Plastics are widely used in industries in human society and because of their structural stability, degradation is a serious global issue. To estimate the degradation of plastic, 31 edible mushrooms were cultured with the selected plastic films (polyethylene [PE], polystyrene [PS], and poly(ethylene terephthalate) [PET]) for 3 months at 25 °C. Measuring the weight of the films showed that four species of mushrooms, namely Porostereum spadiceum, Ganoderma lucidum, Coprinellus micaceus, and Pleurotus ostreatus, exhibited the highest degrees of plastic degradation. In addition, the mushrooms and fungi that exhibited the most significant plastic degradation were cross-cultured to promote this degradation. As a result, cross-cultivation of G. lucidum and Aspergillus niger showed a weight loss of 2.49% for the PET film. For the PS film, Aspergillus nidulans showed a weight loss of 4.06%. Cross-cultivation of A. nidulans and C. micaceus, which showed a weight loss of 2.95%, was noted as an alternative for PS biodegradation, but is harmful to humans. These bio-degradation effects of edible mushroom will contribute to the development of alternatives for eco-friendly plastic degradation.
This research aimed to examine the effects of grapefruit seed extract (GSE) at various concentrations on the microbial safety and physicochemical characteristics of onion puree (0.01~0.1%). The onion puree was kept at 4℃ for 14 days. The results of the study indicated that the addition of GSE did not cause any significant changes in the sample’s brix degree and viscosity in onion puree (p<0.05). However, as the concentration of GSE increased, the pH level decreased. On the other hand, as GSE was added, the lightness of the onion puree increased, while the redness and yellowness decreased. Compared to pure onion puree, the GSE-incorporated onion puree had higher levels of total flavonoid and total polyphenol content, indicating that it helps to maintain antioxidant activities. Based on the microbial safety test, aerobic bacteria, yeast, and mold were absent until day 14 of storage. In conclusion, the study suggests that the addition of GSE to onion puree increases its antioxidant activity and shelf-life.
UV and O3 are materials used in the water treatment process, and many studies have been reported to remove organic matters, contaminants, and microorganisms. In this study, we were investigated effects of Chirnomidae (Chironomus flaviplumus, Chironomus riparius), which are contamination indicator species to exposure UV and O3 for the survival rate, body color change and gene expression response. The survival rate of C. flaviplumus exposed to UV decreased to about 70% after 24 hours, and C. riparius about 50%. There was no change in the survival rate of C. flaviplumus exposed to O3, and C. riparius decreased to 95% after 10 minutes of exposure, but there was no change during the subsequent exposure time. In addition, UV and O3 exposure to the two species in body color faded in a time-dependent. In the HSP70 gene expression, C. riparius showed an increase in expression after UV exposure compared to the control group, and a significant difference was shown 12 hours after exposure (P<0.05). C. flaviplumus exposed to O3 showed a relatively low expression compared to the control group, and showed a significant difference at 10 minutes and 1 hour after exposure (P<0.05). These results reported the ecotoxicological effects on Chironomidae according to UV and O3 exposure. Therefore, the results of this study can be used as basic data to understand the effects of UV and O3, which are disinfectants used in water treatment plants, on Chirnomidae entering plants. The objective of this study was to achieve biological control of green mold disease in Pyogo mushrooms using antagonistic microorganisms. Bacillus subtilis BSM320 cells inhibited mycelial growth by 48–60% against three Trichodermaisolates including T. hazianumisolated from the substrates of Lentinula edodes, showing their antifungal activity.The bacteria were cultured to a high density of 4.2 x 109±113.7 cfu/mlin aqueous extract of composted spent mushroom substrates of L. edodes containing 1% glucose and showed a higher growth rate than that observed when using the commercial medium, Luria-Bertani broth. The bacterial culture showed a 75% protective effect without damaging the mushroom fruiting bodies. These results suggest that B. subtilis BSM320culture is suitable for biological control of green mold disease during mushroom cultivation.
V. parahaemolyticus causes waterborne and foodborne disease such as acute diarrhea. In this study, V. parahaemolyticus isolates from seawater, fish tanks, and distributed fishery products in Jeju were investigated for potential toxin or species-specific genes (tdh, trh, tlh, and toxR) using RT-PCR and their genetic characteristics were analyzed using Pulsed-field gel electrophoresis (PFGE). Overall, V. parahaemolyticus of 90 strains (36.7%), including 33 strains from seawater, 8 strains from fish tanks, and 50 strains from fishery products, were isolated from 245 samples. All V. parahaemolyticus strains did not detect the tdh gene, whereas all strains detected tlh or toxR genes. In addition, trh genes were detected in 3 strains from seawater and 1 strain from fishery products. Monthly quantitative testing of seawater revealed that V. parahaemolyticus was positively correlated with water temperature. The 90 strains of V. parahemolyticus obtained in this study showed by gene homology between types, ranging from 64.0–97.3%. Among these, thirteen types showed 100% homology between genes. These results indicate that continuous monitoring is needed to facilitate food poisoning epidemiological investigations because some isolated V. parahaemolyticus strains harbored toxin genes and V. parahaemolyticus strains isolated from seawater, fish tanks, and distributed fishery products showed genetic similarity.
