        This study was done in order to monitor the quality properties of the granule using Bokbunja (Rubus coreanus Miquel) extracts. In order to prepare the granule depending on operational parameters such as content of Bokbunja extract (X1, 0.4∼1.2 g), sugar content (X2, 6∼10 g) and citric acid content (X3, 0.1∼0.3 g), a response surface methodology was applied to monitor the optimum recipes on the organoleptic properties and Hunter's color. The optimum recipe on the organoleptic color showed extract content of 0.96 g, sugar content of 7.05 g and citric acid content of 0.232 g. The optimum recipe on the organoleptic flavor showed extract content of 0.86 g, sugar content of 6.04 g and citric acid content of 0.215 g. The optimum recipe on the organoleptic taste showed extract content of 0.92 g, sugar content of 6.39 g and citric acid content of 0.251 g. The optimum recipe on the overall palatability showed extract content of 0.86 g, sugar content of 6.65 g and citric acid content of 0.272 g. The response surface of the Hunter's color b value was similar to the response of the overall palatability; therefore, the optimum conditions accepted by the consumers were 0.8 g Bokbunja extract content and 0.6 g sugar content in the Hunter's color a value of 6.0.

        This study was carried out in order to investigate sterilization effect and to extend storage periods of the Rubus coreanus by treating with tap water (TW), electrolyzed water (EW) and aqueous chlorine dioxide (ClO2). After each treatment plot was soaked with 10, 50, 100, 200 ppm in each sterilizing solution within 30 sec, each treatment was compared during the storage time at room temperature and refrigerator temperature. As results of total plate count according to temperatures and periods, the microbial sterilizing power of each treatment plot was bigger at EW and ClO2 treatment plots than the TW treatment plot; however, it sharply increased on the high concentration ClO2 treatment plot. Futhermore, the cold storage treatment plot had more outstanding microbial sterilizing power than the room temperature treatment plot. As a result of observing the surface of the Rubus coreanus using scanning electron microscope (SEM), no microbe was seen in EW and ClO2 treatment plot. The results of measuring enzyme activity showed a more significant decrease in EW and ClO2 solutions treatment plot than TW treatment plot but gradually increased with time. The contents of total polyphenol revealed similar values on each treatment. The EW and ClO2 treatment of the Rubus coreanus could be considered as good methods for inhibiting microbial growth in fresh vegetables and fruit, thereby contributing to quality maintenance.

        The objective of this study is to investigate the optimal condition for macerating enzymatic extraction process that leads to the highest yield and the largest extracted amount of bio-active contents from Rubus coreanus Miq. fruit. The optimal extraction conditions were found as the following: The initial amount of the water added to the fruit was 20 ~ 30% by weight. The mixing ratio used for the macerating enzyme was 4 : 1 : 2 (w : w : w) for cellulase:pectinase:amylogucosidase, and the amount of the macerating enzyme added was 2% by weight. The extraction process was done at a temperature of 45~50℃ for 10 hours. The extraction yields on Rubus coreanus Miq. fruit by macerating enzymatic extraction process was increased by 84.3% compared to that of hot-water extraction process. The amounts of organic acids and vitamin found in the extract were also higher. The amount of polyphenol and anthocyanin contents in the extract were 185% and 257% of those from hot-water extraction, respectively. These results suggest that macerating enzymatic extraction is an effective method to boost extraction yield and to increase the amount of extraction of bio-active contents from Rubus coreanus Miq. fruit.

        This study developed a high-utility type of vinegar from Rubus coreanus by optimizing its fermentation conditions. In the alcohol fermentation process, the optimal conditions for the maximization of the alcohol contents were an initial sugar concentration of 15 °Brix, a temperature of 30℃ and 4 days. The optimal conditions for the acetic acid fermentation were 9 days of fermentation at 30℃ and 200 rpm, with 6% alcohol and 2% initial acidity. The sucrose, fructose, and glucose contents were 952.90, 491.01, and 386.62 mg%, respectively. The free organic acids were acetic, malic, succinic, malonic, oxalic, and lactic acids. The total free amino acid content was 104.33 μg/mL, with alanine, glutamic acid, γ-amino-N-butyric acid, and o-phospho-ethanolamine as the major amino acids. The K, Na, and Mg contents were 1,686.10, 172.50, and 69.33 ppm, respectively. The total phenolic and anthocyanin contents were 25.19 and 80.71 mg/100 mL, respectively. The DPPH- and ABTS.+ radical scavenging activities were approximately 65 and 94%, respectively. Moreover, the vinegar’s β-carotene bleaching activity and reducing power showed that it had strong anti-oxidant properties. These results show that Rubus coreanus vinegar has anti-oxidant properties and may be used as functional food.

