        Colon cancer is one of the most common malignant tumors, but there are still a few validated biomarkers of colon cancer. Exosome-mediated microRNAs (miRNAs) have been recognized as potential biomarkers in cancers, and miRNAs can regulate a variety of genes. Recently, Fusobacterium nucleatum was discovered in the tissues of human colon cancer patients. Its role in colon cancer was highlighted. F. nucleatum may contribute to the progression of colon cancer through the mechanism of exosome-mediated miRNAs transfer. However, the exosomal miRNAs regulation mechanism by F. nucleatum in colon cancer is not well known. Thus, we performed next-generation sequencing to investigate the overall pattern of exosomal miRNAs expression in the colon cancer cell culture supernatant. We have confirmed the alterations of various exosomal miRNAs. In addition, to investigate the function of exosomal miRNAs, a Kyoto Encyclopedia of Genes and Genomes analysis was performed on the target genes of changed miRNAs. Potential target genes were associated with a variety of signaling pathways, and one of these pathways was related to colorectal cancer. These findings suggested that F. nucleatum can alter exosomal miRNAs released from colorectal cancer cells. Furthermore, exosomal miRNAs altered by F. nucleatum could be potential biomarkers for the diagnosis and therapy of colon cancer.

        The aim of this study was to compare the two in vitro culture systems callus and adventitious root by investigating the biomass and phenolic compounds in calli and adventitious roots induced from four different explants (leaf, root, petal, and ovary) in Camellia japonica. The biomass of calli and adventitious roots was examined after 4 and 8 weeks of cultivation, respectively, and 22 phenolic compounds were analyzed using high-performance liquid chromatography (HPLC). The biomass of the ovary-derived calli (2.0 g・mass-1) was 1.5-fold that of the leaf-derived calli. The dry weight (DW) was highest in ovary-derived calli; however, the highest dry matter content was obtained from leaf-derived calli. Differences in the investigated characteristics depended on the callus origin. In adventitious roots, the highest biomass was achieved in the leaf-derived adventitious root system; its fresh weight was 2.3-fold (89 mg・ea-1) higher, and its DW was 1.8-fold (16 mg・ea-1) higher than those of ovary-derived adventitious root system. Active cell division was detected in petal-derived lines in both the calli and adventitious roots. Results of the HPLC analysis revealed that the total content of 22 phenolic compounds was highest in ovary-derived calli and ovary-derived adventitious roots. Our experiments confirmed that the calli and adventitious roots of C japonica have different cytological characteristics and bioactive compounds depending on the explant origin. In addition, callus culture was a more suitable system than adventitious root for producing phenolic compounds when the duration of the culture period and biomass were considered.

        배경/목적: 췌장암은 일반적으로 불량한 예후를 보이며, 조기 진단이 어렵다는 점이 이에 영향을 미칠 것으로 생각되고 있다. 본 연구는 한국인 췌장암 환자에 있어서 혈장내 대사체 분석을 시행하여 건강 자원자의 대사체 프로파일과 비교 하였다. 방법: 각 10명으로 비한국인들을 대상으로 이루어진 기존 연구에 비하여 대상자 수가 매우 적지만 검진으로 시행한 복부 CT상 췌장 질환의 증거가 없는 건강한 자원자를 모집하였으며, 이는 췌장암의 조기 진단이 어렵다는 점을 감안할 때 기존 연구에 비한 장점이 될 수 있겠다. 결과: 혈장내 대사체 비교 분석상 췌장암 환자에서 L -lysine의 혈장 농도는 1.36배 높고 L -leucine은 0.63배, palmitic acid는 0.93배 낮은 것으로 나타나 이 세 가지의 대사체 프로파일의 조합이 건강자원자와 췌장암 환자를 가장 잘 구분해 주는 것으로 나타났다. 결론: 앞으로 췌장암의 조기 진단 혹은 발생 역학의 규명을 위하여 대사체 분석에 대한 연구가 더 필요하겠다.

        Bemisia tabaci is a species complex consist of about 40 cryptic species. This species is also a vector of at least 100 begomovirus including Tomato yellow leaf curl virus (TYLCV). TYLCV transmitted by B. tabaci in a circulative propagative manner but its molecular mechanism is remain unclear. We investigate the transcriptional response of B. tabaci MED to TYLCV infection using next generation sequencing (NGS) analysis. In total 16757 differentially expressed genes between viruliferous and non-viruliferous whiteflies were analyzed. Among them 289 transcripts found significant, where 116 up-regulated and 173 down-regulated. Our results reveal the whitefly-TYLCV relationship and provide important molecular information about virus transmission of vector insects.

