        This study aimed to investigate the protective effect of enzymatically modified stevia (EMS) on C2C12 cell-based model of dexamethasone (DEX)-induced muscle atrophy to provide baseline data for utilizing EMS in functional health products. C2C12 cells with DEX-induced muscle atrophy were treated with EMS (10, 50, and 100 μg/mL) for 24 h. C2C12 cells were treated with EMS and DEX to test their effects on cell viability and myotube formation (myotube diameter and fusion index), and analyze the expression of muscle strengthening or degrading protein markers. Schisandra chinensis Extract, a common functional ingredient, was used as a positive control. EMS did not show any cytotoxic effect at all treatment concentrations. Moreover, it exerted protective effects on C2C12 cell-based model of DEX-induced muscle atrophy at all concentrations. In addition, the positive effect of EMS on myotube formation was confirmed based on the measurement and comparison of the fusion index and myotube diameter when compared with myotubes treated with DEX alone. EMS treatment reduced the expression of muscle cell degradation-related proteins Fbx32 and MuRF1, and increased the expression of muscle strengthening and synthesis related proteins SIRT1 and p- Akt/Akt. Thus, EMS is a potential ingredient for developing functional health foods and should be further evaluated in preclinical models.

        Globally, colon cancer is increased gradually and known as one of the major causes of cancer death. Stevia, a substitute of sugar, is known to have many components including alpha-tocopherol and anthocyanin etc, as antioxidants. This study's purpose is to investigate whether stevia plant extract can have a protective effect against colon carcinogenesis induced by azoxymethane (AOM) and dextran sodium sulfate (DSS) in mice. Total 30 male ICR mice were divided into 2 groups; AOM/DSS treatment (control group), AOM/DSS + stevia extract (0.5%, in drinking water). After acclimation for 1 week, five weeks old mice received three intraperitoneal AOM (10 mg/kg b.w.) injections weekly for 3 weeks (0–2nd weeks of the experiment) and 2% DSS as drinking water for the next one week. AIN-76A purified rodent diet and 0.5% stevia extract water were supplied to the animals for 6 weeks. The colons of mice were collected and the number of aberrant crypt foci (ACF) and aberrant crypts (ACs) in colonic mucosa were counted after staining with methylene blue. Malondialdehyde (MDA) concentration in feces were determined. The numbers of ACF and ACs were significantly (p<0.01) decreased in stevia-treated group compared with the control group. The MDA concentration in feces was also significantly (p<0.01) decreased in stevia-treated group compared with the control group. In histopathology of colonic epithelium, hyperplasia of colonic epithelium was less observed in steviatreated group. These results indicate that stevia has a protective effect against colon carcinogenesis induced by AOM/DSS in mice and further study needs to illustrate the protective mechanisms.

        This study added stevia leaf powder at ratios of 0.5, 1.0, 1.5, and 2.0 percent to Omija-pyun (Schisandra chinensis Jelly) as a natural low-calorie sweetener instead of sugar which is added to Omija-pyun in considerable amounts to evaluate quality characteristics and antioxidant activities compared to the control group with the addition of sugar. Moisture content of Omija-pyun expanded by increasing the measurement of stevia leaf powder (p<0.001), pH (p<0.01) and sugar content (p<0.001) decreased. L-values and b-values revealed a tendency to increase by adding more stevia leaf powder, but a-value revealed a tendency to decrease (p<0.001). Hardness (p<0.001) and chewiness (p<0.05) decreased by adding more stevia leaf powder. Based on the consumer preference evaluation, Omija-pyun with the addition of 0.5 percent stevia leaf powder was the most preferable in terms of color and flavor (p<0.001). The control group and Omija-pyun with the addition of 0.5 percent stevia leaf powder was the most preferable in terms of taste and texture (p<0.001). There were significant differences in the organoleptic properties except hardness between the samples by quantitative descriptive analysis. The control group revealed the highest preference in terms of redness and transparency, and redness and transparency tended to decrease by adding more stevia leaf powder (p<0.001). Organoleptic properties on bitterness and sourness were enhanced by adding more stevia leaf powder in the principal component analysis (PCA). Regarding antioxidant properties, total phenol compounds and flavonoid contents of Omija-pyun increased by adding more stevia leaf powder, and DPPH radical scavenging capacity also increased (p<0.001). Based on results, it is preferable to serve Omija-pyun with the addition of 1.0 percent stevia leaf powder instead of sugar within the context of quality and antioxidant activity.

