        Ganoderma lucidum has been traditionally used as a medicine for treatment of bronchitis, arthritis, and high blood pressure, and it has been reported to display many biological activities including anticancer and immune activities. Since mushroom mycelium is known to have excellent biological activities together with mushroom fruiting body, studies on biological activities of mushroom mycelium have been actively conducted. Thus, the present study compared the biological activities before and after the cultivation of Ganoderma lucidum mycelium on Atractylodes rhizoma. When the radical scavenging activity was assessed by the DPPH assay, ARGL (ethanol extract of Atractylodes rhizoma mycelium fermented with Ganoderma lucidum) showed radical scavenging activity of 5.58~82.56% at concentrations of 10~500 μg/assay, while AR (ethanol extract of Atractylodes rhizoma) showed radical scavenging activity of 5.27~72.08% at the same concentrations. When measured by using the ABTS assay, ARGL showed higher radical scavenging activity than AR, which was consistent with the result obtained by the DPPH assay. In the MTT assay, the cytotoxicity of ARGL against all cell lines was higher than that of AR. In particular, the cytotoxicities of AR and ARGL against Hep3B at a concentration of 400 μg/assay were 71.81% and 86.40%, respectively. In addition, the result obtained by the SRB assay was consistent with the result obtained by the MTT assay. According to the results mentioned above, there is a high probability that medicinal herb cultures using mycelium can be used as sources of functional foods since the cytotoxicities against cancer cells and antioxidant activities increased when the mycelium was fermented with Atractylodes rhizoma.

        Microglia have multiple functions in regulating homeostasis of the central nervous system. Microglia cells have been implicated as active contributors to neuron damage in neurodegenerative disorders. In this study, medicinal plant extracts (MPEs) were used to evaluate the cell-death induction effect in microglia BV-2 cells. Among 35 MPEs tested in this study, 4 MPEs showed less than a 30% cell survival after 24 hours of incubation. These were Foeniculi Fructus, Forsythiae Fructus, Zingiberis Rhizoma and Hedera Rhombea. The concentration showed that 50% cell death (IC50) occurred with 33, 83, 67 Ed highlight: Please confirm wording, and 81 μ /ml, respectively. For further study, we chose Zingiberis Rhizoma (ZR) which showed a reasonably low IC50 value and an induction of cell death in a relatively narrow range. Western blot analysis showed that ZR-treated cells showed activation of caspase-3 and cleavage of PARP Ed highlight: When an acronym is first presented it needs to be spelled out in both dose- and time-dependent manners. However, the level of Bcl-2 and Bax were not changed by ZR-treatment in BV-2 cells. These results suggest that ZR-induced apoptosis in BV-2 cells occured through caspase-3 activation. The results also suggested that ZR may be useful in developing treatments for neurodegenerative diseases.

        To investigate the effects of coated liposome from Discorea rhizoma extract (DRE) in streptozotocin (STZ)-induced, we evaluated changes in body weight, fasting blood glucose, blood insulin and blood lipid concentrations in mice. Mice were divided into four groups: (DC), diabetic DRE at 10 ㎎/㎏ (DRE-1), diabetic DRE at 50 ㎎/㎏ (DRE-2), and diabetic DRE at 250 ㎎/㎏ (DRE-3). Mice had free access to water and diet (10 weeks). The DC group showed higher blood cholesterol than the DRE-1, DRE-2, DRE-3 groups. In glucose tolerance test, the DRE-1, DRE-2, and DRE-3 groups increased after 30 minutes in decremental glycemic response area under the curve. Fasting blood glucose levels in the DRE groups significantly decreased through 4 weeks. Plasma total cholesterol, triglyceride and LDL-cholesterol concentrations were also lower in the DRE groups. On the other hand, the DRE-1, DRE-2 and DRE-3 groups showed higher HDL-cholesterol and insulin levels than the DC group. Moreover, blood glucose and lipid levels significantly decreased in streptozotocin (STZ)-induced diabetic mice treated with DRE. These results indicate that DRE may reduce elevated blood glucose levels and serum lipid concentrations in hypoglycemic and diabetic mice, suggesting its usefulness as a functional food.