        This study was performed to determined the antioxidant activities, anticancer and immuno-activities of anthocyanin fraction from Rubus coreanus Miquel fruits (Bokbunja). Anthocyanin fraction extracted from Bokbunja revealed the presence of three anthocyanin components, which were tentatively identified as cyanidin 3-O-sambubioside, cyanidin 3-O-xylosylrutinoside and cyanidin 3-O-rutinoside using RP-HPLC/DAD/MS. The anthocyanin fraction from Bokbunja always showed reducing power and high scavenging activities against DPPH, hydroxy radical (OH) and superoxide anion radical (O2-) similar to general synthetic antioxidant and polyphenol compounds from plant origin. Anthocyanin fraction from Bokbunja showed high inhibition on proliferation of LNCaP and A549 cells and did not inhibit the proliferation of other cancer cells. Immuno-activities of Anthocyanin fraction from Bokbunja were investigated, it showed high promotion of human B and T cells growth about 50% and secretion of IL-6 and TNF-α by treatment after 6 days. Over all, the result of the study suggest that anthocyanin fraction from Bokbunja displays antioxidant activity comparable to that general synthetic antioxidant, also, anthocyanin fraction from Bokbunja are expected to be good candidate for development into source of anticaner and immuno-activator agent in food industry.

        This study was carried out to evaluate biological activities to different ethanol extracts from unripened and ripened fruit (Rubus coreanus Miq.). 25% to 75% ethanol extracts of unripened and ripened fruits were similar to extract yield respectively. Yield of ethanol extract of ripened fruit were approximately 3 times higher than that of unripened fruit. 75% ethanol extract of unripened fruit showed the highest contents of total polyphenol (180.04±0.41 mg/g) and total flavonoid (50.43±0.81 mg/g). Contents of total polyphenol and total flavonoid of unripened fruit were about 2 times higher than those of ripened fruit. IC50 values of DPPH radical scavenging activity and linoleic acid peroxidation inhibition activity of BHA and α-tocopherol showed 13.19±0.21 and 18.16±0.23μg/ml, 4.25±0.04μg/ml and 5.56±0.10μg/ml respectively, but 75% ethanol extract of unripened fruit showed the lowest 23.85±0.10μg/ml and 7.34±0.07μg/ml among other all extracts. IC50 values of LDL (low density lipoprotein) oxidation inhibition activity and α-glucosidase inhibitory activity of 75% ethanol extract of unripened fruit showed the lowest 1.04±0.04μg/ml and 7.21±0.13μg/ml among other all extracts respectively. Specifically, 75% ethanol extract of unripened fruit has relatively better biological activities than other ethanol extracts, it could be potentially used as bioactive source for health functional foods.

        본 연구는 한약탕제 찌꺼기를 친환경 퇴비자원으로서 재활용하기 위하여 발효과정을 거친 퇴비(MHWC; medicinal herb waste compost)와 계분퇴비(PMC; poultry manure compost) 및 이들을 혼합한 퇴비(MHWC+PMC, 1:1)를 식재 2년생 복분자 포장에 무처리(UC; untreated control, 0), 20, 40 Mg/ha로 각각 처리하였다. 밭 토양의 화학성과 수확한 복분자 과실의 수량 및 당도변화 조사 결과는 다음과 같다. 토양 pH는 UC보다 모두 증가하여 유의적인 pH 변화 조절기능이 인정되었다. EC는 MHWC 처리구가 UC보다 0.56-0.46 배 감소를 보였으며, PMC 처리구와 차이가 있었다. 유기물 함량과 질소함량은 퇴비 종류와 무관하게 증가하였고, 가장 증가된 퇴비는 MHWC이었다. 유효인산함량은 퇴비 종류 및 처리량에 따른 차이가 있었다. 각 퇴비를 처리한 토양의 중금속 함량은 토양오염우려기준보다 낮았다. MHWC 처리구의 수량과 당도는 다른 처리구에 비해 높은 경향을 나타내었다. 이상의 결과로 보아, MHWC 처리는 복분자 경작지의 토양환경개선과 더불어 복분자 과실의 수량 및 당도를 향상시키는 효과적인 퇴비자원인 것으로 사료된다.

