본 연구에서는 메주에서 분리된 C. allociferrii JNO301 을 이용한 최적 홍삼 ginsenosides의 생물전환 조건을 확인한 결과, 1.0% (v/v)의 홍삼 추출물에 탄소원으로서 2.5%(w/ v)의 galactose, 질소원으로서 1.0% (w/v)의 soytone을 첨가 하여 30oC에서 145 rpm으로 8일간 배양을 최적 생물전환 조건으로 확립하였다. C. allociferrii JNO301에 의해 홍삼 추출물에 존재하는 Rb1, Rb2, Rc, Rf가 생물전환되어 F2, Rh1, Rh2, compound O, compound Mc1, compound K 등 의 minor ginsenosides의 함량이 증가되었다.
O-D-mannopyranosyl-(1→4)-O-D-glucopyranosyl-(1→4)-D-Mannopyranose (MGM) was prepared via the enzymatic hydrolysis of konjac glucomannan and the subsequent elimination of monosaccharides from the resultant hydrolysate using yeast. The enzyme system hydrolyzed konjac glucomannan and produced monosaccharides and MGM without other oligomers at the 48 h reaction. Konjac 20 g was hydrolyzed at 60 o C and a pH 6.0 for 48 h with 200 mL crude enzyme solution from Xylogone sphaerospora. By eliminating monosaccharides from the hydrolysis products with yeast (Candida guilliermondii), 3.8 g of crystalline MGM was obtained, without the use of chromatographic techniques. After 48 h of yeast cultivation, the total sugar content fell from 5.2% to 3.7%, while the average degree of polymerization (D.P.) rose from 2.6 to 3.3.
The purpose of this study is to analyze the distribution of Candida species in patients with oral disease and clarify the distinction of Candida culture test according to its isolation technique. 75 samples was isolated from 42 patients who visited Chonnam National University Dental Hospital due to oral disease from December 2015 to August 2016. For isolating the candida sampling, saliva sampling and oral swabbing were used. Acquired sampling was cultured in CHROMagar Candida Culture Medium, which indicates the candida species as color. Of the 42 patients, C. albicans was the most frequently isolated species in 39 patients. For 17 patients out of 21 who underwent saliva sampling and oral swabbing simultaneously, oral swabbing was quantitatively underestimated comparing to saliva sampling. 12 samples in 21 samples having particular Candida species were not isolated by oral swabbing. Considering the possibility of fungal infection in various oral disease, it is recommended to perform not only oral swabbing but saliva sampling when isolating Candida.
장님마디톡토기는 토양생태계에서 오염물질의 독성영향을 평가하기 위한 국제 공인 표준시험종으로 제안되고 있다. 토양생태계 먹이사슬에서 중요한 역할을 수행하는 톡토기의 특성에 따라 다양한 연구가 시도되고 있으며, 더 민감한 독성영향 종말점이 제시되고 있는 실정이다. 본 연구에서는 대표적인 토양오염물질인 중금속의 생태독성영 향을 평가하기 위하여 장님마다톡토기의 도약능을 이용하였다. 장님마디톡토기는 표준시험법에 따라 중금속 오염토 양에 만성노출되었으며, 이후 생존개체를 수집하여 도약능 평가를 수행하였다. 톡토기의 도약은 일시적인 화학스트레 스를 이용하여 유도되었으며, 이를 회피하기 위한 운동능을 컴퓨터 프로그램을 통해 기록하여 평가하였다. 중금속 오염토양에 노출된 개체는 도약을 통한 회피 경로가 단순화되었으며, 도약 횟수와 길이가 중금속 농도에 따라 통계적으로 유의하게 감소하였다. 이는 중금속 오염토양에서 톡토기의 회피능력이 감소할 수 있음을 의미하며, 생태독성을 평가하기 위한 종말점이 될 수 있을 것이라 판단된다.
