Polyphenism is one of the most intriguing phenomenon observed from various insect species. Polyphenism is changing morphological, behavioral, and physiological phenotypes without changing genome informations. Therefore, polyphenism is thought to be controlled by epigenetic methods. Epigenetic controls are including DNA methylation, histone acetylation, small RNAs, long non-coding RNAs, and protein phosphorylation. To investigate the mechanism involved in polyphenism, various genomic researches have been completed. However, metabolite changes in different phases have not been reported yet. Thus, we studied the metabolite changes and found the changed metabolites. These studies will make an important contribution to reveal the difference between gregrious and solitary phases and reveal eco - friendly control techniques.

To investigate unknown molecualr and cellular mechanisms underlying polyphenisms in the migratory locusts, we performed whole transcriptome analysis from 12 different groups. Significantly differentially expressed coding and long non-coding (LNC) RNAs were present according to habitats, phases, and genders. Those differentially expressed coding and LNC RNAs were confirmed by quantitative-realtime-reverse transcriptase PCR. Those identified coding and LNC RNAs could be a target for developing environmental friendly management tools in the near future.

가루깍지벌레(Pseudococcus comstocki)는 노린재목 가루깍지벌레과로 포도원에서 나무껍질 사이에 알, 약충 및 성충으로 월동하고, 봄철 포도나무 수액이 이동할 즈음 활동을 시작한다. 개체수가 증가하는 7월 중순에 이동을 시작해 결과지와 봉지를 씌운 포도송이 안에서 줄기를 흡즙하고 감로를 배설하여 그을음병을 유발한다, 7월 하순~8월 상순 경에 포도 과실 지경에 알을 낳으며, 포도 수확기인 9월 상순 무렵 약충이 증가되면 그을음이 늘어 포도 상품성을 떨어뜨린다. 유기포도원의 알렉산드리아 같은 청포도 계통의 포도에서는 피해 증상이 뚜렷이 나타나 농가피해가 더욱 심하다. 본 연구는 가루깍지벌레의 피해를 줄이고자 식물로부터 유래하는 기피물질을 이용해 시설포도원에 적용하고자 수행하였다. 선발된 essential oils을 6월부터 8월까지 포도송이 줄기와 만나는 T자 줄기에 털실을 감고 0∼100㎕ 처리한 결과 100㎕ 처리시 기피효과가 높았으며, 6월부터 8월까지 처리한 결과 7월 중․하순에 처리하였을 때 기피효과가 가장 높게 나타났다.

Ethyl formate (EF) and phosphine (PH3) is an alternative fumigant to methyl bromide (MB). The egg, nymph, and adult stages of Frankliniella occidentalis in asparagus were examined for the fumigation activity of EF and PH3. The eggs of F. occidentalis were more tolerance than other stages (adults and nymphs) in both EF and PH3. When the EF was treated for 4 hrs at 5℃ and 20℃, the eggs of F. occidentalis were LCT99=98.70 mg/L and LCT99=61.13 mg/L, and adults were LCT99=11.50 mg/L and LCT99=3.18 mg/L, respectively. However, at 5℃ and 20℃, the eggs were LCT99=83.76 mg/L and LCT99=53.6 mg/L, and adults were LCT99=4.58 mg/L and LCT99=3.44 mg/L, respectively for 4 hrs treatment of PH3. The PH3 was not any external phytotoxic damages at the maximum dose (4 mg/L), but the EF caused significant phytotoxicity in asparagus.

벗초파리(Drosophila suzukii)는 아시아가 원산인 파리목 초파리과의 해충이다. 국내에서의 피해사례는 보고되지 않았지만 북미와 유럽에서는 블루베리, 체리, 포도 등의 작물에 많은 피해를 입히는 중요한 해충으로, 알자스의 포도 수확량이 15% 정도 감소되었다는 피해사례가 보고되었다. 미국, 뉴질랜드 등에 수출을 할 때 검역 해충으로 수출포도단지 및 선과장 등에서의 발생이 민감한 해충이다. 본 연구는 효율적인 초파리 모니터링을 위해 식초의 종류, 식초량에 따른 유인량을 알아보고, 수출포도단지에서의 벗초파리 발생시기를 알아보고자 수행하였다. 사과 식초 30, 70, 100ml를 넣고 초파리 유인량을 비교한 결과 유인량에는 차이가 없었다. 양조식초, 현미식초, 사과식초, 백포도식초, 사과식초+와인(5:5) 처리 결과 사과식초+와인(5:5)에서 유인량이 가장 많았다. 수출포도단지 내 재배포장에서 조사한 결과 노랑초파리는 8월에도 발견되었으나, 벗초파리는 9월 중순부터 발견되었다.

