        본 연구는 부화 온도가 맹꽁이(Kaloula borealis) 알의 부화 기간에 영향을 미치는 여부를 알아보기 위해 수행되었다. 본 연구에서 맹꽁이 알에서 올챙이가 생긴 것을 기준으로 알 부화 과정을 기록했다. 연구 결과 맹꽁이의 모든 알은 산란 후 48시간 이내에 부화하였으며, 28.1%(±10.8, n=52)가 24시간 이내에, 99.9%(±0.23, n=49)가 산란 후 48시간 이내에 부화했다. 수온의 차이에 따라 맹꽁이 알의 평균 부화율은 유의미한 차이를 나타냈다. 산란 후 15~24시간 사이의 평균 부화율은 24.1(±0.2)°C보다 21.1(±0.2)°C의 수온에서 더 높았다. 본 연구 결과는 비교적 낮은 수온에서 빠른 부화가 되는데 이는 비가 오는 계절에 일시적인 연못이나 웅덩이에 알을 낳는 번식 습성으로 웅덩이가 마르기 전에 빠른 부화가 필요하기 때문으로 추측한다. 본 연구 결과는 멸종위기종인 맹꽁이 알의 최적 부화온도를 이해하는데 도움이 된다.

        느타리 ‘흑타리’ 품종의 배양 중 고온스트레스에 의해 발생되는 미발이 현상을 구명하기 위하여 배양온도에 따른 생육차이를 조사하였다. PDA 배지에서‘흑타리’의 적정생육온도는 23~26 ̊C였고, 균사생장속도는 ‘춘추2호’에 비하여 빠른편이었다. 병내 배지온도는 초기에 상승하여 배양 중반에 최고점에 도달한 후 온도가 하강하였다. 배양 온도가 높을수록 배양기간은 짧아졌다. 배양온도 20 ̊C 처리구에서 배양기간은 25일 정도 소요되었으며, 미발이율은 1.8%, 수량은 139.4 g/병을 나타내었다. 배양온도 24 ̊C 처리구에서 배양기간은 20일 정도 소요되었으며, 미발이율은 4.2%, 병당수량은 132.1 g/병을 나타내었다. 배양온도 16 ̊C와 28 ̊C처리구에서는 미발이율이 증가되었고 수량이 감소하였다. 이 결과를 바탕으로 농가에서 배양온도와 미발이율의 관계를 조사하였다. 배양실 온도를 18 ̊C로 설정하고 배지품온을 28 ̊C 미만으로 관리하는 농가는 미발이율이 0.3~0.8%를 나타내었다. 배양실내의 온도가 20 ̊C 이상이며 환기가 잘 이루어지지 않은 농가에서는 미발이율이 3.5% 정도 발생되었다. 배양실 온도가 19 ̊C이며 배지 최고 품온이 31.3 ̊C까지 상승하는 농가는 미발이율이 8.2%로 높게 나타났다. 배양중 병내부 온도가 28 ̊C이상 상승하고 배양실내의 환기가 잘 이루어지지 않을 경우 미발이율이 증가하고 수량이 감소되는 경향을 보였다. 결과적으로, 배양실은 배지품온이 28 ̊C 이상 상승하지 않도록 배양실 내부의 공기를 지속적으로 순환시키고, 배양공간에 맞는 최적의 배양량을 넣어 관리하여야 한다.

