        애긴노린재류(Nysius species)는 세계에 널리 퍼져 있는 해충으로 기주범위는 매우 넓으며 특히 국화과의 주요해충으 로 알려져 있다. 최근 종실 들깨에서 피해가 보고되고 있으며, 들깨를 수확할 때 종실과 같이 다량의 애긴노린재류가 함께 혼입되어 그 발생밀도가 매우 높음을 알 수 있다. 본 연구에서는 이러한 애긴노린재류가 들깨에 미치는 영향을 연구하고자 들깨 종실의 무게변화와 발아율을 조사하였다. 건전한 들깨(들샘) 종실에 애긴노린재류 성충 0, 5, 10, 15, 20, 25마리를 인위적으로 접종하여 3일과 5일 후 들깨 무게를 측정하였다. 측정결과 접종한 노린재 밀도가 5, 10, 15, 20, 25마리로 높아질수록 0마리일 때 보다 들깨 무게가 3일 접종 시에는 각각 2.6, 5.5, 9.0, 10.5, 11.0% 감소하였고, 5일 접종 시에는 각각 2.9, 9.9, 12.0, 17.6, 18.1% 감소하였다. 애긴노린재의 밀도가 높아질수록 종실의 무게가 감소폭은 컸으며, 3일 접종 때보다 5일 접종 때가 들깨의 무게는 더욱 많이 감소하였다. 또한 들깨무게가 감소함에 따라 발아율도 함께 떨어졌다. 노린재 흡즙에 의한 피해로 들깨 종실의 무게가 감소하였다는 것을 추정할 수 있으며, 이로 인해 들깨의 발아율이 떨어진 것으로 생각된다.

        Nysius is one of the most common and widely distributed species in the world. It is one of the emerging pests of perilla crop in the field in Korea. In this study, we investigated the oviposition preference and development of Nysius plebeius and N. hidakai. on five crops seed (Perilla, Sesame, Proso-millet, Sorghum, and Fox-tail millet) using both multiple choice and no-choice tests. Both results found perilla seeds to be the most suitable seeds for oviposition and development. Total developmental period was shortest on perilla seed, and the longest was on rest of crops seeds. The highest of adult emergence, longevity, and adult weight were occurred on the perilla seed. This results suggest that the higher preference and performance of Nysius spp on the perilla seed. The information may facilitate the exploration of resistant genetic materials chemical associated with seeds for successful breeding program in Korea.

        In this study, we examined the protective immunity of a combination of seven Brucella abortus recombinant proteins; superoxide dismutase (rSodC), riboflavin synthase subunit beta (rRibH), 50S ribosomal protein (50s rL7/L12), nucleoside diphosphate kinase (rNdk), malate dehydrogenase (rMDH), arginase (rRocF), and elongation factor (rTsf) cloned in a pMal vector system and expressed in DH5α. Mice groups were immunized thrice with a combined subunit vaccine (CSV-7) at 0, 2, and 5 weeks and subsequently challenged with B. abortus at 5 × 104 CFU at 6 weeks. At four weeks post-infection, the mice were sacrificed and the bacterial burden in their spleens was quantified. Results revealed bacterial log reductions of 0.63 and 0.34 in comparison to PBS and maltose-binding protein (MBP), respectively. Cytokine profiling revealed a marked increase in IFN-γ (interferon-gamma), MCP-1 (macrophage chemoattractant protein-1) and IL-6 (interleukin 6) cytokines at 5-weeks post-immunization. On the other hand, only TNF was heightened at 7-weeks post-immunization. In general, this cytokine profile is consistently reflective of a Th1 immune response, which is beneficial for host immunoresistance.

        This study evaluated the protective effects of a combination of eight B. abortus recombinant proteins that were cloned and expressed into a pMal vector system and DH5α: nucleoside diphosphate kinase (rNdk), 50S ribosomal protein (rL7/L12), malate dehydrogenase (rMDH), DNA starvation/stationary phase protection protein (rDps), elongation factor (rTsf), arginase (rRocF), superoxide dismutase (rSodC), and riboflavin synthase subunit beta (rRibH). The proteins were induced, purified, and administered intraperitoneally into BALB/c mice. The mice were immunized three times at weeks 0, 2, and 5 and then infected intraperitoneally (IP) with 5×104 CFU of virulent B. abortus 544 one week after the last immunization. The spleens were collected and the bacterial burden was evaluated at four weeks post-infection. The results showed that this combination produced a significant reduction of the bacterial burden in the spleen with a log reduction of 1.01 compared to the PBS group. Cytokine analysis revealed induction of the cell-mediated immune response in that TNF (tumor necrosis factor) and proinflammatory cytokines IL-6 (Interleukin 6) and MCP-1 (macrophage chemoattractant protein-1) were elevated significantly. In summary, vaccination with a combination of eight different proteins induced a significant protective effect indicative of a cell mediated immune response.

