Electron spin resonance (ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity of irradiated crabs and the stability of these radicals under 9 weeks of storage. Swimming and small crabs were irradiated with doses of 0, 1, 2, 3, 5 and 7 kGy using a Co-60 irradiator at ambient temperature. A claw, a walking leg and a carapace of the crab pieced and dried were placed in a resonant quart tube within an EPR X-band spectrometer. The irradiated crabs presented an asymmetric absorption in shape at g₁=2.002±0.003 and g₂=1.998±0.005, and were different fro the non-irradiated ones. The intensity of the ESR signals was greatest in the claw, intermediate in the carapace and lowest in the walking leg. Samples given low and high doses of irradiation could also be distinguished. The ESR signal after irradiation was stable, even after a 9-week storage.

Lipid peroxidation is one of the main manifestations of oxidative damage and has been found play an important role in the toxicity and carcinogenesis of many carcinogens. This study was carried out to investigate the effects of aflatoxin B₁ co-administrated with antioxidant vitamins on lipid contents and fatty acids components of liver in mice. For this work, vitamin C and vitamin E, the major antioxidants, were administrated with 10 ㎎/㎏ and 63.8 ㎎/㎏ respectively, through intraperitoneal(i.p) injection to male ICR mice, and 0.4 ㎎/㎏ of the AFB₁ injected by i.p. lhr later. The results were as follows: two fold amounts of free cholesterol, triglyceride, and total cholesterol in serum and liver of mice treated with only AFB₁ were observed, when compared to those of mice co-administrated with antioxidant vitamins. However, the levels of phospholipids in serum and liver of mice treated with only AFB₁ were decreased. Concerning to fatty acids composition of liver from AFB₁-treated mice, P/S ratio was shown more low level in cholesteryl ester, triglyceride, total cholesterol and phospholipid than those of mice co-administrated with antioxidant vitamins. In these data which provide with a reliable evidence on their antioxidantal effects to aflatoxicosis.

The low salt seasoned jeotkal, salted and fermented fisheries product, may has some problems, such as short shelf-life, its putrefaction by mining some microorganism from additives. It was considered that most microorganism in seasoned jeotkal were introduced from red pepper powder. Therefore, it is important to pasteurize red pepper powder for improving its microbial quality. When red pepper powder was pasteurized by ohmic heating, the survival cell concentration in red pepper powder was reduced to 1-log-unit at 500 V/m, 700 V/m, above 80℃. But viable cell counts were reduced from 8.5 × l0^6CFU/g to 2.1 × 10₂CFU/g, i.e. 4.6-log-unit, during ohmic heating at 90 for 40 min. Color values of red pepper powder during ohmic heating with different holding time were not changed significantly. When squid jeotkal was manufactured by using the pasteurized red pepper powder, viable cell counts of the product were decreased by about three log cycles, compare with control product. And also the counts of fungi were significantly decreased.

In this study, ohmic heating was applied for pasteurization of red pepper powder, and investigated its pasteurization effect. After pasteurization, seasoned squid jeotkal was manufactured by using red pepper powder, and its quality changes were investigated. On comparing sensory evaluation in squid jeotkal by pasteurized and conventional red pepper powder during storage at 5, 15 and 25℃, quality changes in squid jeotkal by the pasteurized red pepper powder. But quality change difference between two products were decrease with increasing storage temperature. Viable cell counts in two products stored at 5℃ were increased slowly until 60 days. Viable cell counts in squid jeotkal by conventional red pepper powder were increased up to ca. 10^6 CFU/g at 15, 25℃ after 30, 15 days, respectively, but that by pasteurized were ca. 10^8 CFU/g at 25℃ after 30 days. Changes in pH, VBN and NH₂-N in two products were shown difference significantly, but were decreased by increasing storage temperature. In squid jeotkal by conventional red pepper powder, the main free amino acids were glutamic acid, leucine, glycine, aspartic acid and alanine, and these amino acids held 47.95% of the total free amino acid. But in squid jeotkal by pasteurized red pepper powder, glutamic acid, glycine, aspartic acid, leucine and lysine, and these amino acids held 57.58% of the total free amino acids.

General composition and microbial load of brine recycled to 6 times for Chinese cabbage salting and the quality of Kimchi using every brined cabbage were investigated. The concentration of salt(NaCl) in brine after soaking dropped 1.36-2.49% and pH of the brine changed significantly until 4 times recycling. The acidities were increased as number of recycling increased. The total viable cell count in recycled brine increased from 1.58 × l0^6/mL in the first soaking brine to 2.3 × l0^8/mL in 4th soaking brine which was highest in number. The pure soluble solid and vitamin C content in brine were accumulated to 0.93% and 0.55 ㎎% respectively after 6th recycling. The pH, acidity and sensory evaluation results of Kimchi prepared by Chinese cabbage salted by each brine recycled 6 times showed no significant difference. It means there is a possiblity reusing brine for salting of cabbage for the preparation of Kimchi to 6 times.