        본 연구는 한약탕제 찌꺼기를 친환경 퇴비자원으로서 재활용하기 위하여 발효과정을 거친 퇴비(MHWC: medicinal herb waste compost)와 계분퇴비(PMC: poultry manure compost) 및 이를 혼합한 퇴비(MHWC+PMC, 1:1)를 식재 2년생 복분자 포장에 무처리(0), 20, 40 Mg/ha로 처리하여 꽃이 첫 개화한 시기를 기점으로 15, 20, 25 DAF(day after flowering)를 수확시기로 하여 수확한 복분자 과실의 생리활성을 조사하여 비교한 결과는 다음과 같다. 복분자의 총 페놀성 화합물 함량은 15 DAF 과실에서와는 달리 20, 25 DAF 과실에서 유의성 있는 증가를 보였다(p<0.05). 수확시기별로는 25>20>15 DAF 순이었고, 그 함량은 40 MHWC Mg/ha가 가장 높았다. Flavonoid 함량변화는 총 페놀성 화합물 함량변화와 유사하였다. 전자공여효과는 15, 20 DAF 과실에서는 차이가 없었지만, 25 DAF 과실에서는 낮은 효과를 나타내었다. 아질산염 소거효과는 pH 1.2>4.2>6.0 순으로 pH 의존적인 경향이었으며 15, 20 DAF 과실보다 25 DAF 과실이 저하되었다. Tyrosinase 저해효과는 MHWC 처리구가 가장 우수하고 처리량에 따라 저해효과가 증가되었으며 15, 20 DAF 과실은 25 DAF 과실보다 낮은 효과를 보였다. Xanthine oxidase 저해활성은 25 DAF 과실의 MHWC 처리구에서 가장 높았고, PMC 및 MHWC+PMC처리구와 UC와의 차이는 인정하기 어려웠다. 이상의 결과에서 MHWC 처리에 따른 복분자 과실의 생리활성이 증가경향을 보여 친환경퇴비자원으로서 가치가 있는 것으로 사료되었다.

        Sunlight, and in particular its UV component, is the major environmental trigger that underlies the major signs of human skin and skin cancer in general. Therefore, this study was carried out to investigate the UV protection effects of R. coreanus. R. coreanus was extracted by ultra high pressure extraction process at 500 MPa and 30℃ for 5 and 15 minutes. The cytotoxicity of the extracts extracted by ultra high pressure process on human dermal fibroblast cell CCD-986sk, human kidney normal cell HEK293, and human lung normal cell HEL299 was measured as 17.5%, 16.5% and 14.0%, respectively in adding 1.0 mg/ml of the samples, which was much lower than that from conventional water extraction method at 100℃ as 23.2%, 22.5%, 21.2%. The secretion of NO- from macrophage showed 15.9 μM on the R. coreanus extract from this process, which was higher than others. Prostaglandin E2 (PGE2) production from UV-induced human skin cells was also greatly decreased down to 510 pg/ml, compared to the control. From the results, we considered that the extracts from R. coreanus could be potent natural materials for skin anti-inflammation agent, and could be used as a potential anti-aging for the photo-damaged skin.

