        Background : Agaricus bisporus mushroom, which originated from Europe, is an edible mushroom with a unique taste and flavor and is the most widely cultivated mushroom type worldwide, particularly because of the relatively high protein, dietary fiber, and vitamin D contents of the fruit body, which have human health benefits like anticancer and DNA damage protection. However, to our knowledge, few studies have been conducted on the ability to discriminate mushroom’s bioactive compounds according to regional production sites. Thus, this study was aimed to examine the regional difference of selected bioactive compounds in of Agaricus bisporus mushroom. Methods and Results : The cultivars (cv.) “Saedo” and “Saehan” obtained from five regions of Korea were measured their phenolics and fatty acids profiling by LC/MS and GC-FID system, respectively. Among 54 targeted phenolics, protocatechuic, p-hydroxybenzoic, caffeic, 2,4-dihydrobenzoic, p-coumaric, and salicylic acids were found in the cv. Saedo and cv. Saehan fruit body. Also, tyrosine and phenylalanine, which are precursor metabolites of phemolics, were also observed in the A. bisporus fruit body. In particular, phenylalanine and p-hydroxybenzoic acid were the most abundant phenolic precusor and metabolite present in the A. bisporus, and was significant with the production regions (p < 0.05). The FA content was significant with the production region, but not much differed (p < 0.05). The n-6 FA including linoleic acid was the major FA in the mushroom, accounting > 70% over total FA content. However, the n-3 FA was the n-6 FA was very minor FA metabolites. Conclusion : These preliminary results can extend knowledge of regional difference of certain targeted bioactive compounds in A. bisporus mushroom produced in Korea, contributing to reliable regional difference with potential broader applications for the international mushroom market.

        This paper presents an improved double diffusion model (DDM) for the alkali silica reaction. By this model we could simulate the diffusion process of alkali both in concrete (macro-scale) and aggregate (micro-scale). As the alkali source, either internal or outside cases can be analyzed through this model. Concrete with different size groups of aggregate distribution also can be analyzed. Numerical simulation is utilized to analyze the ion concentration both inside the concrete and aggregate at different moment of penetration.

        By the end of 2012, the recycled proportion of domestic waste tires was 287,330 ton (93.9%) of the amount of waste tires discharged (305,877 ton). The waste tires have been reused for heat supply, material recycling and other purposes; the proportions are 50.1%, 20.7% and 23.1%, respectively. In the case of heat supply, waste tires are supplied to cement kiln (104,105 ton, 68%), RDF manufacture facilities (47,530 ton, 31%) and incinerators (1,923 ton, 1%). Recently, there has been an increase in the use of waste tires at power generation facilities as an auxiliary fuel. Thus, physico-chemical analysis, such as proximate analysis, elemental analysis and calorific value analysis have been carried out to evaluate potential of waste tires as an auxiliary fuel in Korea. The LHV (Lower Heating Value) of waste tires is approximately 20% higher than that of coal, at an average of 8,489 kcal/kg (7,684 ~ 10,040 kcal/kg). Meanwhile, the sulfur content is approximately 1.5wt. %, and balance of plant (e.g. pipe line, boiler tube, etc.) may be corroded by the sulfur. However, this can be prevented by construction and supplementation with refractories. In this study, TDF (Tire Derived Fuel) produced from waste tires was co-combusted with coal, and applied to the CFB (Circulating Fluidized Bed) boiler, a commercial plant of 100 tons/day in Korea. It was combined with coal, ranging from 0 to 20wt. %. In order to determine the effect on human health and the environment, gas emission such as dioxin, NOx, SOx and so on, were continuously analyzed and monitored as well as the oxygen and carbon monoxide levels to check operational issues.