        This research was carried out to find herbal preservatives for Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. When Makgeolli was kept at 25℃ to find the changes in acidity, total microbial cell number, yeast cell number, and bacterial species variety, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, and Makgeolli was changed to complete off-flavor. Acetobacter pasteurianus is the main acidifier in Makgeolli and shows a synergy effect in acid formation when cultured in combination with Lactobacillus casei. Among 12 herbs, the ethanol extract of Sutellaria baicalensis showed antimicrobial activity against A. pasteurianus, whereas the ethanol extract of Coptidis rhizoma showed antimicrobial activity against L. casei. Makgeolli added with Sutellaria baicalensis extracts demonstrated a lower acidity than that with Coptidis rhizoma extracts, which indicates that the inhibition of an acetic acid former is more important than that of a lactic acid former in Makgeolli preservation. Sutellaria baicalensis extracts prolonged the shelf life of Makgeolli by 1~2 weeks at a minimal inhibitory concentration (0.63 ㎎/㎖) during storage at 10℃.

        The present study was undertaken to estimate the antibacterial effect of a combination of C. rhizoma,L. Flos, and P. japonica (1:1:1) extracts (CLP1000) and a combination of the herbal extract mixture and dioctahedral smectite (CLPS1000) against murine salmonellosis. At the concentration of CLP1000 and CLPS10000.5 mg/ml, the antibacterial effect was not showed on Salmonella typhimurium (S. typhimurium). On the other hand,the antibacterial effect against S. typhimurium was observed at the concentration of CLP1000 and CLPS1000 1.0 mg/ml. Oral administration of Smectite, CLP1000, and CLPS1000 at the dose of 10 mg/ml showed a therapeutic effect for S. typhimurium infected BALB/c mice. The mortality of Smectite, CLP1000 and CLPS1000-treated mice was 90%,90%, and 70% at 12 days, respectively, while that of untreated mice was 100% at 9 days after a lethal dose of S. typhimurium infection. The results of our study strongly indicate that CLPS1000 has potential as an effective of salmonellosis.

        In an attempt to find natural sources of antioxidants and whitening agents, Comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex were carried out. Comparison of the four ethanol extracts revealed that, Astragali Radix had the highest electron-donating ability(72.5%) and the highest SOD-like ability(26.1%). The xanthine oxidase experiment exhibited a hindrance effect of 88.5% in Atractylodis Rhizoma Alba, 81.1% in Acanthopanacis Cortex, 75.8% in Astragali Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 42.1% in the Acanthopanacis Cortex, 37.2% in the Atractylodis Rhizoma Alba, 6.0% in the Astragali Radix. Based on these results, we suggest that the ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex can be used as food and cosmetic ingredients.

        In order to fine antioxidant and whitening agent source from nature, the comparisons of antioxidative activity and tyrosinase inhibitory activity were carried out for various ethanol extract on Polygoni Multiflori Radix, Polygonati Rhizoma and Ephedrae Herba. Comparing for three ethanol extracts, the highest electron donating ability was found at Polygonati Rhizoma (86.6%), but, the highest SOD-like ability, at the Ephedrae Herba (47.8%). Xanthine oxidase experiment exhibited 95.7% of hindrance effect in Ephedrae Herba, and 84.0% in Polygonati Rhizoma. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 6.5% in the Polygoni Multiflori Radix, 32.6% in the Polygonati Rhizoma, 64.0% in the Ephedrae Herba. Based on these results, we suggest that the ethanol extracts of Polygoni Multiflori Radix, Polygonati Rhizoma and Ephedrae Herba can be used as food and cosmetic ingredients.

Comparing for three ethanol extracts, the highest electron donating ability was found at Poria Cocos (81.2%), but, the highest SOD-like ability, at the Coptidis Rhizoma(58%). Xanthine oxidase experiment exhibited 23.3% of hindrance effect in Pueraria Radix, and 79.3% in Poria Cocos. To evaluate at the whitening effect, tyrosinase inhibitory activity was conducted. Tyrosinase inhibitory activity was detected at 17.9% in the Puerariae Radix, 5.2% in the Poria Cocos and 83.3% in the Coptidis Rhizoma. From these results, we suggest that the ethanol extracts from Pueraria Radix, Poria Cocos, and Coptidis Rhizoma can be used for cosmetic ingredients.

