        A voltammetric investigation of Au assay was conducted at a low cost, using Nafion and DNA immobilized on a graphite Pencil working electrode (NDP) with a black lead counter and reference. The following optimal parameters were found: 0.4 V amplitude, 500 Hz frequency, -0.7 V initial potential, and 0.015 V increment potential. These optimal conditions were also applied to sand obtained from the river site. The aforementioned technique is simpler and less costly compared to the common voltammetry and spectrophotometric methods.

        Canine herpesvirus (CHV) is a member of the alphaherpesvirus subfamily, which can cause severe hemorrhagic diseases in neonatal pups as well as mild or subclinical respiratory infections in adult dogs. We examined the effects of cold stress on disease progression of CHV, an alphaherpesvirus, in neonatal puppies. Eight puppies were challenged intranasally with CHV suspension and divided into a cold stress treatment group and a hyperthermal group. Four pups were left uninoculated as controls and divided into cold and hyperthermal groups. In the challenged cold treatment group, all pups showed CHV-related disease within 5 days; pathological changes were observed in organs of puppies showing clinical symptoms. Grossly, numerous petechial red foci were scattered throughout lungs, kidneys, livers, and intestines of all CHV-infected puppies exposed to cold stress. Most puppies showed typical clinical signs and macroscopic lesions, and CHV infection was confirmed by isolation of the virus. However, in the challenged hyperthermal group, only one of the pups showed mild symptoms of CHV-induced disease. None of the puppies in the uninoculated group showed abnormal signs, although they were exposed to cold stress. These findings indicate that cold stress can cause rapid disease progression of CHV, an alphaherpesvirus.

        Rhizopus fruit rot was observed on strawberry fruit (Fragaria x ananassa Duchesne) grown in a glass greenhouse at the Gyeongsangnam-do Agricultural Research and Extension Services, Jinju, South Korea, from 2011 to 2013. Symptoms included water-soaking, rapid softening, and rotting. When these symptoms were left untreated, vigorous fungal growth was observed on the surface of infected fruits. Colonies on potato dextrose agar (PDA) at 30°C were white-cottony to brownish-black. Sporangia were globose, black, and 40–210 ㎛ in diameter. Sporangiophores were light-brown and 6–22 ㎛ in diameter. Sporangiospores were globose to oval, brownish, streaked, and 4–12 ㎛ in length. Columellae were light brownish gray, hemispherical, and 80– 120 ㎛ in diameter. To confirm the identity of the causal fungal pathogen, the complete internal transcribed spacer (ITS) ribosomal RNA gene region was amplified and sequenced. Based on these symptoms, mycological characteristics, pathogenicity tests on host plants, and molecular identification, the fungus was identified as Rhizopus oryzae Went & Prinsen Geerligs. This is the first report of Rhizopus fruit rot on strawberry caused by R. oryzae in Korea.

        One of the major problems in the modern pig industry is infectious disease. Susceptibility to infectious diseases is influenced by both constitutional (e.g. genotype, age, gender, and reproductive status) and environmental factors (e.g. nutrition, management, infections, and other forms of stress). Genetic variations within individual animals or herds can cause differences in the execution of immune functions against infectious agents in domestic pigs. The objectives of the present study were to identify genetic factor(s) responsible for piglet survival and mortality under commercial field conditions with infectious diseases as well as determine QTL regions for immune capacity in a pig reference family. Sex ratio was examined between normal grown and presumed dead groups of F2 animals generated by Korean native pigs (KNP) and Yorkshire (YS) breeds. The ratio of males was significantly higher in the presumed dead group than in the normal grown group. In order to study genetic factors associated with presumed mortality under disease outbreak, allelic frequencies of 239 single nucleotide polymorphisms (SNPs) were compared between normal grown and presumed dead groups. Exactly 22 SNPs showed significant differences in their allelic frequencies between the two groups, and four of them were validated with another commercial population divided by normal and emaciated pigs. Furthermore, the relationships between 239 SNPs and immune-related traits were studied. These results demonstrate that identification of genetic components of animal immune systems and susceptibility to infectious diseases is possible and will be useful to improve disease resistance in individuals as well as in breeding programs.