Candida albicans and their associated Candida species are opportunistic pathogens which exists as normal flora in the oral cavities of healthy individuals. In response to physiological changes in the host, these yeasts can become pathogenic, resulting in oral candidiasis. The rapid detection and identification of Candida species in clinical laboratories are extremely important for the management of patients with hematogenous candidiasis. The presently available culture and biochemical methods for detection and species identification of Candida are time-consuming and lack the required sensitivity and specificity. In this study, we have established a seminested PCR (snPCR) using universal and species-specific primers for detection of Candida species in saliva. The universal outer primers amplified the 3end of 5.8S ribosomal DNA (rDNA) and the 5end of 28S rDNA, including the internally transcribed spacer 2 (ITS2), generating 350- to 410-bp fragments from the four commonly encountered Candida spp., viz., C. albicans, C. tropicalis, C. glabrata, and C. parapsilosis. The saliva from 331 healthy and, over 50 years of aged people lived in Dong-gu, Gwangu city, was collected. Total DNA were extracted by Hoffman-Winston yeast total DNA prep. method and performed t he s nP CR. R esults appeared to b e negative on 292 people ( 88.2%), however, 2 6 people ( 7.9%) were p ositive Candida albicans, 6 people (1.8%) were positive Candida glabrata, 5 people (1.5%) were positive Candida tropicalis, and only 2 person (0.6%) were positive Candida parapsilosis. These result showed that detection and identification of Candida species could be established by saliva analysis, so that snPCR on saliva is useful method of diagnosis of clinical fields
The objective of this study was to evaluate the antifungal properties of xanthorrhizol (extract from curcuma xanthorrhiza) against Candida albicans. Some of the commercial products were evaluated for their ability to inhibit growth of C. albicans. OTC product containing povidone iodide and cosmetic grade product with surfactants were tested. Antifungal activity was shown in 1.56% of OTC product, however, there was no antifungal effect in cosmetic product. For the comparison, we tested several materials. Povidone iodide, lemon tea tree oil and xanthorrhizol showed antifungal activities against C albicans in 0.25, 0.062 and 0.007%, respectively. We also tested the antifungal effects of povidone iodide and xanthorrhizol in surfactant base. Test results revealed that 2.5% of povidone iodide and 0.156% of xanthorrhizol showed similar antifungal effects. These findings support the application of xanthorrhizol for vaginal cleanser or personal cares using antifungal effect.
용존 유기물의 농도를 BOD로 신속히 측정하여 폐수 처리 공정에 feed back할 수 있는 Candida BOD sensor를 이용하여 BOD를 측정하였다. Glucose, acetic acid, aspartic acid 및 glutamic acid에 대한 sensitivity는 lactose, sucrose및 lactic acid의 sensitivity보다 더 높게 나타나서 오염물질의 종류에 따라 BOD sensor의 출력 신호가 다르게 제시되었다.
Enzymatic biodiesel using lipase Novozyme-435 instead of using chemical catalyst was studied in this paper for cost reduction, increase of production efficiency and environment friendly product in this paper. In this paper the standard of biodiesel quality was met by using enzyme. The optimal reaction condition of methanol/waste oil molar ratio was 3, which 1 mol of methanol was injected at initial reaction. Then, each of 1 mol of methanol was injected at 4 hours of interval. Other reaction condition was like as follows : Reaction temperature, reaction time, agitation speed and amount of lipase were 55oC, 24 hours, 400 rpm, and 6wt%, respectively.
This study carried out development of a natural antimicrobial agent from Schima wallichii ssp. liukiuensis. Compound I exhibiting potent antimicrobial activity against Candida spp. was isolated from the methanol extracts of Schima wallichii ssp. liukiuensis. The structure of I identified as a sterol glycoside consisted of a trisaccharide and α1-sitosterol. Trisaccharide composed of L-rhamnose, D-galactose and D-glucose residues. The antimicrobial activity of I was selective on yeast rather than bacteria or other fungi. Compound I was demonstrated to be ineffective against toxicity to mouse liver cells where as protective to human dermal fibroblast cells at low concentrations. Thus, it is reasonable to expect a sterol glycoside (I) as a valuable alternative for synthetic antifungal.
