        충청북도 충주지역에서 생육하는 굴참나무림에서 세근에 의한 지하부에서의 연간 탄소축적량을 조사하기 위하여 2001년 4월부터 11월까지 매달 토양샘플러를 사용하여 0-30cm, 30-60cm, 그리고 60-90cm 토양 깊이에서 샘플을 채취하였다. 세근에 의한 탄소생체량(fine root carbon biomass)은 다른 두 개의 조사 토양깊이에서 보다 0-30cm 토양깊이에서 가장 많은 것으로 나타났다. 세근에 의한 순탄소생산량 (kg/㏊/yr)은 0-30cm에서 671kg, 30-60cm에서 599kg, 그리고 60-90cm토양깊이에서 479kg로 나타났으며, 0-90cm토양깊이에서 세근에 의한 총 순탄소생산량은 1749kg이었다 세근고사율은 0-30cm에서 0.43, 30-60cm에서 0.96. 그리고 60-90cm에서 1.03로 나타났다. 세근고사율에 의한 토양으로의 연간 양분투입량은 N의 경우 33.9kg/㏊/yr, P의 경우 1.8kg, K의 경우 11.4kg, 그리고 Ca의 경우 20.1kg으로 나타났다.

        Background : This experiment was conducted to improve the farm income by decreasing the rate of corrosion and increasing the emergence rate by selecting the proper storage method of the medicinal crop seed root. Methods and Result : Ten kinds of medicinal crop seed roots of Glycyrrhiza uralensis Fischer, Salvia miltiorrhiza Bunge, Codonopsis lanceolata, Platycodon grandiflorum, Atractylodes japonica, Angelica acutiloba, Adenophora triphylla, Rehmannia glutinosa, Angelica gigas and Cnidium officinale were used as test materials. In experiment 1, storage experiments were carried out using four treatments: untreated (dry in advance), untreated, filler (dry in advance) and filler. Drying in advance was carried for 2 days in dark place and the filler was vermiculite. For storage, in styrofoam box, seed roots were laid down in 3 replicates of 10 sets, stored in – 1℃ warehouse. The storage temperature was set as long as each seed root was not frozen investigating freezing point, supercooling point. As a result, the rate of corrosion was significantly lower in the 4th treatments (filler), such as A. japonica (30%) and A. gigas (10%), compared with untreated one. Therefore, in experiment 2, six treatments were carried out using filler. There were 6 treatments: filler disinfectant (dry in advance), filler disinfectant, filler biochar (dry in advance), filler biochar, filler disinfectant biochar (dry in advance) and filler disinfectant biochar. The disinfectant was used by metalaxyl-M at a concentration of 0.4 g/ℓ, and a biochar was used at a volume ratio of 50% (v/v). As a result, the rate of corrosion was significantly lower in the 6th treatment (filler disinfectant biochar), such as S. miltiorrhiza (13.3%) and C. lanceolata (6.7%). Conclusion : In storing medicinal crop root, it is better to store using fillers. It was shown that a mixture of filler and disinfectant biochar reduced the rate of corrosion.

        Background : The practice of keeping the medicinal herbs at room temperature causes many problems, but due to lack of sufficient field research and study, it is difficult to improve related regulations and safety management. Methods and Results : The Cnidium Rhizome and Angelica gigas Root were inoculated with Lasioderma serricorne F. and incubated at 28℃ for 2.5 months and 5 months. After five months, the number of Lasioderma serricorne F. in Cnidium Rhizome increased from 30 to 1,429 (about 47 times). In the same period, the number of insects in Angelica gigas Root increased from 30 to 663 (about 22 times). Due to the rapid increase in pest population, hygiene deteriorated, changes in the active ingredient and appearance quality of the herbal medicines, which greatly damaged the value of the herbal medicine. Conclusions : These results show that current regulation requiring only sealing and not specifying the storage temperature do not guarantee quality safety. Therefore, it is necessary to establish appropriate preservation standards and improve management regulations in order to preserve safety.

        Sweet potato has low regeneration capacity, which is a serious obstacle for the fruitful production of transgenic plants. Simple and rapid regeneration method from storage root explants of purple sweet potato (Ipomoea batatas L.) was investigated. The embryogenic callus was observed from 4 cultivars and its highest rate was induced at 1 μM 2,4-D after 5 weeks of culture. Result revealed that a low concentration of 2,4-D and low light intensity was important factors for embryogenic callus formation. After subculture on medium with 5 μM ABA for 4 days, subsequently, occurred the regeneration of shoots within 4 weeks when these embryogenic callus was transferred onto the MS hormone free medium. Regenerated shoots were developed into platelets, and grown normal plants in the greenhouse. We developed a simple and quickly protocol to regenerate plantlets in storage root explants of purple sweet potato. This regeneration system will facilitate tissue culture and gene transfer research of purple sweet potato.

        도라지를 수확 후 진공포장, 밀폐포장, 질소가스포장 후 저온과 상온에 100일 동안 저장하면서 저장 50일과 100일에 도라지 경도와 사포닌 함량을 조사한 결과를 요약하면 아래와 같다.1.저장온도에 따른 도라지의 경도는 저온저장이 상온저장보다 높게 나타났으며, 저장기간이 길어질수록 도라지의 경도는 낮아졌다. 2.저장방법에 따른 도라지의 경도는 저장 50일차 지퍼팩을 이용한 밀봉에서 1,765 g으로 가장 높게 나타났으며, 저장온도와 저장방법과의 상호작용에서는 고도의 유의성(P≤0.01)이 인정되었다.3.저장온도에 따른 platycodin D3, polygalacin D, 그리고 deapioplatycodin D의 함량조성이 다르게 나타났다.4. 저장기간에 따른 platycodin D3와 polygalacin D의 함량은 저장 50일 이후 감소하는 경향을 보였지만, deapio-platycodin D는 저장기간이 길어질수록 증가하는 경향을 보였다.