        The study was conducted to investigate the effects of post-incubation period and temperature treatment conditionsduring incubation on the uniform primordia formation and cultural characteristics of oyster mushroom (Pleurotus ostreatus). Threekinds post-incubation period; 25, 30, 35 days and control were applied for 30 days while two kinds incubation roomtemperature 23oC and 26oC and control were used 20oC. The substrate temperature during pre-incubation was of ‘Suhan No. 1’and ‘Gonji No. 7’. Oyster mushroom varieties tended to increase between 24oC to 26oC at 11 to 15 days after inoculation andthen they were maintained in treatment temperature during post -incubation period. The CO2 occurrence was at the highest at6,500ppm for ‘Suhan No. 1’ and 5,800ppm for ‘Gonji No. 7’ at the time of the highest temperature increase. The ratio of un-uniformal primordia formation and the ratio of non-commercial fruit body were reduced by 40%, 10.5%, respectively comparedto control for ‘Suhan No. 1’ when in the post-incubation temperature was 26oC, and incubated for 10days and 15daystreatment. Also, ‘Gonji No. 7’ was reduced by 19%, 9.5%, respectively when in the post-incubation temperature was 26oC, andincubated for 10 days treatment. Therefore, the higher post-incubation temperature of room and longer post-incubation periodresulted in the higher percentage of primordia formation of two cultivars.

        산업적 가치가 있는 천연 레스베라트롤은 인체에 항산화 작용, 심혈관질환 예방, 항염증 등의 효능이 있다. 레스베라트롤 생합성 벼(익산 526)를 이용하여 레스베라트롤 함량을 높이기 위해 현미의 발아온도(25°C, 30°C)와 처리일수(0, 1, 3 및 5일)의 최적조건을 구명하고자 실험한 결과는 다음과 같다. 레스베라트롤 함량은 25°C와 30°C에서 발아 5일째 각각 1.7배와 2.1배로 가장 높게 나타났으며, piceid 함량은 3.5배와 1.4배 증가하였다. 25°C 발아조건과 비교하여 30°C에서 transresveratrol은 20% 이상, piceid는 70% 이상 증가하였다. 레스베라트롤 유전자 발현량도 25°C와 30°C에서 발아 5일째 가장 높게 나타났으며, 30°C에서는 25°C와 비교하여 5.5배 증가하였다. 발아 전과 비교하여 25°C에서는 2.5배, 30°C에서 14배 증가하였다. 하지만, 익산 526의 레스베라트롤 함량 증가는 항산화 활성에 큰 영향을 미치지 않았다. 따라서, 익산 526 현미를 이용하여 레스베라트롤 함량을 증가시키기 위해서는 30°C에서 5일 동안 배양하는 것이 가장 효과적으로 향후 이를 산업적 소재로 이용 할 수 있는 기초자료로 제공한다.

        Cryopreservation of epididymal spermatozoa offers a potential tool for rescuing genetic material from males of genetically elite populations. Castration, catastrophic injury, sudden death or any other event that makes semen collection or mating impossible may prematurely terminate a stallion reproduction. Stallion epididymal spermatozoa vary widely in the loss of progressive motility, acrosomal integrity, and viability during freezing and thawing. The objective of this work was to investigate the effect of (1) freezing package types on cryopreservation efficiency, (2) thawing temperatures (37, 56 or 70℃) on Computer Assisted Sperm Analysis (CASA) parameters and (3) post-thawing incubation time (0, 1, 2 or 4h) on castrated stallion epididymis. Post-thawed sperm motility ranged between 59.69% and 64.28% (56℃ and 37℃) in various thawing temperatures. When stallion epididymis sperm was frozen, straw was better than freezing tube on VCL (Velocity of Curvilinear Line) and VAP (Velocity of Average Path) parameter. Higher percentage of motility was observed at 37℃ thawing temperature even though no significant difference was observed among various temperatures. The motility, VCL, ALH (Amplitude of Lateral Head displacement), VAP, BCF (Beat-Cross Frequency) and STR (Straightness index) parameter of post-thawed sperm were significantly decreased by increasing the incubation time for all thawing temperatures. The present study showed that type of freezing package (Straw vs. Freezing tube) was not significantly different on cryopreservation efficiency. Furthermore, stallion epididymal spermatozoa frozen-thawed at 37℃ for 1 min resulted the highest proportion of motility and velocity movement. In addition, motility and viability of frozen-thawed stallion epididymal spermatozoa were also decreased by incubation.

