To understand molecular and cellular mechanisms of many gene products in the female reproductive organs including the ovary and uterine endometrium as well as during embryo development, researchers have developed and utilized many effective methodologies to analyze gene expression in cells, tissues and animals over the last several decades. For example, blotting techniques have helped to understand molecular functions at DNA, RNA and protein levels, and the reverse transcription-polymerase chain reaction (RT-PCR) method has been widely used in gene expression analysis. However, some conventional methods are not sufficient to understand regulation and function of genes expressed in very complex patterns in many organs. Thus, it is required to adopt more high-throughput and reliable techniques. Here, we describe several techniques used widely recently to analyze gene expression, including annealing control based-PCR, differential display-PCR, expressed sequence tag, suppression subtractive hybridization and microarray techniques. Use of these techniques will help to analyze expression pattern of many genes from small scale to large scale and to compare expression patterns of genes in one sample to another. In this review, we described principles of these methodologies and summarized examples of comparative analysis of gene expression in female reproductive organs with help of those methodologies.

Successful in vitro embryo production heavily relies on the normal maturation and fertilisation of oocytes. We examined the normal and abnormal fertilisation of zebu cattle oocytes matured in vitro. Immature cumulus oocyte complexes (COCs) from zebu cattle ovaries at slaughter were matured in vitro (IVM) for 24 h. The oocytes were either fixed, stained and examined for nuclear changes or fertilised in vitro (IVF) with Percoll-separated, heparintreated spermatozoa (1.0 /mL) of zebu (n = 7) and crossbred bulls (n = 7). After 18 h of sperm-COCs co-incubation at C with 5% in humidified air, the presumptive zygotes were fixed, stained and examined for pronuclei. The number of oocytes retrieved per ovary was 5.4 0.7. The percentage of matured oocytes was 73.0. The difference in motility of spermatozoa before and after Percoll seperation was significant (p<0.001). The percentages of normal and abnormal fertilisation (polyspermia and oocytes with one pronucleus) varied significantly depending on individual bulls (p<0.05). A protocol for IVF of IVM oocytes in Bangladeshi zebu cattle is developed. A future study may elucidate the capacity of such IVM-IVF oocytes to develop to the blastocyst stage for transfer to surrogate mother.

The objective of this study was to examine the effect of macromolecule in a maturation medium on nuclear maturation, intracellular glutathione (GSH) level of oocytes, and embryonic development after parthenogenetic activation (PA) and somatic cell nuclear transfer (SCNT) in pigs. Immature pig oocytes were cultured in maturation medium that was supplemented with each polyvinyl alcohol (PVA), pig follicular fluid (pFF) or newborn calf serum (NBCS) during the first 22 h and the second 22 h. Oocyte maturation was not influenced by the source of macromolecules during in vitro maturation (IVM). Embryo cleavage and cell number in blastocyst after PA was altered by the source of macromolecule but no difference was observed in blastocyst formation among treatments. Oocytes matured in PVA-PVA medium showed lower rates of oocyte-cell fusion (70.4% vs. 7782%) and embryo cleavage (75% vs. 8690%) after SCNT than those matured in other media but blastocyst formation was not altered (1327%) by different macromolecules. pFF added to IVM medium significantly increased the intracellular GSH level of oocytes compared to PVA and NBCS, particularly when pFF was supplemented during the first 22 h of IVM. Our results demonstrate that source of macromolecule in IVM medium influences developmental competence of oocytes after PA and SCNT, and that pFF supplementation during the early period (first 22 h) of IVM increases intracellular GSH level of oocytes.

This study was designed to investigate the effect of kinetin on in vitro development of parthenogenetic porcine oocytes exposed to demecolcine prior to activation. In vitro matured metaphase II stage oocytes were incubated in 0 or 2 g/ml demecolcine supplemented defined culture medium for 3 h and the oocytes were activated electrically. The parthenogenetic porcine embryos were then cultured in 0 or 200 M kinetin supplemented defined culture medium for 7 days. Regardless of demecolcine treatment, kinetin supplementation increased blastocyst rates significantly (7.0% versus 12.1% and 4.9% versus 8.5%; Control versus Kinetin and Demecolcine versus Kinetin + Demecolcine, respectively, p<0.05). Demecolcine treatment before activation tended to decrease blastocyst rates regardless of kinetin supplementation although it is not statistically significant. Total cell numbers in the blastocysts also tended to be elevated in embryos when supplemented with kinetin, however only the result between Kinetin and Demecolcine groups is statistically significant (37.6 7.2 versus 28.1 9.5, respectively, p<0.05). In conclusion, the present report shows that kinetin enhances developmental competence of parthenogenetic porcine embryo regardless of demecolcine pre-treatment before parthenogenetic activation when they were developed in defined culture condition.