        In order to develop tea by using the leaves of stevia, which is a herbal plant, and to solve the disadvantages of stevia dried leaf tea, we have manufactured the steamed tea, stir-fried tea and fermented tea by changing the manufacturing processes. As a result of the sensory tests, the steamed tea, stir-fried tea and fermented tea received higher evaluations than the dried leaf tea. In terms of efficiency, it is desired that the total number of steaming and stir-frying is only once, but the fermentation is found to be the most desirable for 2 days. There are no trends for changes in the general ingredients, mineral and free amino acid contents of stevia leaf teas by different manufacturing processes. As a result for the measurement of antioxidant activities, the steamed tea and dried leaf tea did not show significant differences, but the stir-fried tea and the fermented tea show significantly low antioxidant activities as compared to the steamed tea. The contents of stevioside in both the stir-fried tea and the fermented tea were less than that in the dried leaf tea, but in the steamed tea, there was no significant difference in the content of stevioside. Base on the present observations, this study supports high potentials of steaming process in order to produce new stevia leaf tea.

        Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low-calorie sweetener and is being developed as a therapeutic agent for diabetes, hypertension, myocardial diseases, and microbial infections. Despite the common use of its leaves and stem, the bioavailability of the components present in S. rebaudiana flowers, when used as ingredients of cosmetics, has not been well investigated. Herein, we investigated the antioxidative and antimelanogenic effects of an aqueous extract of S. rebaudiana flowers (Stevia-F). Total flavonoid and phenolic content in Stevia-F were determined to be 8.64 ± 0.23 ㎎ of quercetin equivalents/100 g and 631.5 ± 2.01 ㎎ of gallic acid equivalents/100 g, respectively. The IC50 values of Stevia-F for reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate radical, hydrogen peroxide, and nitric oxide scavenging activities were 5541.96, 131.39, 466.34, and 10.44 ㎍/mL, respectively. Stevia-F showed inhibitory effects on the tyrosinase (IC50 = 134.74 ㎍/mL) and α-glucosidase (IC50 = 114.81 ㎍/mL) activities. No significant cytotoxicity of Stevia-F was observed in B16F10 cells, treated with up to 100 ㎍/mL of the extract for 24 and 48 h (p > 0.05). Stevia-F (1–100 ㎍/mL) suppressed α-melanocyte stimulating hormone-induced melanin production in B16F10 cells (p < 0.05) and also inhibited the cellular tyrosinase activity (p < 0.05). Overall, our results show that Stevia-F possesses potential for inhibiting tyrosinase and α-glucosidase activities and has significant antioxidant capacity. The antimelanogenic potential of Stevia-F should extend the usage of S. rebaudiana flowers in the development of skinwhitening products.

        Background : Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low caloric sweetener and therapeutic agent for diabetes, hypertension, myocardial, and antimicrobial infections. It has been commonly used leaves and stems because of their high anti-oxidative potential. The present research was carried out to explore anti-oxidative and anti-melanogenic effects of aqueous extract of Stevia rebaudiana Bertoni flos in B16F10 cells. Methods and Results : Anti-oxidant activity of Stevia flos extract (SFE) was determined by using 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging assay, nitric oxide (NO) radical scavenging method, and reducing power method. The results showed that the total phenolic content of SFE was 63.154 ± 0.0002 ㎍·QE/100㎎ and the total flavonoid was 8.64 1 ± 0.002 ㎍·GAE/100㎎. SFE exhibited a big significant effect on NO radical scavenging activity (IC50: 179.6 ㎍/㎖) comparing with standard ascorbic acid (IC50: 368.6 ㎍/㎖), and showed concentration dependent DPPH radical scavenging activity (IC50: 131.8 ㎍/㎖). Anti-melanogenic effect of SFE was also examined with B16F10 melanocytes. The amount of melanin synthesis followed by α-melanocyte stimulating hormone on B16F10 cells were significantly reduced in the presence of SFE treatment (p < 0.05). SFE also suppressed the tyrosinase activity (p < 0.05) and α-glucosidase activity (p < 0.05). Conclusion : These results provide evidence Stevia rebaudiana flos has an antioxidant potency and can be used as an anti-melanogenic agent.