        An evidence suggests that even low-dose irradiation can lead to progressive cognitive decline as well as memory deficits in both humans and experimental animals in part due to hippocampal dysfunction. To determine whether or not green tea (GT) and epigallocatechin gallate (EGCG) could attenuate memory impairment as well as suppress hippocampal neurogenesis, passive avoidance and object recognition memory test as well as TUNEL assay and immunohistochemical detection with markers of neurogenesis (Ki-67 and doublecortin (DCX)) were performed using adult mice treated with relatively low-dose gamma irradiation (2.0 Gy). GT was administered intraperitonially at a dosage of 50 mg/kg of body weight at 36 and 12 hr pre-irradiation and at 30 minutes post-irradiation, or orally at a dosage of 250 mg/kg of body weight/day for 7 days before autopsy. EGCG (25 mg/kg of body weight) was administered intraperitonially at 36 and 12 hr pre-irradiation and at 30 minutes post-irradiation. In the passive avoidance and object recognition memory test, mice trained for 1 day after acute irradiation (2 Gy) showed significant memory deficits compared with sham controls. The number of TUNEL-positive apoptotic nuclei in the dentate gyrus increased by 12 h after irradiation. In addition, the numbers of Ki-67- and DCX-positive cells significantly decreased. GT treatment prior to irradiation attenuated memory defects, blocked apoptotic death, as well as reduced the number of DCX-positive cells. Therefore, GT may attenuate memory defects in adult mice exposed to a relatively low dose of radiation possibly by inhibiting the detrimental effects of irradiation on hippocampal neurogenesis.

        The aim of the present study was to evaluate the use of an immunochromatographic test (ICT) strip using recombinant MPB70 (rMPB70) protein as a complementary tool for the diagnosis of naturally occurring tuberculosis in cattle. The study was performed on 249 cattle from populations known to be free from Mycobacterium bovis (M. bovis) and 119 cattle with M. bovis infection, confirmed postmortem. Compared to reference standards (culture isolation and/or visible lesion), the sensitivity of ICT was 94.12% (95% CI: 89.89∼98.35%) while the specificity was 96.80% (95% CI; 94.62∼96.82%). The findings indicate that the ICT strip is efficient for diagnosing bovine tuberculosis in cattle from Korea.

        Purpose: To evaluate the feasibility of iPad for visual impairment by utilizing the magnification functions on the iPad compared to conventional optical magnifier when looking at near object. Methods: 18 participants (9 males and 9 females, 42 through to 71 years of age, with a mean age of 63.2 ± 12.3 years.)were asked to perform timed tasks of viewing bills and photographs to find the certain information. They were then asked to compare between their optical aid and the iPad in terms of comfort, ease of use and clarity when viewing bills and photographs. Results: Participants spent significantly less time completing the tasks when using their own optical magnifiers compared to the iPad (p<0.01). In the photograph viewing task, optical aids were perceived to be significantly clearer than the iPad (p=0.02). Conclusion: Overall, the iPad were not shown to be significantly more comfortable or offer more visual clarity when compared to traditional optical magnifying systems. Conversely, we also could not show that the iPad is any worse than traditional optical magnifying systems. Due to the likely benefits of training on the outcome, the iPad may have great potential as a low vision aid, but it may also need additional accessory such as stand for better image stabilization.

        This study aimed to investigate the association between carotid artery calcification (CAC) on panoramic radiograph and intima-media thickeness (IMT) measured on ultrasound. Panoramic radiographs which were taken from dental patients aged 50 years and older who visited for dental treatment were screened for the presence of CAC. The study group was composed of seven patients (four males and three females, average age 74.4±4.2 yrs) with CAC detected on panoramic radiographs, and the control group eleven patients (seven males and four females, average age 64.5±10.1 yrs) without CAC. All the patients underwent carotid ultrasonography to measure carotid IMT. The IMT was compared between the groups by nonparametric analysis of covariance (ANCOVA). The range of IMT of the study group was 1.10~2.0 mm, while that of the control group 0.60~1.10 mm. The mean of IMT was 1.50±0.34 mm in the study group and 0.85±0.14 mm in the control group, and there was statistically significant difference between the two groups (p<.01). In conclusion, CAC detected on panoramic radiograph might have an association with atherosclerosis

