Aphis gossypii is a representative pest that transmits plant viral diseases. It is difficult to control with chemical pesticides alone due to their high pesticide resistance. Entomopathogenic fungi are biological control agents that can replace chemical pesticides and have characteristics of high host specificity and safety to humans. Therefore, we investigated the immune pathways of aphids against initial infection by entomopathogenic fungus. We treated aphids with the Beauveria bassiana JEF 544 strain and examined the immune response in early infection by qPCR. furthermore, we also studied changes the molting time of nymphs and changes in adult nymphal production caused by entomopathogenic fungi.
현재 농가에서 이용되고 있는 보광등과 난방등의 특성을 조사하고 하우스내 기상 환경과 절화장미의 개화와 품질에 미치는 영향을 알아보기 위해 실험을 수행하였다. 보광등으로는 LED등(light emitting diode lamp, LED), 메탈할라이드등(metal halide lamp, MH), 고압나트륨등(high pressure sodium lamp, HPS)을, 난방등으로는 나노탄소섬유 적외선등(nano-carbon fiber infrared lamp, NCFI)을 절화장미 재배상 위에 설치하였다. 그 결과 LED와 NCFI의 광도는 각각 6.8μmol·m-2·s-1과 0.4μmol·m-2·s-1으로 매우 낮은 반면, MH와 HPS는 79 ~ 100μmol·m-2·s-1 범위로 높은 편이었다. 또한 LED와 MH는 적외선역에 파장이 없어 발열량이 거의 없었으나, HPS와 NCFI는 적외선역에 파장이 많아 발열이 많은 것으로 나타났다. 동일한 공간에서 칸막이로 실험구간을 나누어 실험이 수행되었음에도 불구하고 MH, HPS, NCFI에 의해 온도는 2°C 정도 상승되는 결과를 얻었으나, 습도에서는 처리간 차이가 보이지 않았다. 광도에 상관없이 모든 전등들은 절화장미의 개화를 촉진시켜 무처리구에 비해 적게는 5 ~ 7일, 많게는 12 ~ 14일 개화가 촉진되는 것으로 나타났다. 광량이 충분한 봄철에는 전조처리에 의해 개화소요일수가 짧아지는 만큼 절화장도 대조구보다 짧아졌지만, 광량이 부족한 겨울철에는 전조처리에 의해 절화장이 길어져 품질이 향상되는 결과를 얻을 수 있었다.
Ganoderma lucidum has been reported to have various biological activities including antioxidant activity. The objective of this study was to compare the antioxidant effects between Ganoderma lucidum (GL) and antler-shaped fruiting body of Ganoderma lucidum (AGL). In vitro antioxidant activities were examined by 2,2-diphenyl-picrylhydrazyl hydrate (DPPH) and 1,2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities of GL and AGL ethanol extracts. In the DPPH and ABTS radical scavenging activities of GL and AGL ethanol extracts, antioxidant activities of AGL extracts (IC50, 66.94 μg/ml and 131.23 μg/ml) was showed higher than GL extracts (IC50, 83.93 μg/ml and 164.54 μg/ml). Total polyphenol content and total flavonoid content (38.00 g GAE/kg extract and 11.58 g NE/kg extract) of AGL were found higher as compared to GL (34.23 g GAE/kg extract and 3.46 g NE/kg extract). In summary, the results of this study demonstrate that AGL extracts had higher antioxidant activities to GL.
The purpose of this study was to investigate whether or not the antler-shaped fruiting body of Ganoderma lucidum (GL) has an anti-inflammatory effect on lipopolysaccharide (LPS)/interferon-γ (IFN-γ)-activated RAW 264.7 macrophage-like cells. To evaluate the anti-inflammatory effects of GL, we examined the inflammatory mediators such as the production of nitric oxide (NO) and the expression of mitogen-activated protein kinase (MAPK), activator protein 1 (AP-1), inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), interleukin-1β (IL-1β), and interleukin-6 (IL-6). LPS/IFN-γ-induced cellular NO production was significantly decreased in GL-treated RAW 264.7 cells. Moreover, Western blotting analysis results demonstrated that reduced protein expression of MAPK families (such as extracellular signal-regulated kinase, c-Jun amino-terminal kinase, and p38 MAPK) and AP-1-targeting inflammatory enzymes (iNOS, COX-2, IL-1β, and IL-6). These results indicated that GL modulates the MAPK/AP-1 signal pathway in inflammatory process. In conclusion, the present study provides important evidence that GL can potentially be used to reduce LPS/IFN-γ-induced inflammatory response by inhibiting the MAPK/AP-1 signaling pathways.