        This study was performed to improve antioxidant activities and skin-whitening effects of Rubus coreanus Miquel extracts by nano-encapsulation. R. coreanus was extracted at 60℃ and encapsulated by lecithin and gelagine. Nano-encapsulated extracts showed highest free radical scavengering effect as 97.62% in adding sample (500 μg/ml), compared to the extracts from conventional processes. It was showed strong inhibition effect on melanin production test by Clone M-3 cells as 55.23%. High inhibitory potency on tyrosinase was also measured as 155.8% by adding nano-sample of 1 mg/ml. The improvement of biological activity was demonstrated by real time confocal microscope. We could consider that the water soluble extracts of R. coreanus could be definitely enhanced by nano-encapsulated as a potent natural resources for antioxidant and skin-whitening agent.

        The purposes of this study were to search for the ways to activate the Gochang Blackberry Festival by examining the visitors to the festival and utilizing the results to make efficient use of the local resources and vitalize the area. The results indicate that the visitors were aware of Gochang blackberry's superiority to that of other regions in quality and commercial value but lacked in the recognition of its related products, which suggests that the Gochang government should make diverse efforts to heighten their recognition of the products. There was a great demand for improving and supplementing the local festivals ad experience programs related to Gochang blackberry. It's thus required to reinforce new experience programs and develop various programs by incorporating blackberry into other local resources. The study has its limitations since its subjects were restricted to the visitors of the Gochang Blackberry Festival and it failed to investigate the preference factors of general potential tourists with the small sample. In future study, they need to use a bigger sample of general public to generalize the research findings.

        This study was performed to improve immune activities of Rubus coreanus Miquel by encapsulation of nanoparticles. Immuno-activities of R. coreanus were investigated through aqueous extracts associated with process of water at 60℃. It showed high promotion of human B and T cells growth about 50%, compared to the case of other conditions. The secretion of IL-6 and TNF-α was also enhanced as 2.44×10-4pg/cell and 1.94×10-4pg/cell, results by adding nano samples. NK cell activation was improved up to 29% higher than the conventional extraction process. The secretion of NO from macrophage showed 14.9 μm on the nano-encapsulation process extracts, which was higher than others. The size of nanoparticles was in the range of 50~300 nm, which can effect the penetration into the cells. It was clearly observed by real time confocal microscope.

        본고에서는 초고압 추출 공정의 활성 증진 효과를 알아보기 위해 복분자의 면역활성 실험을 실시하였는데 그 결과는 아래와 같다. 인간의 정상 신장세포인 HEK293을 이용한 세포 독성은 추출물은 1.0 mg/ml의 농도에서 최고 19.5%의 세포독성을 나타냈다. 항암활성은 AGS와 A549 암세포를 이용하였는데, 초고압을 실시할 경우 1.0 mg/ml의 농도에서 80%이상의 높은 억제활성을 보였으며, 암세포의 생육활성에 대한 정상 세포의 세포독성의 비로 나타낸 선택적 사멸도는 모든 고농도에서 모두 3 이상으로 나타났다. 모든 처리구 중 15분간 초고압 추출한 것이 가장 높은 활성을 보였으나 5분간 초고압 추출한 것과 큰 차이가 나지 않아 5분간 초고압 추출을 실시하는 것이 효율적인 것으로 보인다. B cell과 T cell을 이용한 면역세포의 생육은 배양 6일째에 15분간 초고압으로 추출한 처리구에서 각각 13.5×104cells/ml, 14.6×104cells/ml로 가장 높은 세포농도를 나타냈다. 면역세포를 이용한 cytokin의 분비량 측정실험에서는 HPE15 추출물이 배양 시간에 따른 cytokine의 분비가 가장 높게 나타나는 것을 확인할 수 있었다. 각 세포의 TNF-α와 IL-6의 분비량은 6일째 최고의 분비량을 나타내었다. 또한 NK cell의 면역활성은 모든 추출물에서 배양 시간에 따라 유의적으로 증가하는 것을 확인하였다. 그 중 HPE15 추출물에서 B cell의 경우 15.8×104cells/ml, T cell의 경우 14.8×104cells/ml으로 가장 높은 생육 활성을 나타내었다. 본 실험 결과를 통하여 초고압으로 인해 기존의 추출 방법으로는 추출되지 않았던 유용생리활성 물질들이 초고압으로 조직과 세포막 파괴로 인해 유용성분의 용출량이 증가하고 새로운 물질의 용출로 인해 높은 활성을 나타낸 것으로 판단되었다.