        최근 수목 병해충을 방제, 예방하기 위해 나무주사법이 많이 사용되고 있다. 나무주사법은 기상에 상관없이 약제처리가 가능하며, 약제사용량 감소와 천적등 비표적 생물에 대한 영향을 줄일 수 있다는 장점이 있다. 하지만 현재 국내에서 나무주사법은 산림 해충을 대상으로만 등록, 적용되어 있을 뿐, 과수에 등록된 제품은 없다. 과수에 나무주사법이 적용되기 위해서는 농약잔류와 약효에 대한 연구가 필요하다. 본 연구에서는 복숭아(천중도)에 Acetamiprid 10% SL을 나무주사 후, 잔류성과 약효를 조사하여 실용가능성을 확인하였다. 약제가 나무주사된 나무는 복숭아 벚나무깍지벌레(Pseudaulacaspis prunicola)에 대하여 방제효과가 있으나, 가지별로 불균일한 약효와 잔류성을 보여 향후 수형, 흉고직경 등을 고려한 나무주사제의 처리법 최적화에 대한 연구가 지속적으로 필요할 것으로 생각된다.

        Xyrosaris Meyrick, 1907 is a small genus of the family Yponomeutidae, established by Meyrick (1907) for its type species Xyrosaris dyropa Meyrick, 1907. A total of nine species were recognized in the world. Most of species of the genus have been described by Meyrick at the early 20th century. Up to date, X. lichneuta Meyrick, 1918 has been reported from Korea. In the present study, we review the genus Xyrosaris from Korea, with two new species: X. sp.1 and X. sp.2. Key to species, illustrations of adult, male and female genitalia, diagnosis and short description are provided.

        Nysius Dallas, 1852, is one of the most common and widely distributed genera under the superfamily Lygaeoidae. Species under this genus are hard to identify due to similarity of the species and variability of the coloration. The Nysius species were collected with the help of aspirator and plastic vile in the perilla crop fields in RDA, Miryang, Korea. Korean species of the Nysius were identified, and three species including a newly reported species N. inconspicuus were recognized. Morphological and genetic characteristics of species were illustrated, and a key to species of Korean Nysius was provided. The DNA barcoding information of N. plebeius and N. inconspicuus were recorded.

        The genus Meganola (Nolidae, Nolinae) was established by Dyar (1898), with the type species Meganola conspicua Dyar, 1898, from America (Type locality: Texas, Colorado, Arizona). This genus comprises 80 species described in the Palaearctic and Oriental region. In this study, genus Meganola Dyar is reviewed from Korea, with description of a new species. We redescribed 12 species and one new species. Illustrations of adults and genitalia of all Korean species are provided, with a key to the genus of Meganola based on the male genitalia.

        Callosobruchus chinensis (Coleoptera: Bruchidae) is a cosmopolitan pest of stored grain legumes. This study assessed the effects of short-term exposure to low-temperatures (4, 0, and -4℃) and seven durations of storage (1, 5, 10, 15, 20, 25, and 30 days) on the survival, development, and adult performance of C. chinensis in laboratory. The effects were studied on three life stages (egg, larva [2-3 instar], and pupa) of C. chinensis. The results showed that low-temperature have significant effects on the life variables of C. chinensis. Among the stages, egg and pupa stages were most sensitive to low-temperature. The very low survival rates of eggs and no pupa survived at –4℃ after 10 days storage. Cold exposure at -4℃ not only increased egg and pupa mortality, but the negative effects also transmitted to the larvae and pupa developed from these eggs. Increasing the duration of cold exposure further reduced survival rates. Low-temperature treatments have also affected on the adult performances such as adult weight. However, there were not any negative effects of low-temperatures on the seed germination. The life variables of C. chinensis are discussed in terms of targeting particular susceptibilities to low-temperatures in different storage duration as an alternative to chemical treatments.

        Antibiotic Detection Kit (Combination I), a lateral flow immunoassay (LFIA) developed for detecting antibiotic residues in milk, was utilized for the analysis of antibiotic residues in the muscle tissue of olive flounder. After 5-h treatment of samples by placing them in water dosed with sulfadimethoxine (SDM; 200 g/ton water), the residue depletion of SDM was investigated in 25 cultured olive flounders (Paralichthys olivaceus). Muscles from fish were sampled before treatment and on the 1st, 2nd, 3rd, 4th and 5th days after treatment. The concentration of SDM in the muscle was then determined by LFIA. The absorbance ratio of the sample to the control blank (Bs/Bo) was employed as an index to determine the residue in olive flounder muscle. To investigate the recovery rate, standard solutions were added to muscle samples to obtain final concentrations of 25 and 50 ng/mL in the muscle. The recovery rates of all spiked samples were >96.6% of the spiked value. SDM was detectable in the muscle of fish treated with the drug until the 1st day of the withdrawal period. The present study shows that the LFIA can be easily adopted to detect SDM residues in the tissue of farmed fish.

