채집한 H. longicornis의 16S rRNA 유전자 V3-V4 영역을 PCR로 증폭 후 Illumina MiSeq 플랫폼으로 시퀀싱하였다. Qiime2를 활용한 미생물 다양성 분석을 통해 총 46개의 샘플에서 1,754,418개의 non-chimeric reads를 얻었으며, 평균 126개 의 operating taxonmic unit (OTU) 을 식별하여 총 1,398개의 OTU를 확인하였다. 대부분의 지역에서 Coxiella spp.가 우점종으로 나타났으며, 특히 Coxiella endosymbiont는 가장 높은 우점도를 보이며, Coxiella burnetii와 계통 발생 학적으로 유사한 것으로 확인되었다. 이 연구를 통해 분석된 결과는 각 지역의 H. longicornis 미생물군집 데이터 베이스 구축에 활용되었으며, 이를 통해 지역별 미생물군집의 특이성을 식별할 수 있게 하였다. 이는 한반도의 H. longicornis에 의한 질병 전파 연구와 이를 통한 공중보건 개선에 기여할 것으로 기대된다.
Honey bees are crucial pollinators for agricultural and natural ecosystems, but are experiencing heavy mortality in Korea due to a complex suite of factors. Extreme winter losses of honey bee colonies are a major threat to beekeeping but the combinations of factors underlying colony loss remain debatable. Finding solutions involves knowing the factors associated with high loss rates. To investigate whether loss rates are related to Varroa control and climate condition, we surveyed beekeepers in korea after wintering (2021–2022 to 2022–2023). The results show an average colony loss rate of 46%(2022) and 17%(2023), but over 40% colony loss before wintering at 2022. Beekeepers attempt to manage their honey bee colonies in ways that optimize colony health. Disentangling the impact of management from other variables affecting colony health is complicated by the diversity of practices used and difficulties handling typically complex and incomplete observational datasets. We propose a method to 1) Varroa mite population Control by several methods , and 2) Many nursing bee put in hive before wintering.
This study investigated ethopabate (EPB) residues in edible tissues of broiler chickens given in drinking water and established the withdrawal time (WT) of EPB in poultry tissues. Twenty-four healthy Ross broiler chickens were orally administered with EPB at the concentration of 3.8 mg/L for 14 days (EPB-1, n=24) and 15.2 mg/L for 7 days (EPB-2, n=24) through drinking water, respectively. After the drug treatment, tissue samples were collected from six broiler chickens at 0, 1, 3, and 5 days, respectively. EPB residue concentrations in poultry tissues were determined using LC-MS/MS. Correlation coefficient values ranged from 0.9980 to 0.9998, and the limits of detection and quantification (LOQ) were 0.03~0.09 and 0.1~0.3 μg/kg, respectively. Mean recoveries in muscle, liver, kidney and skin/fat tissues were 95.9~109.8, 108.7~115.3, 89.9~96.6 and 86.7~96.8%, respectively, and coefficient of variations were less than 17.11%. At the end of the drug-administration period (0 day), EPB was detected at levels under the LOQ in all tissues from both the EPB-1 and EPB-2 groups. According to the results of EPB residue in Ross broiler tissues, withdrawal periods of both EPB-1 and EPB-2 in poultry tissues were established to 0 day. In conclusion, the developed analytical method is suitable for the detection of EPB in poultry tissues, and the estimated WT of EPB in poultry tissues will contribute to ensuring the safety of Ross broiler chickens.
In recent years, young Chinese consumers have become more favorably inclined toward products relating to traditional culture. Therefore, this study aimed to develop a textile design that incorporates traditional Chinese patterns and cultural symbolism that will appeal to consumers from the MZ generation (millennials and Generation Z). Through a literature search for traditional Chinese patterns and symbols regarded as auspicious, our design concept and motives were established. We selected peony, lotus, and frog motifs as representative of a “wealth and eternal prosperity” design theme. In textile design work 1, we used hand drawing and watercolor techniques, color transformation with Adobe Photoshop, and colorway and end-use 3D simulation with TexPro. The 3D simulation work suggests that this textile pattern is suitable for women’s outerwear, mufflers, and tote bags. Textile design work 2 conveyed the ‘wealth and eternal prosperity’ design theme and had a graceful mood that embodied the nobility of the lotus flower whilst also encompassing the symbolism of money and status. The end design is a modern reinterpretation of traditional Chinese patterns and motifs. As such, it is hoped that it will satisfy the needs of young consumers for cultural values yet offer a unique new aesthetic distinct from existing textile designs. These qualities can be expected to enhance the competitive market value of textiles bearing this design.