Chrysanthemums (Dendranthema grandiflorum ‘Iwanohakusen’) were grown in a greenhouse with complete nutrient solution system to investigate the effect of silicon (Si) supplement on salt induced deleterious effects in chrysanthemum plants. The experiment was conducted in plastic pots supplemented with a mixture of upland soil : leaf mold : river sand (3:3:4, v:v:v). Si and salinity were treated in combination with two levels of NaCl (0 and 100 mM) and two sources of silicon (K2SiO3, KSi and silicate fertilizer, SiF) at the same concentration (1.8 mM Si) by weekly-drenching for 12 weeks. Chrysanthemum plants supplemented with Si increased in fresh and dry matter enhancing water content and salinity tolerance. The plants grown under salt stress produced less fresh and dry matter than control plant. However, Si supplement to plants under salt stress ameliorated negative effects of salt stress. In soil, EC and NaCl increased by salt stress were mitigated by Si supplement. Salt stress significantly decreased the contents of K and P in leaf, but Si supplement under salt stress significantly recovered the decreased contents with enormous desorption of K and P in soil. Added Si significantly increased content of available SiO2 with its adsorption by salt stress in soil, which was directly related to Si accumulation in leaf. However, Si uptake by roots was suppressed by salt stress irrespective of Si supplement. Si supplement did not ameliorated the negative effects of salt stress on chlorophyll content and membrane integrity in leaf of chrysanthemum plant although significantly increased Si content in leaf, but reversed pest (Liriomyza trifolii) resistance to above-control level.
Background and Purpose: The Ankle-Brachial Index (ABI) is the ratio of blood pressure in the lower legs to that in the arms. The intima-media thickness (IMT) of extracranial carotid arteries determined by B-mode ultrasound is a measurable index of the presence of atherosclerosis. A low ABI and a high carotid IMT are independently related to increased risk of cardiovascular events. This study examined the association between carotid IMT and ABI in patients with ischemic stroke. Materials and Methods: Retrospectively, 116 hospitalized patients with ischemic stroke were recruited. Using a pulse wave velocity ABI device along with carotid duplex sonography, we measured carotid IMT and ABI and investigated the correlation between average values. Results: There was a significant difference in carotid IMT between the normal and abnormal ABI groups (P=0.0262). The group with an abnormal ABI was more than five times as likely to have increased carotid IMT as the group with a normal ABI (age, sex-adjusted OR 5.67 (95% CI 1.85~17.38)). The ABI and carotid IMT showed a weak inverse linear correlation in patients with ischemic stroke (correlation coefficient −0.378 after adjusting for age and sex). Conclusion: Our study suggests that an abnormal ABI is associated with a high carotid IMT in patients with ischemic stroke.
This study was conducted to evaluate the effect of the exercise on elderly balance ability by using hippotherapy and therapeutic ball exercise. 10 patients were assigned to the hippotherapy group and they got with 30 minutes of hippotherapy. Another 10 elderly were assigned to the therapeutic ball group and they got with 30 minutes of therapeutic ball exercise. All procedures were repeated 5 times a week for the total of four weeks. To investigate the participants balancing abilities, the Time“ Up & Go”(TUG) and One Leg Stand Test(OLST) were evaluated. The results of study were significant differences between pre-test and post-test of TUG and OLST(p<.05), and there were no significant differences between hippotherapy and therapeutic ball exercise(p>.05). The conclusion showed that both the hippotherapy and the therapeutic ball exercises were effective on elderly balancing ability. Consequently, it would be better to practice therapeutic ball than hippotherapy for elderly exercise because the more economical and there is less restriction of space than the hippotherapy.