        This study investigated the antibacterial effects of Galla rhois extract (GRE) against Campylobacter jejuni and Campylobacter coli. The minimum inhibitory concentrations (MICs) of GRE against C. jejuni and C. coli were 0.28 and 0.55 mg/mL, respectively, and the corresponding minimum bactericidal concentrations (MBCs) were 4.4 and 5.5 mg/mL. C. jejuni treated with the MIC, MBC or 2×MBC of GRE showed significant inhibition of growth compared with that of the control group during the incubation period, and no viable bacteria were detected at 24 h after incubation. C. coli treated with MIC, MBC or 2×MBC of GRE also showed inhibition of growth compared with that of the control group during the incubation period, and in the C. coli cultures treated with MBC and 2×MBC of GRE, no viable bacteria were detected at 24 h after incubation. In conclusion, GRE is a candidate antibacterial agent against C. jejuni and C. coli, and may have applications for the control of Campylobacter infection in poultry.

        To date, most serodiagnostic methods for brucellosis screening are based on antibodies against lipopolysaccharides of Brucella spp. However, this approach has the drawback of yielding false-positive results due to cross-reactivity with lipopolysaccharides of other related pathogens, especially Yersinia enterocolitica O:9. In this study, Brucella abortus AspC was cloned and expressed by PCR amplification into a pCold TF expression system to obtain recombinant AspC (rAspC). The immunogenicity of rAspC was confirmed by western blotting of Brucella-positive bovine serum. rAspC-based ELISA was performed to determine whether rAspC could be used in the serodiagnosis of bovine brucellosis. rAspC reacted strongly with anti-Brucella antibodies in positive sera in the tube agglutination test (TAT), but did not show strong reaction with most negative samples. In particular, the average OD492 value at the highest TAT titer showed a 1.4-fold increase with respect to the cutoff value. The accuracy, specificity, and sensitivity of rAspC were 71.88%, 78.33%, and 68%, respectively. These findings suggest that rAspC might be valuable for the serological diagnosis of bovine brucellosis.

        Brucellosis is an important and re-emerging zoonotic disease worldwide. The prevention of human infection is achieved predominantly through the control of brucellosis in agricultural animals, which in turn depends on accurate diagnosis and vaccination. However, conventional serological diagnosis of brucellosis has several limitations, and currently available vaccines for animals have several drawbacks, including the ability to cause infection in humans. Phosphoglycerate kinase (Pgk) is one of the specific proteins reactive with mouse sera in the early stage of Brucella infection, and deletion of the pgk gene in B. abortus strain 2308 resulted in extreme attenuation of this strain in vitro and in vivo. Furthermore, the B. abortus pgk mutant has been used as a live vaccine, and in challenge experiments, it induced protection that was superior to that conferred by commercial strains. In this study, the pgk gene from Brucella abortus 544 was successfully amplified and cloned into a maltose binding protein fusion protein expression vector (pMAL). The recombinant protein was expressed in Escherichia coli DH5α and purified. The immunogenicity of purified recombinant B. abortus 544 Pgk (rPgk) was evaluated by western blot analysis using Brucella-positive mouse sera. rPgk could be used as an antigenic component for future serological tests and potential vaccine development.

        본 연구는 오배자 에탄올 추출물 (GRE), 염소산나트륨 (SC) 그리고 오배자 에탄올 추출물과 염소산나트륨 합제(GS)의 B. abortus에 대한 항균효과를 확인하기 위해 수행 되었다. GRE, SC 그리고 GS를 B. abortus에 처리하여 배양한 후, B. abortus의 생존수를 확인하였으며, 마우스 탐식세포 내 감염된 B. abortus의 증식 억제효과를 경시별 (2, 24, 48시간)로 조사하였다. GRE, SC 그리고 GS는 각각 400 μg/mL 이하, 15 mM 그리고 0.6GS (GS 1, GRE 1,000 μg/mL + SC 30 mM) 이하의 농도에서 세포독성을 나타나지 않았다. 모든 처리구에서 B. abortus의 생존율은 용량- 의존적으로 현저하게 감소하는 결과를 나타내었다. 또한, GRE (400 μg/mL), SC (15 mM) 그리고 0.5GS (GRE 500 μg/mL + SC 15 mM)를 처리한 세포에서 배양 48시간 후에, B. abortus의 증식이 통계적으로 유의성 있게 감소하였으며 (GRE, p < 0.01; SC and 0.5GS, p < 0.001), 특히, GS를 처리한 경우, B. abortus의 세포내 증식이 GRE와 SC의 상승작용에 의한 강력한 항균효과를 나타내었다. 결론적으로, GS는 B. abortus에 대한 항균물질로서 유용할 뿐만 아니라, 식육과 우유 위생 분야에 적용할 수 있을 것으로 생각된다.

