        This study analyzed population density changes and taxonomic characteristics of various microorganisms, which play important roles in culture medium fermentation of Agaricus bisporus and examined changes in decompositionrelated enzyme activity secreted by these microorganisms. Various microorganisms such as thermophilic bacteria, actinomycetes, fluorescent pseudomonas spp. and filamentous bacteria are closely related to culture medium fermentation of Agaricus bisporus. The population density of these microorganisms changes and harmful bacteria disappear during thermophilic fermentation. During outdoor fermentation, Psychrobacter sp., Pseudomonas sp., Bacillus sp. and Pseudoxanthomonas sp. showed the highest percentage distribution in the different culture medium while Bacillus sp. and Psychrobacillus sp. were the most dominant after pasteurization. When analyzing the enzyme activity of microorganism during rice straw fermentation, cellulose enzymes become important at the early stage of fermentation. Increase in hemicellulose enzyme follows after decomposing carbon sources. The microorganisms that secretes these enzymes were distributed at the second and third turning stage of fermentation.

        Since the first Korean button mushroom ‘Sae-Ah’ developed in 2010, most farmers had wanted to cultivate a new variety. A new variety, ‘SaeJeong’, was developed in 2011, but this variety was difficult to cultivate in mushroom farm. To solve the variety problem, a new variety ‘Sae-do’ was developed in 2012. The variety, ‘Sad-do’, was made by crossing hetrokaryon A175 and homokaryon ASI1346-15 selected by genetic analysis. The heterokaryon A175 is crossed line between ASI1038-211 and ASI1346-20. The mycelium of ‘Saedo’ on CDA (compost dextrose agar) grew well at 25°C. The optimum pin-heading temperature of it was 13-15°C and optimum growing temperature was 13-20°C. Earliness (days required from casing to first harvesting) was 2 days shorter than control ‘Saejeong’. Even though the diameter of mature cap and the length of stipe were shorter than a control, the individual weight of fruiting body was heavier because of thick pileus and stipe. Therefore, the yield was increased by 1.89 times than control and the hardness of pileus was stronger. The field experiment of ‘Saedo’ variety has been started since 2013. Most farmers evaluated that the variety has good compost colonization and cap shape, and high yield in harvesting. For that reason, ‘Saedo’ has occupied half of the of distribution rate of Korean varieties. Also distribution rate of Korean varieties was increased from 26.1% in 2014 to 37% in 2015.

        The button mushroom, Agaricus bisporus, is one of the major economical crops cultivated in Korea. This mushroom showed the 5th production to 11,493 M/T in 2014. Several fungus are known as the causal agents of diseases of the cultivated button mushroom (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus). Cladobotryum mycophilum is the causal agent of cobweb disease of commercial mushrooms. Early symptoms were noticed as round, fleshy, yellowish brown lesions on mushroom caps. Late symptoms progressed when the parasitic fungus formed white cobweb circular colonies on dead or damaged pinheads, spread on the surface of the casing, and covered entirely fruiting bodies. A Gram-positive bacterium was isolated from mushroom media that markedly showed the antagonistic activity against Cladobotryum mycophilum, the most destructive pathogen of cultivated mushrooms. The HC57 strain was selected as antagonistic bacterium by inhibition zone method and it was identified as Bacillus subtilis by the cultural, morphological and physiological characteristics, and analysis of the 16S rDNA. The isolated bacterium is saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for Cladobotryum mycophilum cell, was sufficient for inhibition in vitro. Inoculation of the isolated bacterium prevents the development of bacterial disease in Cladobotryum mycophilum. Control efficacy of browning disease of strain HC57 treatment was 71% on Agaricus bisporus. The optimal culture medium for the antagonistic bacteria growth was determined as follows: 1.5% Xylose, 2% Soytone, 1% NH4H2PO4, 7 mmol CaCl2, and 0.5% Histidine at pH 6.0 at 25℃. The suppressive bacterium may be useful in future for the development of biocontrol system and the construction of genetically modified edible fungi resistant to the disease caused by Cladobotryum mycophilum.

