Multiple interferon tau (IFNT) genes exist in bovine. An antiluteolytic substance secreted by the bovine conceptus and primarily responsible for maternal recognition of pregnancy is bovine trophoblast protein 1 (bIFNT1), one of new type I interferon tau (IFNT) genes. The objectives of this research were to investigate whether multiple, distinct gene encode bIFNT1 and other type I bIFNT gene in the bovine genome or not and to examine the expression of bIFNT1 and other bIFNTc1 mRNAs during conceptus development. The transcription of these genes could be regulated through caudal-related homeobox-2 (CDX2) and ETS2 and/or AP1(JUN) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. The presence of mRNAs encoded by bIFNT1 and type I bIFNTc1 genes were examined quantitatively via reverse transcription-polymerase chain reaction (RT-PCR) analysis of total cellular RNA (tcRNA) extracted from on the days 17, 20 and 22 bovine conceptuses. bIFNT1 was highly expressed on the day 17 and transcripts were gradually and weakly detectable on the days 20 and 22. However, the other bIFNTc1 gene examined transcripts was highly expressed on the day 20 and transcripts were weakly detectable on the days 17 and 22 bovine conceptuses. Furthermore, human choriocarcinoma JEG3 was co-transfected with an -1kb-bIFNT1/c1-Luc constructs and several transcription factor expression plasmids. Compared to each -1kb-bIFNT1/c1-Luc increased when this constructs were co-transfected with, ETS2, AP1(JUN), CREBBP and/or CDX2. Also, bIFNTc1 gene was had higher effect on activity by alone ETS2, and AP1(JUN) expression factors in choriocarcinoma JEG3 cell. However, bIFNT1 gene expression of the upstream region was not idented. These results demonstrate that these genes display differential, tissue-specific expression and developmental regulation during pregnancy.

Kisspeptin-10 (KP-10) has been reported to act as a tumor metastasis suppressor via its receptor, G protein-coupled receptor 54 (GRP54). The KP-10/GPR54/BMPs signaling pathway plays an important role in embryonic kidney development. However, its function in osteoblast differentiation is unknown. The aim of this study was to confirm the molecular mechanism for the action of KP-10 on osteoblast differentiation. Expression of the Bone morphogenetic protein-2 (BMP2) and osteogenic genes were determined by RT-PCR and real-time PCR analysis in C3H10T1/2 cells. Transient transfection assays were performed to confirm the effects of KP-10 on BMP2-Luc activity. BMP2 and phospho-Smad1/5/9 protein levels were determined by Western blot analysis. Alkaline phosphatase (ALP) staining experiment was performed to evaluate ALP activity. To further confirm the effect of KP-10-induced GPR54, we used GPR54 Knock out (KO) C3H10T1/2 cells. KP-10 significantly increased osteogenic gene such as Runx2, ALP and Dlx5 in C3H10T1/2 cells. The ALP staining levels were also increased by KP-10. Interestingly, BMP2 mRNA, protein expression and promoter activity were also increased by KP-10. However, KP-10-induced BMP2 expressions were not increased in GPR54 KO cells. These results suggest that KP-10 increases BMP2 expression through GPR54. Next, Western blot analysis shown Smad1/5/9 phosphorylation were enhanced in a time-dependent manner by KP-10 treatment. It is well known that BMP2 increased phosphorylation of Smad1/5/9 via BMP2 receptor. In addition, KP-10 increased NFATc4 mRNA levels and NFATc4 overexpression enhance BMP2 mRNA levels. To confirm the KP-10-induced BMP2 action, we used KP-10-treated medium in wild type cells and GPR54 KO cells. The osteogenic genes were not elevated by KP-10-treated medium (GPR54 KO cells) whereas increased expression levels by KP-10 medium (wild type cells). These data indicate that KP-10 induced osteoblast differentiation through NFATc4-mediated BMP2 signaling.