This study was undertaken to develop PCR primers for the identification and detection of Streptococcus anginosus using species-specific forward and reverse primers. These primers targeted the variable regions of the 16S ribosomal RNA coding gene(rDNA). The primer specificity was tested against 12 S. anginosus strains and 6 different species(10 strains) of oral bacteria. The primer sensitivity was determined by testing serial dilutions of the purified genomic DNA of S. anginosus ATCC 33397T. The data showed that species-specific amplicons were obtained from all the S. anginosus strains tested, but not in the six other species. The PCR could detect as little as 0.4pg of the chromosomal DNA from S. anginosus. This suggests that the PCR primers are highly sensitive and applicable to the detection and identification of S. anginosus.
Immuno-MemBlot is a technique for detecting, analyzing, and identifying proteins, similar to the Western blot technique but differing in that protein samples are not separated electrophoretically but are spotted through circular or slot templates directly onto the membrane. Recently we developed a new Immuno-MemBlot (IMB) method applying immunoreactions and coloring procedures directly in the wells of MemBlot apparatus, which were connected by canals to perform drainage for reagent application and buffer irrigation. This IMB method was designed to get theimmunoblot results more rapidly and clearly than the previous immunoblot ones. This study is aimed to evaluate the analytical accuracy of IMB using different biological assay. In the sensitivity test of IMB the monoclonal antibody can clearly detect the 30 ng (about 12 pM) of Mucocidin peptide (35 mer), and is also available to detect at least 10 ng (about 4 pM) of Mucocidin peptide (35 mer). The IMB was effective in the quantitative analysis of methothrexate (MTX) assay for cellular apoptosis. And more, this IMB is useful to screen large number of specific samples with ease and accuracy in a short time. In the screenings for the presence of Mucocidin in saliva the quantitative comparison is conspicuous among 48 persons depend on the different conditions ofgender, drinking and smoking habits, and oral diseases. Therefore, it is presumed that, even though the target proteins were partly degraded, a specific epitope can be detected if a monoclonal antibody was still reactive. Conclusively, these data suggest that the IMB can be useful in the primary qualitativeand quantitative analysis of proteins in various fluids, i.e., blood, saliva, tear, urine, etc.

The primary growth center (MdPGC) of human fetal mandible was conspicuously distinguished in the soft X-ray view of fetal mandibles.1) As the peripheral adaptive growth of mandible advanced during the postnatal period, the MdPGC became overshadowed by condensed cortical bone. However, in the well-processed radiograms of adult mandible a condensed radiopaque image, measuring 0.5-1.0 cm in diameter, can be observed below the apex of first premolar. In this study we aimed to trace a sclerotic sequela of mandibular primary growth center during postnatal period. Panoramic radiograms of two hundreds adults and soft X-ray views of thirty dry mandible were analyzed by statistical methods. The adult MdPGC was clearly distinguishable from the mental foramen. The area of MdPGC was seldom changed in the older persons, even in the edentulous mandibles. Additionally, the benign lesions of odontogenic cysts and tumors hardly destroyed the original structure of MdPGC, while the malignant tumors of squamous cell carcinoma and metastatic cancer rapidly destroyed and resolved the radiopaque area of the MdPGC.
사용후핵연료 저장조는 지진하중에 대하여 운영기간중 자체의 구조적 건전성 및 저장된 사용후핵료의 안전성을 확보할 수 있어야 한다. 본 연구에서는 LRB 면진장치를 설치한 사용후핵연료 면진저장조의 지진시 거동특성을 파악하기 위하여 서로 다른 특성을 갖는 두종류의 지진에 대하여 지진해석을 하여 그 결과를 비면진 저장조에서의 응답과 비교하였다. 면진장치를 사용함으로써 지진시 상부구조로 전달되는 지진력과 응답을 크게 감소시켜 저장조와 저장된 사용후액연료의 안전성 확보에 유리한 것으로 나타났다.