        A new brown button mushroom cultivar, ‘Hogam’, C34 line, was made by crossing homokaryons, ASI1164-37 andASI1175-66, selected by RAPD analysis and by cultivating three times. Mycelium of ‘Hogam’ on CDA (compost dextrose agar)media grew well at 25oC. The optimum pin-heading temperature of new variety and optimum growing temperature was 14-18oC. The thickness of the mature cap and stipe were thicker than a control, ‘Dahyang’ that developed in 2010. The color ofpileus was light brown and lighter than ‘Dahyang’. Days required from casing to first harvesting were three days longer thancontrol strain, but the weight of harvested fruiting body increased by 1.35 times. ‘Hogam‘ cultivar are expected to contribute tothe diversification of domestic mushroom cultivars.

        This study was carried out to analyze of the cause of button mushroom production decrease of Gyoengbukprovince. In 1978, Agaricus bisporus was produced 48,000 ton and exported more than $50 millions. But since 2000, Domesticproduction of button mushroom was decreased by 70%, and button mushroom farm was also decreased by 37%. Cultivationarea was increased by 6%, but Gyeongbuk Province was decreased by 30%. Especially, Production per 3.3ߊ was dramaticallydecreased more than half. There were several causes such as rising labor and material cost, climate changes, and aging ofmushroom cultivation farmers. And there was no effort to develop of domestic button mushroom cultivation equipments. One ofthe main reasons for this reduction was supplied to low quality of button mushroom compost to the farm.

        검정날개버섯파리과에 속하는 긴수염버섯파리(Lycoriella ingenua)는 우리나라 양송이버섯 재배에서 심각한 피해를 주는 주요해충의 하나이다. 본 연구는 양송이버섯 재배에서의 긴수염버섯파리의 온도별 발달과 피해양상을 조사하고자 실시하였다. 버섯파리는 균사를 배양한 페트리디쉬에서 사육하였고 16, 20, 24, 28oC 온도에서 알에서 성충까지 소요기간은 각각 35.2, 25.8, 23.5, 22.2일로 조사되었다. 버섯파리 성충은 버섯재배사를 침입하여 종균이 접종된 배지에 알고 낳고 1-7 mm의 부화된 유충에 의해서 균사와 생육하고 있는 버섯의 갓과 대를 섭식하여 가장 큰 피해를 입힌다. 성충은 자실체의 외관을 더럽히고, 여러 가지 곰팡이와 세균병을 전파시키며, 응애 등을 옮긴다. 적절한 해충방제 없는 조건에서 버섯파리에 의한 피해가 증가하면서 자실체 수량은 27%까지 감소하였다.

        양송이 수확후 배지로부터 lipase 생산균을 분리하여 16S rDNA 유전자 분석을 통해 동정한 결과, Burkholderia cepacia ATCC와 99.8% 상동성을 나타냈다. 분리균 B. cepacia 배양여액 중에 함유된 효소단백질을 70% 황산암모늄으로 침전시켜 crude lipase를 회수하였다. 고정화 효소를 제조하기 위하여 crude lipase(CL)과 Novozyme lipase(NL)을 cross-linking 법에 의해 Silane화된 Silicagel에 고정화 시킨 결과, immobilized CL(ICL)은 61%, immobilized NL(INL)은 72%의 잔존활성을 유지하였다. 중성지방 Canola oil을 알칼리(NaOH) 촉매와 효소(CL 및 ICL) 촉매를 이용하여 지방산(fatty acid)으로 분해한 후, methanolysis에 의한 에스터전이반응(trans-esterification)을 통해 지방산으로부터 전환된 바이오디젤(fatty acid methyl ester, FAME)의 종류와 수율을 비교 하였다. 생성된 총 FAME 함량은 NaOH 781mg L-1, free lipase 681mg L-1, 고정화 lipase 598mg L-1순으로 높았으며, 지방산 조성별 FAME 함량은 linoleic acid(C18:1)가 약 50%로 가장 높았으며, stearic acid(C18:0)가 22%정도의 높은 수준이었다. 또한 반응시간이 증가함에 따라 CL과 ICL 모두 불포화지방산 FAME의 조성비는 감소하고, 상대적으로 포화지방산 FAME의 조성비는 증가하는 경향을 보여 lipase 효소가 transesterification 활성과 interesterification 활성을 동시에 가지는 것으로 여겨진다. 고정화효소의 잔여활성은 반복회수가 증가함에 따라 서서히 감소하여 4회 반복 후, 초기 활성도에 비해 ICL은 34% 와 INL은 21%까지 감소하였다.