        Background : Stevia extracts were reported to contain steviol glycosides like stevioside and rebaudioside A, as well as various polyphenol compounds. However, there are many difference in researchers with extract methods, considering components and antioxidant activity. This study was conducted to select optimum extraction method based on reported methods by measuring the natural sweeteners contents, analyzing antioxidant materials and activity according to extraction methods. Method and Results : Stevia leaves were extracted through vacuum extraction, hot water extraction, soxhlet extraction, high temperature and pressure extraction and ultrasonic extraction to identify the high extraction efficiency. Extraction yield was high in order of hot water extraction, soxhlet extraction, high temperature and pressure extraction and ultrasonic extraction except for vacuum extraction and contents of rebauduoside A and stevioside in stevia leaves was highest in high temperature and pressure extraction followed by vacuum extraction, soxhlet extraction, hot water extraction and ultrasonic extraction. Also, total phenolic contents and flavonoid contents were highest in high pressure extraction and vacuum extraction according to extraction methods. But DPPH radical scavenging activity was highest in ultrasonic extraction and tended to be rather low in high temperature and pressure extraction (ultrasonic extraction > hot water extraction > high pressure extraction > soxlet extraction > vacuum extraction) and there wa no difference in ABTS radical scavenging activity among extraction methods. Conclusion : The high temperature and pressure extraction is considered to be a suitable method for extracting stevia leaves because it showed high extraction yield and had high contents of rebaudioside A, stevioside, total phenolic contents and flavonoid contents.

        Background : Recently, there have been dynamic researches conducted on stevia as natural sweetener subtitute for sugar, However, Researches related to harvest period and parts of plant in stevia are few. Therefore, this study was conducted to select optimum harvest time and parts by measuring the natural sweeteners contents and analyzing antioxidant materials and activity according to harvest times, parts of plant. Methods and Results : Stevia was cultivated in plastic house, The leaves were harvested from April to October and the stem were only harvest in July and September. Stevia leaves and stems were extracted using high temperature and pressure extraction: Dried stevia leaves of 5g were added by 100ml of distilled water equivalent to 20 times of dry weight, and the mixture were extracted by autoclave at 121℃ for 15min. The contents of Rebaudioside A and Stevioside of stevia leaves harvested from April to October showed a tendency to increase gradually from July to October as the temperature increased, but the contents of rebaudioside A and stevioside decreased slightly in August due to excessively high temperature. The extraction yield of stevia leaves were highest in October and September, and there was no significant difference in the other period. In the stevia stems, the extraction yield was lower than that of stevia leaves in general regardless of harvest time. Total phenolic contents and flavonoid contents according to harvest time showed little difference among treatment. Conclution : stevia leaves were better than stevia stems regarding the use of rebaudioside A and stevioside as natural sweeteners. Also, it was confirmed that the stevia leaves of July, September and October, except for the high temperature period of August, had superior in quality and quantity.

        Background : Currently, obesity and adult diseases as a result of sugar intake have been a consistent problem in Korea. Natural sweeteners as sugar substitutes have many advantages over sugar as a small quantity. Therefore, this study was conducted to investigate possibility of industrialization as food additives by using the nature sweetness components of stevia (Stevia rebaudiana Bertoni). Method and Results : Stevia was cultivated in a plastic house. The leaves and stems were harvested at October. They were ground into fine powder using a mill, and were extracted by high temperature and pression extraction method. The extracts were evaporated under vacuum and lyophilized. Three strawberry cultivars of ‘Seolhyang’, ‘Maehyang’, and ‘F22-196’ were cultivated in a plastic house and were harvested from March to May. The fruits harvested were stored at 50℃ until processing. In order to test the processing suitability of stevia extract, the characteristics of the three strawberry varieties (line) were investigated. As a result, the ‘F22-196’ line, which was bred as a processing strawberry, generally contained more antioxidant materials and activity than those of ‘Maehyang’ and ‘Seolhyang’ varieties. Comparing the sugar contents which affects the quality of strawberry jam, the average sugar contents of ‘F22-196’ line was higher than 'Maehyang' and 'Seolhyang' varieties. In the preparation of strawberry jam using ‘F22-196’, strawberry jam was prepared by adding only sugar or stevia extract powder, which was 1/100 of the amount of the sugar in only sugar strawberry jam, to the sensory test. As a result, we identified that sugar jam and stevia jam added stevia extract showed 50 : 50 at the sensory test and stevia jam does not make a difference to the marketing jam at the point of view of general consumers. Conclusion : ‘F22-196’ line represented the best quality for strawberry jam in test caltivars. Stevia powder is judged to be used as a sweetener of sugar substitutes in the production of strawberry jam and processing food.

