This study compared the antioxidant and mineral properties of the leaves and seeds of fifteen Korean adzuki bean (Vigna angularis L.) breeding lines. This study was conducted in an attempt to expand the use of Korean adzuki bean leaves. The potassium, calcium, magnesium, and sodium contents of the leaves were significantly higher than the seeds, in particularly, the potassium content. The leaves had approximately 3.3 times higher potassium content than the seeds. For instance, the potassium content of YA1317 leaves was 21% higher than that of Arari. The total polyphenol content and ABTS activity of Adzuki bean leaves were significantly higher than the seeds, as opposed to the total flavonoid content and DPPH scavenging activity. Among the 15 breeding lines, YA1402 had 1.2~3.2 times higher antioxidant content and activity as compared to the Arari variety. It was concluded that adzuki bean leaves had higher mineral content, antioxidant component and activity as compared to the seeds. Therefore, adzuki bean leaves could be used an ingredient for dishes and as a medicine.
본 연구에서는 팥의 꼬투리와 줄기를 가해하는 Ostrinia속 해충에 대해 종 동정 과정을 기술하였고, 사육하면서 관찰된 발육특성들을 보고 하였다. 수컷의 생식기는 3-lobed uncus 형태였으며, 가운뎃다리 종아리마디에는 털을 많이 갖는 것으로 나타났다. 미토콘드리아 cytochrome oxidase I (COI)과 II (COII) 유전자의 부분 염기서열은 콩줄기명나방(O. scapulalis), 큰섬들명나방(O. zaguliaevi), 큰조명나방(O. zealis bipatrialis)들에 대해 일본과 중국에서 보고된 서열들과 100% 일치를 보이는 종은 없었다. 기주식물 범위는 국내외 보고들 간에 일치하지 않았다. 암컷 성페로몬샘 추출물의 가스크로마토그래피 분석에서 큰섬들명나방과 큰조명나방의 성페로몬 성분인 (Z)-9-tetradecenyl acetate는 검출되지 않았다. 이상의 결과들을 종합하여 고려하였을 때, 본 연구의 팥 해충을 가해하는 곤충 종은 콩줄기명나방(O. scapulalis)일 것으로 추정되었다. 가을 야외에서 채집된 유충들을 야외조건에서 보관하였을 때, 이듬해 6월과 7월 사이에 성충들이 우화하였는데, 이 결과로부터 본 곤충 종은 말령 유충 단계에서 겨울휴면을 하는 것으로 추정되었다. 이외에 반합성 인공사료를 이용하여 실내 사육이 가능하였다.
This study was conducted to identify an insect species in Genus Ostrinia (Lepidoptera: Crambidae) that gave serious damage to the red bean, Vigna angularis. The species has ever been described as O. zaguliaevi in the previous presentation (Jung et al., 2010). Because, however, inconsistent information has been recognized for the species, we reviewed characteristics in morphological, molecular and sex pheromone levels, and host-range. Male genitalia had 3-lobed uncus and tibia of midleg showed massive type. The morphology indicated that the species might be one of O. zaguliaevi, O. scapulalis and O. zealis. Partial nucleotide sequences of cytochrome oxidase subunit Ⅰ(COⅠ) and Ⅱ genes were not identical with those of the 3 species in GenBank, respectively. The deduced amino acid sequence of COⅠ was not identical with that of O. zealis. In the 23 analyses that sex pheromones were checked, (Z)-9-tetradecenyl acetate, which was reported in the sex pheromone components of both O. zaguliaevi and O. zealis, was not detected at all. An intensive study in Japan has reported that the feeding habit of O. scapulalis is polyphagous, while that of O. zaguliaevi is monophagous (only in Petasites japonicus) (Ishikawa et al., 1999). After considering all these information, we concluded that it is reasonable to decide that the insect species in the red bean in Korea is O. scapulalis.
Three lepidopteran insect pests of the legume pod borer, Maruca vitrata (Lepidoptera: Pyralidae), the soybean pod worm, Matsumuraeses phaseoli (Lepidoptera: Tortricidae) and an Ostrinia spp. (Lepidoptera: Pyralidae) attacking adzuki bean, Vigna angularis, were confirmed as the major insect pests during the reproductive developmental stage of adzuki bean, and M. vitrata existed dominantly. Almost all plants in the adzuki bean field were in jured by at least one of the three species, and 15~60% of reproductive organs were injured. Unbloomed inflorescence occurred continuously through the reproductive stage of adzuki bean. While the flower was many at the early stage and its number soon decreased, the pod was few at the early stage, but its number increased soon and maintained at a constant level. The results suggested that the adzuki bean injured by insect pests compensate the flower loss by developing new inflorescence, but the compensation do not cause the development of new pods. While M. vitrata and M. phaseoli were observed in flowers, pods and stem mainly during the first half of reproductive stage of adzuki bean, Ostrinia spp. was observed only in pods and stem during the second half. In addition, while all instars of larvae of M. vitrata were observed, larvae between the third and fifth instars for M. pahseoli and Ostrinia spp. were observed.
