Haemaphysalis longicornis는 사람과 동물에게 여러 심각한 병원체를 전달하는 주요 매개체로, 한반도에 널리 분포하고 있다. H. longicornis는 Rickettsia spp., Borrelia spp., Francisella spp., Coxiella spp., 그리고 중증열성혈소판 감소증후군 바이러스 (SFTS virus) 등을 매개하는 것으로 알려져 있다. 국내에 서식하는 H. longicornis의 미생물 군집과 관련된 연구는 많이 진행되지 않은 것으로 확인되었다. 이 연구는 한반도 내 다양한 지역에서 채집된 H. longicornis의 미생물군집 다양성을 지역별, 성장 단계 및 성별에 따라 분석하였다. 2019년 6월부터 7월까지 질병관리청 권역별기후변화매개체감시거점센터 16개 지역에서 채집한 H. longicornis의 16S rRNA 유전자 V3-V4 영역을 PCR로 증폭 후 Illumina MiSeq 플랫폼으로 시퀀싱하였다. Qiime2를 활용한 미생물 다양성 분석을 통해 총 46개의 샘플에서 1,754,418개의 non-chimeric reads를 얻었으며, 평균 126개 의 operating taxonmic unit (OTU) 을 식별하여 총 1,398개의 OTU를 확인하였다. 대부분의 지역에서 Coxiella spp.가 우점종으로 나타났으며, 특히 Coxiella endosymbiont는 가장 높은 우점도를 보이며, Coxiella burnetii와 계통 발생 학적으로 유사한 것으로 확인되었다. 이 연구를 통해 분석된 결과는 각 지역의 H. longicornis 미생물군집 데이터 베이스 구축에 활용되었으며, 이를 통해 지역별 미생물군집의 특이성을 식별할 수 있게 하였다. 이는 한반도의 H. longicornis에 의한 질병 전파 연구와 이를 통한 공중보건 개선에 기여할 것으로 기대된다.
한강 3개 보 구간 수변부 11개 지점을 대상으로 2023년 총 2회 채집된 저서성 대형무척추동물은 총 100종 평균 426.0 개체/m2였다. 여주보에서 64종으로 가장 많이 출현하였고, 강천보에서 59종, 이포보에서 54종, 한강보상류 에서 41종, 한강보하류에서 32종이 확인되었다. 전체 평균 개체밀도는 강천보에서 944.1 개체/m2로 가장 많았고, 한강보상류에서 282.7 개체/m2, 여주보에서 251.9 개체/m2, 이포보에서 180.6 개체/m2, 한강보하류에서 172.9 개체 /m2 순으로 확인되었다. 한강 보 구간에서는 플라나리아류, 실지렁이, 깔따구류 등이 우점함. 보구간의 특성상 상대적으로 단순한 하상 및 깊은 수심에 적응성이 높은 실지렁이, 깔따구류의 우점은 일반적인 특성으로 볼 수 있으며 일시적으로 수변부에서 밀집된 플라나리아류의 채집이 이뤄져 우점된 결과를 보인 것으로 보임. 한강 보 구간 수변부 및 중앙부 조사지점별 군집분석을 실시한 결과 한강보 하류는 3개 보에 비해 우점도가 높고 다양도 와 풍부도가 낮았으며 전반적으로 상류부에 위치한 한강보상류 등의 지점에서 군집안정성이 확보되고 있는 것으로 보이며 이러한 점은 상대적으로 다양한 미소서식환경에 기인한 것으로 판단된다.
