We developed new two-rowed covered barley ‘Gwangmaeg’ superior to ‘Hopum’ that has been widely cultivated in Korea for using beer-materials. ‘Gwanngmaeg’ was related from the line named as ‘Iksan149’ in 2010 after yield trials test in conducted for three years from 2008. And then it was registered to ‘Plant protection right, No. 5847’ in 2016 after field experiment by KSVS(Korea Seed & Variety Service). ‘Gwangmaeg’ has distinct characteristics in winter type (IV) and mid-tall culm length which makes reduce damage by off-season heading and lodging comparing to those of ‘Hopum’. ‘Gwangmaeg’ have characteristics of cold tolerance and disease resistance to BaYMV(Barley yellow mosaic virus) and Net blotch(Pyrenophora teres). Yield of ‘Gwangmaeg’ was about 611kg/10a and 524kg/10a in upland and paddy field condition, respectively, which higher 2-4% than ‘Hopum’, however, it has no significant difference. In quality examination for beer usage, ‘Gwangmaeg’ has lager grain and higher assortment ratio than those of ‘Hopum’. In malt quality, ‘Gwangmaeg’ showed higher and better quality in malt extract rate and diastatic power than those of ‘Hopum’. ‘Gwangmaeg’ would be suitable for the area above –4℃ of daily minimum temperature in January in Korean peninsula.

Rice production is largely affected by various environmental conditions such as cold, heat and flooding. Here, to identify cold tolerant QTLs at seedling stage in rice, we generated RIL population derived from a cross between Hanareum 2 and Unkwang which are a highly cold sensitive and cold tolerant, respectively. We observed cold phenotype of this population in the growth chamber conditions and natural field conditions. For observation of cold tolerant phenotype of RIL population in the growth chamber, we treated cold stress (5~13℃) for 14 days and recovery for 4 days. When we examined the phenotype of RIL in the field conditions, temperature range in the field conditions was about 6 to 25℃ in 2015~2016. We named QTLs as Seedling Cold Tolerant (SCT) in growth chamber and Cold induced Yellowing Tolerant (CYT) in the field, respectively. Three QTLs for SCT and 5 QTLs for CYT were detected on chromosome 1, 6, 7, 8, 10, 11 and 12. Among these QTLs, qSCT12 on chromosome 12 showed 26.3 LOD score with 25.5% of phenotypic variation. When qSCT11.1 and qSCT12 were combined, cold tolerant was most strongest in our experimental conditions. qCYT10 on chromosome 10 was identified in field experiment on both 2015 and 2016. These results may provide useful information for a marker-assisted breeding program to improve cold tolerance in rice.

This study selected commonly known species of fruit trees, and re-selected the species that endure the stress of extreme cold weather and physiologically restore themselves to the previous state until the following year. Then we could go ahead to propose the species that were appropriate as urban greening plants in weather condition of any part of the country. To do this, we conducted an experiments for six species of fruit trees based on the preference of the general public and recommendation of the experts; Morus alba (English name: mulberries), Diospyros kaki (English name: Persimmon), Prunus persia (English name: Peach), Elaeagnus umbellata var. coreana (English name: Korean Autumn Olive), Malus domestica 'Alps Otome' (English name: Alps Otome), and Prunus mume (English name: Blue Plum). The experiment verifies whether the trees survive without any stress from the cold weather under the national climate conditions (one in the suburbs of Seoul: Yongin city, one in the central Chungcheong region: Daejeon city, and in the southern Gyeongsang region: Jinju city in Korea). The experiment lasted for a year from August 2016 to August 2017. The levels of electrolytic efflux, chlorophyll content, plant height, fresh weight, and dry weight were measured four times (on August of 2016, January, February, and August of 2017) for each tree planted bare ground outdoors. Results showed that Diospyros kaki, Prunus persia, and Malus domestica 'Alps Otome' were proven durable and resistant to winters of all three areas (one in the suburbs of Seoul: Yongin city, one in the central Chungcheong region: Daejeon city, and in the southern Gyeongsang region: Jinju city in Korea). Especially, the increase of chlorophyll content and the reduction of electrolytic efflux were noticeable in Prunus persia than in the other two species, proving itself as the most cold-tolerant among the six species used in the experiment. In addition, interpreting from the physiological restoration data of one-year span before and after getting through winterer, Prunus persia was proven to be the most cold-tolerant species.

This study was carried out to investigate and assess the cold tolerance of 24 species broad-leaved evergreen trees in southern region, South Korea, and propose the selection for urban greening responsive to the climate change. The cold stressed impact of each species was measured and calculated by the electrolyte leakage (EL) method, and then the lethal temperature was predicted by the non-linear regression analysis. The scattered plots and fitted curves of most species tended to show sigmoidal response curve. On assessing the EL values and sigmoidal response curve pattern with different temperature, the differences were obviously showed among all the species. Also, among the species within the same family, the differences were obviously showed. The maximum temperature difference among the species was over 10℃. Between Ilex rutunda and Ilex integra within the same family, Aquifoliaceae, it was over 10℃. The results indicate that there are significant differences in cold tolerance among different species in the same region, which are not affected by any environmental factors but affected by any genetical factors. Thus it is valuable to assess the cold tolerance on most broad-leaved evergreen trees in southern region, South Korea. As a result, Euonymus japonicus, Trachelospermum asiaticum, Dendropanax morbiferus, Ilex integra, Machilus thunbergii, Ilex x wandoensis, Cinnamomum japonicum, Distylium racemosum, and Castanopsis sieboldii may have better cold tolerance and survive the region closer to middle region, South Korea compared to the others.