The adzuki bean (Vigna angularis L.) is a red-grained legume that has a number of essential nutrients and is used in traditional dishes in Asia. Adzuki bean industrial by-products are also a potential low-cost source of some unique bioactive polyphenols. Hence, here, the authors aimed to perform a comparative study of the phytochemical profiles of the leaves and seeds of the adzuki bean and compare their antioxidant, α-glucosidase inhibition, and tyrosinase inhibition activity. The authors assessed antioxidant activity by DPPH, ABTS, FRAP, PR, TPC, and SOD assays, which showed wide variation, respectively. From the relative antioxidant capacity index results, 10 adzuki bean landraces were selected to compare for phytochemicals and bioactivity using leaf and seed extracts. Antioxidant, α-glucosidase inhibition, and tyrosinase inhibition activity in the leaf extracts were higher than in the seed extracts, and there were more flavonols and isoflavones in the leaf extracts than in the seed extracts. This study demonstrated that adzuki bean leaf extracts could be a new natural antioxidant or antidiabetic agent and a skin whitener and can also be used in industrial applications.
장 건강에 유익한 프리바이오틱스 소재를 개발하기 위하여 배변을 촉진하는 효능을 지닌 붉은팥을 식품 발효에 이용되는 Bacillus subtilis KCCM 11965P로 발효하여 다음과 같은 결과를 얻었다. 붉은팥의 일반성분은 회분 3.35±0.04%, 조단백질 21.1±0.19%, 조지방 0.35±0.02% 함 유되었다. 붉은팥 원물 1%, 3%, 5%와 Bacillus subtilis KCCM 11965P 3% (v/v)를 접종하여 0, 24, 48, 72시간 배양 하였다. 배양액의 총균수를 측정한 결과 붉은팥 원물을 3% 첨가한 후 72시간 배양군에서 Bacillus subtilis KCCM 11965P 발효가 가장 적합하였다. 발효 시간에 증가함에 따라 총 폴리페놀 함량과 DPPH 라디칼 소거활성이 증가하였다. Protease 활성은 붉은팥 원물 5% 첨가한 후 72시간 배양한 군(2.69±0.003 unit/mL)에서 활성이 가장 높았다. 발효시간과 붉은팥 원물 첨가 농도가 증가함에 따라 α -amylase 활성도 증가하였으며, 붉은팥 원물 5% 첨가한 후 72시간 배양한 군에서 0시간 배양군(1.0±0.1 unit/mL) 보다 26.0±0.2 unit/mL로 증가하였다. Bacillus subtilis KCCM 11965P로 72시간 배양한 후 유리아미노산을 측정한 결과 leucine은 붉은팥 원물 5% 첨가한 0시간 배양군 5.22 mg/L 에서 67.59 mg/L로 증가하였으며, 비필수아미노산인 tyrosine은 5% 첨가 0시간 배양군 10.08 mg/L에서 259.35 mg/L로 증가하였다. 이와 같이 Bacillus subtilis KCCM 11965P로 붉은팥을 발효하면 항산화 활성, protease 효소활성, 및 α-amylase 효소 활성이 증가하였으며, 유리아미노 산과 유기산이 증가하였다. 붉은팥을 발효하는데 Bacillus subtilis KCCM 11965P가 적합할 것으로 판단되며, 붉은팥은 프로바이오틱스를 활성화시켜 장 건강을 증진시킬 수 있는 프리바이오틱스 소재로 개발할 수 있는 가능성을 시사 하였다.