        We newly sequenced mitogenomes of five skippers belonging to Lepidoptera to obtain further insight into characteristics of butterfly mitogenomes and performed phylogenetic reconstruction using all available gene sequences (PCGs, rRNAs, and tRNAs) from 85 species in 19 families in eight superfamilies. The general genomic features found in the butterflies also were found in the five skippers: a high A/T composition (79.3% - 80.9%), dominant usage of TAA stop codon, similar skewness pattern in various levels, consistently long intergenic spacer sequence between tRNAGln - ND2 (64-87 bp), the ATACTAA motif betweent RNASer(UCN) and ND1, and characteristic features of the A+T-rich region (the motif ATAGA, varying length of poly-T stretch, and poly-A stretch). The start codon for COI was CGA in four skippers as typical, but Lobocla bifasciatus evidently possessed canonical ATG as start codon. Phylogenetic analyses mainly yielded the consensus superfamilial relationships ((((((Bombycoidea + Noctuoidea + Geometroidea) + Pyraloidea) + Papilionoidea) + Tortricoidea) + Yponomeutoidea) + Hepialoidea) with a high support for most nodes, confirming the validity of Macroheterocera and its sister relationship to Pyraloidea. Within Rhopalocera the familial relationships (Papilionidae + (Hesperiidae + (Pieridae + ((Lycaenidae + Riodinidae) + Nymphalidae))) were strongly supported, confirming invalidity of the superfamily Hesperioidea. On the other hand, superfamilial relationships among Noctuoidea, Geometroidea, and Bombycoidea and the familial relationships among Saturniidae, Sphingidae, and Bombycidae were dubious, requiring further representative taxon sampling.

        Panax ginseng, also known as Korean ginseng, has long been used as a broad tonic in Oriental medicine to augment vitality, health, and longevity, particularly in older people. This study investigated the effects of Korean red ginseng (RG) on bone loss in ovariectomized (OVX) mice. C3H/HeN mice (10-weeks-old) were divided into sham and OVX groups. OVX mice were treated with vehicle, 17β-estradiol (E2), RG (oral administration, 250 mg/kg/day), or RG (intraperitoneal administration, 50 mg/kg/every other day) for 6 weeks. Serum E2 concentration and alkaline phosphatase (ALP) activity were measured. Tibiae were analyzed using microcomputed tomography. Biomechanical properties and osteoclast surface level were measured. There was no significant difference in the degree of grip strength, body weight, uterine weight, mechanical property, tibiae length, or tibiae weight between the OVX and RG-treated groups. Compared with the OVX group, the serum ALP level was significantly lower in the RG-treated groups. Serum E2 levels and osteoclast surface levels did not change between the OVX and RG-treated groups. RG could not preserve trabecular bone volume, trabecular bone number, trabecular separation, trabecular thickness, structure model index, or bone mineral density of the proximal tibiae metaphysic. In conclusion, there was no definite effect of RG on OVX-induced bone loss in C3H/HeN mice.

        Toxoplasma gondii (T. gondii) causes a life-threatening opportunistic infection. Despite its clinical importance, very few therapeutic drugs against T. gondii are available. Furthermore, these therapeutic regimens are not always suitable for prolonged treatment due to adverse side effects as well as the potential of clinical failure by selecting drug-resistant parasite variants. Dictamnus dasycarpus is known to have many medicinal properties, including anti-inflammatory, anti-fever, and anti-rheumatic activities. In this study, 70% ethanol extract of Dictamnus dasycarpus showed anti-T. gondii effects. Ethanolic extracts of Dictamnus dasycarpus used to treat T. gondii were tested in vitro for their anti-T. gondii activity and cytotoxicity. The selectivity of Dictamnus dasycarpus extract was 7.52, which was higher than that of Sulfadiazine (2.08). We conducted an in vivo animal test to evaluate the anti-T. gondii activity of Dictamnus dasycarpus extract as compared with that of Sulfadiazine. In T. gondii-infected mice, the inhibition rate of Dictamnus dasycarpus extract was high, similar to that of Sulfadiazine. This indicates that Dictamnus dasycarpus extract may be a source of new anti-T. gondii compounds.