        양송이는 재배사라는 밀폐된 환경에서 재배되는 바 양질의 생산을 위해서는 재배사 내 환경에 대한 정보가 요구된다. 본 연구는 양송이 재배사내 실내환경에 대한 기초 정보를 얻고자 수행되었다. 재배사 내 온도와 습도는 19.75±0.35˚C, 87±3.67%로 세균이 서식할 수 있는 환경조건이었으며, 부유세균의 농도는 3.84×103cfu/m3로서 환경부에서 고시한 실내공기질 오염유지 기준치인 8.0×102cfu/m3을 초과하는 수치였다. 분리된 부유세균은 Advenella kashmirensis, Bacillus vietnamensism, B. licheniformis, Burkholderia sordidicola, Fictibacillus phosphorivorans, Lysobacter daejeonensis, Microbacterium esteraromaticum, Pseudomonas aeruginosa, P. protegens, P. gessardii, P. mosseli 등 7속 11종으로 동정되었다.

        Spawn is the vegetative growth of the mycelium and serves as the inoculum or seed for cultivating mushrooms. Various cereal grains, such as wheat, millet, barley, sorghum, brown rice, rye, and oat were assessed to compare their characteristics and suitability for spawn production of button mushroom (Agaricus bisporus). The mycelial growth rates, density, and the number of completely colonized grains were measured from the twentieth day of inoculation. Wheat grains showed fastest mycelial growth with 8.4 cm followed by rye, oat, barley with 8.2, 7.5 and 7.3 cm, respectively. In the mycelial density, foxtail millet, barley, and sorghum were best compared with that of wheat grains. Especially, the number of grains which were completely colonized by mycelia were greatest in foxtail millet with 5,123 grains followed by proso millet, and wheat with 3,052 and 914, respectively. Based on the results obtained, barley, foxtail millet, and sorghum grains would be appropriate substituting for wheat grain in spawn production of button mushroom.

        Mushroom is cultivated as one of the major economical crops in many areas in Korea. The total production has steadily increased from approximately 198,563 M/T in 2009 to 208,941 M/T in 2012. Several fungus are known as the causal agents of diseases of the cultivated button mushroom (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus). Trichoderma harzianum is the causal agent of brown blotch disease of commercial mushrooms. Colonization of mushroom caps by the bacterium results in development of brown or cream lesions on pileus and stipe. These lesions are slightly concave spots and can be round or spreading. Antagonists against Trichoderma harzianum, CAM33 were selected and their control efficacy of green mould disease was investigated in this study. The CAM33 strain was selected as antagonistic bacterium by inhibition zone method and it was identified as Bacillus methyrotrophicus. by the cultural, morphological and physiological characteristics, and analysis of the 16S rDNA. The isolated bacterium is saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for Trichoderma harzianum cell, was sufficient for inhibition in vitro. Inoculation of the isolated bacterium prevents the development of bacterial disease in Trichoderma harzianum. Control efficacy of browning disease of strain CAM33 treatment was 77% on Agaricus bisporus. The optimal culture medium for the antagonistic bacteria growth was determined as follows: 3.0% Saccharose, 1.5% Soytone, 1.0% (NH4)2HPO4, 10 mmol MgSO4, and 2.0% Glutamic acid at pH 6.0 at 25°C. The suppressive bacterium may be useful in future for the development of biocontrol system and the construction of genetically modified edible fungi resistant to the disease caused by Trichoderma harzianum.

The suppressive bacterium may be useful in future for the development of biocontrol system and the construction of genetically modified edible fungi resistant to the disease caused by Cladobotryum mycophilum.

        The potential ability of Button mushroom compost (BMC) to solubilize heavy metals was estimated with metal contaminated soils collected from abandoned mines of Boryeong area in South Korea. The bacterial strains in BMC were isolated for investigating the mobilization of metals in soil or plant by the strains and identified according to 16S rRNA gene sequence analysis. When metal solubilization potential of BMC was assessed in a batch experiment, the BMC was found to be capable of solubilizing metals in the presence of metals (Co, Pb and Zn) and the results showed that inoculation of BMC could increase the concentrations of water soluble Co, Pb and Cd by 35, 25 and 45% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 27, 25 and 28% respectively in Co, Pb and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction of Co, Pb and Zn from contaminated soils.