        다양한 환경에서 수집한 국내 민들레속 유전자원 수집종의 엽록체 DNA 영역(trnL-trnF와 rps16-trnK) 염기서열을 이용하여 종내․ 간 변이 및 배수성을 구명하여 유전자원 육성의 기초 자료룰 제공하고자 수행하였다. 민들레속 유전자원의 배수성은 털민들레, 서양민들레, 붉은씨서양민들레가 3배체이고, 흰 민들레와 흰노랑민들레는 4배체였다. 염기서열의 길이는 trnLtrnF 영역에서 자생종류인 털민들레, 흰민들레, 흰노랑민들레 가 931 bp에서 935 bp, 서양민들레는 910 bp, 붉은씨서양민들레 는 975 bp로 종간 차이를 나타내었고, 종 특이적 염기서열 88개, 자생종 및 귀화종 특이적 염기서열 41개가 검출되었다. rps16- trnK 영역은 털민들레 882∼883 bp, 흰민들레 875∼881 bp, 흰 노랑민들레는 878∼883 bp 서양민들레 874∼876 bp, 붉은씨서 양민들레는 847∼848 bp로 37개 종특이적 염기서열이 검출되 었다. 염기서열의 유사도는 trnL-trnF 영역에서 0.860∼1.000 사이로 평균 0.949이며, rps16-trnK 영역의 유사도는 0.919∼ 1.000 사이로 평균 0.967이었다. 염기서열을 바탕으로 유연관계를 분석한 결과, trnL-trnF 영역은 크게 자생종류와 귀화종 류로 구분되었으며, 서양민들레와 붉은씨서양민들레는 같은 종 간에 유집되었고, 자생종류는 분리되지 않았으며, rps16-trnK 4개 그룹과 유집되지 않은 5개체로 나뉘었다. 흰노랑민들레는 두 영역 모두 흰민들레와 동일 계통군을 형성하였고, 염기서열 상 두 종간 뚜렷한 차이가 없었다. 유연관계에서 모두 독립적으로 존재한 흰민들레 No. 10 (조계산)과 털민들레 1번(광양)은 민들레 유전자원 육성소재로 활용이 기대된다.

Obtained results indicates that the impact resistances of overlay concrete median barrier was improved as the rebar and shear keys are more closed to the top surface of the CMB.

        본 시험은 논으로부터 밭으로 윤환한 2년차인 답전윤환밭과 연속밭에 풋옥수수 찰옥4호 등 8품종을 사용하여 윤환밭과 연속밭의 병충해 발생, 생육 및 수량특성 차이 구명은 물론 풋옥수수의 체계적인 유기농 답전윤환 재배기술을 위한 기초자료를 확립하고자 수행한 결 과는 다음과 같다. 유기농 답전윤환 시 주요 병해는 전반적으로 연속밭에 비해 윤환밭에서 발병률이 높았으며, 시험품종 중 저항성 품종으로 발병률이 2% 미만인 흑진주찰과 대학찰 로 판단되었다. 유기농 답전윤환 시 주요 충해인 조명나방 유충 피해는 전반적으로 연속밭 에 비해 윤환밭에서 피해주율이 높았으며, 시험품종 중 저항성 품종으로는 피해주율이 낮 은 찰옥4호와 흑점2호로 판단되었다. 유기농 답전윤환 시 수량은 윤환밭에서 연속밭의 85 % 수준으로 감소하였으며, 답전윤환밭에서는 찰옥4호가 789.0 kg/10a로 가장 수량이 많았 다. 내병성, 내충성, 수량을 종합 평가하면 유기농 답전윤환 시 적합 품종으로는 찰옥4호, 흑진주찰, 흑점2호로 판단된다.

        엑소좀은 세포에서 분비되는 작은 소낭체로서, 기원세포와 조직에 따라 다양한 기능을 수행하며, 세포생 존 및 세포 간 커뮤니케이션에 중요한 역할을 한다. 최근 엑소좀을 활용하여 종양연구, 면역질환 개선, 질병진단 bio-marker 개발 등 다양한 분야에서 연구가 진행되고 있으나, 피부세포에서 분비기전 및 피부 생리적 기능에 대한 연구는 미흡한 실정이다. 따라서 본 연구에서 인체피부 유래 각질형성세포(HaCaT)로부터 분리된 엑소좀 이 피부각질형성세포의 증식과 이주에 미치는 영향을 확인하고자 하였다. HaCaT으로부터 ExoQuick-TC를 활 용하여 엑소좀을 분리하고, 열처리 엑소좀(boiled exosome)과 무처리 엑소좀(unboiled exosome)으로 구분하 였다. HaCaT 유래 엑소좀은 농도 의존적으로(0.1 ∼ 20 μ g/mL) HaCaT의 증식을 유도하였으며, 20 μ g/mL에 서 대조군(control)에 비해 각각 186.96 ± 3.87%(열처리) 또는 193.48 ± 10.48%(무처리)의 증식 유도활성 을 나타내었다. 또한 HaCaT 유래 무처리 엑소좀은 농도 의존적인(0.1 ∼ 20 μ g/mL) HaCaT의 이주활성을 나타내었고, 20 μ g/mL에서 대조군에 비해 179.39 ± 4.89%의 이주를 유도하였다. 그러나 열처리 엑소좀은 이주 유도활성을 나타내지 않았다. 뿐만 아니라 무처리 HaCaT 유래 엑소좀은 collagen sprout outgrowth를 농도 의존적으로 유도함을 확인하였다. 이러한 결과를 통해서 HaCaT 유래 엑소좀은 지질 및 열에 안정한 물질 이 세포의 증식을 유도하고, 단백질 또는 열에 불안정한 물질이 세포의 이동 및 sprout out growth 활성에 관여 하는 것으로 확인되었다. 따라서 피부각질형성세포 유래 엑소좀은 피부의 재상피화 및 상처치유 등의 활성을 나타낼 수 있으며, 향후 화장품소재로서 응용 가능성이 확인되었다.

