본 연구는 불법적으로 수산물에 사용될 수 있는 염료 18종에 대한 안전관리 강화를 위해 정량 및 정성 분석이 가능한 LC-MS/MS를 적용하여 검증하기 위해 수행되었다 . 확립된 시험법은 CODEX CAC/GL-71 가이드라인에 따라 직선성, 정밀성 , 정량한계 및 회수율 등을 통해 유효성을 확인하였다 . 대상시료에 1% 아세트산을 함유한 아세토니트릴로 추출 후 C18 과 PSA로 정제하였다 . 본 실험에서 정량한계는 0.002 mg/kg 수준으로 정량한계를 포함한 농도에 따라 검량선을 작성하였고 모두 0.98 이상의 직선성을 확인하였다 . 또한 정확성은 63%-112% 이고, 정밀도는 15% 이하로 재현성이 우수하였다 . 국내 유통 중인 수산물 124 건을 수거하여 개발된 분석법의 적용성 검증과 안전성을 확인하고자 잔류실태조사를 실시 하였고 그 결과 7건이 미량으로 검출 되었고 부적합은 없었다 . 확립된 시험법은 수산물 안전관리에 활용할 수 있을 것으로 사료되는 바이다 .
In this study, two commercial PCR and ELISA test kits were examined for identification of eight animal species (beef, pork, chicken, duck, turkey, goat, lamb, and horse) from raw meat and meat products in Korea. The detection limit in RAW meat ELISA kit® on three types of meat samples blended with beef, pork and chicken, demonstrated that all meat species were differentiable down to 0.2%. RAW meat ELISA kit® on animal species resulted in differentiation rate of 94.5% for beef, 93.3% for pork, 90% for lamb, and 100% for chicken, duck, turkey, goat, and horse. In contrast, Powercheck Animal Species ID PCR kitTM resulted in 100% specificity at 0.05% limit of detection for all meat species. The detection limit of Cooked Meat ELISA kit® on mixed meat samples heat-treated with different temperatures and times, resulted in 0.1% for all heat-treated mixed meat except for chicken at 1.0%. Additionally, ELISA kit on sixty meat products resulted in specificity of 31.8% for ham, 13.6% for sausages, and 12.5% for ground processed products, and relatively low rate for more than 2 types of mixed meats. On the contrary, meat species differentiation using PCR kit showed higher percentage than that using ELISA kit®: 50.0% for ham, 41.7% for sausages, and 28.6% for ground processed meat. Futhermore, PCR kit on 54 dried beef meats detected pork genes in 13 products whereas ELISA kit showed negative results for all products. Hence, the possibility of cross-contamination during manufacturing process was investigated, and it was found that identical tumblers, straining trays, cutters and dryers were used in both beef and pork jerky production line, suggesting the inclusion of pork genes in beef products due to cross-contamination. In this study, PCR and ELISA test kits were found to be excellent methods for meat species differentiation in raw meat and heat-processed mixed meat. However, lower differentiation rate demonstrated in case of meat processed products raised the possibility of inclusion of other species due to cross-contamination during manufacturing process.
This study was carried out to investigate the consumption behaviors of environmentally-friendly agricultural products, including satisfaction of purchases. Questionnaires completed consumers were statistically analyzed. Regarding purchase frequencies of environmentally-friendly agricultural and animal products, consumers consumed them 1 time per month. No significant differences in consumption by age, living area, family type, or BMI were found. Exactly 38.2% of consumers preferred to purchase items in the packing amount of 0.2-0.5 kg, whereas 39.7% of consumers preferred amounts from 0.5-1.0 kg. Significant differences in packing amount were found according to age (p<0.05) and family type (p<0.01). However, there were no significant differences according to living area or BMI group. Many consumers made decisions based on the outer packing label (69.6%). Significant differences by age and living area were not found, whereas there was significant difference according to family type (p<0.05). Consumers answered that they purchased products in a general supermarket (24.7%) and cooperative (24.2%). Significant differences were found by age and living area (p<0.01) but not by family type. Consumer satisfaction of purchases scored 3.39 out of 5 points. In conclusion, significant differences in consumer satisfaction were not found according to age, living area, family type, or BMI.
This study investigated the effects of a healthy image on the preference and intake frequency of meat and animal products. The study looked into beef, pork, chicken, sausage, mackerel, cutlass fish, croaker, tuna, squid, shrimp, clams, fish cakes, eggs, milk, yogurt, ice cream, and cheese. A total of 359 usable surveys given to elementary school students, college students, and adults were collected using a convenient sampling method. While milk had the healthiest image, sausage had the least healthy image. The respondents preferred yogurt the most and sausage the least. The intake frequency of eggs was the highest and clams the lowest. The healthy image, preference, and intake frequency for all studied foods showed significant differences across both gender and age. The relationship between healthy image and preference was significant for all foods, and a healthy image always had a positive influence on preference. The relationship of healthy image and intake frequency was significant in 14 foods except for mackerel, cutlass fish, and tuna. Also a healthy image created a positive effect on the intake frequency of 14 foods.