This study aimed to determine the elemental compositions of selected edible wild plant species, Hemerocallis fulva, Allium victorialis, Syneilesis palmata and Ligularia fischeri. The samples were dried, crushed, and subjected to microwave-assisted digestion. The macro and micro elements were analyzed by inductively coupled plasma-optical emission spectrometery (ICP-OES), and ICP-mass spectrometry (ICP-MS), respectively. The macro elements in the analyzed species decreased in the order K Ca P Mg S Fe Zn Na, and the micro elements ˃ ˃ ˃ ˃ ˃ ˃ ˃ followed the order Mn˃Ba˃Rb˃Cu˃Ni˃Ga˃Li˃Cr˃V˃Co˃Be˃Se. The percentage ratio of calcium content for potassium in the samples was 42.9% (A. victorialis) > 42.4% (S. palmata) > 33.8% (L. fischeri) > 25.3% (H. fulva). The calcium content was 13.7, 10.9, 6.4, and 2.9 times higher than the phosphorus content in S. palmata, L. fischeri, A. victorialis, and H. fulva, respectively (p<0.05). Manganese was the most predominant among the trace minerals, and it followed the order of A. victorialis > H. fulva > L. fischeri > S. palmata. In general, these wild plants are richer in calcium as compared to other common vegetables, and hence can be considered a good source for calcium that is lacking in Korean food products.
This study was conducted to compare the volatile flavor compounds of Artemisia annua L. after extraction by simultaneous steam distillation extraction (SDE) and solid-phase micro extraction (SPME) followed by gas chromatography-mass spectrometry (GC-MS) analysis. Via SDE and SPME processes, 79 (1,254.00 mg/kg) and 39 (488.74 mg/kg) compounds were identified respectively. The compounds extracted by SDE included 27 alcohols, 13 aldehydes, 22 hydrocarbons, 3 esters, 12 ketones, 1 oxide and 1 N-containing compound, on the other hand, using the SPME method, 7 alcohols, 5 aldehydes, 1 ester, 18 hydrocarbons, 7 ketones, and 1 oxide were extracted. The major volatile flavor compounds of Artemisia annua L. isolated by the two methods were caryophyllene oxide, -caryophyllene, camphor, -selinene, -muurolene, 1,8-cineol, (E)-pinocarveol and pinocarvone. β β γ The sesquiterpene named caryophyllene oxide was the most abundant volatile flavor compound with relative contents of 234.16 mg/kg and 195.44 mg/kg obtained by the SDE and SPME methods, respectively. Among the identified volatiles, sabinene, β-pinene, α-terpinene, γ-terpinene, yomogi alcohol, myrtenol, (Z)-nerolidol, p-cymen-8-ol and eugenol were detected by the SDE method only while (E)-anethole and α-cubebene were detected by the SPME method only. This study confirmed that the composition and contents of the volatile flavor compounds vary between different extraction methods. More volatile flavor compounds were identified using the SDE method than the SPME method.
Background : Ganoderma lucidum is a non-toxic, medicinal mushroom, which is known to possess anti-inflammatory and immunomodulating activities. However, the effects and mechanism of action of Ganoderma lucidum on lipopolysaccharide (LPS) and interferon-gamma (IFN-γ)-induced nitric oxide (NO) production and its-related cytokine expression are not yet fully understood. This study aimed to evaluate the effect of Ganoderma lucidum on NO production and NO-mediated pro-inflammatory cytokine expression in LPS/IFN-γ-induced RAW 264.7 macrophage-like cells. Methods and Results : The results showed that Ganoderma lucidum inhibited inducible NO synthase (iNOS) expression of RAW 264.7 macrophage-like cells at non-cytotoxic concentrations probably through the reduction of reactive oxygen species (ROS) production. After pre-treatment of cells with non-toxic doses of Ganoderma lucidum; NO production was significantly decreased. Moreover, Ganoderma lucidum treatment suppressed LPS/IFN-γ -stimulated pro-inflammatory cytokine secretion, including interleukin-1β and interleukin-6, in a dose-dependent manner. Conclusion : Taken together, these results indicate that the anti-inflammatory activation of Ganoderma lucidum in LPS/IFN-γ-stimulated macrophages might be due to abrogation of NO-dependent cytokine release by impairment of iNOS expression via ROS generation.