        본 논문은 교량의 예방적 유지관리 효과를 생애주기비용 절감 측면에서 분석 하였다. 예방적 유지관리는 현재 실시되고 있는 대응 적 유지관리 전략과 대비 되는 유지관리 전략으로 교량의 열화지연을 통해 궁극적으로는 교량의 사용수명을 연장 시키며 생애주기비용을 절 감시키는 유지관리 전략이다. 이러한 예방적 유지관리 효과를 분석하기 위해 교량의 공용연수에 따른 건전도 점수 변화 모델과 FHWA 자료를 바탕으로 한 국내 고속도로 교량의 건전도 점수에 따른 보수·보강 비용 모델을 제시 하였다. 제시한 복수의 모델과 국내외 참고 문헌을 바탕으 로 제안된 예방적 유지관리 항목(청소와 도장)의 비용과 주기를 활용하여 사용수명 100년까지의 표준교량(1,730 m2/교량) 당 생애주기 비용과 향후 20년간의 고속도로 총 유지관리 비용을 산정 하였다. 분석 결과 100년간 예방적 유지관리 활동으로 인해 교량 당 5억원의 절감효과를 볼 수 있으며 현재 예방적 유지관리 활동을 실시할 경우 20년 후(2035년)에는 총 1,837억원, 즉 연간 약 92억원의 비용 절감효과를 볼 수 있는 것으 로 분석 되었다.

        The morphological characteristics and genetic relationships among 32 germplasms of Zanthoxylum schinifolium and Zanthoxylum piperitum collected from two farms in Korea were investigated. The traits with the most variability were seed color, leaf size, and spine size. The intraspecific polymorphism of Z. schinifolium and Z. piperitum was 96.5% and 60.3%, respectively. The genetic diversity and Shannon’s information index values ranged from 0.11 to 0.33 and 0.19 to 0.50, with average values of 0.26 and 0.42, respectively. Two ISSR primers (UBC861 and UBC862) were able to distinguish the different species. The genetic similarity matrix (GSM) revealed variability among the accessions ranging from 0.116 to 0.816. The intraspecific GSM for Z. schinifolium and Z. piperitum was 0.177–0.780 and 0.250–0.816, respectively. The GSM findings indicate that Z. schinifolium and Z. piperitum accessions have high genetic diversity and possess germplasms qualifying as good genetic resources for cross breeding. The clustering analysis separated Z. schinifolium and Z. piperitum into independent groups, and all accessions could be classified into three categories. Z. Schinifolium var. nermis belonged to independent groups. Comparison of the clusters based on morphological analysis with those based on ISSR data resulted in an unclear pattern of division among the accessions. The study findings indicate that Z. schinifolium and Z. piperitum accessions have genetic diversity, and ISSR markers were useful for identifying Z. schinifolium and Z. piperitum.

        Background: In recent years, adjuvants have received increasing attention owing to the development of purified subunit and synthetic vaccines which are poor immunogens and require additional adjuvants to evoke an immune response. Therefore, immunologic adjuvants have been developed and tested. Plant polysaccharides have been recognized as effective biological response modifiers with low toxicity.Methods and Results: In this study, the polysaccharide from the aboveground part of Astragalus membranaceus Bunge containing immunomodulating arabino-3,6-galactan was evaluated for its hemolytic activity and adjuvant potential in the specific cellular and humoral immune responses to ovalbumin. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge was co-immunized with the purified Vi capsular polysaccharide of Salmonella typhi vaccine in mice. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge did not induce any hemolytic activity or side effects at doses up to 500㎍/㎖. The concanavalin A-, lipopolysaccharide-, and ovalbumin-induced splenocyte proliferation and serum ovalbumin-specific IgG, IgG1 and IgG2b antibody titers in immunized mice were significantly enhanced by AMA. Pharmacological data revealed that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge increased antigen-specific antibody levels in immunized mice. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge-adjuvanted purified Vi capsular polysaccharide of Salmonella typhi vaccine improved the proliferation of splenocytes and macrophages as well as stimulated cytokine production.Conclusions: These results suggest that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge-adjuvanted vaccines enhanced humoral and cellular immunity and that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge is a safe and efficacious adjuvant candidate suitable for use in prophylactic and therapeutic vaccines.