        양송이 재배농가는 복토재료로 흙을 사용하고 있으며, 대부분의 농가는 채취된 그대로 사용하고 있어 흙속에 있는 각종 병원균의 감염으로 버섯재배에 실패하는 농가가 많다. 복토 살균온도에 따른 미생물의 밀도변화를 조사한 결과 호기성세균은 양송이 생육주기가 길어질수록 증가하였으며 2주기 수확후 부터는 급격히 증가하였다. 사상균은 복토 8일째 무처리에서 급격히 증가하였고, 살균 처리구에서는 증감을 반복하다가 2주기 수확후 부터 급격히 증가하였다. 푸른곰팡이균도 증감을 반복하다가 2주기 수확후부터 급격히 증가하였다. 형광성 Psuedomonas속은 살균온도가 높은 처리구에서 높은 밀도를 보였으며, 복토 22일에는 100˚C-30분과 60분에서 다른 처리구보다 급격히 증가하였다. 중온성 방선균은 복토 15일 약간 증가하는 경향을 보였다가 1주기 수확후부터 급격히 증가하였다. 고온성 방선균은 복토 15일에 급격히 증가하여 복토 22일에 가장 높은 밀도를 보였지만, 1주기 수확후 부터 급격히 감소하였다. 양송이버섯의 수량과 품질은 80˚C-90분에서는 상품 및 중품의 수량이 가장 높았다. 병해충 발생은 100˚C-30분 처리에서 가장 낮았지만 양송이의 수량이 낮았으며, 100˚C-60분 처리구에서는 양송이 수량은 높았으나, 병해충의 발생이 높았다. 따라서 양송이 재배를 위한 복토의 적정 살균조건은 80˚C-60분, 80˚C-90분이 가장 좋을 것으로 판단된다.

The HC42 strain was selected as antagonistic bacterium by inhibition zone method and it was identified as Bacillus safensis. by the cultural, morphological and physiological characteristics, and analysis of the 16S rDNA.. The isolated bacterium is saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for P. agarici cell, was sufficient for inhibition in vitro. Inoculation of the isolated bacterium prevents the development of bacterial disease in Agaricus bisporus. Control efficacy of browning disease of strain HC42 treatment was 66% on Agaricus bisporus. The optimal culture medium for the antagonistic bacteria growth was determined as follows: 1.5% D-galactose, 1.5% yest extract, 1% NH4Cl, 1.5% KCl, and 1.0% L-asparagin at pH 6.0 at 25℃. The suppressive bacterium may be useful in future for the development of biocontrol system and the construction of genetically modified edible fungi resistant to the disease caused by P. agarici.

        Commonly known as the button mushroom, Agaricus bisporus is one of the most widely cultivated mushroom species of edible fungi. In the breeding of new button mushroom, Seolwon was developed by crossing two homokaryons. Because of the predominantly pseudohomothallic life cycle, only a small percentage of homokaryotic meiospores are produced, which do not fruit. Homokaryotic cultures derived from these types of single spores produce a vegetative mycelium that contain a variable number of genetically identical nuclei per cell. After crossing two homokaryons, hybrids were cultivated on a small scale and on a commercial scale at a farm. The spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of Seolwon on CDA was better at 25oC when it was compared with that of Seolgang. The mature cap shape of new strain Seolwon is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 39.7 mm on average. In comparison with white strain Seolgang, the strain had a yield that was 11% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 9.7% firmer with a good shelf life. Days of fruiting body were 1-2 days later than those of Seolgang. The physical characteristics such as springiness, chewiness, adhesiveness, gumminess were better than that of Seolgang.

