This study evaluated the physicochemical and microbial quality characteristic of seasonal commercial kimchi for hygienic safety levels. The pH of seasonal commercial kimchi was 3.84-6.36 and the titratable acidity and salinity of the samples were 0.21-1.16 and 1.19-1.54%, respectively. The content of nitrate and nitrite in the commercial kimchi were lower in the spring and summer, which was affected by acidic condition of the kimchi depending on fermentation. Heavy metal contents in commercial kimchi are not an issue because they were detected only at very low levels. The total aerobic bacteria and coliforms counts ranged from 5.25 to 8.44 Log CFU/g and 0.00 to 5.08 Log CFU/g, respectively. The total aerobic bacteria and coliforms were detected more in summer than in the other seasons. E. coli was detected in three of the samples tested. Food-borne pathogens were not detected in any of the samples except for B. cereus. B. cereus was detected in the fall in more than 70% of samples. These results suggest that commercial kimchi distributed in the fall maintain the quality properties and the microbiological safety of kimchi compared to the other seasons. Therefore, further studies as an effective distribution system for the particular seasons will be needed to guarantee the hygienic safety levels of commercial kimchi required by the consumers.
본 연구에서는 김치 제조시 철저한 재료 세척의 필요성을 확인하고, 더불어 비가열 식품인 김치의 위생 안전성 실태를 파악하여 효율적 미생물 저감화 방안을 마련하고자 국내에서 유통되는 김치 원·부재료 200건의 세척 전과 후의 비교분석과 시판 김치 100건에 대하여 미생물 분석을 실시하였다. 김치 원·부재료 및 완제품에 대하여 일반 세균수, 대장균 및 대장균군을 모니터링 하였으며, 병원성 미생물 9종(Bacillus cereus, Clostridium perfringens 정성·정 량, Staphylococcus aureus, Enterohemorrhagic Escherichia coli, Campylobacter jejuni/coli, Yersinia enterocolitica, Salmonella spp., Vibrio parahaemolyticus, Listeria monocytogenes 정성)을 분석 하였다. 모니터링 결과, 김치 원· 부재료 및 완제품에서 일반세균은 1.4~9.0 log CFU/g 수준 으로 확인되었으며, 원·부재료 중 생강에서 8.8 log CFU/g, 완제품 중 총각김치에서 9.0 log CFU/g으로 일반세균이 가장 높게 나타났다. 대장균군은 0.5~7.3 log CFU/g으로 확 인되었으며, 생강에서 7.3 log CFU/g으로 가장 높게 나타 났고, 병원성 미생물 9종의 분석결과, Bacillus cereus를 제 외한 나머지 8종의 식중독균은 모든 시료에서 검출되지 않았다. 김치 제조 시 사용되는 김치 원·부재료들의 세척 공정 후 미생물 수는 일반세균수 0.2~3.2 log CFU/g, 대장 균군 0.3~2.7 log CFU/g, Bacillus cereus 1.0~3.9 log CFU/g 감소하였다. 따라서, 김치 원·부재료의 세척 공정으로 미생물 오염도를 감소 시켰으며, 이 결과를 통하여 김치 완 제품의 위생 및 저장성 증진에 기여 할 수 있을 것으로 확인되었다.
MEN1 환자에서 췌장 NET는 30-80% 동반된다. 증상 및 악성 가능성이 있는 경우 수술적 절제가 예후에 중요한 영향을 미친다. 수술이 불가능한 췌장 NET에 대해 EUS-EI를 고 려할 때는 합병증과 불완전 치료 및 재발 가능성에 대한 주 의 깊은 고려가 필요하다.