        Brucellosis is a notorious zoonotic disease with global implications. Efforts to control the spread of the disease have been restricted to the agricultural livestock. Increasing incidences of accidental human infection have motivated researches to start working on alternative vaccines. At present, live attenuated vaccines are the only accepted type of vaccines used in developed countries for the prevention of brucellosis. Although serodiagnosis is occasionally unreliable, some countries have already claimed to have eradicated the disease, based on this testing. Live attenuated vaccines are not suitable for use in pregnant and immune-depressed animals. Moreover, these vaccines are not tolerated in humans. Therefore, many researches have been striving to discover alternative methods of vaccination. Most research has focused on the generation of subcellular, subunit, and DNA vaccines that are as efficient as the live attenuated vaccines. At present, none of the available vaccines has been able to replace the live attenuated vaccines. Therefore, additional research is necessary in order to discover a new brucellosis vaccine that is suitable for human use.

        This study investigated the therapeutic effects of Galla rhois (GR) ethanol extract (GRE), sodium chlorate (SC), and a combination of GRE and SC on mice infected with Brucella abortus (B. abortus). Mice were infected intraperitoneally with B. abortus and then treated with GRE, SC, and a combination GRE and SC in drinking water for 14 days. Then, serum antibodies were used in a tube agglutination test (TAT), after which the weight and CFUs from each spleen were measured. In addition, histopathological changes in each liver were examined at 14 days post-infection. At 14 days post-infection, negative reactions of serum antibodies in PC (positive control), SCT (SC 1.6 g/L drinking water), GRT (GRE 200 mg/L drinking water), and GST (GRE 200 mg + SC 1.6 g/L drinking water) were 0, 40, 60, and 80%, respectively. The average spleen weight was not significantly different between the groups. At 14 days post-infection, bacterial numbers in all treated groups were significantly lower compared to to that of the PC (GRT and SCT, P<0.05; GST, P<0.001). In terms of histopathological changes in the livers, there were numerous multifocal microgranulomas in the PC, whereas this number successively decreased in the SCT, GRT, and GST groups. Conclusively, a combination of GRE and SC exhibits therapeutic effects on mice infected with B. abortus. These results suggest the potential efficacy of a mixture of GRE and SC in the treatment of brucellosis.

        Brucellosis is an important bacterial zoonotic infection worldwide responsible for economic losses in livestock industries which represents a considerable and increasing public health burden. The disease is caused by Brucella which possess the ability to invade and replicate within phagocytes and may result to chronic infection, leading to difficulty in medical therapy of the disease. The treatment for brucellosis employs conventional principles that have been applied for a long time and a combination of antibiotics is currently used due to low efficacy of monotherapies. However, vital effects with respect to health and safety are neglected. Nevertheless, the preventive methods and treatments for brucellosis using traditional medicine have not yet been thoroughly studied, hence, alternative therapies such as the use of natural plant extracts as traditional medicine that are safe, efficient and economical should be explored in order to identify candidates that eliminate complications due to brucellosis. Coptis chinensis Franch (Huanglian) is a traditional Chinese medicinal herb and its extract has been known to possess strong antibacterial activity. In this study, the antibacterial effects of C. chinensis Franch extracts, and the major components of the herb namely berberine and palmatine, were investigated on B. abortus. The C. chinensis Franch ethanol extracts (CCFE) showed bactericidal effects at 1,000 μg/ml concentration and berberine at 100 μg/ml concentration. However, C. chinensis Franch and its components did not affect invasion and intracellular growth of B. abortus in RAW 264.7 cells. In conclusion, C. chinensis Franch ethanol extracts, water extract and its major components such as berberine and palmatine would be a beneficial antimicrobial agent without affecting phagocytic pathway within macrophages, and further study for the precise mechanisms of antibacterial effect would be necessary.

        Brucella spp. are facultative intracellular pathogens that have the ability to survive and multiply in professional and nonprofessional phagocytes and cause abortion in domestic animals and undulant fever in humans. Brucella species can survive in a variety of cells, including macrophages and their virulence and chronic infections are thought to be due to their ability to avoid the killing mechanisms within macrophages. Inhibition of phagosome-lysosome fusion has been proposed as a mechanism for intracellular survival of Brucella in professional and nonprofessional phagocytes. Toll-like receptors (TLRs) are part of a skillful system for detection of invasion by microbial pathogens. Recognition of microbial components by TLRs triggers signaling pathways that promote expression of genes and regulate innate immune responses. Recent studies for the interaction between TLRs-Brucella have indicated the importance of control of Brucella infection. Here, we review selected aspects of TLRs-Brucella interaction, which may be helpful to understanding the mechanism of Brucella pathogenesis.