        A galactose fermentation bacterium producing lactose from red seaweed, which was known well to com-promise the galactose as main reducing sugar, was isolated from button mushroom bed in Buyeo-Gun, Chungchugnam-do province. The lactic acid bacteria MONGB-2 was identified as Lactobacillus paracasei subsp. tolerans by analysisof 16S rRNA gene sequence. When the production of lactic acid and acetic acid by L. paracasei MONGB-2 was inves-tigated by HPLC analysis with various carbohydrates, the strain MONGB-2 efficiently convert the glucose and galactoseto lactic acid with the yield of 18.86 g/L and 18.23 g/L, respectively and the ratio of lactic acid to total organic acidswas 1.0 and 0.91g/g for both substrates. However, in the case of acetic acid fermentation, other carbohydrates besidesgalactose and red seaweed hydrolysate could not be totally utilized as carbon sources for acetic acid production by thestrain. The lactic acid production from glucose and galactose in the fermentation time courses was gradually enhancedupto 60 h fermentation and the maximal concentration reached to be 16-18 g/L from both substrates after 48 h of fer-mentation. The initial concentration of glucose and galactose were completely consumed within 36 h of fermentation, ofwhich the growth of cell also was maximum level. In addition, the bioconversion of lactic acid from the red seaweedhydrolysate by L. paracasei MONGB-2 appeared to be about 20% levels of the initial substrates concentration and thisresults were entirely lower than those of galactose and glucose showed about 60% of conversion. The apparent resultsshowed that L. paracasei MONGB-2 could produce the lactic acid with glucose as well as galactose by the homof-ermentation through EMP pathway

        Y-Aminobutyric acid(GABA) is a four carbon non-protein amino acid that has several well-known phys-iological functions, such as a postsynaptic inhibitory neurotransmitter in the brain and induction of hypotensive and tran-quilizer effects. A lactic acid bacterium was isolated from button mushroom bed, which is showing high GABAproductivity by TLC or HPLC analysis. The strain was identified as Lactobacillus hilgardii by analysis of 16S rDNAgene sequence. When the maximum production of GABA by L. hilgardii was investigated with various concentration ofmonosodium glutamate, the yield of GABA reached to be 53.65mM at 1% mono sodium glutamate (MSG) in flask cul-tivation. A Glutamate decarboxylase (GAD) enzyme, which was known to convert MSG to GABA, was purified froma cell-free extract of L. hilgardii and the molecular weights of purified GAD was estimated to 60,000 by SDS-PAGE.The optimum pH and temperature of GAD were at pH4.6 and at 37°C, respectively. The GAD activity was increasedby the addition of sulfate ions such as ammonium sulfate, sodium sulfate and magnesium sulfate, indicating that theincrease of hydrophobic interaction causes the increase of GAD activity.

Pseudomonas agarici is the causal agent of browning disease of commercial mushrooms. Colonization of mushroom caps by the bacterium results in development of browning lesions on pileus. Antagonists against P. tolaasii, HC42 were selected and their control efficacy of browning disease was investigated in this study. After proceeding antagonistic test, HC42 was selected as a strong antagonist against P. agarici and the HC42 strain was identified as P. safensis with the cultural, physiological and biochemical properties and analysis of the 16S rRNA.         Mushroom is cultivated as one of the major economical crops in many areas in Korea. The total production has steadily increased from approximately 186,400 M/T in 2007 to 190,111 M/T in 2011. The button mushroom, Agaricus bisporus, showed the 5th production to 13,052 M/T in 2011. Several bacteria are known as the causal agents of diseases of the cultivated button mushroom (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus). Pseudomonas agarici is the causal agent of browning disease of commercial mushrooms. In this study, antagonistic bacteria to P. agarici were selected in vitro tests using confrontation bioassay and paper disk diffusion assay. The most active bacteria, HC12 were selected and their control efficacy of brown blotch disease was investigated in this study. After proceeding antagonistic test, HC12 was selected as a strong antagonist against P. agarici and the HC12 strain was identified as Alcaligenes sp. with the cultural, physiological and biochemical properties and analysis of the 16S rRNA.         The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, ‘Seolgang’ was developed by crossing two monokaryons ‘CM020913-27’ and ‘SSU423-31’. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of ‘Seolgang’ on CDA was better at 20℃ and 25℃ when it was compared with that of ‘505 Ho’. The mature cap shape of new strain ‘Seolgang’ is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain ‘505 Ho’, the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of ‘505 Ho’. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of ‘505 Ho’. Genetic analysis of the new strain ‘Seolgang’ showed different profiles compared to ‘505 Ho’, CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.