Agricultural by-productsis a rich source of dietary fibers including arabinoxylans, which is found mainly in water-insoluble form and reported to have immunomodulatory activities. We investiglatedwhether solublearabinoxylan (AX)-enriched byproducts alters macrophage activity.Agricultural by-products wereorally given to mice for 4 weeks. Peritoneal macrophages from mice were assayed for scavenger receptor expression, phagocytosis, and lipopolysaccharide (LPS)-stimulated responses. Serum levels of inflammatory cytokines were determined 1 hour after intraperitoneal injection of LPS. Agricultural by-productstreated mice showed a higher expression of SRA and CD36, accompanied by enhanced phagocytosis of latex beads, relative to control mice. Upon stimulation with LPS ex vivo, macrophages from the byproducts group showed lower levels of CD86 and CD40 at the surface protein level and lower production of NO, tumor necrosis factor (TNF)-α, and interleukin (IL)-6, compared with the control group. Theagricultural by-products group showed a decrease in serum levels of LPS-induced TNF-αand IL-6, while mice given crude byproducts, which was prepared without enzymatic hydrolysis, showed a reduction only in serum TNF-α,indicating that soluble arabinoxylans contributes to the immunomodulatory effect ofagricultural by-products. Our data present evidence that soluble arabinoxylan-enriched agricultural by-products can be an immune enhancing functional food.

Fucoidan is a sulfated polysaccharide that is purified from brown algae, such as Fucus vesiculosus. This compound has multiple biological activities including immune-stimulating, and anti-viral activities. We recently demonstrated that the cytotoxicity of fucoidan can be dependent on the batch of its production and its molecular weight. In a previous study, fucoidan B exerted cytotoxicity toward mouse spleen cells. To confirm the biological activity of Fucoidan B, we cultured HL-60 cells, a human leukemia cell line, and treated then with fucoidan. The metabolic activity of the HL-60 cells decreased in response to treatment by fucoidan. Moreover, the morphology of HL-60 treated by fucoidan changed. To investigate the fucoidan’s effects, we analyzed the size and level of Annexin V/propidium iodide staining of HL-60 cells using a flow cytometer. Fucoidan consistently induced cell death, including apoptosis of HL-60 cells. As potential mechanisms, fucoidan destabilized the mitochondrial membrane potential and altered the production of reactive oxygen species in HL-60 cells. Taken together, these results suggest that fucoidan has anti-tumor activity on HL-60 cells via destabilization of mitochondrial membrane potential. The present study demonstrates that fucoidan can be used as an anti-cancer agent for leukemia.

The study examined qualities including chlorophyll, total phenol, total flavonoid content and antioxidative activity in 7-year-old mistletoe that was artificially cultivated with Japanese apricots as the host. The chlorophyll a content of the artificially cultivated Japanese apricot mistletoe leaves was 7.67 mg/g (old leaves) and 3.17 mg/g (new leaves), while the chlorophyll b content was 0.80 ~ 3.53 mg/g and 1.31 ~ 3.56 mg/g. The carotenoid content of the Japanese apricot mistletoe leaves were in the range of 0.29 ~ 2.48 mg/g, and the old leaves (1.85 mg/g) had a carotenoid content 2.1 times higher than the new leaves (0.89 mg/g). The total polyphenol content of the leaves and nodes of the 7-year-old mistletoe was 60.22 mg/100 g TAE for the leaves and 35.50 mg/100 g TAE for the nodes. Among the host trees, polyphenol content was highest (81.44 mg/100 g TAE) in the old mistletoe leaves of the Quercus mongolica, while the total flavonoid content of the Japanese apricot mistletoe was higher in the new leaves (47.89 mg/100 g RUE) than the old leaves (42.19 mg/100 g RUE). The DPPH radical scavenging ability of the Japanese apricot mistletoe scored a low 22.4 ~ 28.5% in the leaves, but was relatively high in the nodes at 52.4 ~ 80.1%. In terms of 1mg/mL concentration, the new leaves (69.1%) and old leaves (82.3%) of the Quercus monglica mistletoe displayed the highest inhibition rate. Chestnut mistletoe displayed an inhibition rate of 21.9 ~ 31.9% for the leaves, and 35.0 ~ 63.8% for the nodes. These results indicate that Japanese apricot mistletoe would be substitute oak tree mistletoe as a source of bio-active property for functional food and medicine.