        This study was carried out to know the active fraction of Dryopteris rhizoma on antimicrobial activity against some food spoilage microorganisms. Also, antimicrobal activities were investigated for the aqueous and ethanolic extracts of four herbs such as Terminaliae fructus, Eugeniae flos, Salviae miltiorrhizae radix and Dryopteris rhizoma. Antimicrobial activities of three herbs except for Terminaliae fructus showed higher activities in 75% ethanolic extracts than in aquoeus extract. Ethanolic extract of Dryopteris rhizoma showed the highest antimicrobial activity among extracts of four herbs. Antimicrobial activity intensities of solvent fractions of Dryopteris rhizoma extracted by 75% ethanol were order to CHCl₃ fraction$gt;EtOAc fraction $gt;BuOH fraction$gt;H₂O fraction. The CHCl_(3+) EtOAc and BuOH fractions also inhibited growth of food spoilage microorganisms as the concentration increased, respectively. In case of EtOAc fraction, 1000 ppm of fraction almost inhibited completely the growth of microorganisms tested.

        지황(地黃)의 주(主) 약효성분(藥效成分)으로 알려져 있는 catalpol의 iridoid골격(骨格)과 glycosidic bond를 형성(形成)하는 유리당류(遊離糖類)를 확인(確認)하기 위하여 생육시기(生有時期)에 따른 무기성분(無記性分), 유리당류(遊離糖類), 회분(灰分), 엑기스 및 catalpol의 함량(含量)을 조사(調査)하였다. 수확기(收穫期)가 가까워질수록 P, K, Ca, Mg, Cu 및 회분(灰分)의 함량(含量)은 감소(減少)하였으나, Fe 및 엑기스의 함량(含量)은 증가(增加)하였다. 수확기(收穫期)가 가까워질수록 전당(全糖), sucrose 및 galactose의 함량(含量)은 감소(減少)하였으나, fructose 및 catalpol의 함량(含量)은 증가(增加)하였다.

        같은 천남성과에 속하면서도 정유의 함유성분 및 함량이 서로 다른 수창포 정유와 석창포 정유가 토끼 적출장관 운동에 미치는 영향과 차이점, 또 어떠한 작용양식에 의하여 이루어지는가를 알아보고저 carbachol pilocarpine, histamine 및 barium chloride등과의 상호작용을 비교, 검토하였던 바 다음과 같은 결론을 얻었다. 1. 수창포 정유와 석창포 정유는 모두 토끼 적출장관 운동을 이완시켰으며, 농도를 증가시킬수록 이완정도는 현저하였고, 장관운동의 이완되는 정도는 두 정유 모두 같은 농도에서는 거의 비슷하였다. 2. ED50에 해당하는 농도는 수창포 정유는 0.0045% 이었고, 석창포 정유는 0.0035%이었다. 3. 수창포 정유와 석창포 정유에 의해 이완된 토끼의 적출장관은 carbachol pilocarpine, barium chloride에 의하여 정유 투여전의 상태로 회복하였으나, histamine에 의해서는 부분적인 회복을 보였다. 4. 수창포 정유와 석창포 정유는 carbachol pilocarpine, histamine 및 barium chloride에 의하여 수축된 토끼의 적출장판을 이완시켰다. 5. 이상의 결과로 보아서 수창포 정유와 석창포 정유는 다같이 향신경성작용 및 향근육성작용에 의해 토끼 적출장관 운동을 이완시킨다고 사료된다.