From 2020, Korean Animal and Plant Quarantine Agency has reset the withdrawal time (WT) for veterinary drugs typically used in livestock in preparation for the introduction of positive list system (PLS) program in 2024. This study was conducted to reset the MRL for amprolium (APL) in broiler chickens as a part of PLS program introduction. Forty-eight healthy Ross broiler chickens were orally administered with APL at the concentration of 60 mg/L (APL-1, n=24) for 14 days and 240 mg/L (APL-2, n=24) for 7 days through drinking water, respectively. After the drug treatment, tissue samples were collected from six broiler chickens at 0, 1, 3 and 5 days, respectively. Residual APL concentrations in poultry tissues were determined using LC-MS/MS. Correlation coefficient (0.99 >), the limits quantification (LOQ, 0.3~5.0 μg/kg), recoveries (81.5~112.4%), and coefficient of variations (<15.5%) were satisfied the validation criteria of Korean Ministry of Food and Drug Safety. In APL-1, APL in all tissues except for kidney was detected less than LOQ at 3 days after drug treatment. In APL-2, APL in liver and kidney was detected more than LOQ at 5 days after treatment. According to the European Medicines Agency’s guideline on determination of withdrawal periods, withdrawal periods of APL-1 and APL-2 in poultry tissues were established to 3 and 2 days, respectively. In conclusion, the developed analytical method is sensitive and reliable for detecting APL in poultry tissues. The estimated WT of APL in poultry tissues is longer than the current WT recommendation of 2 days for APL in broiler chickens.
An induction melting facility includes several work health and safety risks. To manage the work health and safety risks, care must be taken to identify reasonably foreseeable hazards that could give rise to risks to health and safety, to eliminate risks to health and safety so far as is reasonably practicable. If it is not reasonably practicable to eliminate risks to health and safety, attention have to be given to minimize those risks so far as is reasonably practicable by implementing risk control measures according to the hierarchy of control in regulation, to ensure the control measure is, and is maintained so that it remains, effective, and to review and as necessary revise control measures implemented to maintain, so far as is reasonably practicable, a work environment that is without risks to health or safety. The way to manage the risks associated with induction melting works is to identify hazards and find out what could cause harm from melting works, to assess risks if necessary – understand the nature of the harm that could be caused by the hazard, how serious the harm could be and the likelihood of it happening, to control risks – implement the most effective control measures that are reasonably practicable in the circumstances, and to review control measures to ensure they are working as planned.
For transport containers for radioactive wastes, a drop test should be performed at a height of 0.3– 1.2 m on a rigid target depending on the weight as a normal condition in the regulation. In the drop test, a strain gauge is commonly used to measure the local strain, and the position of the strain gauges is determined by the experiences of the engineer in advance of the test. For this reason, the strains can be measured at only predetermined points. The DIC (Digital Image Correlation) method using highspeed cameras can be used to measure the change in strain over the region of interest. In addition, it is possible to measure effectively even in areas with high strain gradients that are difficult to measure with strain gauges. Therefore, the DIC method can measure the strain change according to time over the entire load path. When the drop test of the transport container is performed, the impact load is delivered through the lower corner fittings-corner posts-upper corner fittings-lids. In this study, white spray was sprayed on these main load path, and black speckles were created on the spayed surface to trace the rigid motion of speckles. The images taken during the drop test can be used to create a strain field over region of interest.