        Exosomes are Nano-sized lipid vesicles secreted from mammalian cells containing diverse cellular materials such as proteins, lipids, and nucleotides. Multiple lines of evidence indicate that in saliva, exosomes and their contents such as microRNAs (miRNAs) mediate numerous cellular responses upon delivery to recipient cells. The objective of this study was to characterize the different expression profile of exosomal miRNAs in saliva samples, periodically isolated from a single periodontitis patient. Unstimulated saliva was collected from a single patient over time periods for managing periodontitis. MicroRNAs extracted from each phase were investigated for the expression of exosomal miRNAs. Salivary exosomal miRNAs were analyzed using Affymetrix miRNA arrays and prediction of target genes and pathways for its different expression performed using DIANA-mirPath, a web-based, computational tool. Following the delivery of miRNA mimics (hsa-miR-4487, -4532, and -7108-5p) into human gingival fibroblasts, the expression of pro-inflammatory cytokines and activation of the MAPK pathway were evaluated through RT-PCR and western blotting. In each phase, 13 and 43 miRNAs were found to be differently expressed (|FC| ≥ 2). Among these, hsa-miR-4487 (|FC|=9.292005) and hasmiR- 4532 (|FC|=18.322697) were highly up-regulated in the clinically severe phase, whereas hsa-miR-7108-5p (|FC|= 12.20601) was strongly up-regulated in the clinically mild phase. In addition, the overexpression of miRNA mimics in human gingival fibroblasts resulted in a significant induction of IL-6 mRNA expression and p38 phosphorylation. The findings of this study established alterations in salivary exosomal miRNAs which are dependent on the severity of periodontitis and may act as potential candidates for the treatment of oral inflammatory diseases.

        To identify genes that commonly respond to the treatment of different insecticides and are responsible for the toleranceenhancement, transcriptomic profiles of larvae treated with sublethal doses of the five insecticides were compared withthat of untreated control. A total of 117,181 transcripts with a mean length of 662 bp were generated by de novo assembly,of which 35,329 transcripts were annotated. Among them, 125, 143, 182, 215 and 149 transcripts were determined tobe up-regulated whereas 67, 45, 60, 60 and 38 genes were down-regulated following treatments with these five insecticides.The most notable examples of commonly responding over-transcribed genes were two cytochrome P450 genes and ninecuticular protein genes. In contrast, several genes composing the mitochondrial energy generation system were significantlydown-regulated in all treated larvae. Considering the distinct structure and mode of action of the five insecticides tested,the differentially expressed genes identified in this study appear to be involved in general chemical defense at the initialstage of intoxication. Their possible roles in the tolerance/resistance development were discussed.

        To identify genes that commonly respond to the treatment of different insecticides, 3rd instar larvae of the diamondback moth, Plutella xylostella, were treated with sublethal doses (<LC10) of chlorantraniliprole, cypermethrin, dinotefuran, indoxacarb and spinosad via leaf dipping. Then, transcriptomic profiles of treated larvae were compared with that of untreated control. A total of 117,181 transcripts in average with a mean length of 662 bp were generated by de novo assembly, of which 35,329 transcripts were annotated. Among them, 207, 153, 336, 360, and 262 transcripts were determined to be up-regulated whereas 117, 47, 92, 115, and 81 genes were down-regulated following treatments with chlorantraniliprole, cypermethrin, dinotefuran, indoxacarb and spinosad, respectively. Finally, with the criteria of >10 X fold change (FC) and p < 0.05 or >4 X FC, p < 0.05 and q < 0.2, the genes commonly over-transcribed in all treated insects were selected and their over-transcription levels were confirmed by quantitative PCR. These commonly responding genes included three cytochrome P450 genes (Cyp303a1, Cyp6a20 and CYP9E2), three cuticle protein genes (LM-8, LM-19 and TM-A3A), lavesin-1, acyl-CoA D11 desaturase, glucose dehydrogenase, nose resistant to fluxetine protein 6, chorion peroxidase and protein yellow. As the five test insecticides have distinct structure and mode of action, the genes identified in this study were suggested to be involved in general chemical defense at the initial stage of intoxication. Their possible roles in tolerance and resistance development were further discussed.