        Brucellosis is an important bacterial zoonotic infection worldwide responsible for economic losses in livestock industries which represents a considerable and increasing public health burden. The disease is caused by Brucella which possess the ability to invade and replicate within phagocytes and may result to chronic infection, leading to difficulty in medical therapy of the disease. The treatment for brucellosis employs conventional principles that have been applied for a long time and a combination of antibiotics is currently used due to low efficacy of monotherapies. However, vital effects with respect to health and safety are neglected. Nevertheless, the preventive methods and treatments for brucellosis using traditional medicine have not yet been thoroughly studied, hence, alternative therapies such as the use of natural plant extracts as traditional medicine that are safe, efficient and economical should be explored in order to identify candidates that eliminate complications due to brucellosis. Coptis chinensis Franch (Huanglian) is a traditional Chinese medicinal herb and its extract has been known to possess strong antibacterial activity. In this study, the antibacterial effects of C. chinensis Franch extracts, and the major components of the herb namely berberine and palmatine, were investigated on B. abortus. The C. chinensis Franch ethanol extracts (CCFE) showed bactericidal effects at 1,000 μg/ml concentration and berberine at 100 μg/ml concentration. However, C. chinensis Franch and its components did not affect invasion and intracellular growth of B. abortus in RAW 264.7 cells. In conclusion, C. chinensis Franch ethanol extracts, water extract and its major components such as berberine and palmatine would be a beneficial antimicrobial agent without affecting phagocytic pathway within macrophages, and further study for the precise mechanisms of antibacterial effect would be necessary.

        The anti-proliferative efficacy of t,t-conjugated linoleic acid (t,t-CLA), c9,t11-CLA, and t10,c12-CLA was compared in several human cancer cell lines. Gastric NCI-N87, liver Hep3B, pancreas Capan-2, and lung NCI-H522 cancer cells were incubated with 50 μM CLA isomers over a period of 6 days. The t,t-CLA inhibited the growth of all cancer cell lines to different extents, but c9,t11-CLA and t10,c12-CLA inhibited or stimulated the growth of the cancer cell lines. NCI-N87 cells were the most sensitive to growth inhibition and apoptosis from all CLA isomers tested. In NCI-N87 cells, CLA isomers reduced the release of arachidonic acid (AA) via the inhibition of cytosolic phospholipase A2 (cPLA2 ) activity, consequently reducing the production of PGE2 through the inhibition of cyclooxygenase-2 (COX-2). The efficacies of CLA isomers were in the following order (from most to least effective): t,t-CLA, t10,c12-CLA and c9,t11-CLA. Overall, these results imply that the anti-proliferative efficacy of t,t-CLA on cancer cells, especially NCI-N87 cells, was greater than other CLA isomers due to its induction of apoptosis through the inhibition of cPLA2 and COX-2 activities.

        The multi-tactile stimulation system was developed to analyze cognitive function characteristics. The developed system consists of three units; control unit, drive unit and actuator that simultaneously and separately supports the vibration, heat, and pressure stimulation. From the results, vibration stimuli have a wide frequency range of 0-300 Hz comprising 20 levels, and stimulation intensity comprising 40 levels. The control temperature and stimulation temperature used for heat stimulation comprise four and eight levels, respectively. Pressure stimuli in the range of 0-600 mmHg can be stimulated comprising 12 levels. Therefore, the developed stimulator can be used for integrative tactile perception and cognition studies on various vibration, heat and pressure sensations.

        In this paper, we developed a power amplifier system for driving a solenoid coil. The developed system consists of three units; control unit, drive unit and coil. The control unit generates a drive signal of sinusoidal wave type, monitors an operating status of the drive unit and vibration characteristics of the coil. The drive unit drives a solenoid coil in response to command from the control unit. The coil was fabricated with the size of diameter in 1.5 cm and length in 1.7 cm using electric copper wire. 4 types of drive unit were tested with thermal and vibration characteristics of the coil using temperature sensor and 3-axis accelerometer, respectively. From the result, the developed system has many advantages with respect to simple control algorithm, small size, and operates stably up to 68 watts driving condition.

        Glyphodes quadrimaculalis (Lepidoptera: Crambidae) feed on a root tuber of Cynanchum wilfordii (Asclepiadaceae) that is one of the most famous traditional medicines in Korea. The genus Glyphodes includes ~130 species distributed worldwide, so the complete mitochondrial genome (mitogenome) would be helpful for bio-identification, biogeographic studies, and multigene-based phylogeny. The 15,255-bp long G. quadrimaculalis genome comprises 37 typical genes and one large non-coding region, with the typical arrangement found in Lepidoptera. Of the 13 protein coding genes (PCGs), 12 begin with typical start codons found in insect mitochondrial PCGs, but the COI gene starts with atypical CGA. One of the noteworthy features of the genome includes the presence of a 51-bp long non-coding space sequence located between tRNAGln and ND2 that reveals high sequence homology (71.4%) to the neighboring ND2 gene, indicating the origin of the region by partial duplication of the ND2 gene.