In this study, a drop analysis of metallic disposal containers for radioactive wastes is performed according to accident scenarios at the disposal site. The weight of the disposal container is about 8 tons, and the ingot-type wastes are loaded in the disposal container. To simulate the floor of the disposal site as the impact target, the reinforced concrete pad is modeled. High impact energy of the disposal container due to their heavy weight and high drop height causes excessive deformation and failure of the concrete target having relatively weak strength. Dynamic growth of cracks due to such failures causes penetration and delamination of concrete. Since the impact force delivered to the container strongly depends on the failure of the concrete pad, it is important to properly simulate the failure of the concrete in the drop analysis. A material erosion method can be used to simulate the concrete failure. In the case of applying erosion based on the finite element method (FEM), the element is deleted when the element exceeds a certain criterion, which causes material and energy loss problem. To solve this problem, mesh-free methods such as smoothed particle hydrodynamics (SPH) can be commonly used, but the mesh-free method has the disadvantage of incurring high numerical cost. Therefore, an adaptive method combining SPH and FEM-based SOLID elements is used for concrete target modeling to simulate excessive deformation and failure of the concrete target. In the adaptive coupling method of SPH and SOLID, the concrete target is first modeled as a solid element. When the damage of concrete exceeds the failure criterion, the solid element is eroded and the SPH element replacing the solid element is activated. Since the activated SPH element continues to participate in the impact, the problem of loss of materials and energy can be effectively solved. In this way, analysis results consistent with actual physical phenomena can be obtained.
The bacterial soft-rot disease is one of the most critical diseases in vegetables such as Chinese cabbage. The researchers isolated two bacteria (Pseudomonas kribbensis and Pantoea vagans) from diseased tissue samples of Chinese cabbages and confirmed them as being the strains that cause soft-rot disease. Lactic-acid bacteria (LAB), were screened and used to control soft-rot disease bacteria. The researchers tested the treatments with hypochlorous acid water (HAW) and LAB supernatant to control soft-rot disease bacteria. The tests confirmed that treatments with the HAW (over 120 ppm) or LAB (Lactobacillus plantarum PL203) culture supernatants (0.5 mL) completely controlled both P. kribbensis and P. vagans.
In order to extend the business viability of carbon nanotubes (CNTs), research on CNT dispersion in a solvent as well as in polymer matrix should be established. Herein, three kinds of dispersing agents, sodium deoxycholate (DOC), sodium dodecylbenzene sulfonate (NaDDBS), polyvinyl pyrrolidone (PVP), are selected and applied to quantify the dispersibility and dispersion stability of CNT aqueous dispersion. The dispersibility of CNT dispersion with the PVP, evaluated via viscosity and particle size analyses, are superior to those with the DOC and NaDDBS dispersing agents. CNT aqueous solution dispersed with PVP showed slightly higher viscosity and narrower particle size distribution than those with DOC and NaDDBS dispersing agents. In addition, the dispersion stability of CNT dispersion with the PVP, measured via lumisizer analyses, are superior to those with the DOC and NaDDBS dispersing agents. HR-TEM analysis verifies that the outstanding dispersibility and dispersion stability of CNTs in aqueous solution are due to the effect of the robust polymer wrapping of the PVP dispersing agent on the CNT surface. From the results of this study, the guidelines for the selection of the suitable dispersing agents and the systematic evaluation of dispersibility and dispersion stability of CNT dispersions can be suggested.
A solid-phase competition enzyme-linked immunosorbent assay (ELISA), recombinant VP2 (rVP2) protein, and monoclonal antibody (mAb) were developed for the specific and sensitive detection of porcine parvovirus (PPV) antibodies in pig sera. A total of 1,544 sera samples were collected from breeding pig farms located in the Gyeongsangbuk-do Province in the Republic of Korea. The optimal operating conditions of SC-ELISA were as follows. The concentration of rVP2 proteins coated on the wells was 4 μg/mL, the swine sera were diluted 1:2, and the HRP-conjugated PPV VP2 mAb (9A8 clone) was used at 500 ng/mL. These results suggest that the SC-rVP-ELISA assay may be a valuable alternative to the current diagnostic tools used to detect PPV-specific monoclonal antibodies and broadly monitor PPV infections in domestic pigs at different breeding stages.