To investigate the effects of bacterial endotoxin on lipids and cytokines, and to explain the relationships between their changes, we carried out this study using experimentally Escherichia coli (E. coli) endotoxin-induced septicemic rabbits. The triglyceride and cholesterol concentrations in endotoxin groups were generally high (p$lt;0.01 or 0.05) at all sampling time points (from 3hr to 24 hr), while phospholipid concentrations in endotoxin groups elevated in dose-dependent fashion at 3hr and 6hr after endotoxin injection compared to control group (p$lt;0.05). There were significant negative correlations between lipids changes and cytokines (TNF-α and IL-1β) each other (p$lt;0.05), and endotoxic rabbits having higher triglyceride of cholesterol levels shown relatively better conditions compared with others. As a result, the alterations in the lipid compositions caused by endotoxin may imply an active exhibition of the host defense mechanism rather than the disturbances of lipid metabolism.

This study was carried out to know the active fraction of Dryopteris rhizoma on antimicrobial activity against some food spoilage microorganisms. Also, antimicrobal activities were investigated for the aqueous and ethanolic extracts of four herbs such as Terminaliae fructus, Eugeniae flos, Salviae miltiorrhizae radix and Dryopteris rhizoma. Antimicrobial activities of three herbs except for Terminaliae fructus showed higher activities in 75% ethanolic extracts than in aquoeus extract. Ethanolic extract of Dryopteris rhizoma showed the highest antimicrobial activity among extracts of four herbs. Antimicrobial activity intensities of solvent fractions of Dryopteris rhizoma extracted by 75% ethanol were order to CHCl₃ fraction$gt;EtOAc fraction $gt;BuOH fraction$gt;H₂O fraction. The CHCl_(3+) EtOAc and BuOH fractions also inhibited growth of food spoilage microorganisms as the concentration increased, respectively. In case of EtOAc fraction, 1000 ppm of fraction almost inhibited completely the growth of microorganisms tested.

Induction of DNA fragmentation of rat embryonic midbrain cells was studied to see whether apoptosis plays a role in OTA-induced microcephaly observed in cultured rat whole embryos during embryogenesis. We first cultured whole embryos (prepared from day 9.5 gestation rats) for 48 hrs with OTA and found that OTA induced microcephaly in cultured rat whole embryos. We also examined whether the microcephaly seen in cultured whole embryos is partially related to the increase of apoptosis of undifferentiated embryonic midbrain cells. Embryonic midbrain cells were prepared from day 12 gestation rat embryos, and cultured in the mixture media of Dulbecco's modified eagle's medium nutrient and Ham's F12 (1 : 1) containing 10% Nuserum, 100 ㎍/㎖ of streptomycin and 100 units/ml of penicillin for 96 hrs. Induction of DNA fragmentation was increased by 0.25-1 ㎍/㎖ OTA in a dose dependent manner in the embryonic midbrain cells. We also tested whether increase of apoptosis by OTA would be associated with change of apoptosis-related proteins (TNF-α and P^(53)) level in embryonic rnidbrain cells. OTA also increased TNF-α and P^(53) levels. These results show that OTA induced microcephaly in cultured whole embryos and this effect may be at least a part due to the induction of apoptosis and apoptosis-related protein levels of undifferentiated embryonic midbrain cells.

Totally, twenty seven specimens of tiger puffer, Fugu rubripes rubripes were collected at Jagalchi fish market in Pusan, Korea during January, April and September in 1995. Anatomical distribution of pufferfish toxin in tiger puffer was examined by mouse bioassay. The frequency rate of toxic specimens containing ≥ 10 MU/g was 14.8% in liver; 16.7% in gonad; and 14.8% in skin, and no toxin was detected in muscle. The highest toxin level found was 160 MU/g in liver, 600 MU/g in gonad and 26 MU/g in skin, and each average toxin level (mean ± standard error) was 7 ± 6, 50 ± 35 and 5 ± 1 MU/g, respectively. Some specimens collected in January and April were toxic, while none of the specimens collected in September showed its toxicity. Although toxicity of tiger puffer showed the seasonal variation, tested tiger puffer was evaluated as a safe seafood for consumption, in that an acceptable level of toxin was found in the edible muscle and skin.

In the 5'-flanking region of the D-AAT, AspAT and AlaDH gene, I found three or two pairs of sequences(designated as P1, P2, P3) which show significant similarity to the E.coli consensus sequences of -35 and -10 for promoters. The spacing between -35 and -10 is 16 to 18bp in all the three putative promoters Pl, P2 and P3 which is in good agreement with the preferred spacer length in E.coli and in B.subtilis. Therefore, the putative promoters may also function to increase the efficiency of transcriptional initiation. The most stable, double-helical $quot;Shine-Dalgarno$quot; pairing is formed with a free energy change(△G) of -13.0 ㎉/㏖, -9.6 ㎉/㏖, -15.8 ㎉/㏖.

In order to develop a antimicrobial agent, dried bamboo trunk was extracted by high temperature suction and then antimicrobial activities against Staphylococcus aureus ATCC 2825 and Escherichia coli DH 5α were investigated as compared with tea tree oil and BHA. The minimal inhibitory concentration(MIC) of the extracted substance against microorganisms were also examined. The results are as follows: 1. By disc diffusion method, BHA showed the strongest antimicrobial activity on Gram-positive bacteria such as S. aureus ATCC 2825 but bamboo essential oil showed the strongest antimicrobial activity on Gram-negative bacteria such as E. coli DH 5α. 2. By broth dilution method, the minimum inhibitory concentration of the BHA, tea tree oil and bamboo oil were not detected against S. aureus ATCC 2825(MIC, 6.0 ㎕/㎖) and E. cola DH 5α(MIC, 6.0 ㎕/㎖)