        Background : Brain waves are generated by the neurons in the brain. Many scientists fixed the electrodes of the electroencephalogram (EEG) on the scalp to record the signal expression of continuous cerebral activity. EEG is widely used in clinical diagnosis. It can be used to screen epilepsy, depression, ementia, stroke, encephalitis, ADHD and other brain diseases. Also, many local and foreign studies on aromatherapy focused on the discussion of human skin absorbing essential oils. This study discusses the level of relaxation efficacy of inhaled essential oils via the human olfactory organs. Thus, this study intended to observe the effects of essential oil of Aucklandiae Radix and Cyperi Rhizoma on EEG activity. Methods and Results : We evaluated the effects of essential oil of Aucklandiae Radix and Cyperi Rhizoma, on human EEG activity. For this purpose, essential oil was extracted from Aucklandiae Radix and Cyperi Rhizoma by the supercritical extraction method. The effect of its inhalation on EEG activity was evaluated by measuring the EEG power spectrum in 20 healthy participants. EEGs signals were recorded using QEEG-8 system (LAXTHA Inc., Daejeon, Korea) from 8 grounding electrodes. The subjects were instructed to sit quietly, close their eyes and to breathe normally during the measurement. The fragrance stimulus was dipped in a paper (14 ㎝2) then placed about 3 cm in front of the subject's nose. EEG was recorded before and during the fragrance exposure for 30 seconds. The mean power values (μV) calculated for fast fourier transform (FFT) analysis indicators. The results were as essential oils of Aucklandiae Radix and Cyperi Rhizoma decreased the overall brain relative theta (θ waves), but increased relative beta (β waves), absolute gamma (γ waves), spectral edge frequency (SEF) and spectral edge frequency 95% (SEF95). Conclusion : The changes in the EEG activities following inhalation of Aucklandiae Radix and Cyperi Rhizoma suggest that the oil can improve psychological well-being by increasing attention and relaxation.

        Background : Curcumae Longae Rhizoma (薑黃) is listed in「The Korea Pharmacopoeia (K P)」as the original plant of Curcuma longa L (Zingiberaceae). Meanwhile, Zeodariae Rhizoma (莪朮) is listed in 「The Korea Pharmacopoeia (KP)」as the original plant of C. phaeocaulis, C. aromatica and C. Kwangsiensi (Zingiberaceae). Due to the morphological similarities of the dried roots of this plant to those of C. phaeocaulis, C. aromatica and C. Kwangsiensis which is used as a substitute herbal for C.longa, distinguish these four species is extremely difficult. Methods and Results : A total of 90 collected samples were used in this study, In order to clearly distinguish of Curcumae Longae Rhizoma and Zeodariae Rhizoma were analysis based on sequence of the chloroplast DNA (trnK, rbcL, trnL-F, atpB-rbcL) and nuclear ribosomal DNA (ITS2). The present study aimed to analyze the percent of variable sites were provided the highest trnK (2.3%), in oder to develop a species-specific primer that can distinguish C.longa form C. phaeocaulis, C. aromatica and C. Kwangsiensis. In addition, the complete chloroplast genome of C. longa were sequenced by a 454 sequencing platform, and the structure of the obtained chloroplast genome was also analyzed. the result used that INDEL (insertion/deletion) marker for distinguish C.longa form C. phaeocaulis, C. aromatica and C. Kwangsiensis. Conclusion : The INDEL markers were developed based on the divergence of each sequence, and it is possible now to identify the four species of Curcumae Longae Rhizoma with just a single performance of PCR. This will not only prevent misused of the plant, but also to maintain the quality of the herbal medicine as well as to verify and guarantee safety for public health.

        Background : Two major forest medicinal resources, Korean angelica (Angelica gigas Nakai) and Cnidii rhizoma (Cnidium officnale Makino) were used as medicinal herbs for long times and in these day consumed as cosmetic materials. For keep pace with a mass demand and supply, pesticides were should be sprayed for control of pests and plant diseases. However, the social concerns on well-balanced life, pesticides safety, and continuous and stable production of clean non-timber products were increased with the times. Therefore, environmental-friendly cultivation was increased before decade. This study performed for selection of effective control agents to hyphal growth of leaf spot and anthracnose in two medicinal resources. Methods and Results : The antifungal effects of eugneol, pine needle E.O, and 7 natural product extracts (Sophora root, green tea, neem, eucalyptus, grapefruit seed, derris, and 36% rosin) were evaluated against two plant pathogens (Phoma sp., and Colletotrichum gloeosporioides). After re-loading of each 20 ㎕ natural product extracts to the PDA plate which loaded two spore dilution (106 spores/㎖) 100 ㎕, the hyphal growth control effects to pathogens were confirmed. Among 9 control agents, eugenol and pine needle E.O were the most effective, and then Sophora root, green tea, and neem. However, 4 agents (eucalyptus, 36% rosin, grapefruit seed, and derris) were not effective. Conclusion : It confirmed that possibility as a control agents of eugenol and pine needle E.O to leaf spot (Phoma sp.) of Korean angelica and anthracnose (Colletotrichum gloeosporioides) of Cnidii rhizoma, respectively. We will be developed as commercial products safe in the forest lands