Bangladeshi medicinal plants (BMP) have a history of traditional use in treating chronic inflammatory diseases, but a BMP bark’s antioxidant and anti-inflammatory effects remain largely unexplored. This study assessed methanolic extracts’ antioxidant and anti-inflammatory properties from the bark of 15 medicinal plant species native to Bangladesh. The methanol extracts of BMP bark were evaluated for their total antioxidant activity and ability to counteract inflammation induced by lipopolysaccharide (LPS) in RAW 264.7 macrophages. Among the 15 bark extracts from BMP, Albizia odoratissima (A. odoratissima), Engelhardia spicata (E. spicata), and Shorea robusta (S. robusta) showed the highest total phenolic contents and total antioxidant capacity by effectively scavenging free radicals. In particular, these three bark extracts significantly reduced the mRNA expression of LPS-induced inflammatory cytokines and enzymes inducible by inflammation in macrophages. Also, the mRNA expression of NADPH oxidase 2 was significantly suppressed by the three bark extracts in LPS-induced RAW 264.7 macrophages. These results suggest that out of the 15 bark extracts obtained from medicinal plants in Bangladesh, the extracts from A. odoratissima, E. spicata, and S. robusta exhibit substantial total antioxidant capacity by efficiently scavenging free radicals and also inhibit LPS-induced inflammation in macrophages.
Wolsong Unit 1, a domestic heavy water reactor nuclear power plant, was permanently shut down in December 2019. Accordingly, Wolsong Unit 1 plans to prepare a Final Decommissioning Plan (FDP), submit it to the government by 2024, receive approval for decommissioning, and begin full-scale decommissioning. One of the important tasks in the decommissioning of Wolsong Unit 1 is to determine the decommissioning strategy. It is necessary to decide on a decommissioning strategy considering various factors and variables, secure the technical background, and justify it. The selection of a decommissioning strategy is best achieved through the use of formal decisionmaking assistance techniques, such as considerations related to influencing factors. It is very important to understand the basic decommissioning strategy alternatives and whether sufficient consideration has been given to situations where only a single unit is permanently shut down in a multi-unit site like Wolsong Unit 1, while the remaining units are in normal operation. As a process for selecting a decommissioning strategy, first, all considerations that could potentially affect decommissioning presented in the KINS Decommissioning Safety Review Guidelines were synthesized, influencing factors to be used in the decision-making process were determined, and the concept was defined. In order to select the most appropriate decommissioning strategy by considering various evaluation attributes of possible decommissioning alternatives (immediate dismantling and delayed dismantling), the Wolsong Unit 1 decommissioning strategy was evaluated by reflecting the AHP decision-making technique.
The effect of various physicochemical processes, such as seawater intrusion, on the performance of the engineered barrier should be closely analyzed to precisely assess the safety of high-level radioactive waste repository. In order to evaluate the impact of such processes on the performance of the engineered barrier, a thermal-hydrological-chemical model was developed by using COMSOL Multiphysics and PHREEQC. The coupling of two software was achieved through the application of a sequential non-iterative approach. Model verification was executed through a comparative analysis between the outcomes derived from the developed model and those obtained in prior investigations. Two data were in a good agreement, demonstrating the model is capable of simulating aqueous speciation, adsorption, precipitation, and dissolution. Using the developed model, the geochemical evolution of bentonite buffer under a general condition was simulated as a base case. The model domain consists of 0.5 m of bentonite and 49.5 m of granite. The uraninite (UO2) was assigned at the canister-bentonite interface as the potential source of uranium. Assuming the lifetime of canister as 1,000 years, the porewater mixing without uranium leakage was simulated for 1,000 years. After then, the uranium leakage through the dissolution of uraninite was initiated and simulated for additional 1,000 years. In the base case model, where the porewater mixing between the bentonite and granite was the only considered process, the gypsum tended to dissolve throughout the bentonite, while it precipitated in the vicinity of bentonite-granite boundary. However, the precipitation and dissolution of gypsum only showed a limited effect on the performance of the bentonite. Due to the low solubility of uraninite in the reduced environment, only infinitesimal amounts of uranium dissolved and transported through the bentonite. Additional cases considering various environmental processes, such as seawater or cement porewater intrusion, will be further investigated.