This study investigated the changes in yields, antioxidant compounds, and antioxidant activities among 11 adzuki bean cultivars cultivated in a paddy field with somewhat poorly drained soil. The adzuki bean cultivars were cultivated in the paddy field from 2015 to 2016 in Milyang, Gyeongsangnam, Korea. Average soil moisture content was 16.5% in 2015 and 31.0% in 2016 at the experimental site during adzuki bean cultivation. As the soil moisture content increased, most of the adzuki bean cultivars showed deceases in stem height, first setting pod node, number of pods, 100 seed weight, and yield. Chungju-pat produced the greatest yields among the 11 cultivars in 2015 and 2016, whereas Hongeon had the smallest yields. Adzuki bean yields after paddy field cultivation was lower for all cultivars than for adzuki bean yields from the same cultivars after upland field cultivation. Chungju-pat and Chilbo-pat showed the smallest decreases in yields after paddy field cultivation, whereas Hongeon and Kumsil showed the greatest yield reductions. There were no significant differences in proximate composition. Some mineral components (P2O5, Ca, and Mg) were statistically different across cultivars. Chungju-pat had the highest Ca and Mg contents, but antioxidant components (polyphenol and flavonoids) and antioxidant activities (ABTS and DPPH) were highest in Saegil and lowest in Jungbu-pat. PCA and clustering analyses, based on the growth, yield, and antioxidant component measurements, performed to identify which variables contributed the most to separating adzuki bean cultivars or to grouping cultivars with similar characteristics. These analyses showed that the antioxidant components and antioxidant activities had the most influence on grouping cultivars together. Among the 11 cultivars, Saegil was statistically different from the other cultivars, but the other 10 cultivars were not significantly different under paddy field cultivation. Soil moisture content affected adzuki bean yield and antioxidant component contents. An increase in soil moisture led to a decrease in yield, but an increase in antioxidant components. These results provide information that will improve the selection of an appropriate adzuki bean cultivar for use in paddy fields.
Field emergence of Azuki bean is poor due to hard seed coat as compared to other legumes. In this study, an attempt was made to develop prediction method with regression analysis based on various seed vigor tests in laboratory for field emergence of azuki bean. Azuki bean seeds artificially aged to provide various levels of seed quality were evaluated by the standard germination test (SGT), cold germination test (CT), cool germination test (CGT), complex stressing vigor test (CSVT), tetrazolium(TZ) vigor test and electroconductivity test. The SGT was suitable for predicting the field emergence in the unaged high vigor seeds. The abnormal seedling percentage and shoot length in the CGT were highly correlated with field emergence of moderate vigor seeds artificially aged for 2 days. Electroconductivity, seed viability in the CSVT, and vigor and predicted germinability in the tetrazolium vigor test were also useful for predicting field emergence. Percent of ungerminated seed in the CSVT was correlated with field emergence in the low vigor seeds artificially aged for 4 days. In a stepwise multiple regression analysis, seed viability in the SGT, normal seedling percentage and dry matter weight in the CGT accounted for 86.9% of the predicted value of field emergence in azuki bean.
국내 전역에서 수집된 팥 150 유전자원을 공시하여 작물학적 형질 특성조사를 통한 품종육성의 기초자료를 제공코자 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 신육형 빈도를 보면 무한신육형이 96.6%(145자원)를 차지하여 가장 많았으며, 유한신육형 및 반무한신육형은 각 각 2.0%(3자원), 1.3%(2자원)를 차지하였다. 2. 화색은 황색이 98.0%(147자원)를 차지하였으며, 협색은 노란색, 갈색, 농갈색, 흑황색, 혼합색의 5가지의 색으로 확인되었다. 이중 가장 많은 협색은 황색으로 64.3%(96자원)를 차지하였다. 종피색은 갈색, 농갈색, 흑색, 흰색, 혼합색, 적갈색, 황색 및 녹색으로 8가지의 립색을 나타내었으며, 적갈색 및 갈색계통이 전체의 66.5%를 차지하였다. 3. 생육일수는 89.0~142.0일의 분포를 보여 변이의 폭이 상당히 컷으며, 평균은 115.0일이었다. 생육일수의 빈도는 131.0~140.0일로 이에 속하는 자원이 37.2%(56 자원)를 차지하였으며, 다음은 121~130일 사이가 27.3%(41자원)를 차지하였다. 4. 협장은 5.9~10.1 cm를 나타내어 변이 폭이 컷으며, 평균은 7.8 cm이었다. 협장은 7.1~8.0 cm 사이에 속하는 자원이 45.1%(68 자원)로 가장 많았으며, 다음은 8.1~9.0 cm에 속하는 자원이 32.2%(48 자원)를 차지하였다. 5. 개체당 평균 협수는 28.5협이었으며 150 유전자원의 협수분포를 보면 5.0~132.5협을 나타내어 변이의 폭이 컷다. 개체당 협수가 가장 많은 자원은 11.0~20.0협 사이로 30.8%(46 자원)를 나타내었으며, 다음은 21.0~30.0협에 속하는 자원이 28.6%(43자원)를 차지하였다. 6. 협당립수는 5.3~11.3립을 나타내어 변이의 폭이 컷으며, 평균은 8.2립이었다. 협당립수가 가장 많은 그룹은 7.1~8.0립으로 32.2%(48자원)를 차지하였으며, 다음은 8.1~9.0립 사이에 속하는 자원이 28.2%(42자원)를 차지하였다. 7. 100립중은 범위가 5.7~23.0 g으로 변이 폭이 컷으며, 평균은 12.9 g이었다. 100립중의 분포는 9.01~13.0 g사이에 속하는 자원이 전체의 43.4%(65 자원)로 가장 많았으며, 다음은 13.01~17.00 g이 28.5%(43 자원)를 차지하였다.