To investigate the epidemiological characteristics of 68 Listeria monocytogenes isolates, including 11 reference strains and 57 isolates from imported US beef, domestic meats (beef, pork, chicken meat), raw milk, and milk plants. L. monocytogenes was to evaluate the production of virulence proteins, such as hemolysin (LLO) and lecithinase (LCP), the adsorption of Congo red (CRA), and to detect virulence genes using the polymerase chain reaction (PCR). In the study of virulence protein production, 68(100%), 62(91.2%), and 54(79.4%) of the 68 L. monocytogenes strains were positive for LLO production, the LCP test, and the CRA test, respectively, while strains of other species, such as L. innocua, L. gray, L. murrayi, and L. welshimeri, were not. There were no significant differences between L. monocytogenes serotypes and the ability to produce LLO or LCP. L. monocytogenesstrains had very high hemolytic titers (2 to 16 fold), while the other Listeria species, other than L. ivanovii and L. seeligeri, did not. The hemolysin activities of L. monocytogenes, L. ivanovii, and L. seeligeri usually exceeded 1.0 HU/mg, while those of other Listeria spp. were less than 0.04 HU/mg. In the PCR assay, all of the L. monocytogenes strains contained the hlyA, plcA, plcB, inlA, and inlB virulence genes and produced a product of the expected size. In the PCR of the actA gene, the expected 385-bp product was seen in 39(57.4%) L. monocytogenesstrains, while an unexpected 268-bp product was seen in 29(42.6%) strains. Most L. monocytogenes strains isolated from Hanwoo beef produced the 385- bp actA gene product, while strains of imported US beef usually produced the 268-bp actA gene product. By contrast, no virulence gene products were amplified in the other Listeria spp.
The major purpose of the present study is to survey vegetables, meats, seafoods, processed foods and imported foods for the presence. of Listeria spp. and to prevent listeriosis caused by the contamination of Listeria monocytogenes. Listeria spp. was isolated from 6.0% of meats, 3.4% of seafoods, and 11.4% of imported foods but was not found in vegetables and processed foods. The optimum growth condition of isolates indentified as Listeria monocytogenes was pH 7.0 and 37℃. The antimicrobial effect of grapefruit seed e×tract (GFSE) was observed in the level of more than 100ppm by disk method. When lml(2.5× 10^6 CFU/ml) of Listeria monocytogenes was inoculated and incubated for 3 days at 30, the total cell number of the organism was 4.5 × 10^9 in the control, 7.2 × 10³ in 100 μg/ml of GFSE medium, and 3.5 μg/ml of GFSE medium. Direct visualization of microbial cells by using both transmission electron microscope and scanning electron microscope showed microbial cell membrane the function of which was destroyed by treating with the dilute solutions of GFSE. It could be confirmed that GFSE completely inhibited the growth of the test strain of Listeria monocytogenes.
Levels of sterols including δ7-dehydrogenase isolated from the tissues of marine animal products (20 species) were determined on 1.5% OV-17 columm of gas-liquid chromatography. The composition showed that the mussels and clams contained various sterols in their tissues : cholesterol, brassicasterol. 24-methylenecholesterol with some minor components such as 22-trans-norcholesta-5,22-dien-3β-ol, 22-cis-dehydrocholesterol, 22-trans-dehydrocholesterol, desmosterol, 7-dehydrocholesterol, campesterol, stigmasterol, β-sitosterol, isofucosterol, and 7-dehydrocholesterol which could be converted into vitamin D3 in the skin tissue of animal was present in the muscle of oyster, Crassostrea gigas. On the other hand, the others including gastropoda were predominantly composed of cholesterol. The minor sterols such as 24-methylenecholesterol, stigmasterol and β-sitosterol in the fish intestines are supposed to be derived from dietary plankton. Cholesterol δ7-dehydrogenase which could convert cholesterol into δ7-dehydrogenase was present in the pickles of Tricurus haumela intestine.
이 연구는 고지방 및 고 콜레스테롤식이(HD)를 먹인 rat에서의 저지질혈증 및 항염증 효과에 영향을 미치는 카제인과 유당(cMRP) 그리고 락토바실러스 fermentum H9 (FcMRP)에 의해 발효된 cMRP에 의해 반응된 마일라드 반응 생성물의 효과를 조사했다. HD 식이를 실시한 rat은 정상적인 식이를 먹인 rat과 비교했을 때, 간의 트리글리세리드 농도가 유의적으로 증가했다. 이는 HD 그룹에 비해서 Simvastatin, L. fermentum H9(H9), cMRP, FcMRP의 처리기간의 총 트리글리세리드를 감소시킨 것으로 나타났다. 조직학적 분석에서 cMRP, F-cMRP 및 H9-를 먹인 rat에서 간과 대동맥 조직의 지질 축적이 감소한 것으로 관찰되었다. 또한, F-cMRP와 cMRP는 HD 그룹에서 내중막 두께를 감소시켰다. 게다가 H9, Cmrp, F-cMRP 처리는 ICAM-1 및 VCAM-1의 발현수준을 유의적으로 감소시켰지만, MCP-1의 발현 수준은 유의하게 감소시키지 못했다. 특히, ICAM-1과 VCAM-1의 발현은 HD 그룹에 비해 F-cMRP 그룹에서 유의적으로 감소했다. 이 연구 결과는 낙농 식품의 cMRP와 F-cMRP가 심혈관계 질환, 특히 죽상 동맥 경화증을 예방하거나 치료하는데 잠재적으로 사용될 수 있음을 시사한다.