Rice is a staple food crop for more than half of the world population. Severe losses of rice production was caused by various environmental conditions such as cold, heat and flooding annually. Rice is a highly sensitive to low temperature below 15-20 ℃ because of originating from tropical or subtropical climates. Especially, seedling of rice is easily damaged to low temperature and result in seedling yellowing, growth retardation, reduced tillering and yield losses at last. We used a recombinant inbreeding lines (RIL) population of 384 individuals derived from a cross between Hanareum 2, a highly cold sensitive variety and Unkwang, a cold tolerant variety for molecular mapping of QTLs related to cold tolerance. Seedling discoloration of each lines and parents caused by cold response were investigated in field condition after transplanting. And leaf samples of RIL population were collected for evaluation of chlorophyll content using 80% acetone extraction. The seedling of each lines and parents was subjected to low temperate by 5~13 ℃ during 14 days. The cold recovery score (CRS) of RILs was recorded after 4 days recovery period according to standard evaluation system (SES, IRRI). Total of eight QTLs were detected on chromosome 1, 7, 8, 10, 11 and 12 using cold tolerance traits, chlorophyll content, seedling discoloration and cold recovery score in 384 RILs. The qCRS12, which detected on chromosome 12 between two flanking markers id12002113, id12002563 (1.1 Mbp) showed 25 LOD score with 26% of phenotypic variation of cold recovery score in RILs population. The positive allele contributing to cold tolerance came from the cold tolerant parent Unkwang. The result may provide useful information for a marker-assisted breeding program to improve cold tolerant in rice.

Anthocyanins are responsible for vivid colors of flowers, fruits and vegetative tissues and biosynthesis of it is primarily controlled by several structural and regulatory genes. The regulatory mechanism of this pathway is still unknown. This study identified 19 transcription factors of Brassica rapa and investigated their regulatory function in anthocyanin biosynthesis pathway genes and cold and/or freezing tolerance in B. rapa. Expression analysis of these genes in the pigmented and non-pigmented portion of leaves of different lines of B. rapa revealed that BrMYB2-2 and BrTT8 showed responses contrasting with anthocyanin accumulation and cold stress. Sequences of these genes were analyzed and compared with similar gene sequences from other species and a high degree of homology with their respective functions was found. Co-regulated cis -elements were found in promoters of BrPAL1, BrCHS, BrF3H1, BrF3’H1, BrFLS, BrBAN, BrDFR8, BrANS1, and BrMYB2-2 and BrTT8 had binding sites of the promoters of those structural genes. Thus, the above results suggest the association of BrMYB2-2 and BrTT8 with regulation of anthocyanin biosynthesis pathway genes and cold and freezing stress tolerance and might be useful resources for development of cold resistant Brassica crops with desirable colors as well.

To obtain a better insight into a possible common regulatory mechanism shared in response to heat and cold stresses in model monocot plant, expression profiling for several selected transcripts of rice (O. sativa L.) was examined from meiotic stage panicle samples of a number of different cultivars including both heat/cold tolerant and susceptible varieties. Induction of several of these genes in response to heat stress treatment was observed across all different rice varieties tested, but no differential induction kinetics between the heat-tolerant and susceptible varieties was discernable for these genes. On the other hand, a few genes that are known to be involved in ROS signaling showed different induction kinetics between the tolerant and susceptible varieties, suggesting that these genes might be playing a key role in conferring the heat/cold stress tolerance at reproductive stage of tolerant rice varieties. This approach was also augmented by comparative gene expression data analyses, utilizing expression data of other monocots and dicots model plants available in the database. Based upon these results, a preliminary gene pyramiding effort to estimate contributing factor of several selected alleles after putting these genes together is underway. This work was supported by a grant from the Next-Generation BioGreen 21 Program (Plant Molecular Breeding Center No.PJ009076), Rural Development Administration, Republic of Korea.