        To have a better understanding of pluripotency, whole gene expression of embryo-derived stem cells (EdSCs) in bovine species was investigated. EdSCs were established from the embryos produced by in vitro fertilization, parthenogenesis and somatic cell nuclear transfer. Then, the microarray was performed and analyzed. Differently expressed genes (DEGs) were also confirmed by Real-time PCR. Among 10,203 DEGs, little difference was found in gene expression among three kinds of EdSCs. Conversely, all EdSCs have an immensely different gene expression when compared with somatic cells, consistent with scatter plat results. To investigate shared pathways for pluripotency in all EdSCs, 2,415 co-DEGs were identified which compared with somatic cells. By KEGG database, there were 54 signaling pathways in co-DEGs and some of them were related with pluripotency maintenance such as TGFβ, WNT and JAK-STAT signaling. In TGFβ signaling, BMP family and SMAD family were involved in co-up-regulated DEGs. In WNT signaling, WNT family and receptors were included in co-up-regulated DEGs, while inhibitors of WNT signaling were associated with co-down-regulated DEGs. In JAK-STAT signaling, STAT3 belonged to co-down-regulated DEGs. These DEGs were also confirmed by Real-time PCR. Taken together, BMP and WNT pathways may be activated and paly central roles to retain pluripotency in bovine EdSCs, whereas the LIF/STAT3 pathway may not be operated well.         This paper analyzes the simulation operation flow of Bullet physics engine. Based on this analysis, four kinds of multi-rigid-body game characters are designed. This paper also profiles the performance metrics such as the CPU utilization, the memory usage, and the computation time by multi-rigid-body character simulations. For the CPU utilization, the Tongs Vehicle is the best and provides 45.1% less than the other character simulations. The computation times of the Four-leg robot and the Dragon are longer than those of the others. The memory usage of the Dragon simulation is the largest, which is average 1.32 times more than the others. Because all parts of Dragon are composed of triangular mesh models in 3DMax. The performance profiling with the criteria such as reducing the computation time and the computing resources, the complexities of the collision shapes, and the number of rigid bodies takes an important role in the design of the multi-rigid-body game characters.

        최근 유럽 의회와 이사회가 합의한 유럽연합 개인정보보호 규칙안은 정보처리기술의 발달에 따른 다양한 개인정보 활용 태양에 대하여 기존의 유럽연합 개인정보보호지침보다 세분화된 규정들 을 두고 있다. 이 중 빅데이터의 활용과 관련하여 특히 주목할 만한 부분은, 목적외 이용과 프로파 일링을 비롯한 자동화된 개인정보처리 및 이에 근 거한 개인에 대한 중요 결정에 대한 취급에 관한 사항들이다. 이에 본고에서는 현행의 유럽연합개 인정보보호지침과 새로운 규칙안, 그리고 우리나 라의 정보통신망법과 개인정보보호법의 관련 규 정을 비교해 보았다. 특히 규칙안과 우리나라법을 비교하여 볼 때, 규칙안은 목적외 이용에 관하여 우리나라 법보다 다소 유연한 규정을 두고 있으며, 프로파일링은 물론 프로파일링에 근거한 개인에 대한 자동화된 개별 결정에 대한 규정을 따로 두고 있다. 또한, 목적외 이용 및 프로파일링, 자동화된 개별 결정 과 관련하여 정보주체의 권익을 보호하기 위한 보호조치를 강조하고 있다. 이와 같은 접근은 정 보처리기술의 발달로 다양한 방식 및 내용의 개 인정보처리가 가능해지고, 이로 인하여 정보주체 의 권익에 미치는 영향이 커지는 현실을 감안할 때 우리 법의 입법론으로서도 참고할 만하다고 생각된다.