Pseudococcus orchidicola and Pseudococcus longispinus, a notorious cosmopolitan pest species of mealybugs, known to be distributed indoors in Korea. The effect of temperature on the development of P. orchidicola and P. longispinus was studied. The duration of nymphal period of the two mealybugs at 24 and 28℃ have been recorded. Developmental duration of P. longispinus was longer than P. orchidicola in both females and males. The adult longevity was estimated to 74.9 and 91.1 days at 24℃ for P. orchidicola and P. longispinus, respectively. The average fecundity per female was greater at 28℃. The survival of P. orchidicola and P. longispinus during winter at outdoor was failed.

Ethyl formate (EF) and phosphine (PH3) is an alternative fumigant to methyl bromide (MB). For applying import sweet pumpkins in Korea, the efficacy of EF and PH3 was evaluated on Tetranychus urticae adult and egg stage which is pest in sweet pumpkins. The eggs of T. urticae were more tolerance than adults in both EF and PH3. When T. urticae eggs were treated with EF for 4 hrs, LCT99=107.63 mg/L and LCT99=45.37 mg/L at 5°C and 20°C, respectively. PH3 treatment at 5°C and 20°C for 24 hrs, eggs were controlled LCT99=49.44 mg/L and LCT99=17.23 mg/L, respectively. The mixed treatment of EF and PH3 showed no significant synergistic effect on T. urticae. However, EF (80 mg/L) and PH3 (4 mg/L) treatment did not cause any external phytotoxicity damages in sweet pumpkin even when treated with the maximum amount of fumigant at 5℃ and 20℃ for 24hrs.

The bean bug, Riptortus pedestris, is a pest of leguminous crops. Recent studies have revealed an interesting relationship between R. pedestris and a gut symbiont Burkholderia sp.. R. pedestris mainly acquires Burkholderia sp. during 2nd instar period, which leads to enhancement of growth and reproductive rate of the host. Nevertheless, how R. pedestris acquires its symbiont is not yet clearly described. Therefore, we investigated whether 2nd instar nymphs have preference on Burkholderia sp. compared to untreated control in laboratory settings. In the choice test, no significant difference was found in the preference by R. pedestris between the two treatments. To further investigate single individual’s behaviors (its first choice, approaching frequency to each treatment, and residence time on the treatment). We video-recorded single individuals in the choice setting for 12 hours. Again, there was no significant difference in any of the behavioral traits recorded. Further choice tests will be conducted in the Y-tube olfactometer systems and semi-field conditions.

본 연구는 최근 소비자의 소비패턴 변화로 인체에 무해한 친환경 농산물 요구도가 증가하고 있고 천적을 이용한 해충방제를 통한 안전농산물 생산과 시설토마토에 발생하는 주요해충의 천적을 이용한 방제기술 개발을 통한 천적시장 활성화에 목표를 두고 진행되었다. 춘천지역의 농가에서 반촉성재배(2~6월)시기에 트랩과 육안조사를 통해 해충발생과 천적 등을 이용한 방제효과를 조사 하였으며, 농가에서 주로 사용하는 약제를 대상으로 천적에 대한 저독성 약제와 유기농자재의 선발시험도 병행하였다. 그 결과 반촉성재배시 발생되는 주요 해충으로는 온실가루 이와 총채벌레이었으며 온실가루이의 경우 4월 중순경 발생되어 5월 중순이후 발생이 증가하였고 총채벌레는 4월 상순경에 발생되어 6월 이후에 증가되는 경향을 보였다. 또한 비교적 천적에 저독성 약제인 스피로메시펜액상수화 제, 피리프록시펜유제 및 유기농자재 등도 선발할 수 있었다. 따라서 시설채소 반촉성재배시에 효과적인 해충의 관리방안으로 발생초기에 천적인 온실가루이좀벌과 미끌애꽃노린재 등을 활용하고 해충의 발생증가 시기에는 천적에 저독성인 약제와 유기농자재 등을 활용하여 관리할 수 있는 모델을 작성하였다.

In insects, serine proteases are involved in a variety of physiological processes including digestion, development, and immunity. Bombyx mori serine protease homolog BmSPH-1 regulates nodule melanization and is recruited into nodules from the hemolymph by B. mori lipopolysaccharide-binding protein. Here, we show the dual role of BmSPH-1 in development and immunity of B. mori. BmSPH-1 was expressed in the hemocytes during larval-pupal transformation and localized to the cuticle of silkworms, which indicates that BmSPH-1 is secreted from hemocytes and then transported to the cuticle via the hemolymph. BmSPH-1 was proteolytically activated in the cuticle during larval-pupal transformation and the early pupal stage. BmSPH-1 RNAi resulted in the arrest of larval-pupal transformation and pupal cuticular melanization. Furthermore, the expression of BmSPH-1 was up-regulated in the hemocytes during infection. Taken together, we found that BmSPH-1 is involved in larval-pupal transformation and pupal cuticular melanization as well as the innate immunity of silkworms, which indicates that BmSPH-1 is responsible for either development or immunity.