연구에서는, 전기방사법을 이용하여 산화철-산화그래핀(Fe3O4/GO, metallic graphene oxide; MGO)이 도입된 PVdF/MGO 복합나노섬유(PMG)를 제조하였으며, 이를 활용하여 비소제거에 대한 특성 평가를 진행하였다. MGO의 경우 In-situ-wet chemical 방법으로 제조하였으며, FT-IR, XRD분석을 진행하여, 형태와 구조를 확인하였다. 나노섬유 분리막의 기 계적 강도 개선을 위하여 열처리과정을 진행하였으며, 제조된 분리막의 우수한 기계적 강도 개선 효과를 확인할 수 있었다. 그러나, PMG 막의 경우, 도입된 MGO의 함량이 증가할수록 기계적 강도가 감소되는 경향성을 보여주었으며, 기공크기 분석 결과로부터, 0.3~0.45 μm의 기공크기를 가진 다공성 분리막이 제조되었음을 확인할 수 있었다. 수처리용 분리막으로의 활용 가능성 조사를 위해, 수투과도 분석을 실시하였다. 특히, PMG2.0 샘플의 경우 0.3 bar 조건에서, PVdF 나노섬유막(91 kg/m2h)에 비해 약 70% 향상된 결과값(153 kg/m2h)을 나타내었다. 또한, 비소 흡착실험 결과로부터, PMG 막의 경우, 비소3 가와 5가에 최대 81%, 68%의 높은 제거율을 보여주었으며, 흡착등온선 분석으로부터, 제조된 PMG 막의 경우 비소3가, 5가 모두 Freundlich 흡착거동을 따른다는 것을 확인하였다. 위 모든 결과로부터, PVdF/MGO 복합 나노섬유 분리막은 비소제거 및 수처리용 분리막으로 충분히 활용할 수 있을 것으로 판단된다.
Multiple starters consisting of two Bacillus amyloliquefaciens strains (MJ1-4 and EMD17), Pichiafarinosa SY80, and Rhizopus oryzae were used for Doenjang making. Bacillus strains were selected based on their abilities to inhibit toxinogenic fungi and Bacillus cereus, fibrinolytic activity, and their ability to confer good flavor to Cheonggukjang. P. farinosa SY80 and R. oryzae, previously isolated from soy sauce, were selected because they were not inhibited by two bacilli. Doenjang was prepared by inoculation of multiple starters (A1 Doenjang). Control Doenjang was prepared by inoculation of B. subtilis KACC 16750 (Natto strain) and Aspergillus oryzae KCCM 60166 (A2 Doenjang). Another control (A3 Doenjang) was prepared by inoculation of microorganisms present in rice straw. Doenjang samples were fermented for 70 days at 20℃. pH of 3 samples decreased from the initial value of 6.4 to 5.8~6.0 and titratable acidity (TA) increased from 0.6 to 1.1~1.3. The amount of amino-type nitrogen increased during fermentation. There were slight differences in moisture, crude-protein, and crude-fat contents after 70 days. Contamination of fungi was observed only in A3 Doenjang and B. cereus was not detected from all 3 samples. A1 Doenjang showed the highest fibrinolytic activity and A2 Doenjang the second. These results indicate that Doenjang made with carefully selected starters was functionally improved and microbially more safe.
본 연구는 전통대두발효식품에서 분리한 Bacillus amyloliquefaciens EMD17, HCD2, 그리고 MJ1-43 균주들과 B. subtilis W42 균주의 혈전용해능과 식염저항성을 조사함으로써 대두발효식품 제조에 종균으로 유용한 균주들을 확보하는 것이다. EMD17과 HCD2는 본 연구를 통해 B. amyloliquefaciens 균주들로 동정되었고 우수한 혈전용해능을 지님이 4 종류의 배지에서의 배양을 통해 확인되었다. B. amyloliquefaciens MJ1-4와 B. subtilis W42 균주들은 각각 항진균력과 항균력이 우수한 균주들로 보고된 바 있으나 혈전용해능은 알려지지 않았다. 본 연구를 통해 이들 2 균주도 혈전용해능을 지님이 확인되었다. 항균력과 혈전용해능이 우수한 바실러스 균주들을 종균으로 사용하는 것은 비단 식중독균 오염을 줄여줄 뿐 아니라 혈전용해능과 같은 기능성도 높여줌으로써 궁극적으로 고품질의 대두발효식품 제조에 기여할 것이다.