        Odontogenic cells express many genes spatiotemporally through complex and intricate processes during tooth formation. Therefore, investigating them during the tooth development has been an important subject for the better understanding of tooth morphogenesis. The present study was performed to identify the genetic profiles which are involved in the morphological changes during the different stages of rat tooth development using the Agilent Rat Oligonucleotide Microarrays. Morphologically, the maxillary 3rd molar germ at 10 days post-partum (dpp) was at the cap/bell stage. In contrast, the maxillary 2nd molar germ showed the root development stage. After microarray analysis, there were a considerable number of up- or down-regulated genes in the 3rd and the 2nd molar germ cells during tooth morphogenesis. Several differentially expressed genes for nerve supply were further studied. Among them, neuroligin 1 (Nlgn 1) was gradually downregulated during tooth development both at the transcription and the translation level. Also, Nlgn 1 was mostly localized in the dental sac, which is an important component yielding the nerve supply. This genetic profiling study proposed that many genes may be implicated in the biological processes for the dental hard tissue formation and, furthermore, may allow the identification of the key genes involved in the nerve supply to the dental sac.

        The more the marketplace become competitive, the more clear and distinct market segments the marketers need to identify. A minority of consumers takes important roles in the marketplace as market influencers or diffusers of information to others, for instance, market maven or opinion leaders. Market mavens tend to have overall market-related knowledge, while opinion leader and fashion leader possess product class-specific information. Fashion leaders are more likely to adopt a product at the early stage, but opinion leaders or market mavens are not necessary. Despite significant roles as reference groups, limited research has examined the differences in the essential traits of three influential groups. The purpose of this research is to examine and compare the differences of psychological attributes in market maven, opinion leaders, and fashion leaders with respect to consumer self-confidence, clothing involvement(INV), status consumption(STATUS), and price consciousness(PRICE). The instrument was modified based on the previous studies(Bearden, et al., 2001; Clark & Goldsmith, 2005; Feick& Price, 1987;Goldsmith, et al., 1991) and each item was measured by seven-point Liker type scales. A total of 857 data were collected through the internet survey method. About 50.3% of respondents were female, 39.2% were single, and age ranged from 20 to 59 years old. Exploratory factor analysis confirmed the differences of the measurement in three influential groups, explaining 70.76% of variances. Consumer self-confidence was generated into five factors, information acquisition & consideration-set formation (IA&CF), personal outcomes(PO), social outcomes (SO), persuasion knowledge(PKN), and marketplace interfaces(MI). Cronbach's alpha was ranged between .78 and .93. In order to investigate the effects of psychological attributes on three influential groups, five factors of self-confidence (IA&CF, PO, SO, PKN, and MI), INV, STATUS, and PRICE were entered as the independent variables in the regression model respectively. In explaining market maven, IA&CF(β=.37) and SO(β=.35) showed the strong positive effects, and STATUS, PKN(-), MI(-), and PRICE were also significant in order (F=107, adj. R2=.498). Opinion leaders were significantly related with SO(β=.84), and PO, INV, and PRICE presented the minor effects(F=496.2, adj. R2=.822). SO(β=.38), STATUS(β=.37), INV(β=.34), and PKN were significant predictors for fashion leaders (F=289.3, adj. R2=.729). When analyzing the influence of market maven, opinion leader, and fashion leader on buying behaviors, market maven and fashion leaders were significantly, positively related with impulse buying behavior (F=69.28, adj. R2=.193), and overall satisfaction(F=38.21, adj. R2=.115). The implications were discussed.

        Vitellogenins (Vgs) are precursors of the major egg storage protein, vitellin (Vn), in many oviparous animals. Insects Vgs are large molecules (200-kD) synthesized in the fat body in a process that involves substantial structural modifications (e.g., glycosylation, lipidation, phosphorylation, and proteolytic cleavage, etc.) of the nascent protein prior to its secretion and transport to the ovaries. However, the extent to which Vgs are processed in the fat body varies greatly among different insect groups. We were cloned Vgs partial genes PaVgs and BgVgs from Periplaneta americana and Blattella germanica. Real-time quantitative PCR shows that PaVgs and BgVgs were differential-regulated with aging. In insects, glutathione S-transferases (GSTs) are enzymes involved in detoxification of insecticides. We were cloned GST partial genes PaGST and BgGST from Periplaneta americana and Blattella germanica. Real-time quantitative PCR shows that PaGST and BgGST were up-regulated with aging, and the mRNA level of PaGST and BgGST was higher in 4℃ and 37℃ than room temperature. The expression level of PaGST and BgGST exposure to temperature stress suggests that PaGST and BgGST are up-regulated after exposure low and hige temperature treatments.