본 연구에서는 수산물 시료 중 Salmonella spp. 검출을 위해 단시간의 전배양(2시간 이내)과 탈염과정을 포함한 DNA 추출법을 사용하여 분자생물학적 검출을 위한 수산 물 전처리 방법에 대해 연구하였다. 배양 시간에 따른 증 균 효율을 탐색하기 위해 100, 101 및 102 CFU/mL농도 의 Salmonella spp. 5종을 NB 0.5에 접종하여 증균 전, 1시간 및 2시간 동안의 증균 효율을 비교하였다. 그 결 과, 2시간 동안 모든 농도에서 약 1 log CFU/mL가 증균 되어 초기 농도와 유의적인 차이가 나타났다. 또한 지역 별 패류시료에 S. Typhimurium을 인위적으로 감염시킨 뒤 DNA를 추출하여 염농도를 측정한 결과, 모든 시료의 염농도가 0%로 DNA 추출과 동시에 탈염이 이루어진 것 을 확인하였다. 이후 추출한 DNA를 사용하여 PCR을 수 행한 결과 모든 시료에서 S. Typhimurium의 특이적 양성 밴드가 확인되었다. 다음으로 수산물 시료 중 Salmonella spp. 검출을 위한 증균 과정과 탈염을 포함한 DNA 추출 방법의 검증을 위해 멸균 홍합시료 및 비멸균 홍합시료 에 Salmonella spp. 5종을 인공적으로 약 100, 101, 102 CFU/g의 농도로 오염시켜 전배양과 DNA를 추출하여 PCR로 특이적 증폭 밴드의 여부를 확인한 결과, 모든 농 도의 Salmonella spp. 5종에서 특이적 밴드가 확인되었다 . 결과적으로 본 연구에서 제시한 전배양 및 DNA 추출 방법을 포함한 전처리 방법과 PCR을 사용하여 수산물 시 료에서 10 CFU/g 미만의 Salmonella spp.를 검출하였으 며, 시간과 비용면에서 효율적이며 과정이 복잡하기 않기 때문에 수산물의 처리 현장에 활용될 수 있을 것으로 기 대된다.
Cyanobacterial resting cells, such as akinetes, are important seed cells for cyanobacteria’s early development and bloom. Due to their importance, various methods have been attempted to isolate resting cells present in the sediment. Ludox is a solution mainly used for cell separation in marine sediments, but finding an accurate method for use in freshwater is difficult. This study compared the two most commonly used Ludox methods (direct sediment treatment and sediment distilled water suspension treatment). Furthermore, we proposed a highly efficient method for isolating cyanobacterial resting cells and eDNA amplification from freshwater sediments. Most of the resting cells found in the sediment were akinete to the Nostocale and were similar to those of Dolichospermum, Cylindrospermum, and Aphanizomenon. Twenty times more akinetes were found in the conical tube column using the sediment that had no treatment than in the sample treated by suspending the sediment in distilled water. Akinete separated through Ludox were mainly spread over the upper and lower layers in the column rather than concentrated at a specific depth in the column layer. The mibC, Geo, and 16S rDNA genes were successfully amplified using the sediment directly in the sample. However, the amplification products of all genes were not found in the sample in which the sediment was suspended in distilled water. Therefore, 5 g to 10 g of sediment is used without pretreatment when isolating cyanobacterial resting cells from freshwater sediment. Cell isolation and gene amplification efficiency are high when four times the volume of Ludox is added. The Ludox treatment method presented in this study isolates cyanobacterial resting cells in freshwater sediment, and the same efficiency may not appear in other biotas. Therefore, to apply Ludox to the separation of other biotas, it is necessary to conduct a pre-experiment to determine the sediment pretreatment method and the water layer where the target organism exists.
본 연구에서는 한국 남해안의 해상가두리에서 북방전복 Haliotis discus hannai 부착생물의 종류 와 밀도에 관한 자료의 축적과 부착생물 제거에 따른 영향을 평가하고자 하였다. 해상가두리에 서 북방전복의 부착생물은 굴, 태형동물, 따개비가 우점하였으며, 이들의 평균 부착생물 면적비 는 약 57.5%였다. 성장률과 간췌장의 건강도는 대조구에 비해 부착생물 제거구에서 좋은 결과 를 보였다. 따라서 전복의 부착생물 제거는 개체의 성장과 출하 시 상품성 향상을 위해 필요한 것으로 판단된다.
This study compares and analyzes the drogue shape applied to aerial refueling with the U.S. Navy drog shape through basic analysis of the model based on the flying fish shape, a marine creature. Flying fish is a creature that is advantageous for non-powered downhill, and this shape is applied to the probe feeding method to minimize the burden on the pilot and the need for refueling technology, which are disadvantages of the probe method. To this end, the results through analysis of basic studies were confirmed, and the superiority and insufficiency of the flying fish shape model and the US Navy drogue model were compared. Therefore, as confirmed through analysis, the flying fish model had a 14.1% lower maximum air pressure and a 9.1% faster maximum speed. Based on this, it is hoped that the advantages and disadvantages of flying fish shape models will be confirmed through actual flight tests in the future.