        유사미소중력, 진동 그리고 저온과 같은 물리적 스트래스하에서 4일간 자란 약용식물인 스테비아(Stevia rebaudiana Bertoni)는 대조구에 비해 3.6%, 21% 그리고 8.7%의 생체 중량의 감소를 보였다. 합성 항산화물질인 AA(ascorbic acid)와 BHA(beta-hydroxyacetic acid)는 비교적 약한 22%와 27%의 항산화 활성을 보였다. 유사미소중력하에서 스테비아의 DPPH(2,2-diphenyl-1-picryl hydrazal)의 음이온 소거효과는 대조구보다 지속적으로 더 높게 나타났으나, 진동과 저온처리의 처리효과는 급히 증가했다가 진동의 경우는 6시간 후에 감소되고 저온처리의 경우는 24시간 후에 대조구 수준으로 감소되었다. 유사미소중력하에서 SOD활성은 대조구에 비해 147%, 진동과 저온처리는 각각 121%와 125%로 증가를 보였다. 이상의 결과로부터 유사미소중력스트레스는 다른 스트레스보다 스테비아의 항산화 활성에 더 효과적인 영향을 미치는 것이 분명해졌다.

        천연 저 칼로리 감미료로써 stevia의 소비가 증가하고 있는 시점에서 재배 및 기능성 식품소재로 이용하기 위하여 수확시기별 특성을 조사한 결과는 다음과 같다. 가용성 고형분 함량과 추출수율은 성장 기간이 길어질수록 증가하였고, pH는 감소하는 경향을 보였다. 색도와 투과율은 큰 변화가 없었으나, 성장 시기가 길어질수록 L, a, b값은 유사한 감소 경향을 나타내었고, 갈색도는 증가하였다. 성장기간 중 stevioside와 rebaudioside

        자가불화합성 작물인 스테비아는 순도를 유지하기 위하여 삽목에 의해 번식하고 있으나 그 소요기간과 증식률이 낮다. 본 실험에서는 액아로부터 완전한 식물계로 분화시켜 대량 증식을 위한 기내배양조건을 알고자 하였으며 그 결과를 요약하면 다음과 같다. 1. Murashige-Skoog(MS) 기본 배지에 NAA와 kinetin을 단독 처리할 때 액아로부터 shoot분화가 가장 좋은 경우는 NAA는 0.01-0.05mg/의 범위였고, kinetin은 0.5-1.0mg/의 범위이었다. 2. MS배지에서 shoot분화는 NAA 0.01-0.05mg/와 kinetin 0.5-1.0mg/을 조합처리하였을 때 좋았다. 3. MS배지의 salt strength를 달리할 때 1 MS가 12XMS보다 shoot분화와 생육에서 좋을 결과를 보였다. 그러나 root분화는12XMS에서 높았다. 4. 이상의 결과로보아 스테비아는 단시일 내에 많은 양을 증식시킬 수 있으며 shoot분화와 root 분화시 배지의 염류농도와 홀몬농도를 달리하는 것이 효율적이다.

