Regulations in the EU, Japan, Korea, etc. require that foods and feeds made of or derived from genetically modified organisms (GMOs) should be approved and labeled according to a threshold. Recently, disease resistant transgenic rice was developed in Korea, which resulted from the transformation events involving choline kinase gene, OsCK1. In order to monitor unintended release of the developed GM rice in the near future, as well as to meet GM-labeling requirements, the development of a reliable method for detection of disease resistant GM rice is requisite. Here, specific primer pairs for the detection of GMO was designed on the basis of a introduced gene and the flanking junction sequences between a plant DNA and a integrated gene construct, and also SPS gene was used as an endogenous reference material. Specificities of all designed primers were tested through qualitative PCRs. Clearly, target specific amplicons could be detected from disease resistant GM rice event. In addition, the limits of detection (LOD) using the event-specific primers were approximately 0.1% for the disease resistant GM rice line. This result indicated that the developed detection method is suitable for the traceability of disease resistant GM rice, because of using the primer specifically corresponded to the junction site between plant genomic DNA and inserted DNA. Keywords: genetically modified organisms, disease resistant GM rice, PCR detection, event-specific primer

To obtain basic information of diseases resistant degree of Korean wheat cultivars, we investigated the diseases occurrence and examined field resistance of the materials against to the major diseases. Disease occurrence was investigated in southern regions from March to May, 2012. And the resistance degree was tested to 30 Korean wheat cultivars seeded in autumn. After wintering season, Soil-borne wheat mosaic virus(SBWMV) was infected as 29.4% in the regions. The sharp eyespot was infected about 43% in stem base even if it was not showed distinct symptoms yet. Three Fusarium spp. were identified from rotten stem base and root crown. After heading, mainly fungal damage such as powdery mildew, leaf rust, sharp eyespot and Fusarium head blight were prevalent. Around the harvest season, discoloration of grain by precipitation also occurred and which was different infection degree depending on the each cultivar. 14 cultivars as Geurumil, Dajoongmil showed resistant leaf yellowing spot symptoms. In powdery mildew resistance investigation, recently developed cultivars were more susceptible comparing to none or a little symptom manifestation of older varieties.

Three-year-old seedlings of susceptible Pinus densiflora and resistant Pinus x rigitaeda were each inoculated with the pinewood nematode, Bursaphelenchus xylophilus, to compare disease development. Needle dehydration was evident on seedlings of P. densiflora by 20 days after inoculation, 10 days earlier than this symptom was observed on P. × rigitaeda. Xylem drying was more frequent in seedlings of P. densiflora than in that of P. × rigitaeda between 20 and 60 days after inoculation. No significant differences were found between P. densiflora and P. × rigitaeda for stem water content or for stem and leaf relative water content in current-year branches after nematode inoculation, but the average number of B. xylophilus recovered from stems differed significantly between the two groups. The number of B. xylophilus recovered from stems was negatively correlated with the stem water content and with stem and leaf relative water content. By the time the experiment was terminated at 60 days after inoculation, all 3 of the last group of P. densiflora seedlings had died, but 2 of the 3 remaining P. × rigitaeda hybrid seedlings were still alive. Additional studies are needed to further explore the specific mechanisms preventing nematode multiplication in the seedlings of resistant P. × rigitaeda.

KNU corn breeding team in collaboration with International Corn Foundation (ICF) is dedicated to develop super corn hybrids for developing countries like Nepal and Cambodia. The team works to develop high bio-energy maize hybrids for Korea. Crossing of temperate with tropical germplasm shows new heterotic patterns. Four tropical inbred lines from IITA, Nigeria, ten sub-tropical lines from Nepal and two temperate lines (P45 and 69B) from Korea were crossed to produce single-cross hybrids. Besides, four single-cross hybrids from Nepalwere crossed with P45 and 69B to produce three-way crosses. Gray leaf spot (Cercospora zea-maydis) being the most important problem of corn in mid-hills of Nepal, GLS resistant materials from IITA were crossed with Nepal materials in Cambodia to produce GLS tolerant hybrids. Single-cross hybrids were evaluated at ICF Agriculture Center in Cambodia and KNU farm, Gunwi in Korea in 2008. The three-way crosses were planted at Gunwi farm and the GLS resistant materials were planted at Khumaltar station of Nepal Agricultural Research Council, Kathmandu. Crosses of Tzi-25 with RML-4 and RL-111 and RL-105xNML-2 were found outstanding in Cambodia. Four tropical x subtropical hybrids (Tzi-18xRML-4, Tzi-18xRL-14, Tzi-25xRL-14 and RL-47xTzi-25), four temperate x subtropical hybrids (P45xRL-111, P45xRML-8, RML-5x69B and 69BxRL-105). P45xGaurab was the most suitable for both grain and biomass production.