Frankliniella intonsa (Trybom) (Thysanoptera: Thripididae) is an important pest of field and greenhouse crops such as pepper and strawberry. Chemical attractants are commonly used for monitoring thrips, but few are available for F. intonsa. Thus, we evaluated three attractants of Frankliniella occidentalis known from literatures for their attraction of F. intonsa in Y-tube olfactometer. Fed unmated adult females of F. intonsa showed higher initial response to (S)-(−)-verbenone than other attractants, i.e. aggregation pheromone of F. occidentalis and methyl isonicotinate. Unfed mated adult females of F. intonsa showed similar initial response. These results suggest that (S)-(−)-verbenone also can be used for monitoring and further in behavioral control tactics of F. intonsa.

Insect immulectins are involved in various aspects of the innate immunity, including encapsulation, melanization, and phagocytosis. Although the silkworm Bombyx mori immulectin (BmIML) has been reported previously, the ligand and functions of BmIML have not been investigated. Here, we show the dual roles of BmIML in cuticular melanization and immunity of B. mori. BmIML recognizes carbohydrates in a Ca2+-dependent manner and binds to Gram-negative and Gram-positive bacteria, fungi, and yeast. BmIML was expressed in the fat body during infections and localized to the hemocytes of silkworms. Additionally, BmIML was expressed in the epidermis during the prepupal stage and localized to the cuticle of silkworms. After treatment with E. coli, dopa, dopamine, or tyrosine injections, BmIML production was induced in the fat body but not in the epidermis of silkworms. Treatment with BmIML RNAi resulted in the arrest of pupal cuticular melanization. Therefore, we concluded that BmIML is involved in pupal cuticular melanization and innate immunity responses of silkworms, suggesting dual roles for BmIML.

Estimating compensation payment for environmental damages by chemical accidents has been controversial because accurate quantification of damages of chemical accident is so complicated and there is lack of scientific studies about impact of chemical accidents. In this study, for supporting chemical accidents compensation criteria, the 7 days acute and 28 days chronic toxicity test was conducted with 4 major chemical accident substances (Sulfuric acid, Nitric acid, Methanol and Methylethylketone) according to the OECD 232 guidelines. Methylethylketone was most toxic chemical followed by methanol, nitric acid and surfuric acid. Further study of comparison between pH and chemical effects was needed.

방충포장 제조용 친환경 기피물질 조사와 적용 가능성 검증을 위하여 식물에서 발현되는 2차대사물질을 함유하는 식품공전에 등재된 식용, 약용식물 선정을 위한 후보군 84종을 조사하였다. 이것을 토대로 저장해충에 방충 및 기피효과를 보인다고 알려진 몇 종의 한약재를 선발한 후 추출물을 이용, 대표적 저장해충인 화랑곡나방과 어리쌀바구 미에 대한 기피효과를 조사하였다. 대상해충에 대한 기피효과의 적합한 분석을 위하여 기피물질 후보군의 추출방법과 물질농축도 등을 다르게 하여 한약재로부터 물질을 추출한 후 기피물질을 확보하였으며, 기피효과 시험은 여지확산법 을 이용하였다. 선정된 기피물질에 대한 기피효과는 화랑곡나방에서는 뚜렷하지 않았으나 어리쌀바구미에서는 7.1~69.3%의 기피효과를 나타냈다. 이러한 결과는 동일한 물질이라 하여도 해충 종에 따라 기피효과가 달라질 수 있음을 보이는 것이다.

To assess the temperature change effects on Collembola communities, we selected 9 Korean mountains in different latitudes. Top soil was collected and Collembola extracted from 3 sites in different elevation in each mountain. Extracted Collembola were sorted to genera and this data were summarized as total abundance, family richness and diversity. Additionally, soil temperature in each plot, physical and chemical properties of the soil was investigated and that correlated with Collembola community data. Average soil temperature and soil properties affected patterns in Collembola abundance and family richness. The results indicate that temperature change and soil properties strongly affected Collembola communities. Therefore, Climate change has high potential to affect Collembola communities.