Virus-like particles (VLPs) are similar to pathogenic viruses, but because they have no nucleic acid, they have excellent safety and immunogenicity and are used as a good vaccine material. However, in the selection of various structural proteins of pathogenic viruses to form VLPs, all expression systems consume a lot of time in common. Among them, the baculovirus expression system causes additional time consumption to construct the recombinant baculovirus. Therefore, there is a need for a system that can rapidly determine the structural proteins required for effective VLP production. This study aims at solving this problem by constructing a BmNPV inducible expression platform through the construction of vectors induced by BmNPV. The platform was evaluated for overexpression using EGFP. We also confirmed the formation of virus-like particles through overexpression of canine parvovirus structural proteins.
Viral particles of Porcine epidemic diarrhea virus (PEDV) consist of a four structural proteins. Among them Spikeprotein mediated responsible for receptor binding and membrane fusion during viral infection and therefore the main targetof neutralizing antibodies. Virus-like particles (VLPs) are consisted of one or more viral structural proteins, and theirmorphologies closely resemble those of the native virus. VLPs have no virulence and can elicit robust immune responsesas compared with inactivated or live-attenuated virus vaccines. Thus, in this study, we tried two methods for VLP constructionin Bombyx mori, one is traditional method and the other is chimeric VLP method using the influenza matrix protein.Both methods could produce successfully PEDV VLPs.
Recombinant proteins including a polypeptide fusion partner, termed affinity tag, to facilitate the purification of the target polypeptides are widely used. However, the affinity tag must be removed from the target after the purification process. Recently, the Synechocystis sp. PCC6803 DnaB mini-intein (Ssp DnaB mini-intein) is widely used in Escherichia coli expression systems as the solution of this problem. The Ssp DnaB mini-intein can be induced simply by shifting of pH and temperature, offering a benefit to cleave a peptide bond without using a protease or chemical reagent. Although the utility of this novel tag is widely studied in E. coli, there is no report yet in baculovirus expression vector system (BEVS). In this study, we generated several recombinant baculoviruses to express foreign proteins with Ssp DnaB mini-intein. In conclusion Ssp DnaB mini-intein was good tag also in BEVS with more advantages.
The Black Soldier Fly (BSF, Hermetia illucens) was widely distributed throughout Korea. This insect was mainly found in the vicinity of and in cattle sheds, manure sheds, living waste dump grounds, and food waste dump grounds. This fly is a kind of a beneficial fly because BSF adults do not go into houses, they do not regurgitate on human food, they do not bite, bother or pester humans in any way and they are not associated in any way with the transmission of disease. But their greatest attribute lies their ability to eat and digest raw waste. They can devour, for example, a large, raw, Irish potato and others in just a few hours. Unlike many other flies, since the BSF larvae have very powerful mouth parts and digestive enzymes, they can ingest raw waste far more efficiently than any other known species of fly. On this study, to investigate whether feeding strategy of the BSF larvae involves extra-oral digestion or not, and to better understand this process, the salivary glands and a few tissue from the BSF were dissected and subjected to morphological and preliminary enzyme characterization.
μ방lt emitting diodes (LED) devices are commercially introduced as an alternative for low-Ievellaser therapy (11ι,T) , and it has several advantages over lasers such as a safe, efficient, and less-expensive altemative to treat wounds. And LED irradiation at the same biostimulatory wavelength has similar bíochemical effεαs. In the present study, to asses whether the I핑ht-emitting diode (LED) irradíation can stimulate bone regeneration, irradiated bony defects with or without grafting materials on rat calvaria were compared to corresponding nonirradiated control. Fifty male Sprague-Dawly rats weighing about 150g, were used. Factors for present study were designed as follows, 1) presence or absence of grafting materials, 2) with or without irradiation, and 3) number of irradiation. Two weeks after operation, rats were sacrifìced. Radiologic and 비stomorphologic fmdings were evaluated. Macrospically, there were no incidents of infection, dehiscence, hematoma and necrosis during study. Radiological findings showed greater radiopacity in the graft group and radiopacity increased as the number of irradiation increase. And microscopically, new bαle formation was great in the graft group and increased as the number of irradiation increase, Present study has shown that LED irradiation improved bone regeneration through radiologic and histomorphologic fmdings in rat.