        Temperature and humidity are the vital environmental factors influencing the distribution and abundance of ectothermal organisms. Also, these variables often determine the efficacy of biological control of spider mites using phytoseiid mites N. californicus, N. womersleyi and P. persimilis are promising biocontrol agents of spider mites. The combined effects of temperatures (15,20,25,30 & 35oC) and relative humidity (33,51,75 & 95%) on egg hatch, incubation period from single egg and cannibalism from five eggs experiment were tested for three phytoseiid mites. Photoperiod was set to 16L:8D and neither water nor diet was provided. The result showed that higher temperatures (30&35oC) and lower relative humidity (33&51%) caused increased egg mortality by shrinking and desiccation. The egg hatch of N. californicus and P. persimilis were higher than N. womersleyi in all conditions with the exception of 35oC and 95% RH treatment where P. persimilis did not hatch at all. The incubation periods of egg decreased as the humidity level increased even under the same temperature. The cannibalism, habit of feeding the same species was found from protonymphal and deutonymphal stages under higher humidity levels (75&95%) in the three phytoseiid species. This tendency was higher in N. californicus and N. womersleyi than in P. persimilis. The results partially explain the lower predator performance during hot summer by P. persimilis and the possibility to sustain the predator population when the prey population is low.

        On June 14, 2008 (the first experiment) and July 24, 2008 (the second experiment), the shores of the Boseong River and the sandy beaches, Seokgok-myun, Moksadong-myun, Gokseong-gun in Jeollanam Province were investigated and a total of 29 soft-shelled turtle (Tryonyx sinensis) eggs in the natural spawning nest eggs were collected (13 eggs were collected in the first experiment and 16 eggs in the second experiment). The temperatures in the natural spawning nests were 25.9- 36.9±0.5℃, the depth of the eggs was 5.2-7.5±0.5 cm as the distance of the average 6.4±0.5 cm. 29 eggs were scattered at least 0.2 cm interval. Artificial incubation of 29 eggs was conducted in artificial nest boxes in thermo-plastic composition of the incubator, and then incubated at 26.5-35.5±0.5℃, and an average constant temperature was 31.2-32.1±1.0℃. The incubation days ranged from 53 to 55. In case of most turtles, incubation at 31℃ (higher temperatures) generally produces all or mostly females, while incubation at 25℃(cooler temperatures) produces all or mostly males. Exceptionally, in case of genus Trionyx, the sex ratio of female : male of T. sinensis of a freshwater soft-shelled turtle was approximately 1:1, which differs from other genera of turtles and makes T. sinensis Strauch only turtles presently known to lack temperature-dependent sex determination.

        Omija-cheong, concentrated extracts from sugar-treated Omija fruit (Schisandra chinensis Baillon), is produced by traditional manner in Korea. The quality characteristics of Omija-cheong processed at low temperature with a pilot-scale were investigated to optimize the incubation time. With increasing incubation time in processing Omija-cheong, the pH level of Omija-cheong remained constant, while titratable acidity and organic acids increased. Fresh Omija fruits contained citric, malic and succinic acids, most of which were extracted into concentrated extracts after 37 days of incubation and reached to the stable concentration after 47 days of incubation. Titratable acidity in Omija-cheong gradually increased from 1.18% to 2.71%, and also was correlated with total concentration of organic acids. About 80% of supplemented sucrose for manufacturing Omija-cheong was converted into glucose and fructose until 68 days of incubation, and the composition of free sugars was maintained to be stable up to 138 days of incubation. The contents of total flavonoids and phenolic compounds in Omija-cheong were 24.1 mg-GAE/L and 1,635 mg-QE/L at 57 days of incubation, which were more than 9 and 5 times higher than those in Omija fruits, respectively. From the quality characteristics in processing Omija-cheong by low-temperature incubation, more than 60 days of incubation is required for the constant quality and value-added beverage.