The aim of this study was to evaluate 30 phenolic compounds in adzuki bean germplasm. Adzuki 21653 had the highest content of total phenolics compounds (6597~;~mug~;g-1 ) while 104372 had the lowest concentration. The average total phenolic content of Japanese (2432~;~mug~;g-1 ) adzuki beans was higher than that of Korean (2256~;~mug~;g-1 ) adzuki beans. The average total phenolic contents were 2507~;~mug~;g-1 in small sized adzuki beans from Japan and 2459~;~mug~;g-1 in those from Korea. In large sized adzuki beans, the average total phenolic contents were 1315~;~mug~;g-1 in Japanese seeds and 1232~;~mug~;g-1 in Korean seeds. The average total phenolic contents in medium seeds were 2369~;~mug~;g-1 in Japanese adzuki beans and 1397~;~mug~;g-1 in Korean ones. In small seeds, the total phenolic contents of adzuki beans varied from 524~;~mug~;g-1 to 6597~;~mug~;g-1 in Japanese ones and from 375~;~mug~;g-1 to 6569~;~mug~;g-1 in Korean ones. Japanese and Korean adzuki beans were divided into landraces and wild adzuki beans. In this study, the wild adzuki beans showed higher contents of total phenolics than the native varieties. Specifically, the wild adzuki beans from Korea had the highest concentration of phenolics (3403~;~mug~;g-1 ). All adzuki bean germplasms were measured for their color and were classified into four groups accordingly: A; L < 30, +a, +b; B; L < 30, +a, -b, C; L > 50, +a, +b, D; L > 50, +a, -b. Especially, group B had the highest concentration of total phenolic compounds (2827~;~mug~;g-1 ), whereas group C had the lowest concentration (1882~;~mug~;g-1 ).
우리나라에서 수집된 재래종 팥 361개의 작물학적 특성을 조사한 결과 수집재래종 팥의 초형은 직립형이 77.8%로 제일 많고 중간형이 15.3%, 만성형이 6.9%로 제일 적었다. 엽형은 원형, 타원형, 점선형 순으로 많았고 화색은 황색 이 89.5%로 가장 많았고 담황색 9.4%, 농황색 1.1% 순이었다. 종피색은 적색 60.6%, 회색 26.9%이며 그 외 녹색, 백색, 담흑색, 적지흑, 암지흑, 백지갈색 은 1.4∼3.3%로 매우 적었다. 개화일수는 69∼75일, 결실일수는 41∼50일, 생육일수는 111∼120일인 수집종들이 가장 많았다. 경장은 41∼60 cm에서 50,2%, 개체당 협수는 16∼20개에서 40.2%, 100립중은 8-10g이 30.5%로 가장 분포비율이 높았다.
The toxic effects of aluminium (Al) on growth, chlorophyll content, δ-aminolevulinic acid dehydratase (ALAD) activity and anatomy of root and shoot were investigated in 7-day-old azuki bean (Vigna angularis) seedlings. Significant depressions in root elongation was observed in the low concentrations of Al (50, 100 ㎛) and increasing Al concentrations caused a sharp decline of root and shoot growth. The degree of inhibition was dependent upon Al supply. Exposure to 50 ㎛ Al or more inhibited root elongation within 1 day. In the 50 ㎛ Al treatments, a recovery of root growth was seen after 7 days exposure. In contrast, lateral root initials was little affected by Al exposure. Al toxicity symptoms and growth responses were more well developed in the roots than in the shoots. Analysis of Al localization in root cells by hematoxylin staining showed that Al entered root apices and accumulated in the epidermal and cortical cells immeadiately below the epidermis. There was a good positive correlation between the level of chlorophyll and ALAD activity. Increasing Al concentrations caused a decrease in total chlorophyll contents, accompanied by proportional changes in ALAD activity, suggesting a coordinated reduction of a photosynthetic machinery. Al exerted specific influence on the morphology of root and shoot. At higher concentrations of Al the roots induced drastic anatomical changes. The epidermal cells were disorganized or destructed while the cortical cells exhibited distortion of cell shape and/or disintegration. The diameter of root and transectional area of cortical cells decreased considerably with Al treatment. In the shoot Al also enhanced reduction of diameter of shoot and cell size. Gross anatomy of leaves treated with Al did not differ significantly from the controls, except for fewer and smaller chloroplast. Our results indicate that toxic effect of Al appear to be manifested primarily in roots and secondarily on shoots, and changes in root morphology are related to changes in the root growth patterns. Results are further discussed in relation to the findings in other plant species, and it is concluded that Al causes morphological, structural and, presumably, functional damage to the roots of the species investigated.