Despite the recent increase in the consumption level of the processed meat-byproducts, the health and safety issue has consistently been raised in the processes of production, distribution and consumption. The purpose of this study is to analyze and evaluate the microbiological hazard elements in the Korean sausage, “Sundae,” to present not only the safety standard of meat by-product vendors based on HACCP (Hazard Analysis Critical Control Point), but also the quality control criteria and sanitary arrangements of small manufacturers. For the study, the microbiological hazards in 24 raw materials, 7 manufacturing processes, 40 facilities and tools, 17 workplace environment, and 12 workers were analyzed. The analysis revealed the hazardous elements in the initial stages with 6.28 and 4.07 log CFU/g of total aerobic count and coliforms, respectively, detected from the porcine blood and 3.23 log CFU/g of coliforms from the porcine small intestines. The result also showed that the total aerobic counts and coliforms in the process of mixing and filling process exceeds the standards in the hygiene guidelines by Natick with the total aerobic counts of 5.23, 5.45 log CFU/g, and the coliforms of 3.25, and 3.31 log CFU/g, respectively. Although the detected total aerobic count and the coliforms in the filling and washing rooms exceeded the standards, it was found that the total aerobic count was significantly reduced by 98% after cleaning and disinfecting and no coliforms was detected in any process thereafter. In order to achieve high level of safety in the manufacturing processes of Sundae, the separation of washing and disinfection room from the other sections and the sanitation control of the workers must be preceded, along with strict monitoring in the storage and distribution processes. The study raises necessity for additional studies for the safety evaluation of the processed meat-byproducts and further researches on the validity of the critical limits.
본 연구는 전남 여수시 소재 거문군도에 분포하는 박달목서의 개체군 분포특성과 개체군 동태, 보존생물학적 연구를 통해 박달목서 개체군의 보전방안을 제시하기 위해 수행되었다. 거문군도 내 4개의 도서에 분포하는 박달목서는 고도에 31개체, 동백섬 41개체, 서도 49개체, 동도 1개체로 총 122개체가 조사되었으며, 서도와 동백섬에서는 개체 수가 점차 증가할 것으로 추정되었다. 122개체 중 성숙목은 81개체였으며, 41개체는 미성숙목 이었다. 성숙목 중 암나무는 39개체, 수나무 42개체로 성비는 1.08로 추정되었다. 유효집단크기는 23.0~30.9으로 성숙목 집단의 크기보다 작은 것으로 나타났다. 암․수나무의 공간분포는 암나무가 수나무에 비해 집중분포하는 경향을 보였다. 박달목서의 유전적 다양성은 10개의 isozyme에서 12개의 유전자좌가 산출되었고, Nei의 유전적 다양성은 평균 E=0.148로 다소 낮은 수준이었다. 그리고 아집단간 유전적 분화 정도는 Fst=0.078로서 매우 낮은 수준이었으며, 유전자 이동(Nm)은 2.94로서 높은 결과를 보였다. 거문군도에 분포하는 박달목서의 분포특성과 개체군 동태 및 보존생물학 적 연구결과를 종합하여 박달목서 개체군의 보전방안을 제시하였다.
Recently, the purchase of fresh-cut produce and meal kits has increased. Ready-to-eat (RTE) fresh-cut products have potentially hazard of cross-contamination of various microorganisms in the processes of peeling, slicing, dicing, and shredding. There are frequent cases of protozoa food poisoning, such as Cyclospora and Cryptosporidium, caused by fresh-cut products. The objective of the study is to investigate the microbiological qualities of various types of RTE fresh-cut products in the domestic on/offline markets. RTE fresh-cut fruits cup (n=100), fresh-cut vegetables (n=50), and vegetables in meal kits (Vietnamese spring rolls and white radish rolls kits, n=50) were seasonally analyzed. The contamination levels of hygienic indicator organisms, yeast and mold (YM), and foodborne pathogens (Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Salmonella spp., and Escherichia coli O157:H7) were monitored. Overall, the lowest microbiological qualities of meal kits vegetables were observed, followed by RTE fresh-cut fruits cup and fresh-cut vegetables. Contamination levels of total aerobic bacteria, coliforms, and YM in meal kits vegetables were 5.91, 3.90, and 4.71 logs CFU/g, respectively. From the qualitative analysis, 6 out of 200 RTE fresh-cut products (3%) returned positive result for S. aureus. From the quantitative analysis, the contamination levels of S. aureus in purple cabbage from a meal-kit and fresh-cut pineapple were below the acceptable limit (100 CFU/g). Staphylococcus enterotoxin seg and sei genes were detected in RTE fresh-cut celery and red cabbage from meal-kits, respectively. S. aureus contamination must be carefully controlled during the manufacturing processes of RTE fresh-cut products. Neither Cyclospora cayetanensis nor Cryptosporidium parvum was detected in the samples of RTE fresh-cut products and vegetables from meal-kits from the Korean retail markets.