Due to their environmental and economical consequences, invasive species have become a major concern worldwide. Among them, the yellow-legged hornet, Vespa velutina, is a keen hunter of domestic honeybees. They use olfactory cues from the prey to assess food information. In this study, we investigated the attraction behavior of V. velutina to honeybee pheromone under outdoor conditions. Nine honeybee pheromones were tested and compared in order to find the best attractant. When testing specific compounds, the honeybee queen pheromone, homovanillyl alcohol, proved highly attractive.

식품 제조업체의 해충 저감화를 위한 방안으로 과자류와 캔디류에서의 곤충관련 이물 클레임 발생 상황을 주요 식품 제조업체를 선정, 2010년부터 2016년 까지 약 7년간 1234건의 곤충관련 이물 클레임을 현물, 사진자료 등을 통하여 조사하였다. 비스켓과 스넥 등의 과자류 370건에서는 딱정벌레목의 비율이 25%로 가장 높았으며, 화랑곡나방 을 포함하는 저장해충이 30%를 차지하였고, 곤충이물 발생유형은 치사성충이 49%를 차지하였다. 클레임 발생원인은 유통, 소비자 단계에서 55%로 가장 높게 조사되었다. 또한 캔디, 쵸코릿, 캐러멜 등을 포함하는 캔디류 72건에서는 나비목이 46%로 가장 높았으며, 화랑곡나방과 어리쌀바구미 등의 저장해충이 56%, 곤충이물 발생유형은 치사성충이 31%로 높게 조사되었으며, 클레임 발생원인은 유통, 소비자 단계에서 79%로 해충 오염 발생 비율이 가장 높게 나타났다.

멸강나방 (Mythimna separata) 유충 및 번데기로부터 채집한 긴등기생파리의 알부터 성충우화까지 발육 기간을 담배거세미나방 (Spodoptera litura) 5∼6령 유충을 숙주 곤충으로 하여 7개 항온조건 (16, 19, 22, 25, 28, 31, 34±1℃)에서 조사하였다. 알부터 번데기까지 발육 기간을 보면 16℃ (23.4일)에서 가장 길었고 34℃ (8.3일)에서 가장 짧았으나, 번데기부터 성충까지 발육기간은 28℃ (7.3일)에서 가장 짧았다. 선형 발육율 모델을 이용하여 추정한 알부터 성충우화 까지 발육영점온도와 유효적산온도는 각각 7.8℃, 370.4DD였다. 4개 비선형 발육율 모델 (Briere 1, Lactin 2, Logan 6, Performance) 중에서는 Briere 1 모델 ( = 0.96)이 가장 높은 해석력을 보여주었다. 동일 연령 집단의 발육완료 분포를 설명하기 위해 사용된 3개 모델 (2-parameter Weibull, 3-parameter-Weibull, Sigmoid)은 모두 같은 결정력 ( = 0.90)을 보였다.

The frequency and time of the outbreak of insect diseases were investigated among the insect-rearing farms by regional groups. The purpose of this study was to predict the insect diseases and reduce the spread of diseases. From February to October 2016, 87 of diseased insects were collected in seven regions. It turned out that the pathogens for fungal diseases were identified as Metarhizium anisopliae. But in case of bacterial diseases, the pathogenicity of the bacteria isolated from the diseased insects were not confirmed. Bacterial diseases occurred mainly in June and August in Gyeonggi-do and Gangwon-do, while in Chungcheong-do, bacterial and viral disease raged. In Jeolla-do, both fungal and bacterial diseases occurred in June. Generally, bacterial diseases occurred mainly from June to October, while the frugal diseases occurred from January to March. It was concluded that the entomopathogenic fungi, Metarhizium anisopliae, tended to cause diseases during late-winter and early spring and bacteria, uncertain, was prone to cause diseases during summer. Therefore, it necessary to adequately control depending on the type of the entomopathogen.

In the age of sexual maturity of queen, mating occurred at 6.7% immediately after eclosion and it was the highest as 85.0% at 10 days after emergence. However, a remarkable decrease was occurred at 20 days after the emergence. With regard to oviposition rate, the highest rate was observed as 81.3-81.8% at 6 to 10 days of eclosion. The rate of colony foundation and progeny-queen production were the highest as 43.8% and 37.5%, respectively, at 8 days of eclosion. In the age of sexual maturity of male, mating rate was as high as 38.3% immediately after the eclosion, the highest as 80.0% at 25 days of eclosion, and 76.6% at 8 days of eclosion. The oviposition rate was the highest as 76.9% at 6 days of eclosion, and decreased to 75.0% at 8 days and 72.7% at 10 days of eclosion. Summarized, our results indicate that sexual maturity for mating of B. terrestris is most favorable 6-8 days after eclosion for queen and 6-10 days after for male.