        The role of an expansin gene (IbEXP1) in the formation of the storage root (SR) was investigated by expression pattern analysis and characterization of IbEXP1-antisense sweetpotato (Ipomoea batatas cv.Yulmi) plants in an attempt to elucidate the molecular mechanism underlying SR development in sweetpotato. Transcript level of IbEXP1 was high in the fibrous root (FR) and petiole at the FR stage, but decreased significantly at the young storage root (YSR) stage. IbEXP1-antisense plants cultured in vitro produced FRs which were both thicker and shorter than those of wild-type (WT) plants. Elongation growth of the epidermal cells was significantly reduced, and metaxylem and cambium cell proliferation was markedly enhanced in the FRs of IbEXP1-antisenseplants, resulting in an earlier thickening growth in these plants relative to WT plants. There was a marked reduction in the lignification of the central stele of the FRs of the IbEXP1-antisense plants, suggesting that the FRs of the mutant plants possessed a higher potential than those of WT plants to develop into SRs. IbEXP1-antisense plants cultured in soil produced a larger number of SRs and, consequently, total SR weight per IbEXP1-antisense plant was greater than that per WT plant. These results demonstrate that SR development was accelerated in IbEXP1-antisense plants and suggest that IbEXP1 plays a negative role in the formation of SR by suppressing the proliferation of metaxylem and cambium cells to inhibit the initial thickening growth of SRs.

        The antimicrobial activities of Smilax china L. against spoilage bacteria isolated from Mang-gae rice cake were investigated and the storage stability of the Mang-gae rice cake was enhanced. Spoilage bacteria, which cause Mang-gae rice cake to rot, were isolated from commercial Mang-gae rice cake, and most of the isolated strains were identified as Bacillus sp. After the leaves, roots, shoots, and stalks of the Smilax china L. were extracted using 50% ethanol as the solvent, their antimicrobial activities were investigated using the paper disc method by treating them with 50 μL of Bacillus cereus, which is known as a major pathogenic micro-organism in foods that contain starch, as the test organism. The antimicrobial activities of the extracts were compared according to the size of the clear zones around the paper discs. The root extract showed significant antimicrobial activities. When red beans, which are used as stuffing for Mang-gae rice cake, were treated with the root extract of the Smilax china L., the viable cell count of the Mang-gae rice cake was 5.04 Log CFU/g after 48-hr storage, and the cake showed significantly slower growth of bacteria than with commercial products. These results show that treatment of red beans with Smilax china root extract could improve the storage stability of Mang-gae rice cake.

        The purpose of this research is to develop bellflower (Platycodon grandiforum) root Jeonggwa as a health food. The Omija (Schizandra chinensis Baillon) water extract with 0-8% (w/w) Jeonggwa added was stored at for 12 weeks. The quality characteristics of Jeonggwa during storage were investigated. The Jeonggwa showed a lower pH and a higher acdity as the amount of Omija water extract that was added increased. The moisture content of Jeonggwa were rose from 15-18% to 16-22% within 2 weeks of storage, and remained at that level up to the end of storage. The total viable cells of bellflower root Jeonggwa were 2.1~2.3 log CFU/g and increased in number during storage but never exceeded 4 log CFU/g. The shelf life of the Jeonggwa was extended when the Omija extract was added.         This study was carried out to obtain basic information for optimum storage method of leaf, stem and root of Angelica acutiloba Radix, which is being utilized as major cultivated herbal medicine materials, root of Angelica acutiloba was stored for 3 months in different packing materials and sealing methods, such as punched packing, sealing and vacuum packing. The results are summarized as follows. The loss in weight as influenced by packing materials and sealing degree was the lowest at vacuum packing and complete sealing condition with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between polyethylene films, but it was significantly different between sealing conditions. Therefore, vacuum packing and complete sealing with polyethylene film are desirable storage condition for Angelica acutiloba Radix.

        수도의 직파재배에 있어서 가장 취약점인 출아.입묘율을 향상시키는데 많은 영향을 미치는 중배축 신장에 대하여 검토하였던 바 그 결과를 요약하면 다음과 같다. 1. 품종의 기원별 중배축장은 다수형 품종이 9.6mm로 재래종과 일본형 각각 4.4, 3.2mm 보다도 훨씬 컸다. 2. 생태형별 중배축 신장은 tjereh, aman, aus, boro 및 bulu 순으로 tjereh가 29.3mm로 buld의 5.4mm에 비해 약 5배나 컸다. 3. 품종간 중배축장은 재래종에서 백경조, 다다조 및 해조가, 일본형은 상풍벼, 팔달 및 간량도가, 다수형 품종에서는 원풍벼, 가야벼, 밀양30호 및 수정벼가 컸다. 4. 종자 숙도별 중배축장은 개화 1주후 채종 종자가 39.3mm로 가장 컸고, 성숙일수가 길어짐에 따라 중배축장은 점차 작아졌다. 5. 종자 저장기간이 길어질수록 종자숙도와 관계없이 중배축장은 점차 감소하는 경향이었고, 저장온도 5℃ 구가 15℃ , 25℃ 에 비하여 중배축장의 감소정도가 작았다.