This study was conducted to investigate the total polyphenol, total flavonoid and the antioxidant activity in different origin and parts of Moringa olerifera cultivated in Cheorwon. The extraction yield from leaf, root and stem were 25.5, 23.0, 12.3%, respectively. The total polyphenol content was high in the leaf extracts (15.77 mg/g) followed by root extracts (11.23 mg/g) and stem extracts (3.50 mg/g) but the difference of origin was not observed. The total flavonoid content was high in the leaf extracts (14.78 mg/g) followed by stem (1.36 mg/g) and root (1.14 mg/g). The total flavonoid content of Philippines and Thailand leaf were statistically high compared to india leaf. The DPPH radical scavenging activity was high in the leaf extracts at the 1 mg/ml, 0.25 mg/ml but the difference of origin was not observed. The DPPH radical scavenging activity may be attributed to the presence of several compounds such as polyphenol and flavonoid. Hence consumption of diet supplemented with Moringa olerifera leaves could protect the human against diseases induced by oxidative stress.

Background: Many previous studies recommended the side-lying hip abduction (SHA) exercise for targeting the gluteus medius (Gmed) and gluteus maximus (Gmax) muscle activity while the decreasing tensor fasciae latae (TFL) activation. Mischoice of hip position and angle in SHA may increase the risk of lower extremity injuries and undesirable muscle activation. However, information is limited on the effect of composite hip flexion angles and hip rotation on the gluteal muscle activity during SHA. Objects: This study aimed to compare muscle activity (Gmed, TFL, and Gmax) and activity ratios (Gmed/TFL, Gmax/TFL, and Gmed/Gmax) using surface electromyography (EMG) during SHA exercise at three different hip flexion angles either with or without internal rotation (IR) in subjects with Gmed weakness. We hypothesized that applying hip flexion and IR during SHA would increase gluteal muscle activity and decrease TFL activity. Methods: Muscle activity and activity ratios in 20 volunteers with Gmed weakness during 6 different SHA were investigated with surface EMG. One-way repeated-measures analysis of variance was used to determine the statistical significance. Results: Significant differences were found among the six different exercises for Gmed (F2,41=11.817, p<.001) and Gmax (F3,52=5.513, p=.003) muscle activity, and Gmed/TFL (F3,54=8.735, p<.001) and Gmax/TFL (F2,37=4.019, p=.028) activity ratios. Conclusion: Applying hip flexion is an effective method for increasing gluteal activity, and it elicits great Gmed/TFL and Gmax/TFL activity ratios during SHA in subjects with Gmed weakness.

The effect of drying temperature and steaming time on the browning and antioxidant activity of dried Platycodon grandiflorum was investigated. Thirteen treatment conditions were constructed using central composite face-centered design containing 5 center points. Drying temperature and steaming time (as factors) were 45-75oC and 15-45 min. According to treatment conditions, dried Platycodon grandiflorum was assessed for color characteristic, degree of browning, total polyphenol content, and DPPH and ABTS free radical scavenging (as responses). When increasing drying temperature within a given steaming time, dried Platycodon grandiflorum exhibited decreased lightness, increased redness, degree of browning, and total polyphenol contents, and enhanced antioxidant activities. Except for total polyphenol contents and antioxidant activities, steaming time within a given drying temperature exhibited similar effects to those observed in drying temperature. However, steaming time did not likely influence total polyphenol contents and revealed the opposite trends observed for the effect of drying temperature on their antioxidant activities. The overall results suggested that drying temperature was the main factor for changes in the browning and antioxidant activity of dried Platycodon grandiflorum.