        교량의 안전은 점검에 의해 보장된다. 그리고 교량의 유지관리는 점검결과를 토대로 시작된다. 따라서 점검결과는 ‘교량의 안전’과 ‘효율적인 유지관리’를 위한 가장 중요한 정보이다. 따라서 본 논문에서는 교량의 점검신뢰도를 평가하고 이를 제고 할 수 있는 방안을 제시하 고자 한다. 이를 위해 본 논문에서는 교량의 점검신뢰도를 평가 할 수 있는 세 가지 점검신뢰도를 제시하였다: 명목점검신뢰도, 실질점검신뢰 도, DS명목점검신뢰도. 교량의 점검신뢰도 분석 방법은 정밀안전진단결과를 참값으로 간주하고 당해년도 정밀(정기)점검과 차년도 정밀안전 진단결과를 비교하였다. 인적오류를 고려한 DS명목신뢰도를 기준으로 현재 고속도로교량의 점검신뢰도 85%로 양호한 편으로 평가 된다. 이 는 현재 고속도로 교량의 평균공용연수가 13년으로 대부분의 교량이 ‘A’와 ‘B’등급에 집중적으로 분포되어 상태변화가 크지 않기 때문인 것 으로 판단된다. 점검신뢰도를 제고하기 위해서는 점검자의 역량을 강화하고 전문점검인력 육성이 필요하며 점검결과에 대해 QC(Quality Control, 품질관리)를 실시하여 점검결과의 신뢰도 향상이 필요하다.

        The subject of this experiment is to supply basic data to inhibit non-productive tillers which are uneconomical for mechanical harvesting and to evaluate optimum planting density and sowing date in central district. Total number of tillers and effective first and second of tillers according to different planting density was higher in 80 ㎝ ridge than 60 ㎝ ridge in proso millet. The wider between plant distance on the ridge, the more increased total number of tillers and effective first and second of tillers. The highest effective tillers (91.7 %) in the first tillers was obtained from the second sowing date (23 May) among different sowing date and next is in the order of 3 rd (13 June, 89.8%) > 1 st (2 May, 85.6%) > 4 th (4 July, 85.2%). The percentage of effective tillers in the second tillers was decreased in the order of 2 May (53.7%), 23 May (40.7%), 13 June (22.2%), 4 July (0%) as the sowing date was delayed. There was no significant difference in days to heading and days to ripening according to different planting density. Although culm length was increased as planting density was increased, whereas number of tillers, stem diameter, ear length, grains per ear and 1000 grain weight was decreased. In the growth and yield characteristics of proso millet according to different sowing date, days to heading and days to ripening, culm length, stem diameter, ear length, grains per ear and yield per 10a were decreased. After the sowing date of 13 June, the reduction of growth and yield characteristics were higher because of excess-moisture injury.

        The present study proposes the definition of service life and the end-of- life criterion for bridge maintenance. Bridges begin to deteriorate as soon as they are put into service. Effective bridge maintenance requires sound understanding of the deterioration mechanism as well as the expected service life. In order to determine the expected service life of a bridge for effective bridge maintenance, it is necessary to have a clear definition of service life and end-of-life. However, service life can be viewed from several perspectives based on literature review. The end of a bridge’s life can be also defined by more than one perspective or performance measure. This study presents definition of service life which can be used for bridge maintenance and the end-of life criterion using the performance measure such as a damage score. The regression model can predict an average service life of bridges using the proposed end-of-life criterion.

        Background: Turmeric (Curcuma longa L.) that is used as a food material has antioxidant, anticancer and anti-inflammatory properties. Recently the demand for functional foods and drugs has increased. The present study was carried out to determined of contents of residual sulfur dioxide, heavy metals, ash, acid insoluble ash and curcuminoids in turmeric from the Seoul Yak-ryeong market. Methods and Results: A total of 31 samples were obtained. Residual sulfur dioxide was not detected in any samples. Heavy metals (arsenic, cadmium, lead and mercury) were analyzed by ICP-MS (Inductively Coupled Plasma Mass Spectrometer) and a mercury analyzer and were detected in the ranges of 0.00 - 0.28, 0.00 - 0.07, 0.00 - 0.29 and 0.002 - 0.027㎎/g respectively. No significant difference were observed between the average heavy metal contents of domestic and imported tumeric. However, average content of ash in domestic samples (7.8%) were significantly higher than that in imported samples (6.1%), whereas that of curcuminoids was significantly higher in imported samples (47.6㎎/g) than in domestic samples (11.2㎎/g). The average content of acid insoluble ash was not significantly different between two sample types (0.9% in each). Conclusions: There are no specific standards for turmeric used as food materials. Therefore, this study can be provided as basic data for the establishment of quality standards for turmeric.