Oral squamous cell carcinoma is the 1st most common malignancy in oral and maxillofacial area. HPV 16 has been strongly linked to progression of cervical carcinoma. E6 and E7 as a small DNA virus encoding two major oncoproteins of HPV 16 can act together to produce efficient immortalization of primary human epithelial cells. Thus it is important to pursue the development of Immortalized human oral keratinocyte(IHOK) culture model which could be related to the pathogenesis of oral squamous cell carcinoma. If we establish IHOK transfected by E6/E7 genes, IHOK will be accepted as a model system for HPV-linked oral carcinogenesis. The purpose of this study were to culture primary normal human oral keratinocyte(NHOK), and to establish IHOK for studying oral carcinogenesis in the future. NHOK was primarily cultured under normal culture condition, and transformed into IHOK by transfection of E6/E7 genes. After 100 passages depend on Ca++ condition, cultured IHOK was confirmed by growth curve, cornified cell envelope measurement, TGase 1activity, mRNA detection, tumorogenecity and anchorage independence assay. After 100 passages, cultured IHOK showed most basal cell and monolayer of polyhedral cells under 0.15mM Ca++, and small area of stratification and flattened epithelial cells with irregular border under 1.2mM Ca++. The cultured IHOK showed relatively resistant growth under high calcium condition. The E6/E7 mRNA was detected in cultured IHOK by RT-PCR. During the terminal differentiation in cultured IHOK, increased insoluble cornified cell envelope formation was accompanied with induction of TGase 1 activity. But the cultured IHOK showed less CEM and TGase 1 activity than those of cultured NHOK. Cultured IHOK showed non-tumorogenecity, but slight anchorage independence. We had developed a technique to transform NHOK into IHOK by transfection of E6/E7 genes. Cultured IHOK was established as intermediate stage cell to study the pathogenesis of human oral squamous cell carcinoma.
The purpose of this study was to investigate the motivation, healing purpose, healing process and healing effect of forest through experiential essays. These essays of 34 forest experience were collected from 2014 to 2015 at Forestry Administration Office. Five steps of analysis were used including self-observation, self-description, selfdiscovery, equality and differentiation, and verification. The results of the study are as follows. First, there were psychological instability factors such as depression, bipolar disorder, helplessness, social phobia, and social maladjustment, and these factors were related to physical illness or family history of patients. Second, the process of self-discovering by self-observation and self-description in forest experience was expressed, and this experience is interpreted to strongly support the effects of various forest healing proved empirically in previous studies. Third, the patients experienced similar sufferings such as depression, anxiety, and frustration, but they described as experiencing the process optimized for each unique characteristic in the motivation, necessity, accessibility, and program utilization of each experience. Ultimately, the participants felt positive change in senses and emotions such as refreshing, comfort, relaxation, and euphoria through the natural vitality of forests that is inhaled through the five senses. They learnt to reflect on themselves and shame, and they could rediscover the value and meaning of life while meaningfully recognizing the problems attributed to them.
Background: This study represents the first report that the anti-obesity activity of ethanol extracts of Aronia melanocarpa can be enhanced through ultrasonification at a frequency of 120 ㎑ at 60℃ (UE).
Methods and Results: The amounts of cyanidin-3-O-galactose (cya-gal), a major anthocyanin in A. melanocarpa were higher by up to 402.4 ㎎/100 g, as compared with 221.4 ㎎/100 g and 322.1 ㎎/100 g, for hot water at 100℃ and 70% ethanol at 80℃ respectively. This result should cause the higher antioxidant activities of the UE than extract of hot water and ethanol in DPPH free radical scavenging. It was confirmed that the high antioxidant activity of UE could play an important role in inhibiting the production of proteins related to adipocyte differentiation, such as peroxisome proliferator activated receptor-γ (PPAR-γ) and sterol regulatory element binding protein 1 (SREBP1).
Conclusions: Ultrasonification at a frequency of 120 ㎑ at 60℃ should result in better anti-obesity activity than that observed using other processes. It was also observed for the first time that the anti-obesity activity of A. melanocarpa was associated with its antioxidant activity, possibly due to the higher elution of intact cya-gal, owing to efficient low temperature ultrasonification extraction. These results could also be applied to improve other biological activities of medicinal herbs that contain many types of heat-labile bioactive substances.