        The quality control of natural products is principal key to guarantee the Good Manufacturing Practices (GMP) and Good Clinical Practices (GCP) for the functional food, pharmaceuticals and cosmeceuticals in the industry. In this study, we examined the quantitative analysis of berberine as marker substance of Coptidis Rhizoma by high performance liquid chromatography-photodiode array detector (HPLC-DAD). The HPLC method was validated and met all the requirements for the quality control analysis recommended by FDA and ICH. The berberine was separated on a Xterra C18 column (5 ㎛, 4.6 × 250 ㎜) using mobile phase consisting of distilled water and acetonitrile with KH2PO4 (3.4 g) and Na2SO4 (1.7 g). Calibration curve of berberine has been estimated (y = 42293.47x-41589 with the correlation coefficient 0.9999). The amount of berberine was calculated as 4.25%. And berberastine, palmatine, columbamine, jatrorrhizine, epiberberine, berberine and coptisine in the Coptidis Rhizoma were identified by high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-ESI-MS) method.

        Background : This study of investigate the quality characteristics of Korean Dioscoreae Rhizoma by species and various processing methods. We used two types of species D. Rhizoma, which were the long type shape of D. batatas and short type shape of D. japoninca, respectively. Methods and Results : The reaction of D. Rhizoma with acetic acid and iodine showed light blue to blue colours, respectively. When compared with loss of drying between two species, the long type species was about 2% more higher than that of short type of species. When compared with processing methods, the both steamed and dried samples was more higher 2.0 - 3.0% than that of non-steamed samples. The contents of acid-insoluble ash samples with peel was relatively higher than that of samples containing no-peel. The results of TLC analysis also showed the same Rf and colours when compared with the standard of D. Rhizoma. However, both dried sample with peel (DWP) showed more clear spot than those of dried sample except peel (DEP) and both steamed and dried sample except peel (SDEP). Conclusion : We sincerely hope that this study will be contributed to the standardization and quality control of korean herbal medicines.

        Background : Undulatum Rhubarb, commonly produced in domestic, is rhizome of Rheum undulatum L. that belongs to the family Polygonaceae. It also can be used as a substitute of R. palmatum L., R. tanguticum Maximowicz ex Balf., and R. officinale Baillon which completely depend on import system. However, there should be clear clarification among Undulatum Rhubarb and Rhubarb, because Undulatum Rhubarb contains rhaponticin as marker compound that is not indicated at Rhubarb. Some of the recently imported Undulatum Rhubarbs have been found to be Rhubarb. Also, it is known that only Undulatum Rhubarb is cultivated at domestic environment. But some plant origins of Rhubarb are grown in Korea, too. Further study are needed to clarify clear origin between Undulatum Rhubarb and Rhubarb. Thus, we collected some domestically cultivated samples and identified them. Methods and Reseults : Rheum undulatum L., Rhubarb, Rheum tanguticum Maximowicz ex Balf. which were cultivated in Gangwondo Agricultural Research and Extension Services in Cheorwon were collected and anayzed the DNA sequences. We also compared DNA sequences in Rhubarb collected from England and R. rhabarbarum L., R. undulatum L., and R. franzenbachii on NCBI. As a result, two kinds of rhubarb cultivated in the test plantation were identified as R. rhabarbarum and R. officinale. In addition, R. undulatum (plant origin of Undulatum Rhubarb) was identified as Rhubarb (Rheum rhabarbarum) in England with 99 - 100% identical in nuclear ITS gene region. Conclusion : R. undulatum, plant origin of Undulatum Rhubarb, is reported as synonym of R. rhabarbarum, R. franzenbachii. Rheum speices which are cultivated as tester in Gangwondo Agricultural Research and Extension Services in Cheorwon are estimated as R. undulatum and R. officinale. Therefore, not only Undulatum Rhubarb but Rhubarb could be grown in Korea.