Cold stress at the seedling stage is a major threat to rice production. Cold tolerance is controlled by complex genetic factors. We used an F7 recombinant inbred line (RIL) population of 123 individuals derived from the cross of a cold-tolerant japonica and a cold-sensitive indica cultivars, for QTL mapping. Phenotypic evaluation of the parents and RILs in an 18/8oC (day/night) cold-stress regime showed continuous variations for cold tolerance or sensitivity. Six QTLs for seedling cold tolerance were identified on chromosomes 1, 2, 4, 10, and 11 with percent phenotypic variation (R2) ranging from 6.1% to 16.5%. Three main-effect QTLs (qSCT1, qSCT4, and qSCT11) were detected in all cold-tolerant RILs which explained high sum of phenotypic variation (SPV) ranging from 27.1% to 50.6%. Two QTLs (qSCT1 and qSCT11) on chromosomes 1 and 11 were fine mapped. The marker In1-c3 from ORF LOC_Os01g69910 of the BAC clone B1455F06 encoding calmodulin-binding transcription activator (CAMTA) and another marker, In11-d1 from ORF LOC_Os11g37720 (Duf6 gene) of the BAC clone OSJNBa0029K08, co-segregated with seedling cold tolerance. These two InDel markers amplified 241-bp and 158-bp alleles, respectively, in cold-tolerant RILs, and in the cold-tolerant donor Jinbu, which were absent in cold-sensitive parent BR29 and cold-sensitive RILs.

QTL analysis for cold tolerance-related traits was conducted using 75 introgression lines (IL) developed from a cross between a japonica weedy rice and Tongil-type rice. A molecular linkage map consisting of 136 SSR markers was constructed to identify QTLs associated with cold tolerance. 75 ILs and the parents were evaluated for three traits associated with cold tolerance: seedling height and SAPD values at the seedling stage. The plants were grown for 15 days in the low temperature condition (13/20℃ day/night) and the control condition (25/20℃ day/night) in the growth chamber. A total of six QTL were identified for two traits and phenotypic variance explained by each QTL ranged from 4.3% to 35.7%. Among two QTL for seedling height, one QTL, sh1 for seedling height was detected at both conditions. The other QTL on chromosome 6 was detected in the low temperature condition. Four QTL were identified for SPAD value and two were detected on chromosomes 2 and 5. At these loci, Milyang 23 alleles increased the SPAD value. The other two QTL on chromosomes 1 and 4 were detected at the low temperature plot. At these loci, Hapcheonaengmi 3 alleles increased the SPAD values. These results indicate that Hapcheonaengmi 3 alleles might increase tolerance to low temperature in the Milyang 23 background because SPAD value is positively correlated with chlorophyll content and N content in rice. The markers linked to low temperature tolerance at the seedling stage would be useful in selecting for lines with enhanced cold tolerance in a breeding program.

Cold tolerance at every growing stage of rice(Oryza sativa L.) is one of the main determinations for the stable growth in temperature and high elevate area. In the current study, a 181 lines of BC population derived from a cross of Gayabyeo, a Tongil type sensitive to cold and Chhamrong a tolerant to cold were evaluated for cold tolerance with cold water irrigation(17℃) at seeding stage as well as low temperature germinate at 13℃, respectively. The resulting linkage map consists of 157 marker loci, covering all of 12 rice chromosomes and spanning 910cM(Haldane function) with an average interval of 76cM between markers. Three main-effect QTLs were identified. The comparison of the OTLs identified in this cold treatments resulted in an intriquing finding that each treatments were controlled by a major QTL. The QTL qCWI-4 on chromosome 4 was found to increase its additive effect to -0.84 as the cold water irrigate stress was given. In the meanwhile, the QTL qLTG-8 was detected with a LOD score of5.54, explaining up to 13% of the phenotypic variation controlled by Gayabyeo allele. In addition, the QTL qLTGV-3, controlled by Gayabyeo allele with a LOD score of 5.19 explaining about 12.5% of the variation was also identified. These results would favor our better understanding of the genetic control of cold tolerance in rice and be important for the development of rice cultivars with a broaden climatic adaptation.

Cold sensitivity has been shown to vary and the cold tolerance from vegetative to reproductive stage appears to be independent during the life cycle. In the current study, cold tolerance under high elevation rice growing area were evaluated using F4 generation of 181 lines derived from Gayabyeo*2/Chhamrong. Five main-effect QTLs related to days to heading, panicle exertion and spikelet fertility were identified. The QTL qDH-3 on chromosome 3 showed the peak LOD score of 6.3, explaining up to 16.5% of phenotypic variation with additive effect of -2.6. Moreover, the QTL qPE-3 and qSF-3 on chromosome 3 were coincided with the QTL qDH-3 showing an opposite allele effects. Thus, the region harboring marker RM523-RM14281 could be helpful for the selection of cold tolerance genotypes in marker assisted selection(MAS) of rice breeding program.

This study was carried out to identify the quantitative trait loci (QTLs) for traits related to cold tolerance using an introgression lines (ILs) derived from a cross between a japonica weedy rice and a Tongil-type rice. Among the 80 RILs, one line (CR1835) showing tolerance for cold tolerance related to traits such as panicle exsertion and discoloration in the cold water plot was selected and backcrossed to the recurrent parent, Milyang 23. This line possessed Hapcheonaengmi 3 segments on chromosomes 1, 5 and 11. By two backcrosses to Milyang 23 and selfing, a total of 88 BC3F5 progenies were developed. The 88 ILs were evaluated for traits related to agricultural performance in cold water and in control plots. Cold tolerance was measured as difference of the culm length, spikelet fertility between two plots and panicle exsertion in cold water plot. The 88 ILs showed decreased culm length and increased anthocyan content in cold water plot than in the control water plot.