Serial ultrasonography was performed to measure the normal appearance of uterine during estrous cycle and to determine whether the unterine appearance was related to the sex hormone, progesterone and estrogen. The uterine appearances, shape, diameter and echogenicity were daily monitored with ultrasonography in 9 Miniature Schnauzer dogs undergoing II estrous cycles. During proestrus and estrous, the uterus became hypoechoic but developed hyperechoic luminal echo. In the longitudinal view, the shape of the uterus occasionally changed from rectangular to coiled or serpentine, compared to other stages of the cycle. The diameter of the uterus during proestrus and estrous was larger (range: 0.600.86 mm) than other stages (range: 0.480.62 mm) of the cycle. The rising estrogen concentrations (range: 14.5116.86 pg/ml) in plasma during proestrus correlated with changes in the uterus (p<0.05). Progesterone concentrations were 0.080.15 ng/ml at the onset of proestrus, but rose 1.061.26 ng/ml at the end of proestrus. There was no relation to progesterone concentration from onset of estrus (p>0.05). There was dramatical changes in normal uterus and sex hormone during estrous cycle. Especially, the appearance, shape and diameter of uterus were related to plasma estrogen concentration during proestrus, correlated with other stages of the cycle.

In the present study, effects of concentration of cryoprotectant solutions on the nuclear maturation of vitrifiedthawed porcine oocytes were examined. Oocytes were cultured in TCM-199 medium supplemented with 5% FBS at C in 5% and air. The percentage of monospermy in the toxicity group and vitrification group (22.0 3.0% and 31.5 3.5%) was decreased compared with that of the control group (44.0 4.0%). The percentage of in vitro development to blastocyst in the toxicity group and vitrification group (12.0 2.5% and 14.8 2.8%) was decreased compared with that of the control group (28.0 3.0%, p<0.05). The survival and in vitro developmental rate of oocytes vitrification-thawed with EDS and EDT + TCM-199 medium supplemented with 0.1% PVA were 46.3 3.0%, 54.5 3.8% and 14.8 2.5%, 16.4 2.7%, respectively. This results were lower than the control group (28.0 3.5%). The in vitro developmental rate of embryos vitrified with EDS and EDT supplemented PVA did not have a significant difference. The survival and in vitro developmental rate of vitrified-thawed morula and blastocyst embryos were 44.2 3.5%, 17.3 3.0% and 48.1 4.2%, 18.5 3.5%, respectively. Vitrified morulae and blastcyst embryos had a lower survival and developmental rates than their control counterparts.

In the present study, effects of concentration of cryoprotectant solutions on the nuclear maturation of vitrifiedthawed bovine oocytes were examined. Also, the developmental capacity of vitrified-thawed immature oocytes following ICSI was investigated. Oocytes were cultured in TCM-199 medium supplemented with 5% FBS at C in 5% and air. The in vitro maturation rate of vitrified oocytes was 24.5 4.2%. The in vitro maturation rate of vitrified oocytes was lower than that of the control (72.0 3.5%, p<0.05). The in vitro maturation rate of vitrifiedthawed oocytes incubated in TCM-199 medium supplemented with 1.05.0 ug CB were 26.7 3.2%, 35.7 3.2%, 54.0 3.0%, 42.5 3.6%, respectively. The in vitro maturation rate (57.0 3.0%) of the vitrified-thawed oocytes treated with 3.0 g CB for 20 min was the highest of all vitrification groups, although the maturation rate were significantly (p<0.05) lower than those of fresh oocytes. The in vitro maturation rates of the vitrified-thawed (with EDS and EDT) oocytes were 53.8 3.4%, 51.1 3.5%, respectively. This results were lower than the control group (72.0 3.0%). The in vitro developmental rates of the vitrified-thawed oocytes following ICSI were 28.6 4.5%, 25.6 4.3%, respectively. This results were lower than the control group (40.0 4.0%).

The purpose of this study was to determine the effect of cows parity and calving seasons on the subsequent reproductive performance of the herd of Korean native cows raised under the same condition. With the parity of the cows ranged 1 to 4 (mean: 1.9), significant associations were found between parity and calving interval (p<0.05). Calving interval of the primiparous cows group was 395.0 16.5 days, which was the longest calving interval among the four groups. On the other hand, calving interval of the second parity group was 333.7 3.6 days. The primiparous cows had tendencies that long interval from calving to conception and small number of service per conception relatively when compared with the multiparous cows. In the case of calving season, the interval from calving to first service was short in summer and winter relatively. The interval from calving to conception in summer was the shortest in four seasons. The number of service per conception was larger in spring and winter and smaller in summer and autumn. Calving in spring showed delayed reproductive performance and calving in summer showed desirable reproductive performance.