        스테비아는 Paraguay 산중에 야생하는 국화과에 속하는 다년생 초본식물로서 그 엽에는 감미성분 Stevioside가 6~12% 함유되어 있다. Stevioside는 배당체로써 감미가 설당과 유사하적, 감미도는 설당의 약 300배가 된다. 우리나라에서는 당원료(사탕무우, 사탕수수)를 전연 생산하지 않으며 또한 합성감미료의 유해성이 논란되어 그 사용이 제한되므로써 새로운 감미료 개발의 필요성이 절실하다. 그리하여 1973년 우리 나라에 도입된 스테비아를 감미자원식물로서 정착시키기 위하여 재배에 관한 시험을 실시하였는데 그 결과를 요약하면 다음과 같다. 1. 정식에서 발뢰기까지 일수는 공시품종 모두 14시간 일장보다 단일조건에서 단축되었으며, 12시간과 14시간 일장간에서 발뢰기의 차이가 가장 크게 나타났다. 77013은 단일에서 발뢰촉진정도가 크며 수원002, 77067은 감광도가 둔한 계통이었다. 2. 12시간 일장을 발뢰기를 결정하는 변곡점(turning point)이었으며 한계일장은 12시간 내외라고 할 수 있다. 단일에서는 조기 발뢰에 따른 생육일수 불1족으로 생육양이 현저하게 낮았다. 3. 삽본묘는 발근전에도 단일에 감응 하는데 40일 양묘에서 단일감응이 예민하여 발뢰까지 일수가 가장 짧은 20일이있으며 양묘일수가 짧으면 단일감응 후발뢰가 지연된다. 4. 실생묘의 출아에서 발뢰까지 일수는 출아후 20일 단일 처리에서 가장 짧았으며 20일보다 단일처리 시기가 빠르거나 늦을수록 발뢰까지 일수는 길어졌다. 5. 삽목기가 3월 20일에서 5월 20일로 늦어질 수록 초장, 분지수 및 건물중이 현저하게 낮아졌으며, 3월 20일 삽목에서는 40~50일, 4월 20일 삽목에서는 30~40일, 5월 20일 삽목에서는 30일 육묘에서 건엽수량이 높았다. 6. 건엽수량과 재식주수와는 Asymptotic relation을 보였으며 10a당 5,000~10,000주까지는 건엽이 급증하였고, 10.000~20,000주는 그 증가가 수만하며 20.000주 이상에서는 증가율이 더욱 둔화되었다. 따라서 20,000주가 적정재식주수라고 할 수 있다. 7. 수원, 청주, 목포, 제주에서 스테비아 활착과 생육은 양호하였으나 생육성기의 한발은 수량감수원인이 되었다. 정식기가 6월 20일로 늦으면 적기보다 건엽수량이 현저하게 감수되었다. 8. 3월 20일 정식의 보온단일구에서 단일감응으로 건엽수량이 낮았으나 보온 장일 및 보온 자연일장은 보온효과가 크게 나타났다. 4월 20일 이후 안식에서는 보온효과는 인정되지 않았다. 9. Stevioside 함량은 상부엽에서 높았으며 지상 20cm 부위는 현저하게 낮았다 건엽과 Stevioside 수량은 대체로 60~120cm에 집중되어 있었으며 그 집중정도는 품종에 따라 큰 차이가 있었다. 10. 발뢰까지는 수확기를 지연시킬 수록 건엽수량은 현저하게 증가되었으며 발뢰 이후에는 수확기를 지연시켜도 수량 차이는 인정되지 않는다. Stevioside 함량은 발뢰기에 가장 높았고 이보다 빠르거나 늦게 수확할 때 감소하는 경향을 보였다. 건엽수량과 Stevioside 함량을 고려한 수확적기는 발뢰시부터 개화전까지이고 수원 지방에서는 9월 10일~9월 15일경이었다. 11. 영양계의 Stevioside와 Rebaudioside 함량범위는 각각 5.4~l4.3%, 1.5~8.3%로 변이폭이 크며 Stevioside와 Rebaudioside 함량간에는 일정한 경향이 없었다.

        본 시험은 제주도에서 stevia (Stevia rebaudian a Bertoni M.)의 효과적인 번식, 월동방법 및 재식거리에 따른 생육과 적응성을 알고져 실시 하였던바 몇가지 얻어진 결과를 요약하면 다음과 같다. 1) 식물생장조절제처리에 의한 발근율은 I.B.A=atonic(equation omitted)N.A.A>tachigalen>rooton>centre의 순위였고 발근수는 I.B.A>N.A.A>rooton>tachigalen>atonic>control 순위였으며 식장은 I.B.A(equation omitted)N.A.A>rooton>tachigalen>atonic>control 순위였다. 2) 상토별 효과에서는 발근율이 송이>심토>냇모래 (천사)>모래의 순위였고 발근수는 송이(equation omitted)모래>심토(equation omitted)냇모래의 순위였으며 근장은 모래>송이>냇모래>심토의 순위였다. 3) 월동효과는 polyethylene film tunnel이 가장효과가 좋았고 다음으로 polyethylene 피복이 효과적이었으며 볏짚과 왕겨피복은 재(회) 피복이나 무처리에 비해서는 효과적이나 지온상승에는 효과가 크지 못하였다. 4) 재식거리별 효과는 밀식 할수록 전체중이나 건엽중도 많았다. 5) 초장신장은 기온이 높을때 비례적으로 증가하며 7월하순부터 8월하순에 걸쳐 신장량은 급진되었다.
